A label-free electrochemical immunosensor for the detection of cancer biomarker α-fetoprotein (AFP) based on hydroxyapatite induced redox current

Jisan Sun,Dazhi Tian,Qingjun Guo,Li Zhang,Wentao Jiang,Minghui Yang

A sensitive and selective electrochemical immunosensor for label-free detection of cancer biomarker α-fetoprotein (AFP) was developed based on hydroxyapatite (HAP) nanoparticles induced electrochemical current. HAP nanoparticles and graphene oxide (GO) were co-immobilized onto an electrode surface to modify the electrode. The phosphate groups contained in the HAP nanoparticles can react with molybdate to form redox molybdophosphate precipitates on the electrode surface, generating electrochemical current. GO enhanced the electrochemical current intensity due to its good conductivity. The following immobilization of the anti-AFP antibody and capture of AFP resulted in the formation of an immuno-complex layer on the electrode. This immuno-complex layer prevented the reaction of hydroxyapatite nanoparticles with molybdate, leading to the decrease of the electrochemical current, and the current change is proportional to the concentration of AFP detected. A wide linear range (0.01–10 ng mL−1) and low detection limit (5 pg mL−1) towards AFP detection were obtained. The reliability of the proposed immunosensor was evaluated by the detection of AFP in human serum samples. This study demonstrated the potential of the developed immunosensor in ultrasensitive detection of AFP or other biomarkers.

Fabrication of core–shell Ag@pDA@HAp nanoparticles with the ability for controlled release of Ag+ and superior hemocompatibility

Keling Chen,Kenan Xie,Qin Long,Lijun Deng,Zhiqiang Fu,Huanhuan Xiao,Lu Xie

Silver nanoparticles (Ag-NPs) are a type of crucial bactericide due to their excellent antibacterial properties. However, the application of Ag-NPs in clinics and in bone tissue engineering is hampered because of their unabiding antibacterial efficacy and undesirable biological properties such as cytotoxicity and inferior hemocompatibility. In this study, Ag-NPs were modified with polydopamine (pDA) and hydroxyapatite (HAp) to prepare novel and multifunctional core (Ag-NPs)–shell (pDA)–shell (HAp) nanoparticles (Ag@pDA@HAp-NPs). The Ag+ release rate was reduced by about 80% on the first day, and then Ag+ was slowly released, which implied that the core–shell Ag@pDA@HAp-NPs could effectively control the release of Ag+. The core–shell Ag@pDA@HAp-NPs could obviously inhibit the growth of E. coli and S. aureus, and the bactericide rate of the Ag@pDA@HAp-NPs was up to 99.99% after being cultured for 24 h. Furthermore, the synthesized Ag@pDA@HAp-NPs exhibited superior hemocompatibility due to the favorable blood compatibility of the pDA coating and HAp shell as well as the controlled release of Ag+. Thus, the core–shell Ag@pDA@HAp-NPs, with the ability for the controlled release of Ag+ and superior hemocompatibility, are promising for reducing cytotoxicity and achieving long-term antibacterial properties, and could have potential applications in clinics and in bone tissue engineering.

Synthesis, modification, and chlorhexidine loading of polymer modified-HM-HAP particles: In vivo antibacterial analysis and cell cytotoxicity assessment

Farishta Shafiq,Chenyu Liu,Simiao Yu,Yongxin Pan,Min Ji,Qingzhao Shi,Weihong Qiao

Hydroxyapatite (HAP) is commonly known as an excellent biocompatible and a potential solution to be used in drug delivery applications. This paper examines the synthesis, modification, and loading of chlorhexidine (CHD) drug into hollow mesoporous hydroxyapatite (HM-HAP) particles to increase the antibacterial efficacy for oral infections. HM-HAP particles were synthesized and modified with different weight polymers that is, polyethylene glycol (PEG (1000, 2000, 4.6k)) and polyethyleneimine (PEI (1200, 1800)), to improve the surface properties and drug loading ability. Chlorhexidine was effectively loaded onto both unmodified and polymer-modified HM-HAP samples, achieving loading efficiencies up to 81.67% for HM-HAP and 77.85% for PEG-1000/HM-HAP verified by UV-vis analysis. The CHD-loaded samples were then eventually incorporated into sodium polyacrylate (PAAS) gels to improve their sticking properties, hence enabling effective adherence to the treatment site and enhancing the local antibacterial effect. The antibacterial activity of both CHD-loaded polymer-modified HM-HAP samples and CHD-loaded polymer-modified HM-HAP gel samples was evaluated both in vitro and in vivo. In in vitro tests, PEI-1200/HM-HAP and PEG-1000/HM-HAP gels exhibited the largest inhibition zones of ∼1.6 cm against Escherichia coli and ∼1.5 cm against Staphylococcus aureus , respectively. In vivo study evaluated the antibacterial efficiency of the CHD-loaded polymer modified HM-HAP gel samples by isolating oral bacteria from mouse teeth, with Gel-PEI-1200/HM-HAP@CHD and Gel-PEG-1000/HM-HAP@CHD samples demonstrating superior antibacterial performance. These findings suggest that CHD-loaded polymer-modified HM-HAP is a promising therapeutic drug delivery system for oral health applications, with PEI-modified HM-HAP exhibiting the slight quickest release and PEG-modified HM-HAP showing sustained activity.

Preparation of CaSiO3 porous scaffold via DLP 3D printing and its surface modification through in situ growth of HAp

Yang Liu,Ruixue Sun,Jianmin Han

Calcium silicate (CaSiO 3 , CS) bioceramic has received widespread attention in the field of bone repair due to its excellent bone conductivity, osteoinductivtiy, and degradability. In this study, the porous CS scaffold was firstly prepared by digital light processing (DLP) 3D printing. The effects of solid loading, pore size, and sintering temperature on the compressive strength, porosity and shrinkage of the CS scaffold were thoroughly investigated. When the solid loading is 35 vol.%, the pore size is 500 µm, and the sintering temperature is 1200°C, the prepared CS scaffold has high porosity (48.96 %) and high compressive strength (32.13 MPa). To solve the problems caused by the rapid degradation of CS, surface modification of the prepared CS scaffold was further conducted through in situ growth of hydroxyapatite (HAp) on its surface. A large amount of HAp nanorods homogeneously grow on the surface of the CS porous scaffold when the concentration of KH 2 PO 4 in hydrothermal solution is 0.01 mol/L. Moreover, the phase composition and morphology of HAp grown on the surface of the CS scaffold can be controlled through controlling the concentration of KH 2 PO 4 .

HAp@GO drug delivery vehicle with dual-stimuli-triggered drug release property and efficient synergistic therapy function against cancer

Rui Sang,Min Chen,Yuanyi Yang,Yunfei Li,Jiacheng Shi,Yi Deng,Xianchun Chen,Weizhong Yang

Nanoscale hydroxyapatite (HAp) is an optimal candidate material in biomedical area for its good biocompatibility and bioactivity. In this study, HAp nanorods are prepared via hydrothermal method and combined with monolayered graphene oxide (GO). The obtained HAp@GO with excellent biocompatibility is revealed to have high drug loading capacity (698.7 μg/mg) for anticancer drug doxorubicin (DOX) and efficient photothermal conversion property. And the drug release property of DOX loaded HAp@GO (HAp@GO-DOX) is demonstrated to be controlled by pH and near-infrared light, which is favorable for cancer therapy. in vitro studies on cancer therapy demonstrate that the combined treatment, compared with either chemotherapy or photothermal therapy alone, has better synergistic therapeutic effect. These findings prove the great potential application of the nanocomposites for cancer therapy.

Organized intrafibrillar mineralization, directed by a rationally designed multi-functional protein

Hang Ping,Hao Xie,Bao-Lian Su,Yi-bing Cheng,Weimin Wang,Hao Wang,Yucheng Wang,Jinyong Zhang,Fan Zhang,Zhengyi Fu

Taking lessons from the structure-forming process of biominerals in animals and plants, one can find tremendous inspirations and ideas for developing advanced synthesis techniques, which is called bio-process inspired synthesis. Bone, as a typical representative of biominerals, is constituted of mineralized collagen fibrils, which are formed under the functions of non-collagenous proteins (NCPs). Intrafibrillar mineralization is the consequence of a synergy among several NCPs. In the present study, we have designed a multi-functional protein, named (MBP)–BSP–HAP, based on bone sialoprotein (BSP) and hydroxyapatite binding protein (HAP), to mimic the intrafibrillar mineralization process in vitro. The three functional domains of (MBP)–BSP–HAP provide the artificial protein with multiple designated functions for intrafibrillar mineralization including binding calcium ions, binding collagen, and binding hydroxyapatite. Platelet-like hydroxyapatite crystals periodically arranged inside the collagen fibrils have been achieved under the function of (MBP)–BSP–HAP. The mechanism of intrafibrillar mineralization directed by the multi-functional protein was proposed. This work may not only shed light on bio-process inspired approaches for more economic and efficient biomimetic synthesis, but also be helpful in understanding the natural process of bone formation for bone regeneration and tissue repair.

Effects of hydroxylapatite microtubules/polylactic acid guided tissue regeneration membrane on periodontal bone

Junyan Wang,Liang Liang,Wei Shang,Noor Huda Ismail,Raja Azman Awang

The main goal of periodontal therapy is to fully regenerate the bone that supports teeth while preventing non-osteogenic tissues from interfering with bone regeneration . There is no double-functional repair material in the existing treatment method. Hydroxyapatite (HAp) has been used as a bone substitute in biomedical applications. Polylactic acid (PLA) can effectively inhibit the growth of adherent cells and play a protective role as a barrier. It is possible to combine these two methods to prepare a guided regeneration membrane for guiding periodontal bone regeneration. In this study, HAp microtubules were prepared through hydrothermal synthesis , and bilayer films were prepared through self-evaporation with PLA. The HAp microtubules/PLA membrane had better tensile and breaking strengths and lower fracture productivity than the PLA membrane. The expansion rate reached 34% after 2 h, and the degradation rate exceeded 25% after 40 days. The results showed that the membrane composite had the clinically quoted mechanical requirements. The results of the cell experiments also showed that the membrane had no cytotoxicity. The HAp microtubule side can promote the expression of genes and proteins related to osteogenesis, while the PLA side can inhibit cell adhesion . The HAp microtubules/PLA membrane has stability and good mechanical properties. The double-layer structure has the function of a biological barrier and promotes bone formation.

Functionalization of SF/HAP Scaffold with GO-PEI-miRNA inhibitor Complexes to Enhance Bone Regeneration through Activating Transcription Factor 4

Lingling Ou,Yong Lan,Zhiqiang Feng,Longbao Feng,Junjie Yang,Yu Liu,Liming Bian,Jiali Tan,Renfa Lai,Rui Guo

Evidence indicates that microRNAs (miRNAs) play vital roles in regulating osteogenic differentiation and bone formation. Methods : Here, we show that a polyethyleneimine (PEI)-functionalized graphene oxide (GO) complex efficiently loaded with the miR-214 inhibitor is assembled into silk fibroin/hydroxyapatite (SF/HAP) scaffolds that spatially control the release of the miR-214 inhibitor. Results : SF/HAP/GO scaffolds with nanosized GO show high mechanical strength, and their hierarchical microporous structures promote cell adhesion and growth. The SF/HAP/GO-PEI scaffolds loaded with mir-214 inhibitor (SF/HAP/GPM) were tested for their ability to enhance osteogenic differentiation by inhibiting the expression of miR-214 while inversely increasing the expression of activating transcription factor 4 (ATF4) and activating the Akt and ERK1/2 signaling pathways in mouse osteoblastic cells (MC3T3-E1) in vitro . Similarly, the scaffolds activated the osteoblastic activity of endogenous osteoblast cells to repair critical-sized bone defects in rats without the need for loading osteoblast cells. Conclusion : This technology is used to increase osteogenic differentiation and mineralized bone formation in bone defects, which helps to achieve cell-free scaffold-based miRNA-inhibitor therapy for bone tissue engineering.

Fabrication of an Asymmetric PCL-HAP/CS Bilayer Composite Membrane with Dual Osteogenic and Fibrogenic Functions for Guided Bone Regeneration

Guijuan Li,Enhui Wang,Kangkang Zhao,Wei Xia,Fuli Tian,Yanqiang Li

This study develops a PCL-HAP/CS bilayer membrane to address limitations in current bone regeneration materials. Traditional single-component materials often exhibit poor biocompatibility, mechanical mismatch, and insufficient osteogenic capacity. To overcome these issues, a composite structure was engineered by combining hydroxyapatite (HAP) synthesized via co-precipitation and silane-treated for enhanced compatibility, polycaprolactone (PCL) for mechanical strength, and chitosan (CS) for bioactivity. The bilayer design features a PCL-HAP layer fused with a CS layer, mimicking natural bone’s stratified architecture. Material characterization confirmed the composite’s structural integrity and functional properties. In vitro tests demonstrated selective cell compatibility: osteoblasts thrived on the mineral-rich PCL-HAP side, while fibroblasts showed enhanced adhesion on the CS layer. Rat skull defect experiments revealed dual functionality—the membrane provided mechanical support for bone regeneration while delivering antibacterial effects to surrounding soft tissues. The synergistic combination leverages PCL’s durability, HAP’s osteoinduction, and CS’s antimicrobial properties, effectively compensating for individual material weaknesses. This asymmetric membrane design shows significant potential for clinical bone repair applications, particularly in oral surgery, by simultaneously addressing hard tissue regeneration and soft tissue protection requirements.

HAp Thermosensitive Nanohydrogel Cavities Act as Brood Pouches to Incubate and Control-Release NSCs for Rapid Spinal Cord Injury Therapy

Min Hao,Dapeng Zhang,Wenhan Wang,Zhijian Wei,Jiazhi Duan,Jianlong He,Xiaohong Kong,Yuanhua Sang,Shiqing Feng,Hong Liu

Limitations such as the cell life span, cellular activity, and cellular diffusion outside the lesion make the direct repair of spinal cord injury (SCI) by neural stem cell (NSC) transplantation challenging. Here, an injectable hydroxyapatite (HAp) thermosensitive nanohydrogel is proposed to achieve stem cell storage and release, as well as fate regulation, for therapy. The cavities formed in situ functioned as brood pouches for cell proliferation. The enzyme-induced collapse of cavities released mouse NSCs (mNSCs) gradually and continuously. The endocytosis of HAp nanorods by encapsulated mNSCs enhanced the property of mNSC neuronal differentiation. The HAp thermosensitive nanohydrogel with mNSCs exert an inspiring therapeutic effect on a complete transected SCI model and has prospects in material design to improve SCI therapy with stem cells.

Studies of a novel bone-targeted nano drug delivery system with a HAP core-PSI coating structure for tanshinol injection

Fengbo Yu,Hongyuan Wang,Qiang Wang,Fengguo Zhai,Jinghua Wang,Chunming Huang,Liao Cui

Tanshinol (Tan) has good therapeutic effects on osteoporosis, fracture, and bone trauma repair. However, it is easily oxidised, has low bioavailability and a short half-life. To solve these problems, the study aimed to develop a novel bone-targeted nano-sustained-release drug delivery system PSI-HAPs for the systemic administration of Tan. This proposed system has hydroxyapatite (HAP) as the core to load drug and polysuccinimide (PSI), PEG-PSI (Polyethylene glycol, PEG), and ALN-PEG-PSI (Alendronate sodium, ALN) as the coating materials to form nanoparticles. The article examines the various PSI-HAPs’ entrapping efficiency (EE, %), drug loading capacity (DLC, %), and distribution to determine the best PSI-HAP formulation in vivo. The in vivo experiment showed that the ALN-PEG-PSI-HAP (ALN-PEG/PSI molar ratio = 1:20) was the best preparation due to its higher distribution on bone (120 h) and lower distribution in the other tissues. The determined preparation was a uniformly spherical or sphere-like nanoparticle with a negative zeta potential. Additionally, it exhibited pH-sensitive drug release in PBS based on an in vitro drug release test. The proposed PSI-HAP preparations were prepared in the water solution using a facile preparation process without ultrasound, heating, and other conditions, which can significantly affect the stability of drugs.

Microstructure, mechanical properties, in vitro degradation behavior and cytocompatibility of biodegradable Zn-3Fe-HAP composites prepared by vacuum heating-press sintering

Ruifeng Shan,Runhua Yao,Hui Wang,Lin Liu,Yuyu Zhao,Xiaohong Yao,Yonghua Sun,Ruiqiang Hang

Zinc (Zn) is a promising biodegradable material, and have attracted widespread attention due to its befitting degradation rate compared to magnesium (Mg) and iron (Fe). However, the inferior mechanical strength of pure Zn limits its application for bone repair. Therefore, there is an urgent need to improve its mechanical properties , regulate its degradation rate, and endow it with optimal biocompatibility. The alloying of metallic elements and preparation of Zn-based composites can be a promising approach for this purpose. In this study, the pure Zn and Zn-3 wt%Fe-xHAP (x = 0, 5, 10, 15, 20 wt%) samples were fabricated by vacuum heating-press sintering. The microstructure, compressive mechanical property, in vitro degradation behavior, antibacterial property and in vitro cytocompatibility of different samples were investigated. We confirmed that the secondary phase Zn 13 Fe significantly improved the mechanical property, and micro-galvanic between Zn and Fe accelerated degradation of the samples. Compressive yield strength of all samples ranges from 85.65 MPa to 301.99 MPa, compressive elastic modulus varies between 8.26 GPa and 15.03 GPa, and corrosion rates are significantly improved which are about 0.005–3.065 mm/y. Moreover, Zn-3Fe-xHAP samples exhibited better biocompatibility and antibacterial property than pure Zn, especially the sample of Zn-3Fe-5HAP. In summary, biodegradable Zn-based composites prepared by vacuum heating-press sintering could be promising material for bone repair.
