Methyl jasmonate enhances isoflavone biosynthesis and antioxidant activities in Fusarium sulawense-infected soybean sprouts

Xiangyu Sun,Yining Cao,Yanxia Wang,Siqi Wu,Trust Beta,Xinchi Shi,Suyan Wang,Daniela D. Herrera-Balandrano,Pedro Laborda

Soybean sprouts are a popular and nutrient-dense traditional food in several countries; thus, they are susceptible to numerous infectious diseases during germination. This study examined the impact of methyl jasmonate (MeJA) at concentrations of 100 and 300 μM on isoflavone levels, antioxidant activities, and disease index against Fusarium sulawense in soybean sprouts. The application of 300 μM MeJA resulted in a 26 % reduction in the disease index of F. sulawense -infected soybean sprouts after 7 d of germination. The isoflavone content in MeJA-treated soybean sprouts increased, particularly on day 5 (7.71–9.57 g kg −1 ). Ten isoflavones were quantified, with genistin (∼1.3 g kg −1 ) being the most abundant and acetyl daidzin (∼0.19 g kg −1 ) the least abundant. On day 5, superoxide dismutase (SOD) and catalase (CAT) exhibited peak levels of 117 and 193 kU kg −1 , respectively, in F. sulawense -infected soybean sprouts when treated with 100 μM MeJA. The total phenolic content (TPC) in MeJA-treated soybean sprouts increased by 2-fold from day 1 to day 7, except for F. sulawense treatment. MeJA boosted the expression of several isoflavone biosynthesis-related genes in germinated soybeans, including PAL , C4H , 4CL , CHS , IFS , and CHI . Collectively, this study suggests that the application of MeJA in F. sulawense -infected soybean sprouts may enhance isoflavone biosynthesis and antioxidant activities.

Enhancement of gibberellic acid production from Fusarium fujikuroi by mutation breeding and glycerol addition

Peng Xiao-Lun,Zhao Wei-Jun,Wang Yuan-Shan,Dai Ke-Lei,Cen Yu-Ke,Liu Zhi-Qiang,Zheng Yu-Guo

Gibberellic acid (GA 3 ) is a natural plant growth hormone that has been widely used in agriculture and horticulture. To obtain higher GA 3 producing strains, the method of screening the strains for resistance to simvastatin was used after treatment with nitrosoguanidine (NTG) and gamma rays. The rationale for the strategy was that mutants showing simvastatin resistance were likely to be high GA 3 producers, as their activity of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase is relatively more effective. GA 3 yield of mutant S109 increased by 14.2% than that of the original strain. The GA 3 production ability in mutant S109 remained relatively stable after ten generations. With the addition of 0.4 g glycerol on the 5th day during the fermentation process in Erlenmeyer flask, maximum GA 3 production of 2.7 g/L was achieved by this mutant, exhibiting 28.6% increase compared with original strain. Furthermore, we also achieved 2.8 g/L GA 3 and had a 33.3% increase with addition 20 g glycerol on the 5th day during the fermentation process in a 5-L bioreactor. Our results indicated efficient GA 3 production could be achieved on the condition that the supply of glycerol at the suitable conditions. This study would lay a foundation for industrial production of GA 3 .

Metabolite interactions mediate beneficial alliances between Bacillus and Trichoderma for effective Fusarium wilt control

Xie Jiyu,Sun Xinli,Wen Tao,Bai Yaoqiang,Qian Tong,Hu Shunjuan,Chen Lihao,Wang Pan,Miao Youzhi,Zhang Ruifu,Kovács Ákos T,Xu Zhihui,Shen Qirong

Bacteria-Fungi Interactions play a crucial role in soil nutrient cycling and plant disease suppression. Bacillus and Trichoderma exhibit antagonism when inoculated on laboratory media, global soil sample analysis reveals a positive correlation between these two genera in addition to enhanced plant-pathogen Fusarium oxysporum suppression and plant growth promotion. Here, we assess cross-kingdom interactions within artificial model communities of Bacillus velezensis and Trichoderma guizhouense. Transcriptomic profiling revealed that in the presence of fungi, the key stress sigma factor of B. velezensis activates expression of biosynthetic genes for antimicrobial secondary metabolite production. Among these, surfactin induces T22azaphilone production in T. guizhouense that hinders oxidative stress. Both surfactin and T22azaphilone contribute to Bacillus and Trichoderma maintenance in soil in the presence of F. oxysporum. Finally, F. oxysporum-secreted fusaric acid temporarily inhibits B. velezensis growth whereas it is efficiently degraded by T. guizhouense. These metabolite-mediated interactions reveal how competing soil microorganisms could form effective alliances that ultimately enhance plant protection against soil-borne pathogens.

Tunable Release Poly(butylene oxalate) Fungicide Nanocarrier for Long-Term Fusarium Crown Rot Management

Haonan Zhang,Donglai Zhang,Bin Yu,Qiuyu Xiong,Yun Fang,Zifeng Yang,Zerui Li,Jingli Cheng,Jinhao Zhao

The design of intelligent pesticide delivery systems based on the characteristics of soil-borne diseases is of great significance for improving pesticide utilization efficiency and achieving long-lasting disease control. Herein, a cost-effective and degradable functional poly(butylene oxalate) (PBO) material was constructed and applied for the encapsulation of metconazole (MEZ) to develop a sustained-release pesticide delivery system (MEZ@PBO). The optimized MEZ@PBO was mainly distributed within the 200–300 nm range, with a fungicide-loading capacity reaching 55.7%. Release experiments confirmed the regulatory effect of the PBO molecular weight on the release rate, with higher molecular weight resulting in a longer sustained release period, thereby prolonging the duration of efficacy. MEZ@PBO significantly enhances the photostability of the fungicide while preventing pesticide loss caused by leaching. Moreover, pot experiments indicated that MEZ@PBO achieved long-lasting control of the Fusarium crown rot. After 14 days, the inhibition rate against Fusarium crown rot was 37.9 and 22.4% higher than those of the metconazole technical material and commercial formulation, respectively. Biosafety evaluation and degradation tests further confirmed the environmental friendliness and crop safety. This work offers a promising strategy for enhancing the stability, release control, and efficacy of fungicides in sustainable agriculture.

Effective assessment of mixed strains against Fusarium oxysporum f. sp. lycopersici causing tomato wilt

Zhe Han,Shengyi Wang,Ao Gao,Meiqi Dong,Yufeng Xiao,Hongyu Pan,Lining Zheng,Hao Zhang

Tomato wilt disease, caused by Fusarium oxysporum f. sp. lycopersici , is a major soil-borne disease affecting tomato cultivation. In this study, two strains exhibiting significant control effects on F. oxysporum f. sp. lycopersici and tomato growth-promoting effects were isolated from the rhizosphere. Through physiological and biochemical identification, as well as 16S rRNA identification, these strains were identified as Enterobacter quasihormaechei L33 and Klebsiella oxytoca TL2, respectively. And the two strains of bacteria were confirmed to have no antagonistic effect through antagonistic compatibility tests. Notably, when these strains were mixed in a ratio of 5:1, they demonstrated remarkable antifungal activity. The mixed strains induced mycelium swelling, necrosis, and rupture in F. oxysporum f. sp. lycopersici , resulting in a 56.65 % inhibition of mycelial fresh weight. Additionally, they inhibited spore production with an inhibition rate of 787.5 %, and the inhibition rate of the lipopeptide produced reached 93.80 % at 50 mg/mL. Furthermore, in potting experiments, the mixed strains reduced the disease incidence by 36.25 % after 8 weeks. Additionally, compared to the control group, the mixed bacterial strain group enhanced fresh weight, plant height, chlorophyll a and b levels, and root activities by 96.31 %, 18.51 %, 184.48 %, 53.03 %, and 44.31 %, respectively. In addition, mixed strains can inhibit the cell wall degrading enzymes produced by Fusarium oxysporum f sp., induce plant defense enzymes, including increased activity of peroxidase, catalase, and superoxide dismutase, and reduce the decrease in malondialdehyde levels. The mixed strains could colonize tomato plant roots. Overall, these findings suggest a potential biocontrol agent deriving from the mixed strains against tomato wilt disease.

Dynamicomic analysis reveals the biocontrol activity of a Streptomyces avermitilis SM12 against Fusarium spp

Xian Ma,Chenbo Jia,Jianyu Su,Wenyu Lu,Rui Zheng,Sijun Yue

Streptomyces avermitilis is widely recognized for its remarkable ability to produce diverse secondary metabolites with broad-spectrum antifungal activity. In this study, we employed an integrated multi-omics approach to investigate the antifungal mechanisms of S. avermitilis SM12 and explore its potential as a biocontrol agent. Genomic analysis revealed 39 biosynthetic gene clusters (BGCs), nine of which are predicted to encode antimicrobial compounds. Notably, a gene cluster showing high homology to the oligomycin biosynthetic pathway was identified. Metabolomic profiling via LC–MS and HS–SPME–GC–MS detected several antifungal metabolites, including oligomycin, phthoxazolin, ectoine, and the volatile compound 2,3-hexanedione. The cell-free supernatant obtained after 4 d fermentation significantly suppressed the growth of Fusarium solani and Fusarium oxysporum ( P  < 0.05). Subsequent in vitro assays validated the potent antifungal activity of oligomycin, ectoine, and 2,3-hexanedione. Integrated transcriptomic analysis demonstrated temporal activation of the oligomycin and phthoxazolin homologous BGC during the peak antifungal production phase, indicating its crucial role in biocontrol activity. Furthermore, time-series transcriptomics uncovered multifaceted regulatory mechanisms operating at post-transcriptional and translational levels. These results provide key insights into the coordination between BGC expression and metabolite synthesis in S. avermitilis SM12, highlighting promising targets for genetic engineering to enhance antifungal efficacy. This study systematically reveals the molecular basis of biocontrol activity in S. avermitilis SM12, supporting its potential application in sustainable agriculture.

Isolation, Identification, and Culture Optimization of a Novel Glycinonitrile-Hydrolyzing Fungus—Fusarium oxysporum H3

Gong Jin-Song,Lu Zhen-Ming,Shi Jing-Song,Dou Wen-Fang,Xu Hong-Yu,Zhou Zhe-Min,Xu Zheng-Hong

Microbial transformation of glycinonitrile into glycine by nitrile hydrolase is of considerable interest to green chemistry. A novel fungus with high nitrile hydrolase was newly isolated from soil samples and identified as Fusarium oxysporum H3 through 18S ribosomal DNA, 28S ribosomal DNA, and the internal transcribed spacer sequence analysis, together with morphology characteristics. After primary optimization of culture conditions including pH, temperature, carbon/nitrogen sources, inducers, and metal ions, the enzyme activity was greatly increased from 326 to 4,313 U/L. The preferred carbon/nitrogen sources, inducer, and metal ions were glucose and yeast extract, caprolactam, and Cu 2+ , Mn 2+ , and Fe 2+ , respectively. The maximum enzyme formation was obtained when F. oxysporum H3 was cultivated at 30 °C for 54 h with the initial pH of 7.2. There is scanty report about the optimization of nitrile hydrolase production from nitrile-converting fungus.

Essential oil from Sabina chinensis leaves: A promising green control agent against Fusarium sp.

Zhang Jianyun,Zhao Ziyi,Liang Wenyu,Bi Jingyi,Zheng Yuguang,Gu Xian,Fang Huiyong

Sabina chinensis is a woody plant with important ecological functions in different regions of China, but its essential oils (EO) against plant pathogenic fungi remain largely undetermined. The purpose of our study was to assess the chemical composition and antifungal activity of S. chinensis EO based on optimization of the extraction process. In this study, an actionable and effective model with the experimental results and identified optimum conditions (crushing degree of 20 mesh, liquid–solid ratio of 10.1:1, immersion time of 9.1 h) was established successfully to achieve an extraction yield of 0.54%, which was basically consistent with the theoretical value. A total of 26 compounds were identified using headspace gas chromatography–mass spectrometry (GC–MS) and showed that the major constituent was β-phellandrene (26.64-39.26%), followed by terpinen-4-ol (6.53-11.89%), bornyl acetate (6.13-10.53%), etc. For Petri plate assays, our experiments found for the first time that S. chinensis EO revealed high and long-term antifungal activity against the tested strains, including Fusarium oxysporum and Fusarium incarnatum, at EC50 values of 1.42 and 1.15 µL/mL, which especially reached approximately 76% and 90% growth inhibition at a dose of 0.2 µL/mL, respectively. Furthermore, the antifungal activity of EO from different harvest periods showed remarkable variation. The orthogonal partial least-squares discriminant analysis (OPLS-DA) method revealed 11 metabolites with chemical marker components, and 5 of its potential antifungal activities, terpinen-4-ol, α-terpineol, α-elemol, γ-eudesmol, and bornyl acetate, were strongly correlated with the mycelial inhibition rate. In total, this study explored the antifungal activity of EO against root rot fungus as a potential fungicide and provided valuable information into developing potential products from natural agents.

Antagonistic and Detoxification Potentials of Trichoderma Isolates for Control of Zearalenone (ZEN) Producing Fusarium graminearum

Tian Ye,Tan Yanglan,Yan Zheng,Liao Yucai,Chen Jie,De Boevre Marthe,De Saeger Sarah,Wu Aibo

Fungi belonging to Fusarium genus can infect crops in the field and cause subsequent mycotoxin contamination, which leads to yield and quality losses of agricultural commodities. The mycotoxin zearalenone (ZEN) produced by several Fusarium species (such as F. graminearum and F. culmorum) is a commonly-detected contaminant in foodstuffs, posing a tremendous risk to food safety. Thus, different strategies have been studied to manage toxigenic pathogens and mycotoxin contamination. In recent years, biological control of toxigenic fungi is emerging as an environment-friendly strategy, while Trichoderma is a fungal genus with great antagonistic potentials for controlling mycotoxin producing pathogens. The primary objective of this study was to explore the potentials of selected Trichoderma isolates on ZEN-producing F. graminearum, and the second aim was to investigate the metabolic activity of different Trichoderma isolates on ZEN. Three tested Trichoderma isolates were proved to be potential candidates for control of ZEN producers. In addition, we reported the capacity of Trichoderma to convert ZEN into its reduced and sulfated forms for the first time, and provided evidences that the tested Trichoderma could not detoxify ZEN via glycosylation. This provides more insight in the interaction between ZEN-producing fungi and Trichoderma isolates.

Study on the Antifungal Activity of Gallic Acid and Its Azole Derivatives against Fusarium graminearum

Yilin Zheng,Yuqi Geng,Wenlong Hou,Zhe Li,Caihong Cheng,Xiuping Wang,Yuedong Yang

The wheat scab caused byFusarium graminearum(F. graminearum) has seriously affected the yield and quality of wheat in China. In this study, gallic acid (GA), a natural polyphenol, was used to synthesize three azole-modified gallic acid derivatives (AGAs1–3). The antifungal activity of GA and its derivatives againstF. graminearumwas studied through mycelial growth rate experiments and field efficacy experiments. The results of the mycelial growth rate test showed that the EC50of AGAs–2 was 0.49 mg/mL, and that of AGAs–3 was 0.42 mg/mL. The biological activity of AGAs–3 onF. graminearumis significantly better than that of GA. The results of field efficacy tests showed that AGAs–2 and AGAs–3 significantly reduced the incidence rate and disease index of wheat scab, and the control effect reached 68.86% and 72.11%, respectively. In addition, preliminary investigation was performed on the possible interaction between AGAs–3 andF. graminearumusing density functional theory (DFT). These results indicate that compound AGAs–3, because of its characteristic of imidazolium salts, has potential for use as a green and environmentally friendly plant-derived antifungal agent for plant pathogenic fungi.

A new plant SUMO ligase, MPEL1, synergizes with MAPK16 to regulate resistance against Fusarium pathogens

Feng Xuanjun,Zhang Weixiao,Guan Huarui,Yang Haolan,Wu Na,Chen Yuting,Mao Xin,Xiao Huihui,Li Mengling,Xiong Hao,Li Yinzhi,Zhang Zhaoyu,Liu Muyan,Jia Li,Cha Shuang,She Yizhen,Zhang Xuemei,Liao Haicheng,Sun Guangchao,Liu Yaxi,Wang Wenming,Xu Yunbi,Zhang Yuelin,Lu Yanli

SUMOylation is critical for plant growth and defense, and the specific substrate recognition mediated by SUMO ligases dictates the biological processes in which SUMOylation functions. However, only four SUMO ligases involved in SUMOylation have been reported in plants. Here, we report MPEL1 as a new SUMO ligase in maize, which paradoxically exhibits SUMO ligase activity despite its sequence homology to SUMO-targeted ubiquitin ligases (STUbL). MPEL1 stabilizes the plant-specific mitogen-activated protein kinase 16 (MAPK16) via SUMOylation at Lys526 within C-terminal domain (CTD). MAPK16 directly binds to JAZ20 via its C-terminal domain and phosphorylates JAZ20 at Thr12/Ser13 residues, thereby triggering its proteasomal degradation and activating broad-spectrum resistance against two Fusarium pathogens. These pathogens cause devastating maize ear rot and stalk rot, leading to severe yield losses and serious food safety concerns worldwide. Notably, MAPK16 overexpression and JAZ20 knockout enhance disease resistance without yield penalty, highlighting their potential for crop improvement. These findings expand the plant SUMO ligase family and provide targets for breeding Fusarium-resistant crops.

Comparative antifungal activity of three essential oils against mycotoxigenic fusarium graminearum and multi-omics analysis

Yating Cheng,Hang Yu,Longxiang Wang,Guangshuang Tu,Shah Meer Muhammad,Xiaoyuan Gong,Xinyu Lei,Jiarui Dong,Xia Fan,Shuai Wang

Carvacrol, cinnamaldehyde and citral inhibited F. graminearum development and DON production, outperforming SDA. • Three essential oils (EOs) downregulated key TRI genes, whereas SDA paradoxically upregulated TRI11 and failed to effectively suppress DON production. • Three EOs disrupted cell walls, membranes and mitochondrial integrity. • Multi-omics revealed EOs disruption of terpenoid and energy metabolism. • Carvacrol-cinnamaldehyde mixtures reduced wheat crown rot and DON. This study aims to identify eco-friendly essential oils (EOs) with dual antifungal and anti-mycotoxigenic activity against Fusarium graminearum , a destructive cereal pathogen associated with yield loss and deoxynivalenol (DON) contamination, compare their bioactivity and elucidate the underlying mechanisms. Among the seven tested EOs, carvacrol, cinnamaldehyde and citral were the most effective, markedly inhibiting mycelial growth, conidial germination, and DON production. These EOs showed greater inhibitory activity than sodium diacetate (SDA), which upregulated key TRI genes. Multi-omics analyses revealed that the tested EOs induced cell wall and membrane damage, mitochondrial dysfunction and perturbations in critical metabolic pathways, including terpenoid biosynthesis and energy metabolism. Wheat seedling assays further demonstrated that carvacrol, cinnamaldehyde and their synergistic combinations substantially reduced crown rot and DON accumulation, whereas SDA exacerbated the disease. These findings identify promising EO candidates and provide a robust scientific basis for developing EO-based sustainable biocontrol strategies against F. graminearum in wheat.
