Sentinel lymph node imaging is important for breast tumor staging and prediction of postoperative metastasis . However, clinical sentinel lymph node imaging has limitations such as low specificity, low contrast, and short retention time. The combination of bio-conjugates chemistry and luminescence technology may achieve the specific targeting effect. In this research, we designed a dual-targeting composite nanoprobe (∼50 nm) using a metal-organic framework (MOF) as carrier, loaded with lanthanide and ICG , and combined with hyaluronic acid and folic acid to detect metastatic lymph nodes. The coupled hyaluronic acid and folic acid can target to the tumor cells and dentritic cells with a dual-targeting effect. The FA-HA/ [email protected] nanoprobes can accumulate rapidly in sentinel lymph node with a stronger luminescence intensity (1.6 times) than that of normal popliteal lymph nodes in vivo , thus distinguish metastatic sentinel lymph node from normal effectively. Furthermore, due to the MOF carrier, the integrated lanthanide and near-infrared dye by transferring the absorbed excitation energy from ICG to Nd 3+ can enhance the signal-to-background ratio of NIR II imaging and have long retention time in vivo imaging. Finally, the FA-HA/ [email protected] @ZIF-8 nanoplatform increased the penetration depth and contrast of imaging, prolonged the retention time, and achieved the sentinel lymph nodes surgical resection. This study has important implications for lymph node imaging and surgical navigation.

As a wound management strategy, this study developed a nanocomposite hydrogel based on the photothermal therapy and photodynamic therapy of nanomaterials, as well as the effects of Bletilla striata polysaccharide (BSP). In this research, MXene nanosheets loaded with indocyanine green were embedded into a hydrogel doped with BSP. By combining the photothermal effect and photodynamic effect, the treatment of wounds infected with Staphylococcus aureus was achieved under 808 nm near-infrared light irradiation (NIR). The characterization results of the hydrogel showed that it had good adhesiveness with an adhesive strength of 629 KPa ± 15.52 KPa and excellent biocompatibility; the swelling rate after 48 h was 485.53 % ± 0.26 %. In terms of antibacterial performance, the hydrogel had an antibacterial rate of over 90 % against Staphylococcus aureus . In addition, it could significantly promote the deposition of collagen and angiogenesis in infected wounds. When applied to wound treatment, the wound closure rate reached 98 % after 14 days, the collagen fraction of the healed wound was 87.63 % ± 1.81 %, and the epidermis thickness was 130.33 μm ± 1.25 μm. This indicated that the nanocomposite hydrogel dressing could effectively promote wound healing.

Thermo-adaptive injectable silk-hydrogel conforms dynamically to irregular wound beds. • Au-TiO 2 /ICG nano-assemblies for image-guided localization • NIR-triggered hyperthermia/ROS generation synergistic antibacterial treatment • IVIS-guided tracking of the hydrogel's delivery and retention • Multifunctional platform helps improve wound repair with excellent biocompatibility. Infection and chronicity of wounds pose an intimidating problem in clinical practice, necessitating a system that would be able to dynamically adjust to the changes in the wound bed, delivering antibacterial effects and monitoring them simultaneously. Herein, a thermo-adaptive injectable hydrogel using Au–TiO 2 –PF127–ICG (ATI) nanocomposites loaded in a silk hydrolysate-PEG 400 -gelatin (SPG) matrix has been developed, integrating mechanical adaptability, phototherapy, biocompatibility, and imaging functionality in a single construct. The hydrogel is stable at room temperature while undergoing gel-sol phase transition at physiological temperature, thus allowing simple injection and in situ shaping within irregular wounds. Rheological analysis confirmed a distinct gel–sol transition near physiological temperature, validating its thermo-adaptive nature. Upon NIR 808 irradiation, the incorporated indocyanine green (ICG) efficiently converted light into heat, while the Au–TiO 2 nanocomposites along the ICG produced reactive oxygen species (ROS), resulting in a dual photothermal/photodynamic antibacterial effect against E. coli and S. aureus . Infrared thermal imaging and IVIS fluorescence visualization further confirmed localized photothermal responsiveness and sustained hydrogel retention at the wound site, enabling fluorescence-assisted image-guided localization and monitoring. Furthermore, in vivo studies revealed accelerated wound closure, enhanced collagen deposition, and favorable histological regeneration under NIR stimulation, signifying the hydrogel's regenerative potential. Overall, this ATI-SPG hydrogel demonstrates a harmonious combination of injectability, self-adaptivity, fluorescence-enabled image-guided localization, NIR-triggered multimodal antibacterial therapy, and wound healing capability, providing a promising platform for next-generation light-responsive materials for wound management.

Rheumatoid arthritis (RA), a common inflammatory disorder of the joints characterized by synovitis and pannus formation, often results in irreversible joint erosion and disability. Methotrexate (MTX) is the first-line drug against RA, but the therapeutic effects are sub-optimal due to its poor retention at the target sites and systemic side effects. Multifunctional nanoparticles are highly promising agents for minimally invasive, traceable and effective targeted therapy. Methods: This study developed iRGD peptide-functionalized echogenic liposomes (iELPs) which encapsulates MTX and indocyanine green (ICG) fluorescent probe through the thin film-hydration method. Results: The resulting iELPs showed high affinity for endothelial cells overexpressing αvβ3 integrin, favorable acoustic response and fluorescence tracking properties. Also, near-infrared (NIR) fluorescence imaging of iELPs and ultrasound-triggered drug release of MTX were proved in a mouse RA model, greatly improving the therapeutic efficacy and reducing MTX side effects. Histological assessment of the articular tissues further revealed significantly lower inflammatory cell infiltration and angiogenesis in the iELPs-treated and sonicated mice. Conclusion: Our study provides a promising nanoplatform for integrating ultrasound-controlled drug release and NIR fluorescence imaging for RA treatment.

Doxorubicin (DOX) is considered one of the most effective chemotherapy drugs and is used as a first-line drug for many cancers. However, it has shown serious side effects such as cardiotoxicity and colonic ulcers , which limit its clinical use. Targeted delivery to tumor cells by nanocarriers or decreasing DOX dosage is considered to be an effective way to reduce the toxic and side effects of DOX on normal human tissues. In this paper, the mannose-functionalized N-2-HACC nanoparticles (MN NPs) loaded with DOX and Indocyanine Green (ICG) (DOX/ICG@MN NPs) were synthesized by the ion-crosslinking method, which were used for the in vitro targeted photothermal and chemical therapy. The results of FT-IR, particle size analysis, and SEM showed that ICG and DOX were successfully loaded, and DOX/ICG@MN NPs had great dispersibility and size. The results of the cell safety test showed that MN NPs had good safety. The photothermal stability tests revealed that the ICG in DOX/ICG@MN NPs had excellent thermal stability and photothermal conversion efficiency than the free ICG. The DOX release of DOX/ICG@MN NPs is pH-responsive, which promoted the effective release in slightly acidic environments at the tumor site. Owing to the specific targeting ability of mannose , DOX/ICG@MN NPs could be effectively uptake into HCT116 cells. In the presence of Near-Infrared irradiation (NIR), DOX/ICG@MN NPs exhibited synergistic treatment effects against tumor cells, which could significantly reduce the dosage of DOX and reduce the toxic side-effects on normal tissues. Therefore, this co-delivery system is considered to have significant potential for targeting tumors and chemo-photothermal therapy.

This work introduces the rational design of a hierarchically structured, multi-enzyme integrated therapeutic platform, referred to as HCuS/Ag/Pt/ICG/DOX (CAPID). The platform is constructed by depositing dual-enzyme-active Ag/Pt on copper sulfide nanoparticles (CuS NPs) to remodel the tumor microenvironment (TME), followed by loading indocyanine green (ICG) as a near-infrared (NIR) fluorophore and doxorubicin (DOX) as a chemotherapeutic payload. The CAPID platform utilizes catalase to decompose H 2 O 2 into oxygen (O 2 ), which synergizes with endogenous H 2 O 2 to initiate a Fenton-like reaction, generating cytotoxic hydroxyl radicals (·OH). The redox cycling between Cu 2 + ions and intracellular thiols depletes GSH, amplifying oxidative stress cascades. NIR irradiation achieved 88.9 % tumor inhibition via spatiotemporal tri-therapy (chemo/PDT/PTT) synergy, thereby addressing the limitations of monotherapy through nanoplatform engineering.

Targeting tumor-specific characteristics of acidic microenvironment, elevated H2O2 levels, and thermosensitivity, this study proposed and developed a copper-doped ZIF-8 nanoplatform coloaded with indocyanine green (ICG) and doxorubicin hydrochloride (DOX), designated as Cu-ZIF-8@ICG&DOX (CZID), to establish a multimodal therapeutic system integrating chemodynamic therapy, photothermal therapy, and chemotherapy. Results showed that the 25% copper doping level optimized the structure, achieving 7.46% actual doping content while limiting the average nanoparticle size to 99.6 nm. Dual loading of ICG and DOX induced morphological transition to spherical core–shell architectures, as confirmed by zeta potential reversal, with maximum drug loading capacities of 23.01 μg/mg for ICG and 122.43 μg/mg for DOX. The ZIF-8 framework exhibited pH-responsive degradation under acidic conditions. Released Cu2+ ions mediated continuous hydroxyl radical generation through glutathione-depletion redox cycling, confirming chemodynamic efficacy. Under 808 nm laser irradiation, ICG enabled enhanced photothermal conversion, showing concentration- and power-dependent temperature elevation. Cellular assays revealed efficient CZID internalization by MCF-7 cells, generating significantly higher intracellular reactive oxygen species levels under irradiation and more intense dead cell staining compared to CZI and CZD groups. Such an enhancement was attributed to photothermally accelerated ZIF-8 degradation, promoting Cu2+ ions and DOX release, thereby strengthening chemodynamic–chemotherapy synergy. At the working concentration of 50 μg/mL, CZID induced 95.36% apoptosis in MCF-7 cells while maintaining over 70% viability in MCF-10A normal cells, validating its precise antitumor potential.

To overcome the critical limitations of poor spatiotemporal coordination and inefficient energy delivery in photodynamic-thermodynamic (TDT-PDT) tumor therapy, we propose a free radical spatiotemporal-matching strategy to enhance therapeutic synergy. An optimized photoelectric therapy needle (PTN) and indocyanine green (ICG)-Cu(II)-AIBI infinite coordination polymer nanoparticles (ICP NPs) were rationally engineered to implement this strategy. The PTN enables precise codelivery of laser irradiation and electric current to the tumor core, achieving spatial overlap of photodynamic and electrothermal activation and free radical generation. The ICP NPs exhibit a high dual-drug loading (85.2%) and pH-responsive release, promoting tumor-selective accumulation and activation. This strategy ensures both spatial overlap and temporal alignment of singlet oxygen and alkyl radical production, maximizing oxidative damage. The ICP NPs display a high maximum tolerated dose (352.10 mg kg–1), markedly reducing systemic toxicity. In vitro, ICP NPs exhibit potent cytotoxicity against 4T1 cells (IC50 = 4.14 μg mL–1, combination index = 0.13), indicating strong synergism. In vivo, under low-energy stimulation and without chemotherapy, this strategy achieves complete tumor inhibition, a low recurrence rate (25% at 60 days), and negligible systemic toxicity. This work establishes a precise, low-toxicity paradigm for spatiotemporally matching dynamic therapy with strong translational potential.

Effective nano-platforms that integrate multiple therapeutic modalities are urgently needed to improve breast-cancer outcomes. Therefore, we engineered a breast cancer cell membrane (CM)-mimetic modified hollow cerium oxide nanoparticles (CM@H-CeO2/ID) for photothermal-chemo-immunotherapy of breast cancer. This platform achieves immune escape and active tumour targeting through homologous CM encapsulation; simultaneously, the hollow H-CeO2 carrier is utilised to co-carry ICG and DOX. Upon NIR irradiation, ICG produces hyperthermia that kills tumour cells and releases antigens; DOX then induces immunogenic cell death, and the Ce3+/Ce4+ redox cycling of H-CeO2 scavenges ROS to relieve immunosuppression. This tri-modal synergy markedly promotes dendritic-cell maturation and cytotoxic-T-cell infiltration, elicits systemic anti-tumour immunity and efficiently halts breast-tumour progression, offering a new paradigm for multimodal breast-cancer therapy.

In this work, multifunctional polystyrene@chitosan@gold nanoparticles–Fe3O4 nanoparticles–folic acid/indocyanine green (PS@CS@Au–Fe3O4–FA/ICG) nanocomposites that can integrate diagnosis and therapy into one nanoplatform was fabricated. The constructed PS@CS@Au–Fe3O4–FA/ICG nanocomposites possessed excellent computed tomography, magnetic resonance and fluorescence imaging performances which could be used to diagnose tumor and monitor the nanocomposites’ localization in vivo. Moreover, PS@CS@Au–Fe3O4–FA/ICG nanocomposites can be delivered in a targeted manner to cancer cells by folic acid in a receptor-mediated way, and tumor growth could be efficiently inhibited, even completely ablated, under NIR irradiation. That is, the constructed PS@CS@Au–Fe3O4–FA/ICG nanocomposites had great potential for computed tomography, magnetic resonance and fluorescence imaging performances, which could be used to diagnose tumors and to monitor local therapy and fluorescence imaging-guided targeted therapy of cancer cells using nanocomposites.

Effective tumor immunotherapy is hindered by an immunosuppressive tumor microenvironment (TME), especially in triple-negative breast cancer. Though phototherapy could induce immunogenic cell death (ICD) to increase antitumor immunity, the simultaneous upregulation of indoleamine 2,3-dioxygenase (IDO) induces the negative immunomodulatory effect termed as the “immune-metabolism” loop to compromise immunotherapeutic efficacy. Herein, we developed IMMGP consisting of biomimetic IND-Mn@PM (IDP) and ICG-MnO2@PM (IMP), which combines the phototherapy-induced ICD and metabolic reprogramming to solve the dilemma. During the light-on phase, IMP effectively kills cancer cells with potent photodynamic ROS generation with the assistance of MnO2-produced oxygen and induces ICD to reverse the immunosuppressive TME. In the light-off phase, Mn2+ (from IDP and MnO2-based redox reaction) elicits a Fenton-like reaction to relay ROS generation, which is further orchestrated with continuous exhaustion of intratumoral GSH by the conversion of Mn3+ to Mn2+, and promotes dendritic cell maturation. Moreover, the released indoximod (IND) downregulated IDO to inhibit kynurenine metabolism, which reinvigorates T cell-mediated antitumor immunity. Collectively, IMMGP amplifies the immune response by breaking the “immune-metabolism” loop and sustaining the “immunologically hot” state after phototherapy, thus leading to nearly complete tumor inhibition (94.25%). Thus, IMMGP-mediated dual-phase photodynamic immunotherapy offers a novel approach in cancer nanomedicine.

Cell-membrane-coated nanoparticles are widely studied due to their inherent cellular properties, such as immune escape and homologous homing. A cell membrane coating can also maintain the relative stability of nanoparticles during circulation in a complex blood environment through cell membrane encapsulation technology. In this study, we fused a murine-derived ID8 ovarian cancer cell membrane with a red blood cell (RBC) membrane to create a hybrid biomimetic coating (IRM), and hybrid IRM camouflaged indocyanine green (ICG)-loaded magnetic nanoparticles (Fe3O4-ICG@IRM) were fabricated for combination therapy of ovarian cancer. Fe3O4-ICG@IRM retained both ID8 and RBC cell membrane proteins and exhibited highly specific self-recognition of ID8 cells in vitro and in vivo as well as a prolonged circulation lifetime in blood. Interestingly, in the bilateral flank tumor model, the IRM-coated nanoparticles also activated specific immunity, which killed homologous ID8 tumor cells but had no effect on B16-F10 tumor cells. Furthermore, Fe3O4-ICG@IRM showed synergistic photothermal therapy, resulting in the release of whole-cell tumor antigens by photothermal-induced tumor necrosis, which further enhanced antitumor immunotherapy for primary tumor and metastatic tumor by activating CD8+ cytotoxic T cells and reducing regulatory Foxp3+ T cells. Together, the biomimetic Fe3O4-ICG@IRM nanoparticles showed synergistic photothermal-immunotherapy for ovarian cancer.
