Identification of Specific N6-Methyladenosine RNA Demethylase FTO Inhibitors by Single-Quantum-Dot-Based FRET Nanosensors

Yan Zhang,Qing-nan Li,Kaiyue Zhou,Qinfeng Xu,Chun-yang Zhang

The fat mass and obesity-associated enzyme (FTO) can catalyze the demethylation of N6-methyladenosine (m6A) residues in mRNA, regulates the cellular level of m6A modification, and plays a critical role in human obesity and cancers. Herein, we develop a single-quantum-dot (QD)-based fluorescence resonance energy transfer (FRET) sensor for the identification of specific FTO demethylase inhibitors. The FTO-mediated demethylation of m6A can induce the cleavage of demethylated DNA to generate the biotinylated DNA fragments, which may function as capture probes to assemble the Cy5-labeled reporter probes onto the QD surface, enabling the occurrence of FRET between the QD and Cy5. The presence of inhibitors can inhibit the FTO demethylation and consequently abolish FRET between the QD and Cy5. The inhibition effect of inhibitors upon FTO demethylation can be simply evaluated by monitoring the decrease of Cy5 counts. We use this nanosensor to screen several small-molecule inhibitors and identify diacerein as a highly selective inhibitor of FTO. Diacerein can inhibit the demethylation activity of endogenous FTO in HeLa cells. Interestingly, diacerein is neither a structural mimic of 2-oxoglutarate (2-OG) nor a chelator of metal ions, and it can selectively inhibit FTO demethylation by competitively binding the m6A-containing substrate.

Cy5 labeled single-stranded DNA-polydopamine nanoparticle conjugate-based FRET assay for reactive oxygen species detection

Lina Ma,Zhen Lei,Fuyao Liu,Zhenxin Wang

This work reports on a simple and feasible fluorescence resonance energy transfer (FRET) assay for detecting reactive oxygen species (ROS) both in solution and living cell using polydopamine nanoparticle (PDA NP) as energy acceptor and Cy5 labeled single-stranded DNA (Cy5-ssDNA) as energy donor. The Cy5-ssDNA and PDA NPs form self-assembled conjugates (Cy5-ssDNA-PDA NP conjugates) via π-stacking interactions. In the presence of ROS, the PDA NP adsorbed Cy5-ssDNAs can be effectively cleaved, resulting in the release of Cy5 molecules into solution and recovery of fluorescence emission of Cy5. In order to obtain ROS solution, the glucose oxidase-catalyzed oxidation reaction of glucose with O 2 is employed to generate hydrogen peroxide for Fenton-like reaction. The formation of ROS in Fenton-like reaction can be detected as low as glucose oxidase-catalyzed oxidation of 100   pM glucose by the Cy5-ssDNA-PDA NP conjugate-based FRET assay. The recovery ratio of Cy5 fluorescence intensity is increased linearly with logarithm of glucose concentration from 100   pM to 1   μM, demonstrating that the FRET assay has wide dynamic range. In particular, intracellular ROS has been successfully detected in chemical stimulated HepG-2 cells by the Cy5-ssDNA-PDA NP conjugate-based FRET assay with a fluorescence microscopy, indicating that this approach has great potential to monitor ROS in living cells.

Silver nanoparticle plasmonic enhanced förster resonance energy transfer (FRET) imaging of protein-specific sialylation on the cell surface

Tingbi Zhao,Ting Li,Yang Liu

A large amount of proteins are post-translationally modified with a sialic acid terminal oligosaccharide, and sialylation directly affects the function of glycoproteins and adjusts relevant biological processes. Herein, we developed a method for imaging analysis of protein-specific sialylation on the cell surface via silver nanoparticle (AgNPs) plasmonic enhanced Förster resonance energy transfer (FRET). In this strategy, the target monosaccharide was labelled with the FRET acceptor of Cy5 via bioorthogonal chemistry. In addition, aptamer linked AgNPs were combined with the Cy3 fluorophore by DNA hybridization as the FRET donor probe, which could be conjugated to the target glycoprotein based on specific aptamer-protein recognition. The Cy5 fluorescence signal was obtained under the Cy3 excitation wavelength via FRET. Moreover, the FRET fluorescence signal was obviously enhanced owing to the plasmonic effect of AgNPs at an appropriate distance to Cy3 on the cell surface. Hence, the protein-specific sialic acids were detected with high contrast. The results showed that the AgNP plasmonic enhanced FRET method was not only superior to the bare FRET method but also can be used to evaluate the expression of sialoglycoproteins in different cell types under pharmacological treatments. The AgNP plasmonic enhanced FRET method provides a valuable tool in the research of glycan metabolism biological processes, the active site of glycoproteins and drug screening.

A Sensitive “Turn-On” Fluorescent Sensor for Melamine Based on FRET Effect between Polydopamine-Glutathione Nanoparticles and Ag Nanoparticles

Li Tang,Shi Mo,Shi Gang Liu,Yu Ling,Xiao Fang Zhang,Nian Bing Li,Hong Qun Luo

In this work, Ag nanoparticles (AgNPs) were synthesized quickly by a one-step method utilizing polydopamine-glutathione nanoparticles (PDA-GNPs) as a reducing agent. The PDA-GNPs and the generated AgNPs acted as the energy donor and acceptor, respectively. Accordingly, the fluorescence of PDA-GNPs was quenched on the basis of fluorescence resonance energy transfer (FRET). In the presence of melamine, the preferential combination of Ag(I) and melamine to form Ag(I)–melamine complex prevents Ag(I) from forming AgNPs, together with fluorescence enhancement compared with the absence of melamine. Under the optimal conditions including the concentration of AgNO3, reaction time, reaction temperature, and pH, the fluorescence enhancement efficiency has a linear response to the concentration of melamine from 0.1 to 40 μM with a detection limit of 23 nM for melamine. The proposed method is simple, time-saving, and low-cost, which was further applied to detect melamine in real milk products with satisfactory results.

Smartphone-assisted ratiometric FRET aptasensor based on quantum dots and gold nanoparticles for point-of-care testing of zearalenone in cereals

Lianjun Song,Meijun Lu,Puye Liang,Tiange Li,Xianqing Huang,Yan Ma,Libin Wan,Tianlin Wang

Herein, a ratiometric fluorescence aptasensor was established for zearalenone (ZEN) detection based on fluorescence resonance energy transfer (FRET) between aptamer-modified CdTe quantum dots (QDs) as donor and gold nanoparticles (AuNPs) as acceptor. The fluorescence of CdTe QDs quenched by AuNPs based on FRET, while Si QDs with blue fluorescence as reference. The aptamer-modified CdTe QDs were adsorbed to ZEN in the presence of ZEN, and the distance between the energy receptor and the donor increased, which prevented the FRET and led to the fluorescence recovering of CdTe QDs. The aptasensor exhibited a limit of detection of 3.5 pg/mL with a linear range of 0.01–100 ng/mL. In addition, a portable detection device was constructed by integrating with smartphone for point-of-care testing (POCT) of ZEN with satisfactory results, which could provide a promising application for visual POCT of ZEN in cereals.

A FRET-based aptasensor for ochratoxin A detection using graphitic carbon nitride quantum dots and CoOOH nanosheets as donor-acceptor pair

Xiaoya Bi,Lijun Luo,Libo Li,Xiaohong Liu,Bainian Chen,Tianyan You

A new fluorescence aptasensor for Ochratoxin A (OTA) analysis in corn and barley flour was developed owing to the favourable quenching function of cobalt oxyhydroxide (CoOOH) nanosheets . The first combination of graphitic carbon nitride quantum dots (g-CNQDs) and CoOOH nanosheets as efficient energy donor-acceptor pair was reported, and the quenching mechanism was proved by investigating the fluorescence lifetime of g-CNQDs. The aptamer-modified g-CNQDs (g-CNQDs-apt) were adsorbed onto CoOOH nanosheets surface by van der Waals force . Consequently, the Förster resonance energy transfer (FRET) from g-CNQDs-apt to CoOOH nanosheets was initiated, leading to quenched fluorescence. With the addition of OTA, the linear aptamer specifically bound with OTA to form G-quadruplex, which had relatively weak interaction with the CoOOH nanosheets and separated from the nanosheets surface. Thus, the FRET process between g-CNQDs-apt and CoOOH nanosheets was hindered, leading to the fluorescence of g-CNQDs-apt recovered clearly. The developed aptasensor exhibited acceptable detection limit with 0.5 nM and desirable linear relationship from 1 nM to 140 nM. Meanwhile, the aptasensor possessed multiple advantages, including easy operation, rapid detection and high selectivity . Moreover, the aptamer sensing platform was favorably applied for OTA determination in cereal (barley and corn flour), in which the recoveries varied from 94.5% to 101% with the relative standard deviation under 2.24%.

Fluorescence turn-on Cu2-xSe@HA-rhodamine 6G FRET nanoprobe for hyaluronidase detection and imaging

Guojuan Yan,Bo Kong,Jiaqiang Zhao,Huanhuan Ni,Lei Zhan,Chengzhi Huang,Hongyan Zou

The application of nanostructures to design fluorescence resonance energy transfer (FRET) based sensing platforms has been greatly concerned with the demand for sensitive and selective detection of biomolecules. Here, a novel sensitive turn-on fluorescence strategy based on the FRET mechanism has been designed for hyaluronidase (HAase) detection through the modulation of Cu 2-x Se@HA-Rh6G nanoprobe fabricated by self-assembly of rhodamine 6G (Rh6G) together with Cu 2-x Se@HA nanoparticles through electrostatic adsorption. The Cu 2-x Se@HA had extensive localized surface plasma resonance (LSPR) absorption in the wide range of ultraviolet (UV) to near-infrared (NIR) wavelengths and showed good light capture characteristics, which can be acted as good acceptors in the FRET interactions with Rh6G , inducing its efficient fluorescence quenching . In the presence of HAase, the FRET process was disrupted and the fluorescence signal was recovered. In the range of 0.1–10.0 U/mL, the fluorescence recovery of Rh6G showed a good linear relationship with the concentration of HAase, and the detection limit was 0.06 U/mL. The sensing platform has been used for HAase detection in real urine samples and cancer cells imaging.

Novel fluorescent perylene liquid crystal with diphenylacrylonitrile groups: Observation of a large pseudo stokes shift based on AIE and FRET effects

Mingguang Zhu,Youzhen Zhuo,Kaicong Cai,Hongyu Guo,Fafu Yang

A novel perylene liquid crystal with diphenylacrylonitrile groups on 1,7-bay positions was synthesized in high yield. This highly decorated perylene bisimide showed the ordered hexagonal columnar liquid crystalline behaviour between 124.9 and 189.5 °C. Upon excitation of 480 nm, an attenuation of fluorescent intensity was observed with the increase of water fraction in THF/H 2 O mixtures. However, the fluorescence intensity enhanced dramatically and attained a maximum value at water fraction of 40% in THF/H 2 O systems when excited at 330 nm. The fluorescence intensity increased by 10 times and the pseudo Stokes shift achieved 251 nm. These phenomena were well elucidated by the AIE and FRET effect, which were supported by the investigation of fluorescence quantum yield and fluorescence lifetime .

Daylight-driven antimicrobial fabrics: Design strategy for enhancing efficiency based on electrostatic repulsion-FRET synergistic effects of dual photosensitizers

Huayu Han,Liu Hu,Fu Wang,Jiahui Wu,Yi Hu

Designing antimicrobial textile materials with non-specific, broad-spectral absorption and multicolor is of great importance in personal health protection. In this study, a highly efficient, broad-spectrum responsive, and dual-color photodynamic antimicrobial nylon/cotton blended fabric (N/C-RB/ZP) was prepared through a one-bath dyeing process based on photosensitive Rose Bengal (RB) and tetra[ β -(4-sodium sulfonylphenoxy)]-phthalocyanine zinc(II) (ZP). The prepared N/C-RB/ZP shows a visible absorption range of 450–700 nm and a mixed color. Additionally, N/C-RB/ZP-0.5 % exhibits superior singlet oxygen ( 1 O 2 ) production capacity, manifesting a “1 + 1 > 2” synergistic effect. The mechanism of synergistic enhancement of photodynamic efficiency by electrostatic repulsion-fluorescence resonance energy transfer (FRET) process between two kinds of liked-charge dyes on heterogeneous fibers was confirmed. N/C-RB/ZP-0.5 % exhibits excellent antimicrobial activity against Staphylococcus aureus (99.99 %) and Escherichia coli (99.67 %) after 90 min of irradiation under daylight flashlight (14 ± 1 mW/cm 2 ), along with good biological safety. Furthermore, after three cycles or 20 times of standard washing, the antibacterial rate remained above 80 % and 75 %, respectively. This dual-photosensitizers modification method for heterogeneous fibers provides a novel strategy for developing efficient, non-resistant, and color-tunable daylight-driven antimicrobial textiles in the fields of healthcare and personal protection.

Fast detection of E. coli with a novel fluorescent biosensor based on a FRET system between UCNPs and GO@Fe3O4 in urine specimens

Yuan Yao,Guoming Xie,Xin Zhang,Jinshan Yuan,Yulei Hou,Hui Chen

Biosensors based on nanomaterials are becoming a research hotspot for the rapid detection of pathogenic bacteria. Herein, a “turn-on” fluorescent biosensor based on a FRET system was constructed for the fast detection of a representative pathogenic microorganism, namely, E. coli, which causes most urinary tract infections. This biosensor was constructed by utilizing synthesized UCNPs as fluorescent donors with stable luminescence performance in complex biological samples and GO@Fe3O4 as a receptor with both excellent adsorption ability and fluorescence quenching ability. A specific ssDNA selected as an aptamer which could recognize E. coli was immobilized on the UCNPs to form UCNP-Apt nanoprobes. The nanoprobes were adsorbed on the surface of GO@Fe3O4 through the π-stacking interactions between aptamers and GO. In the presence of E. coli, UCNP-Apt nanoprobes detached from GO@Fe3O4 due to the specific recognition of aptamers and bacteria, resulting in obvious fluorescence recovery, and the concentration of bacteria was positively correlated with the intensity of the fluorescence signal; such a “turn-on” signal output mode ensures excellent precision. In addition, the easy magnetic separation of GO@Fe3O4 simplifies the operation process, helping the sensor detect bacteria in 30 minutes with a linear range from 103 to 107 CFU mL−1 and a limit of detection of 467 CFU mL−1. Moreover, recovery test results also showed that the sensor has clinical application potential for the rapid detection of pathogenic microorganisms in complex biological samples.

3D DNAzyme Motor Nanodevice With Self-Powered FRET Amplifier and Self-Supplied H2O2 for Enhancing Human Neutrophil Elastase Profiling and Chemodynamic Therapy in Lung Tumor

Huiyan Du,Ensheng Xu,Yihan Xu,Qingwang Xue,Hongxia Xu,Jibin Song

The development of theragnostic nanosystems integrating FRET (fluorescence resonance energy transfer) imaging and chemodynamic therapy (CDT) for accurate diagnosis and effective treatment of lung tumors is still a big challenge. Herein, a peptide-assembled 3D DNAzyme motor nanodevice is engineered for a self-powered FRET amplifier profiling human neutrophil elastase (HNE) and self-supplied H 2 O 2 enhancing CDT. The nanodevice is prepared by depositing AuNPs on ZIF-8, in which ZIF-8 co-loaded the lysosomal targeting peptide-modified copper peroxides (PCPs) and hairpins (H1, H2, and H3), AuNPs are co-labeled by DNAzyme-peptide (DP) conjugate and H3. In the tumor micro-environment, HNE driven 3D DNAzyme walker followed by an exponential amplification constructed by a synergistic cross-activation between hybridization chain reaction and DNAzyme, generating a self-powered FRET amplifier. The FRET amplifier specifically measures HNE with a sensitivity of 0.026 pM, and successfully images exogenous HNE in living cells and monitors HNE in mouse models. Moreover, the PCPs can target lysosomes, reducing lysosome escape. The self-supplying H 2 O 2 undertaken by PCPs improves the Cu (II)-catalyzed Fenton-like reaction, effectively causing cell apoptosis to inhibit tumor growth. Significantly, the nanodevice successfully screens inhibitors and discriminates the HNE level in normal and lung cancer tissues, suggesting that the nanodevice provides an effective tool for the diagnosis and treatment of lung tumors.

Construction of a dual colorimetric fluorescent imprinting polymer hybrid system for the detection of alpha-fetoprotein based on the multi-FRET effect and modeling of its color response mechanism

Liuyue Xiao,Tianhao Li,Hui Ling,Yanjing Yang,Shian Zhong

The hybrid ratiometric fluorescence sensors (GKHMIPs & RKHMIPs) encompass luminescent substances incorporated within a core-shell architecture. These sensors manifest a diverse array of hues in response to variations in the concentration of alpha-fetoprotein, accomplished through disparities in the quantum dots (QDs)-protein response. This study is devoted to comprehending the principles governing the alteration of fluorescence during the recognition process of the ratiometric fluorescence sensor. A compensation mechanism, rooted in the correlation between fluorescence and protein response, is postulated through a fluorescence interaction model (multi-FRET model). Multi-FRET model can be divided into FRET process and Fluorescence compensate process (FCP), FCP occurs when electron orbits are filled, hindering the fluorescence resonance energy transfer process and leading to the return of excited state electrons to the ground state by alternative pathways. Based on the multi-FRET model we demonstrate the Sherpa formular to calculating the fluorescence-target relation. To assess the visual impact, the CIE-Lab model is introduced to validate the rationale behind the multi-color colorimetric model. The influence of different mixing ratios on the visualization effect is elucidated using the concentration fluorescence response model, and the optimal relationship between wavelength combinations in the tricolor mixing model is proposed.
