EGCG-Releasing Nanofibrous Scaffold Enhances Wound Healing via γδ T Cells Modulation

Tiantian An,Zhaoyu Xu,Yang Yang,Xinhui Li,Shuaidong Chen,Xingmei Yang,Yi Man,Chen Hu,Yili Qu

This study investigates the role of γδ T cells in wound healing facilitated by an EGCG (Epigallocatechin gallate)-releasing nanofibrous scaffold with aligned topography (APEMS). Incorporating core-shell microcapsules for sustained EGCG release over 11 days, the biocompatible scaffold enhances γδ T cell activity to promote advanced wound repair. In a splinted wound excisional model in 6-8 weeks-old male C57BL/6J mice, APEMS significantly improved re-epithelialization and wound closure compared to a control scaffold (APS). Single-cell RNA sequencing revealed a marked increase in γδ T cell in the APEMS group, where γδ T cells accounted for 66.2% of the T cell population, compared to 33.8% in the APS group. Cellchat analysis identified the AREG-EGFR signaling axis as pivotal to γδ T cell-mediated wound repair. APEMS-induced γδ T cells secreted amphiregulin (AREG), which acted on keratinocytes and fibroblasts to enhance keratinocyte proliferation, migration, and fibroblast-driven extracellular matrix remodeling. In immunodeficient mice lacking mature γδ T cells (TCRδ-/-), wound healing was significantly impaired, characterized by delayed re-epithelialization, reduced neo-dermal thickness, and increased inflammation. Inhibition of AREG and EGFR function via monoclonal antibodies against AREG and EGFR antagonist Gefitinib similarly delayed wound healing, the therapeutic advantage of APEMS over APS was abolished, emphasizing its critical role in the process. These findings provide a framework for developing advanced wound management materials and highlight the therapeutic potential of targeting γδ T cells and AREG-EGFR signaling to enhance tissue regeneration.

Reduction in Autophagy by (-)-Epigallocatechin-3-Gallate (EGCG): a Potential Mechanism of Prevention of Mitochondrial Dysfunction After Subarachnoid Hemorrhage

Chen Ying,Huang Liyong,Zhang Huiyong,Diao Xiling,Zhao Shuyang,Zhou Wenke

Mitochondrial dysfunction and subsequent autophagy, which are common features in central nervous system (CNS) disorders, were found to contribute to neuronal cell injury after subarachnoid hemorrhage (SAH). (-)-Epigallocatechin-3-gallate (EGCG), the main biological active of tea catechin, is well known for its beneficial effects in the treatment of CNS diseases. Here, the ability of EGCG to rescue cellular injury and mitochondrial function following the improvement of autophagic flux after SAH was investigated. As expected, EGCG-protected mitochondrial function depended on the inhibition of cytosolic Ca 2+ concentration ([Ca 2+ ] i ) influx via voltage-gated calcium channels (VGCCs) and, consequently, mitochondrial Ca 2+ concentration ([Ca 2+ ] m ) overload via mitochondrial Ca 2+ uniporter (MCU). The attenuated [Ca 2+ ] i and [Ca 2+ ] m levels observed in the EGCG-treated group likely lessened oxyhemoglobin (OxyHb)-induced mitochondrial dysfunction, including mitochondrial membrane potential depolarization, mitochondrial membrane permeability transition pore (mPTP) opening, reactive oxygen species (ROS), and cytochrosome c (cyt c) releasing. Subsequently, EGCG can restore the disrupted autophagy flux after SAH both at the initiation and formation stages by regulating Atg5 , LC3B , and Becn-1 ( Beclin-1 ) mRNA expressions. Thus, precondition EGCG resulted in autophagosomes and more autolysosomes compared with SAH group. As a result, EGCG pre-treatment increased the neurological score and decreased cell death. This study suggested that the mitochondrial dysfunction and abnormal autophagy flux synergistically contribute to SAH pathogenesis. Thus, EGCG can be regarded as a new pharmacological agent that targets both mitochondria and altered autophagy in SAH therapy.

Design and construction of IR780- and EGCG-based and mitochondrial targeting nanoparticles and their application in tumor chemo-phototherapy

Jiahe Bao,Yinan Zhao,Jing Xu,Yuanqiang Guo

An integration combination of phototherapy and chemotherapy to treat carcinoma, solving the inner limitation of individual-modal chemical agent-based therapy or phototherapy, emerges to be a strategy with high prospects for achieving synergistic curative effects. The dye IR780-iodide (IR780) close to infrared radiation is a phototherapy agent with high prospects. However, it is limited in its clinical applications due to poor solubility in water. While epigallocatechin-3-gallate (EGCG), naturally resourced green tea polyphenol, has been extensively proven with intrinsic antitumor activity, but it is largely restricted by its low bioavailability in vivo. Hence, novel multiple-function nanoparticles comprising hyaluronic acid (HA) and IR780 were proposed to deliver EGCG, defined as EGCG@THSI nano-scale particles (EGCG@THSI NPs), thereby rapidly solving limitations of EGCG and IR780. Amphiphilic nano-scale carrier was prepared by triphenylphosphine (TPP), hyaluronic acid (HA), cystamine, and IR780, termed as TPP–HA–SS–IR780, and EGCG was loaded into the amphiphilic copolymer by self-assembly. TPP–HA–SS–IR780 endowed the as-synthesized EGCG@THSI NPs with excellent TPP-mediated mitochondrial-targeted and glutathione-triggered rapid drug release properties. As impacted by the integration of phototherapy and chemotherapy, the EGCG@THSI NPs under NIR laser irradiation showed a prominent anti-tumor effect. Taken together, this study presented a multiple-function nano-scale carrier platform with high prospects in improving the therapeutic efficacy of anti-carcinoma drugs.

Effects of Zein-Lecithin-EGCG nanoparticle coatings on postharvest quality and shelf life of loquat (Eriobotrya japonica)

Lijuan Dong,Qingbo Jiao,Jian Gao,Xin Luo,Yuling Song,Ting Li,Chen Huan,Min Huang,Gerui Ren,Qing Shen,Linglin Fu,Hujun Xie,Zisheng Luo

The loquat ( Eriobotrya japonica ) has high nutritional and economic values. However, loquat easily rots at room temperature, resulting in the loss of commodity value. In this study, zein, lecithin, and (−)-epigallocatechin gallate (EGCG) were used to prepare Zein-Lecithin-EGCG (Z-L/E) nanoparticles, and the effects of these nanoparticles on loquat quality were studied. EGCG addition affected the zeta potential values of the nanoparticles, which increased from −11.80 mV to −23.34 mV. The Fourier transform infrared (FTIR) results showed that electrostatic interactions and hydrogen bonding formed among lecithin, zein, and EGCG. The Z-L/E nanoparticles showed great antioxidant activity with the DPPH radical scavenging rate of 84.76% and the ABTS radical scavenging rate of 80.95% for Z-L/E 4:1 sample, and had good antibacterial ability by decreasing the growth rate of E. coli . The coating treatment with Z-L/E nanoparticles for loquats reduced the weight loss and the decay rate by 13.3% and 1.4%, respectively, during the storage at 25 °C for 22 days, and the decrease of titratable acidity (TA) and vitamin C (VC) content in loquats were also effectively delayed. The coating treatment with Z-L/E nanoparticles for loquats could significantly prolong the storage life of fruits and maintain the quality of fruits.

Whey Protein Sodium-Caseinate as a Deliverable Vector for EGCG: In Vitro Optimization of Its Bioaccessibility, Bioavailability, and Bioactivity Mode of Actions

Ali Korin,Mostafa M. Gouda,Mahmoud Youssef,Eman Elsharkawy,Amgad Albahi,Fuchao Zhan,Remah Sobhy,Bin Li

Epigallocatechin gallate (EGCG), the principal catechin in green tea, exhibits diverse therapeutic properties. However, its clinical efficacy is hindered by poor stability and low bioavailability. This study investigated solid particle-in-oil-in-water (S/O/W) emulsions stabilized by whey protein isolate (WPI) and sodium caseinate (NaCas) as carriers to enhance the bioavailability and intestinal absorption of EGCG. Molecular docking revealed binding interactions between EGCG and these macromolecules. The WPI- and NaCas-stabilized emulsions exhibited high encapsulation efficiencies (>80%) and significantly enhanced the bioaccessibility of EGCG by 64% compared to free EGCG after simulated gastrointestinal digestion. Notably, the NaCas emulsion facilitated higher intestinal permeability of EGCG across Caco-2 monolayers, attributed to the strong intermolecular interactions between caseins and EGCG. Furthermore, the emulsions protected Caco-2 cells against oxidative stress by suppressing intracellular reactive oxygen species generation. These findings demonstrate the potential of WPI- and NaCas-stabilized emulsions as effective delivery systems to improve the bioavailability, stability, and bioactivity of polyphenols like EGCG, enabling their applications in functional foods and nutraceuticals.

Natural P-gp inhibitor EGCG improves the acteoside absorption in Caco-2 cell monolayers and increases the oral bioavailability of acteoside in rats

Fei Zhou,Weisu Huang,Tao Xu,Lipeng Wu,Qi Chen,Jiyu Peng,Xin Liu,Baiyi Lu

Acteoside is one of the most widespread phenylethanoid glycosides with pharmacological activities , including antioxidant, neuroprotective property, etc. However, its bioavailability is poor due to the low absorption and P-gp efflux. This study aimed to select food derived P-gp inhibitors for promoting the acteoside absorption and investigate whether the inhibitors could increase the bioavailability and stability of acteoside. Results showed that EGCG and quercetin significantly decreased the BL-to-AP efflux and promoted the AP-to-BL influx of acteoside across Caco-2 monolayers with optimum concentrations of 320 μM EGCG or 240 μM quercetin adding to 320 μM acteoside. EGCG increased the bioavailability of acteoside to 1.43-fold, but quercetin had no such effect. Further study showed that EGCG and quercetin had no effects on the storage and digestion stability of acteoside. This work revealed that EGCG could improve the acteoside absorption across the Caco-2 monolayers and enhance the bioavailability of acteoside in rats.

A-type dimeric epigallocatechin-3-gallate (EGCG) is a more potent inhibitor against the formation of insulin amyloid fibril than EGCG monomer

Rong-zu Nie,Wei Zhu,Jin-ming Peng,Zhen-zhen Ge,Chun-mei Li

Because fibrillary protein aggregates is regarded to be closely associated with many diseases such as Alzheimer's disease, diabetes, and Parkinson's disease, growing interest and researches have been focused on finding potential fibrillation inhibitors. In the present study, the inhibitory effects of epigallocatechin-3-gallate (EGCG) and A-type dimeric epigallocatechin-3-gallate (A-type EGCG dimer) on the formation of insulin fibrillation were compared by multi-dimensional approaches including thioflavin-T (ThT) fluorescence assay , 1-anilinonaphthalene-8-sulfonic (ANS) fluorescence assay, dynamic light scattering (DLS), transmission electron microscopy (TEM), Fourier transform infrared (FTIR) spectroscopy and circular dichroism (CD) spectroscopy. Our results confirmed that A-type EGCG dimer is a more potent inhibitor against the formation of bovine insulin amyloid fibril than EGCG. In addition, A-type EGCG dimer could not only inhibit insulin amyloid fibril formation, but also change the aggregation pathway and induce bovine insulin into amorphous aggregates. The results of the present study may provide a new guide on finding novel anti-amyloidogenic agents.

Preparation of zein-lecithin-EGCG complex nanoparticles stabilized peppermint oil emulsions: Physicochemical properties, stability and intelligent sensory analysis

Hujun Xie,Fangfang Ni,Jian Gao,Chengzhi Liu,Jieyu Shi,Gerui Ren,Shiyi Tian,Qunfang Lei,Wenjun Fang

Peppermint oil emulsions were prepared by using zein-lecithin-EGCG (Z-L/E) complex nanoparticles as emulsifiers. The preparation conditions of emulsions were optimized via measuring the particle size, surface tension and stability of emulsions, and peppermint oil of 3% (particle size = 375 nm, polydispersity index (PDI) = 0.45), the zein:lecithin ratio of 4:1 ( w/w ) (particle size = 396 nm), and the zein:EGCG ratio of 10:1 ( w/w ) (surface tension = 47.32 N/m) was the optimal condition. The rapid stability analysis showed that the instability mechanism of emulsions was ascribed to creaming and stratification, and the stability mechanism of emulsions was explored, indicating that the complex nanoparticles adsorbed on the surface of oil droplets to give Pickering emulsions. Electronic tongue experiments showed that the Z-E/L 4:1 stabilized emulsion was distinguished from the other three samples due to its good stability. The electronic nose experiment could distinguish the emulsions with different droplet sizes.

Simultaneous determination of the pharmacokinetics of A-type EGCG and ECG dimers in mice plasma and its metabolites by UPLC-QTOF-MS

Jin Li,Jian Zeng,Jinming Peng,Yangyang Jia,Chun-Mei Li

A-type epigallocatechin-3-gallate (EGCG) and epicatechin-3-O-gallate (ECG) dimers have multiply biological activities. In this study, the pharmacokinetics of them were investigated in mice after a single dose intravenous administration, and the metabolites in mice plasma and urine were investigated by ultra-performance liquid chromatography-Quadrupole-time of flight mass spectrometer (UPLC-QTOF-MS). Our results showed that the half-life (t1/2) of A-type EGCG and ECG dimers were 116.37 min and 33.04 min, respectively, and the maximal concentration in plasma was 32.81 μg/mL and 55.59 μg/mL, respectively. It was found that two dimers were firstly experienced by quinone methide (QM) fission to form the EGCG and ECG analogue, and the phase II metabolites were generated subsequently. The main metabolites in plasma and urine were glucuronidation and sulphation derivatives. In addition, small molecule weight of phenolic acids were detected in urine.

Comparative study of the properties of lutein nanoliposomes coated with chitosan/(−)-epigallocatechin- 3-gallate (EGCG) complexes

Hongxu Yan,Yayuan Xu,Zhuqing Dai,Zhongyuan Zhang,Yihong Bao,Da-jing Li

BACKGROUND Numerous positive effects have been attributed to lutein, a lipophilic nutrient, including resisting ultraviolet radiation and protecting retinal pigment epithelial (RPE) cells against blue light damage. It also has preventive effects against cardiovascular disease and cancer. However, its use could be limited by its poor stability and low bioaccessibility in the human digestive system. An encapsulation delivery system was therefore developed to resolve these limitations. In this study, chitosan-modified lutein nanoliposomes (CS-LNLs), chitosan-EGCG covalently modified lutein nanoliposomes (C-CS-EGCG-LNLs), and chitosan-EGCG noncovalently modified lutein nanoliposomes (non-C-CS-EGCG-LNLs) were designed. The average particle size, ζ -potential, and retention of lutein during storage were measured to indicate the physicochemical stability of the modified lutein nanoliposomes. The bioaccessibility of modified lutein nanoliposomes was also investigated to demonstrate the availability of lutein in the human digestive system. RESULTS First, Fourier-transform infrared spectroscopy (FTIR) verified that covalent bonds between chitosan and EGCG were formed. Subsequently, ζ -potential results revealed that C-CS-EGCG-LNLs had a relatively stable structure in comparison with lutein nanoliposomes (LNLs). The retention rate of lutein in CS-LNLs, C-CS-EGCG-LNLs, and non-C-CS-EGCG-LNLs was improved, especially in C-CS-EGCG-LNLs (at around 70% of lutein in initial system). An in vitro digestion experiment illustrated that CS-LNLs, C-CS-EGCG-LNLs, and non-C-CS-EGCG-LNLs presented relatively higher bioaccessibility, especially in C-CS-EGCG-LNLs (at around 33% of luein in initial system), which increased 2.5 and 1.65 times in comparison with free lutein and LNLs, respectively. CONCLUSION Overall, the results showed that C-CS-EGCG-LNLs presented greater physicochemical stability and bioaccessibility than LNLs, CS-LNLs, and non-C-CS-EGCG-LNLs. © 2023 Society of Chemical Industry.

Ultrasound-assisted multilayer Pickering emulsion fabricated by WPI-EGCG covalent conjugates for encapsulating probiotics in colon-targeted release

Xian He,Wanshui Yang,Xinsheng Qin

This study demonstrated the influences of ultrasound-assisted multilayer Pickering double emulsion capsules on the pasteurization and gastrointestinal digestive viability of probiotic (L. plantarum) strain liquid. Firstly, the role of ultrasonic homogenization on the morphology of W 1 /O/W 2 double emulsions were studied. The double emulsion formed by ultrasonic intensity at 285 W had a single and narrow distribution with smallest droplet size. The double emulsion particles were then coated with chitosan(Chi), alginate (Alg), and CaCl 2 (Ca). The multilayer emulsion after pasteurization and gastrointestinal digestion both had the highest viability at 5 coating layers, but its particle size (108.65 μm) exceeded the limit of human oral sensory (80 μm). It could be noted that the deposition of 3–4 layers of coating had similar activity after pasteurization/GIT digestion. And droplets with 3 layers of coating were the minimum and most available formulation for encapsulated probiotics ( L. plantarum ). Hence, the results suggest that the use of ultrasound-assisted multilayer emulsions encapsulated with probiotics in granular food and pharmaceutical applications is a promising strategy.

Multi-strategy dynamic cross-linking to prepare EGCG-loaded multifunctional Pickering emulsion/α-cyclodextrin/konjac glucomannan composite films for ultra-durable preservation of perishable fruits

Hongcai Li,Weiteng Tan,Mengxin Hou,Shuang Yang,Caiyun Liu,Mengzhen Han,Jingjing Liang,Zhenpeng Gao

Developing multifunctional films with antibacterial, antioxidant, and sustained-release properties is a robust strategy for preventing contamination of perishable fruits by foodborne microorganisms. This study engineered a sustained-release biodegradable antibacterial film loaded with EGCG (Pickering emulsion (PE)/α-Cyclodextrin (α-CD)/Konjac glucomannan (KGM)) through multi-strategy cross-linking for fruit preservation. EGCG is stabilized using PE and incorporated into the α-CD/KGM inclusion compound; the unique structure of α-CD enhances EGCG encapsulation, while KGM provides the film toughness and surface adhesion. The composite film's physicochemical properties, antioxidant, bacteriostatic and biodegradability were studied. Results showed that Pickering emulsions with 3 % oil phase exhibited excellent stability. Moreover, α-CD introduction increased the loading and sustained release of EGCG from the film, and its concentration significantly affected the light transmission, thermal stability, mechanical strength, mechanical characteristics and antioxidant capacity of the composite membrane. Antioxidant and antimicrobial activities of the composite film increased significantly with increasing α-CD concentration. Application of the film to tomatoes and strawberries effectively inhibited Escherichia coli and Staphylococcus aureus growth, prolonging the shelf-life of the fruits. Notably, the composite film exhibits superior biodegradability in soil. This EGCG-loaded PE/α-CD/KGM composite film is anticipated to be a multifunctional antimicrobial preservation material with sustained-release properties and biodegradable for perishable food applications.
