Mounting evidence suggests that dietary polyphenols exert health benefits partly through their favorable interactions with gut bacteria. However, little is known about polyphenol’s metabolic regulatory effects towards individual bacteria at the molecular level. Ellagic acid (EA), a polyphenol abundantly present in plant-based foods, was found to exhibit prebiotic properties through differential interactions with probiotic-like bacteria, including the EA-to-urolithin converting species such as Gordonibacter urolithinfaciens ( G. uro ). This study aimed to investigate the crosstalk between EA and EA-responsive beneficial bacteria, including both conventional and next-generation probiotics originating from the human gut, and to understand the underlying mechanism by which EA exerts prebiotic activities in vitro . The influence of EA and urolithins on probiotic bacteria was investigated at the levels of fecal microbiota and individual strains via anaerobic culturomics and metabolomics approaches. Results indicate that dietary-level EA favorably regulated gut microbial composition through the enrichment of probiotic genera (e.g., Bifidobacterium and Akkermansia ) in vitro . Regarding individual bacteria, EA supplementation promoted the growth of Lacticaseibacillus rhamnosus GG and Bifidobacterium infantis . Integrated targeted and untargeted metabolomic analyses of intracellular and extracellular metabolites revealed that EA/urolithins modulated metabolic pathways associated with amino acid metabolism, energy production and oxidative stress. Furthermore, G. uro exhibited strong EA uptake ability, facilitating the urolithin bioconversion and cellular accumulation in a dose-dependent manner. Overall, this study provides in-depth understanding on how dietary polyphenols with prebiotic properties regulate the growth and metabolic functions of probiotic-like bacteria.

This study investigates the antioxidant interactions of γ-tocopherol, β-sitosterol, and ellagic acid in iron walnut oil within ethanol and ethyl acetate, assessing their impact on oxidative stability. In ethanol, γ-tocopherol and ellagic acid display initial synergistic effects, enhancing free radical scavenging beyond their individual capabilities. Conversely, γ-tocopherol and β-sitosterol show antagonistic effects, reducing their combined scavenging abilities. Kinetic analysis indicates ellagic acid's robust scavenging, generally outperforming combinations of antioxidants. Fourier-transform infrared spectroscopy revealed that solvent polarity significantly impacts hydrogen bonding interactions. Ethanol, with its higher polarity, promoted more extensive hydrogen bonding compared to ethyl acetate, which imposed constraints on such interactions due to its lower polarity. Notably, despite its poor solubility in ethyl acetate, ellagic acid exhibits enhanced synergy with γ-tocopherol, likely via electron transfer. These findings underscore the intricate dynamics of antioxidant interactions, further emphasizing the pivotal role of solvent polarity in modulating these interactions. These results indicated that solvent properties significantly influence antioxidant behavior, yet they also introduce novel insights specific to the iron walnut oil system.

Rationale : Chemodynamic therapy (CDT) based on the Fe(II)-mediated Fenton reaction is an emerging tumor treatment strategy. However, the catalytic efficiency in tumors is crucially limited by Fe(II). Herein, an endogenous hydrogen sulfide (H 2 S) accelerated Fe(III)/Fe(II) transformation and photothermal synergistically enhanced CDT strategy based on ellagic acid-Fe-bovine serum albumin (EA-Fe@BSA) nanoparticles (NPs) was developed for colon tumor inhibition. On the one hand, the Fe(III) with low catalytic activity in the EA-Fe@BSA NPs could be rapidly reduced to the highly active Fe(II) by the abundant H 2 S in colon cancer tissues. Thus, a rapid Fe(III)/Fe(II) conversion system was established, wherein highly active Fe(II) ions were continuously regenerated to improve the CDT efficiency. On the other hand, the photothermal effect of EA-Fe@BSA NPs also accelerated the production of hydroxyl radicals (•OH), thereby synergistically enhancing the CDT performance and improving the therapeutic efficacy. Methods : The endogenous H 2 S accelerated Fe(III)/Fe(II) conversion and PTT enhanced CDT were investigated by characterization of the Fe valence state and detection of •OH. T 1 -weighted magnetic resonance imaging (MRI) was tested both in vitro and in vivo . The biocompatibility of NPs were examined via MTT assay, hemolysis analysis and routine blood measurements. The enhanced CDT was investigated in HCT116 colon cancer cells by Calcein-AM/PI staining and MTT assay, and tumor inhibition was demonstrated in HCT116 tumor bearing mice. Results : In this work, EA-Fe@BSA NPs were constructed as a CDT theranostic reagent. The H 2 S accelerated Fe(III)/Fe(II) conversion was confirmed, more degradation of MB and generation of •OH demonstrated the enhanced CDT in vitro . EA-Fe@BSA NPs exhibited good T 1 -weighted MRI performance. More importantly, it displayed strong near-infrared (NIR) absorption and excellent photothermal efficiency, further promotes the production of •OH. Hence, the efficacy of CDT was enhanced, and the tumor growth was inhibited efficiently. Conclusion : All results demonstrate that this strategy based on endogenous H 2 S promoted Fe(III)/Fe(II) transformation together with PTT acceleration permits efficient Fenton-reaction- mediated CDT both in vitro and in vivo , which holds great potential for effective colon cancer theranostics.

β‑amyloid (Aβ) aggregation and tau hyperphosphorylation are considered to be the primary pathological hallmarks of Alzheimer's disease (AD). Targeted inhibition of these pathological processes may provide effective treatments for AD. Accumulating evidence has demonstrated that ellagic acid (EA) exerts neuroprotective effects in several diseases. The present study investigated the effects of EA on AD‑associated learning and memory deficits on APP/PS1 double transgenic mice and the underlying mechanisms. APP/PS1 mice or wild‑type C57BL/6 mice were intragastrically administered EA (50 mg/kg/day) or vehicle for 60 consecutive days. The learning and memory abilities of mice were investigated using the Morris water maze test. Hippocampal regions were examined for the presence of amyloid plaques, neuronal apoptosis and tau phosphorylation. Expression levels of APP, Aβ, RAC‑αserine/threonine‑protein kinase and glycogen synthase kinase (GSK)3β in the hippocampus were determined by western blot analysis and ELISA. The results demonstrated that EA treatment ameliorated spatial learning and memory impairment in APP/PS1 mice and significantly reduced neuronal apoptosis and Aβ deposition in the hippocampus (P<0.05 and P<0.01). In addition, EA significantly inhibited the hyperphosphorylation of tau and significantly decreased the activity of glycogen synthase kinase (GSK)3β (P<0.01), which is involved in tau phosphorylation. Overall, these findings indicated that the beneficial effects of EA on AD‑associated cognitive impairments may be attributed to the inhibition of Aβ production and tau hyperphosphorylation, and its beneficial action may be mediated in part, by the RAC‑α serine/threonine‑protein kinase/GSK3β signaling pathway.

Industrial processing of raspberry juice and wine generates considerable byproducts of raspberry pomace. Ellagic acids/ellagitannins, being characterized by their antioxidant and antiproliferation properties, constitute the majority of polyphenolics in the pomace and are valuable for recovery. In the present study, we developed a novel procedure with sodium bicarbonate assisted extraction (SBAE) to recover ellagic acid from raspberry wine pomace. Key parameters in the procedure, i.e., sodium bicarbonate concentration, temperature, time and solid/liquid (S/L) ratio, were investigated by single factor analysis and optimized subsequently by Response Surface Methodology (RSM). Optimal parameters for the SBAE method here were found to be 1.2% (w/v) NaHCO3, 1:93 (w/v) S/L ratio, 22 min and 100 °C. Under these conditions, the ellagic acid yield was 6.30 ± 0.92 mg/g pomace with an antioxidant activity of 79.0 ± 0.96 μmol Trolox eq/g pomace (DPPH assay), which are 2.37 and 1.32 times the values obtained by extraction with methanol–acetone–water solvent, respectively. The considerable improvement in ellagic acid extraction efficiency could be highly attributed to the reactions of lipid saponification and ellagitannin hydrolysis resulted from sodium bicarbonates. The present study has established an organic solvent-free method for the extraction of ellagic acid from raspberry wine pomace, which is feasible and practical in nutraceutical applications. Keywords:ellagic acid;extraction;raspberry;wine pomace;sodium bicarbonate;response surface methodology

Drug resistance is one of the leading causes of treatment failure in current cancer chemotherapy. In addition to the classical drug efflux transporter-mediated chemoresistance, cancer cells with stemness features play a crucial role in escaping the maximum impact of chemotherapy. To sensitize cancer chemotherapy, in a novel approach, the hedgehog pathway inhibitor ellagic acid (EA) is coordinated with Cu 2+ to develop nanoscale metal–organic frameworks (EA-Cu), which are then loaded with doxorubicin (DOX) and modified with targeted chondroitin sulfate (CS) to form the CS/E-C@DOX nanoplatform (CS/NPs). Notably, EA inhibits stemness maintenance by suppressing the hedgehog pathway, while Cu 2+ further decreases stemness features of tumor cells by disrupting mitochondrial metabolism, effectively enhancing DOX-mediated chemotherapy. Meanwhile, EA can act synergistically with Cu 2+ to cause mitochondrial dysfunction and cuproptosis, which effectively decreases ATP levels and subsequently suppresses the activity of P-glycoprotein (P-gp), thus reducing drug efflux and sensitizing DOX-mediated chemotherapy. Additionally, the attached CS endows CS/NPs with specific tumor targeting properties, whereas EA-Cu endows this nanoplatform with pH/glutathione (GSH) dual-responsive release behavior. Taken together, CS/NPs exhibited excellent antitumor effects by inducing cuproptosis and significantly inhibiting cancer cell stemness, which has great potential for overcoming cancer chemoresistance.

Inflammatory bowel disease (IBD) is a kind of chronic non-specific intestinal inflammatory disease, which accompanied by various abnormal psychological states, such as anxiety and depression. Nevertheless, the pathogenesis of IBD with depression is still unclear. Herein, we synthesized an enable a drinkable, liquid in situ-forming hydrogel, which form directly in the stomach through sequential ingestion of a crosslinker solution of CaCl 2 and ε-poly-l-lysine (ε-PL), followed by a drug-containing polymer solution of sodium alginate (SA). Using olsalazine (Olsa) and antioxidant (ellagic acid (EA) and ZnCl 2 ) as small molecule drug (referred as Olsa/EA/Zn), the resulting hydrogel (referred as Olsa/EA/Zn@SA/ε-PL hydrogel) could form rapidly in gastric fluid and exhibit remarkable stability under acidic conditions, unique low swelling properties and a rational release of drug in entering the intestine. We demonstrated that the oral administration of drug-loaded in situ-forming hydrogel with excellent antioxidant, anti-inflammatory activities, cytocompatibility, intestinal retention capacity of the drug, significantly enhanced the IBD therapeutic outcomes, and relieve accompanying depression-like symptoms in colitis mice. Overall, this designed in situ-forming hydrogel oral delivery system will open up new pathway in combined gastrointestinal therapy and psychological comorbidities for IBD patients, especially those with dysphagia.

Exposure to cadmium sulfate (CdSO4) can lead to neurotoxicity. Nevertheless, the precise molecular mechanisms underlying this phenomenon remain unclear, and effective treatment strategies are scarce. This study explored the protective effects of ellagic acid (EA), a natural polyphenolic compound, against CdSO4exposure-induced neurotoxicity in HT22 cells and the underlying molecular mechanisms. Our findings demonstrated that exposure of HT22 cells to CdSO4resulted in apoptosis, which was effectively reversed by EA in a dose-dependent manner. EA supplementation also decreased reactive oxygen species (ROS) and mitochondrial ROS production, reduced malondialdehyde (MDA) levels, and restored the activities of superoxide dismutase (SOD) and catalase (CAT). Additionally, EA supplementation at 5–20 μM significantly counteracted Cd-induced the loss of mitochondrial membrane potential and the decrease of ATP and reduced the ratio of Bax/Bcl-2 and cleaved-caspase-3 protein expression. Furthermore, EA supplementation resulted in the upregulation of Nrf2 and HO-1 protein and mRNAs while simultaneously downregulating the phosphorylation of JNK and p38 proteins. The pharmacological inhibition of c-Jun N-terminal kinase (JNK) partially attenuated the activation of the Nrf2/HO-1 pathway induced by CdSO4and exacerbated its cytotoxic effects. In conclusion, our findings suggest that ethyl acetate (EA) supplementation offers protective effects against CdSO4-induced apoptosis in HT22 cells by inhibiting oxidative stress and activating the Nrf2 signaling pathway. Furthermore, the activation of the JNK pathway appears to play a protective role in CdSO4-induced apoptosis in HT22 cells.

In this paper, two surface oxygen vacancy-rich TiO 2 composite photocatalysts were successfully constructed by solid thermal reduction ((R-Vo)TiO 2 ) and bimetallic doping (Nd-Fe-(D-Vo)TiO 2 ). The oxygen vacancies were characterized in detail using XPS, Raman, EPR and STEM. The results show that the excellent photocatalytic performance of Nd-Fe-(D-Vo)TiO 2 deriving from its doping energy and surface oxygen vacancies. Nd-Fe-(D-Vo)TiO 2 could degrade 58 % of phenol within 120 min, which was much greater than 15 % of TiO 2 and 27 % of (R-Vo)TiO 2 . Three composite photocatalysts, TiO 2 @EA, (R-Vo)TiO 2 @EA and Nd-Fe-(D-Vo) TiO 2 @EA, with better visible light absorption and photocatalytic performance were further obtained by bonding ellagic acid (EA) to the surface of TiO 2 . Nd-Fe-(D-Vo)TiO 2 @EA could almost completely degrade ethinyl estradiol within 20 min with a high minimization degree of 84 %. The possible sites of ethinyl estradiol attacked by free radicals were predicted by theoretical calculation combining with high-resolution mass spectrometry analysis, and the possible degradation mechanism was proposed.

Ellagic acid (EA), a natural polyphenol, features a highly organized planar structure with an extensive sp 2 carbon-conjugated area, making it favorable for light energy absorption and fluorescent emission. However, this planar structure can easily aggregate through π-π stacking, leading to aggregation-caused quenching of fluorescence. In this research, aniline-doped ellagic acid (EAAn 2 ) was successfully synthesized via a C-N coupling reaction to address this issue. The fluorescence properties of EAAn 2 in various organic solvents were thoroughly investigated. Compared to EA, EAAn 2 exhibited a red shift in fluorescence emission, primarily revealing yellow fluorescence instead of the green fluorescence of EA. Moreover, the fluorescence intensity and the concentration at which aggregation-caused quenching occurs for EAAn 2 were substantially improved due to the two grafted aniline molecules. EAAn 2 was utilized as a molecular fluorescent probe in DMSO to detect three kinds of metal ions: alkali and alkaline-earth metal ions, transition metal ions, and post-transition metal ions, using EA as a control. It was found that the combination of EAAn 2 with these metal ions exhibited stronger fluorescence intensity than EA and effectively resisted the blue shift in fluorescence emission due to aniline doping. Based on these results, Mg 2 ⁺ ions could be clearly distinguished from other alkali and alkaline-earth metal ions by differences in their fluorescence emission, as could Cr 2 ⁺ ions among transition metals. This research effectively enhances the fluorescence properties of EA through C-N coupling and demonstrates its greater potential in the detection of metal ions, providing fundamental and valuable information for future fluorescent applications.

In the present study, a low-cost and environmentally-friendly method for the recovery of ellagic acid (EA) from waste pomegranate peel (WPP) was developed. Thermal hydrolysis assisted extraction (THAE) method was designed to efficiently prepare EA at low economic and environmental costs by the simultaneous extraction and hydrolysis of EA and its derivatives using NaH 2 PO 4 solution as extraction solvent, which could be reused at least 5 times after simple post-treatments. Ethyl acetate was chosen as liquid-liquid extraction (LLE) solvent to directly recover EA from THAE extract and regenerate NaH 2 PO 4 solution. After a series of optimization of operation parameters, the recovery rates of EA in the THAE and LLE processes reached ∼94% and 96%, respectively. As all of the solvents used above belong to cheap, eco-friendly, and edible solvents, this proposed method provides a good way to get safe and inexpensive EA-rich extract (with a purity of ∼89%), which could be used as natural ingredient in food, cosmetic, and medicinal industries, as well as starting materials for the preparation of pure EA.

A Brønsted acid ionic liquid, 3-methyl-1-(4-sulfonylbutyl) imidazolium hydrogensulfate ([HO3S(CH2)4mim]HSO4), was used for the first time for the preparation of a sulfonated carbon catalyst. The catalyst was prepared from the residue obtained after recovery of the essential oil from the leaves of Cinnamomum longepaniculatum. The sulfonated carbon catalyst with an amorphous structure attained high acidic efficiency at a sulfonation temperature of 200 °C for 2 h of sulfonation time, and was characterised. SEM morphologies revealed that the carbon catalyst consisted of uniform carbon microspores. FTIR analysis, elemental analysis, and X-ray photoelectron spectroscopy revealed that the sulfonic acid group was successfully introduced on the surface of the sulfonated carbon catalyst. The result of TG analysis showed that the obtained sulfonated carbon catalyst has high thermal stability. Good acid and catalytic activity of the obtained sulfonated carbon catalyst were observed for the preparation of ellagic acid and gallic acid, which is comparable to those of diluted sulfuric acid and a sulfonated carbon catalyst that had been prepared with concentrated sulfuric acid. The excellent reusability of the sulfonated carbon catalyst was also confirmed by repeated experimental trials. In summary, the sulfonated catalyst derived from the residue obtained after recovery of essential oil from the leaves of C. longepaniculatum is an economic, eco-benign and promising substitute for traditional mineral acid catalysts for acidic catalysis in industrial applications.
