New Formulation to Accelerate the Degradation of Pesticide Residues: Composite Nanoparticles of Imidacloprid and 24-Epibrassinolide

Jingyu Zhao,Rong Song,Hui Li,Qianqi Zheng,Shaomei Li,Lejun Liu,Xiaogang Li,Lianyang Bai,Kailin Liu

Pest control effectiveness and residues of pesticides are contradictory concerns in agriculture and environmental conservation. On the premise of not affecting the insecticidal effect, the pesticide residues in the later stage should be degraded as fast as possible. In the present study, composite nanoparticles in a double-layer structure, consisting of imidacloprid (IMI) in the outer layer and plant hormone 24-epibrassinolide (24-EBL) in the inner layer, were prepared by the W/O/W solvent evaporation method using Eudragit RL/RS and polyhydroxyalkanoate as wall materials. The release of IMI in the outer layer was faster and reached the maximum within 24 h, while the release of 24-EBL in the inner layer was slower and reached the maximum within 96 h. The contact angle of the composite nanoparticles was half that of the 5% IMI emulsifiable concentrate (EC), and the deposition of composite nanoparticles on rice was twice that of 5% IMI EC, which increased the pesticide utilization efficiency. Compared with the common pesticide, 5% IMI EC, the insecticidal effect of the composite nanoparticles was stronger than that of planthoppers, with a much lower final residue amount on rice after 21 days. The composite nanoparticles prepared in this study to achieve sustained release of pesticides and, meanwhile, accelerate the degradation of pesticide residues have a strong application potential in agriculture for controlling pests and promoting crop growth.

Highly sensitive and specific determination of imidacloprid pesticide by a novel Fe3O4@SiO2@MIPIL fluorescent sensor

Wei Xie,Yulong Ju,Jian Zhang,Yiwen Yang,Yanbo Zeng,Hailong Wang,Lei Li

A magnetic Fe 3 O 4 @SiO 2 @MIPIL fluorescent sensor for the determination of imidacloprid (IMD) with high sensitivity and specificity was prepared through surface molecular imprinting technology on the surface of Fe 3 O 4 @SiO 2 by using dialkenyl ionic liquid (C) as a fluorescent polymer monomer . The as-prepared sensor was characterized by thermogravimetric, FT-IR, SEM, TEM, and nitrogen adsorption-desorption analysis. Results showed that IMD rapidly quenched the fluorescence of the sensor within 1 min and had an excellent linear relationship to the fluorescent quenching intensity of the sensor within the range of 1–1000 nM (IMD). The LOD was 0.3 nM (S/N = 3). The sensor had a specific response to IMD and was used to detect IMD in actual spiked environmental water and food samples with satisfactory result.

Electrochemical sensor for imidacloprid detection based on graphene oxide/gold nano/β-cyclodextrin multiple amplification strategy

Jinmei Luo,Shuhuai Li,Yuwei Wu,Chaohai Pang,Xionghui Ma,Mingyue Wang,Chenghui Zhang,Xu Zhi,Bei Li

The key to improving the sensitivity of electrochemical sensors for direct detection is the introduction of catalytically amplified nanomaterials. Accordingly, in this study, a new strategy for the preparation of electrochemical sensors with multiple amplification effects is proposed. Graphene oxide (GO), gold nanoparticles (Au NPs), and β-cyclodextrin (β-CD) were introduced to the surface of a glassy carbon electrode via cyclic voltammetry to prepare a modified electrode sensor with a GO/Au NPs/β-CD thin film. The prepared sensor effectively catalyzed and amplified the reduction current of imidacloprid in a B-R buffer with pH = 5.0 owning to the multiple amplification effects of GO/Au NPs/β-CD. A good linear relationship was observed the level of the reduction current response and the imidacloprid at concentrations ranging from approximately 5 × 10 −10 to 3000 × 10 −10 mol/L; thus, a new imidacloprid detection method using an electrochemical sensor was established. The new method, whose detection limit reached 1.33 × 10 −10 mol/L, was tested on actual samples, achieving recoveries of 92.0 % to 110.0 %.

Evaluating the Impact of Individual and Combined Toxicity of Imidacloprid, Cycloxaprid, and Tebuconazole on Daphnia magna

Yanli Man,Tian Sun,Chi Wu,Xingang Liu,Mingyuan He

The risks posed by chemicals in the environment are typically assessed on a substance-by-substance basis, often neglecting the effects of mixtures. This may lead to an underestimation of the actual risk. In our study, we investigated the effects of three commonly used pesticides—imidacloprid (IMI), cycloxaprid (CYC), and tebuconazole (TBZ)—both individually and in combination, using various biomarkers to assess their impact on daphnia. Our findings indicated that the order of toxicity, from highest to lowest, was TBZ, IMI, and CYC, as determined by acute toxicity as well as reproduction. The effects of the ITmix (IMI and TBZ) and CTmix (CYC and TBZ) combinations on immobilization and reproduction were evaluated by MIXTOX, revealing a higher risk of immobilization at low concentrations for ITmix. The effect on reproduction differed depending on the ration of pesticides in the mixture, with synergism observed, which may be caused mainly by IMI. However, CTmix showed antagonism for acute toxicity, with the effect on reproduction depending upon the composition of the mixture. The response surface also exhibited a switch between antagonism and synergism. Additionally, the pesticides extended the body length and inhibited the development period. The activities of superoxide dismutase (SOD) and catalase (CAT) content was also significantly induced at different dosage points in both the single and combination groups, indicating changes in the metabolic capabilities of detoxifying enzymes and target site sensitivity. These findings highlight the need for more attention to be focused on the effects of pesticide mixtures. Keywords:ecotoxicity;pesticide mixture;combined toxicity;MIXTOX;risk assessment

The combined pollution of imidacloprid and glufosinate-ammonium aggravates toxicity on Hydrilla verticillata and epiphytic microbial community

Xiaodan Zhang,Songhe Zhang,Hongyu Guo,Wenjie Yuan,Xin Lv,Tilang Zhang,Shuaijie Jiang

The combined effects of imidacloprid (ICD) and glufosinate-ammonium (GLAM) on submerged plants and epiphytic microbial community remains unclear, despite the frequent co-occurrence of these pesticides in aquatic environments. In this study, a 24-day experiment was conducted to examine changes in water quality, plant physiology, and epiphytic microbial communities in Hydrilla verticillate -dominated wetlands under ICD alone or together with GLAM. Results showed that GLAM+ICD induced greater oxidative stress and a stronger antioxidant response in H. verticillata compared ICD alone, demonstrating that the mixtures exacerbated ecotoxicological effects on plant. Compared to ICD, many micro-biomarkers were obtained in ICD+GLAM, including Bacteroidota, Comamonadaceae, Proteobacteria and Cocconeis . Redundancy analysis indicated that the ICD concentration was correlated positively to Stigeoclonium, Closterium and Cocconeis , but negatively related with Oedogonium and Chloromonas , GLAM is opposite to ICD. More complex interkingdom interactions among bacteria and eukaryotes were observed in high concentration MIX treatment group compared to others. Co-occurrence networks analysis further demonstrated that GLAM and ICD changed microbial interactions within epiphytic biofilm, and impaired the stability and function of the microbial communities . These results underscore that the combined presence of ICD and GLAM poses a significant ecological risk to Hydrilla verticillate- biofilm system in agricultural regions.

Inner filter effect-based homogeneous immunoassay for rapid detection of imidacloprid residue in environmental and food samples

Fangfang Si,Rubing Zou,Shasha Jiao,Xusheng Qiao,Yirong Guo,Guonian Zhu

This paper reports a novel and sensitive homogeneous immunoassay for rapid detection of imidacloprid residue in environmental and food samples. The assay signal was originated from inner filter effect (IFE) between upconversion nanoparticles (UCNPs) and gold nanoparticles (AuNPs). UCNPs (energy donor) were coupled to the antibody against imidacloprid, and AuNPs (energy acceptor) were used to label the antigen of imidacloprid. The competitive immunoreaction occurred between imidacloprid and antigen-AuNPs binding to antibody-UCNPs. The assay system, sensitivity, selectivity , accuracy, and reliability were investigated. Under the optimal assay condition, the limit of detection for imidacloprid was 0.79   ng/mL and the concentration recovering 50% saturation of the signal (SC 50 ) was 18.7   ng/mL. The linear detection range (SC 10 to SC 90 ) was 1.39–335.81   ng/mL. Furthermore, the proposed method was successfully applied to determine imidacloprid in spiked water, Chinese cabbage, and honey samples, giving recoveries in the range of 78.1–97.9%. Through the blind sample tests, the IFE-based immunoassay demonstrated good correlation with ultra-performance liquid chromatography-tandem mass spectrometry (R 2 = 0.9866). Therefore, the new one-step immunoassay can be used as a promising tool for rapid detection of imidacloprid in field of environment and food safety.

Combined imidacloprid and chlorpyrifos disrupt gut microbiota-host interactions and reduced performance and survival in honey bees

Sihao Chen,Jiasen Wang,Sa Yang,Jincai Han,Shangfeng Zhou,Xiaoyu Shi,Hongmei Li-Byarlay,Dingfeng Luo,Changsheng Ma

Combined exposure to chlorpyrifos and imidacloprid exerted stronger toxic effects on bees than single insecticides. • Co-exposure significantly reduced sucrose consumption, body weight, and survival probability. • Gut detoxification capacity and intestinal integrity were severely impaired under combined exposure. • Multi-omics analyses revealed disruptions in energy metabolism, detoxification, and immune-related pathways. Gut microbiota plays a central role in insect physiology and health. However, how agrochemical exposure perturbs gut microbiota of the host and its downstream functional consequences remains understudied. Here, we employed an integrative multi-omics framework combining microbiome profiling, transcriptomics, and metabolomics to investigate how field-realistic exposures to the insecticides chlorpyrifos (CPF), imidacloprid (IMD), and their combination (IC) affect gut microbial composition as well as sucrose consumption, body weight dynamics, and survival in bees. We found that sucrose consumption was significantly reduced in bees exposed to IMD and IC, but not in the CPF group. Survival probability was significantly lower only in the IC group, whereas IMD and IC treatments caused greater weight loss compared to controls. Furthermore, IC exposure induced pronounced gut dysbiosis, and severe midgut structural damage, including epithelial degeneration and disruption of gut barrier integrity. At the functional level, IC exposure suppressed key detoxification enzymes, including glutathione S-transferase (GST) and cytochrome P450, indicating impaired xenobiotic metabolism. Multi-omics integration revealed 842 differentially expressed genes and 193 metabolites under IC exposure, with alterations in key pathways associated with energy metabolism, oxidative stress response, immune regulation, and host–microbiome interactions. These findings demonstrate that combined pesticide exposure disrupts gut microbiota composition and function, leading to cascading effects on host metabolic and detoxification processes as well as the overall health, and highlight the need for careful pesticide management to mitigate adverse impacts of combined pesticides across the environment. Imidacloprid-Induced Ferroptosis-Associated Injury in Common Carp Hepatocytes: Potential Involvement of the miR-153c/SQSTM1 Axis

Huijie Chen,Yitong Wang,Zhenkai Yao,Jing Li,Peng Li,Lei Diao

The neonicotinoid insecticide imidacloprid (IMD), widely used in agriculture, poses potential risks to aquatic ecosystems, yet its hepatotoxic mechanisms in freshwater fish remain poorly understood. This study investigates the roles of ferroptosis, mitochondrial dysfunction, inflammation, and the miR-153c/SQSTM1-mediated ferritinophagy pathway in IMD-induced hepatotoxicity in carp ( Cyprinus carpio ) hepatocytes. Using the CCK-8 assay, 0.6 μM IMD was selected for subsequent experiments. IMD exposure triggered ferroptosis, evidenced by increased intracellular Fe 2+ accumulation, upregulated pro-ferroptotic gene and protein expression, and downregulated ferroptosis inhibitors (SLC7A11 and GPX4). Mitochondrial dysfunction was confirmed by reduced ATP content, decreased mtDNA levels, and a lowered NADPH/NADP + ratio, along with aberrant expression of mitochondrial fission (Fis1 and Drp1) and fusion (Mfn1 and TFAM) genes. Additionally, IMD induced oxidative stress (elevated ROS and MDA; decreased T-AOC, CAT, and SOD activities) and a pronounced inflammatory response with upregulated pro-inflammatory cytokines. Mechanistically, IMD downregulated miR-153c, with concomitant upregulation of SQSTM1 at both the mRNA and protein levels, and dual-luciferase assays validated SQSTM1 as a direct target of miR-153c. miR-153c overexpression attenuated IMD-induced ferritinophagy, ferroptosis, and inflammation via the miR-153c/SQSTM1 axis. Collectively, these findings demonstrate that IMD induces hepatotoxicity in carp by triggering ferroptosis, mitochondrial dysfunction, and inflammation through the miR-153c/SQSTM1-mediated ferritinophagy pathway, providing mechanistic insights and potential targets for mitigating IMD’s ecological risks.

Modularly integrated synergistic SERS strategy for ultra-trace detection of imidacloprid in fresh fruit and vegetable juices

Xinru Yu,Hongbin Pu,Da-Wen Sun

A reliable IMIPCs–AMoO 2 /C SERS analytical method was established. • Validated results were acceptable for IMI in fresh fruit and vegetable juices. • The method displayed good linear correlation, low LOD, trueness and precision. • The mechanisms of a modularly integrated synergistic strategy were explored. • Rapid detection of trace levels of IMI in the sample was achieved (10 min). Imidacloprid (IMI), a first-generation neonicotinoid insecticide, is taken up by plants and distributed to edible parts owing to its systemic transport properties. This leads to its residues in fruit and vegetable juices, posing a potential food safety risk. Herein, we report a modular integrated synergistic SERS strategy for reliable, efficient and selective detection of IMI. An IMI-imprinted polymer-pesticide carb (IMIPCs) cartridge was designed for recognition, purification, and enrichment. The AMoO 2 /C SERS substrate, optimised through annealing, aligns its bandgap with the excitation wavelength, while IMI adsorption induces band bending and triggers charge-transfer resonance, enabling signal amplification. Under optimised conditions, the method exhibits excellent linearity from 0.500 ng/mL to 50.0 ng/mL, a low detection limit of 0.0904 μg/kg, and high accuracy in spiked typical juice matrices (tomato, celery, apple, and mixed juices), with recoveries of 92.7%–98.1% (RSD%, 0.332%–2.50%) and no significant difference compared with the HPLC method at the 95% confidence level. Application to twenty real samples collected from the market, including fruit and vegetable juice samples such as kiwi, kale, banana, and orange juices and their mixtures, demonstrated good selectivity, reliability, and reproducibility, with residue levels detected in the ppb range. Overall, this LEGO-inspired SERS method provides a simple, efficient and highly selective analytical strategy for monitoring IMI residues in fruit and vegetable juices, and can serve as a supplementary screening tool for routine food safety monitoring of such juice products. Its applicability to complex high-fat or protein-rich matrices, such as milk-based or nut-based beverages, remains to be further validated through subsequent research. Environmentally relevant imidacloprid exposure elicits hippocampal microglial apoptosis via JNK-FOXO3 pathway: Implication for latent central neurotoxicity

Xinyi Wen,Meiyan Li,Zhongyuan Zhang,Jingwen Wang,Hao Hu,Weihong Wang,Jing Jin,Kairong Wang,Jian Sun,Rui Zhang

Imidacloprid (IMI), a widely used neonicotinoid insecticide, has been frequently detected in drinking water and food; however, whether long-term low-dose exposure to environmentally relevant levels of IMI induces neurotoxic effects in the central nervous system remains unclear. In this study, C57BL/6   N mice were exposed to IMI in drinking water for three、six months at doses derived from human exposure estimates (0, 3   μg/kg, 103   μg/kg, and 6.55   mg/kg). Behavioral tests, including the Morris water maze and novel object recognition, revealed no overt cognitive impairment following IMI exposure. Nevertheless, IMI significantly increased microglial apoptosis in the hippocampus. Mechanistic analyses showed that IMI exposure enhanced JNK phosphorylation and promoted nuclear translocation of FOXO3, accompanied by increased expression of pro-apoptotic proteins BIM and BAX and decreased BCL-2 levels. Consistently, IMI induced apoptosis in BV2 microglial cells through activation of the JNK–FOXO3 signaling pathway, whereas FOXO3 knockdown or JNK inhibition with SP600125 markedly attenuated this effect. These findings indicate that environmentally relevant IMI exposure can trigger hippocampal microglial apoptosis via JNK–FOXO3 signaling even in the absence of overt behavioral deficits, suggesting a potential early neurotoxic effect of neonicotinoid exposure.

Metatranscriptomic insights into nitrogen loss mediated by multi-kingdom microbial interactions in periphytic biofilms under imidacloprid exposure

Zuqiang Cao,Juan Chen,Chao Wang,Peifang Wang,Han Gao,Bingbing Feng,Bo Zhang,Rong Wang,Ge Cui

Periphytic biofilms are ubiquitous and drive biogeochemical nutrient cycling in paddy ecosystems. However, the mechanisms of multi-kingdom microbial interactions regulating nitrogen (N) cycling in the biofilms remain unexplored. This study conducted a microcosm experiment to investigate the effects of imidacloprid (IMI), a widely used neonicotinoid pesticide, on N cycling in periphytic biofilms. Throughout the 30-day experiment, 0.02–0.12 mg of N was lost from the water-biofilm microcosm systems, increasing with higher IMI exposure concentrations. Metatranscriptomic sequencing demonstrated 84.3 %–92.2 % downregulation of nitrification genes and 33.9 %–143 % upregulation of denitrification genes in the biofilms. These alterations in gene expression were regulated by microbial competition and cooperation within ecological clusters associated with the biofilm potential nitrification and denitrification activities, respectively. Correlation between the relative abundance of microorganisms in these clusters and gene abundance revealed multi-kingdom nitrifying and denitrifying communities, including Nitrosopumilus (Archaea) and Nitrospira (Bacteria) for nitrification, and Enterococcus (Bacteria) and Aspergillus (Fungi) for denitrification. Partial least squares path modeling indicated that the biofilm-mediated N loss was driven by promoted denitrification (total effect = −0.83) rather than suppressed nitrification (total effect = 0.52). Our findings highlight multi-kingdom microbial interactions regulating N cycling in periphytic biofilms and neonicotinoid pesticide risks.

Ti4O7-C3N4 anode with a low charge transfer resistance value for degradation of imidacloprid in actual wastewater

Shuting Guo,Qiongfang Zhuo,Gang Huang,Liying Lan,Tongyin Zhu,Wenlong Wang,Zehong Yang,Jiasheng Fang,Yongfu Qiu,Bo Yang

Imidacloprid (IMD) is a kind of neonicotinoid insecticide, which is widely used in the agricultural production. There are few reports on electrocatalytic degradation of IMD with the low concentration. In this study Ti 4 O 7 -C 3 N 4 electrode was employed for the first time to electrocatalytically degrade low concentration IMD. Compared with pure Ti 4 O 7 electrode, the oxygen evolution potential was increased from 1.84 V to 2.50 V, the charge transfer resistance (R ct ) was reduced by 66.05 %, and the oxygen vacancies were increased by 2.5 times compared to the original Ti 4 O 7 . Especially, the R ct for Ti 4 O 7 (5.45 Ω·cm 2 ) was only 1/7 of that for boron-doped diamond (BDD) (38.89 Ω·cm 2 ). IMD degradation ratio was up to 98 % in only 1.256 min (75.360 s) using a Ti 4 O 7 -C 3 N 4 . When SongShan Lake water, Wanjiang water, and secondary effluent were treated, the degradation radios of IMD were 70.30 %, 72.33 % and 81.12 %, respectively. In the presence of Cl − coexisting ions, no disinfection by-product of ClO 4 − was detected, which is an advantage for Ti 4 O 7 -C 3 N 4 compared with BDD. The first and 48th degradation ratios were 99 % and 97 %, respectively, indicating the good stability of Ti 4 O 7 -C 3 N 4 . The development of Ti 4 O 7 -C 3 N 4 provides a stable, low energy consumptions and no secondary pollution anode for the treatment of actual wastewater.
