This study assessed the protective effects of konjac glucomannan (KGM) on gut microbiome against the antibiotic perturbation in C57BL/6J mice. The native KGM (1.82 × 10 7 ) was partially hydrolyzed by endo-1,4-β-mannanase, and two hydrolyzed fractions (KGM-eM with 3.82 × 10 5 Da and KGM-eL with 8.27 × 10 3 Da) were characterized and applied to mice with perturbation of antibiotics in comparison with the native KGM. The results showed that the native KGM better maintained the microbial diversity and composition in feces, and increased the production of the individual and total SCFAs in feces and serum with perturbation of antibiotics. In contrast, KGM with lower MW (KGM-eM and KGM-eL) increased the proportion of Lactobacillus and SCFA production with no antibiotics, however, the prebiotic effects were eliminated with perturbation of antibiotics. These results have demonstrated the protective effects of KGM with high MW on gut microbiome against the antibiotic perturbation in vivo.

Background Constipation is a gastrointestinal symptom with high incidence rate and large number of patients. It is becoming one of the urgent medical problems. Poor intestinal motility is one of the important causes of constipation. Current drug treatments for constipation are associated with many side effects; thus, it is necessary to study more effective treatment methods and potential mechanism. Methods A zebrafish model of intestinal motility obstruction was established by loperamide hydrochloride to evaluate the effect of probiotic, food ingredients, and combination on intestinal peristalsis according to intestinal peristalsis frequency counts. The gastrointestinal survival ability of the best probiotics was evaluated by surface hydrophobicity, self-aggregation, acid and bile salt tolerance, and gastrointestinal transit tolerance. Interactions between probiotics and food ingredients were studied in vivo and in vitro. The expression of 5-HT was detected by ELISA and fluorescence immunoassay, and 5-HT related genes were detected by RT-PCR. Key Results We obtained the probiotics, food ingredients, and combination that effectively promoted intestinal peristalsis, X11 and YRL577, P . persica and KGM, KGM + X11, respectively. Both KGM and P . persica promoted colonization of probiotics in vivo. KGM + X11 could effectively promote the increase in 5-HT synthesis in zebrafish via up-regulating gene expression of TPH-1, TPH-2, and 5-HT R and down-regulating gene expression of SERT. The specific in-depth mechanism needs further study. Conclusions and Inferences The combinations of KGM with X11 effectively promoted intestinal peristalsis. We provide a theoretical basis for new modalities that can promote intestinal peristalsis and alleviate constipation.

Gluten (Glu) is important to wheat products by forming a three-dimensional matrix. This study aimed to investigate the physicochemical and structural properties of gluten after conjugation with konjac glucomannan (KGM) through the Maillard reaction. The study revealed that the degree of graft increased with the prolonged reaction time. The Glu-KGM conjugates were possessed of increased β-sheet but decreased α-helix and β-turn, as well as unfolding and loose tertiary structures as the reaction proceeded. Among three different proportions, the Glu-KGM 1:1 conjugate was proved to have the most excellent foaming and emulsifying properties, and could form more rigid and firm gelation structures, which could be related to the decreased particle size and increased zeta potential of the conjugate. Overall, the physicochemical and structural properties of gluten were significantly related to the KGM ratios as well as the reaction period. Keywords:gluten;konjac glucomannan;Maillard reaction;physicochemical properties;structural properties

BACKGROUND:Polysaccharide hydrogel is one of the most important materials for the colon target drug release system. However, the degradation time of polysaccharide hydrogel is much longer than the retention time in the colon. The drugs are expelled from the body before being released. OBJECTIVE:In order to match the degradation of drug carriers and their retention time in the colon, a rapidly degradable konjac glucomannan (KGM) hydrogel was designed for colon target drug release. METHODS:A crosslinker containing azo bond, olsalazine, was used to prepare the rapidly degradable KGM hydrogel. The degradation and drug release of the hydrogels with different crosslinking densities in the normal buffer and the human fecal medium were studied to evaluate the efficiency of colon drug release. RESULTS:More than 50% of the KGM hydrogel by weight was degraded and more than 60% of the 5-fluorouracil (5-Fu) was released within 48 h in 5% w/v human fecal medium. CONCLUSION:The drug was released more rapidly in a simulated colon environment than in a normal buffer. Furthermore, the drug release was controlled by the degradation of the hydrogel. The KGM hydrogel containing azo crosslinker has great potential for colon drug release.

The rapid development of portable electronic devices requires high-performance electromagnetic interference (EMI) shielding materials that exhibit excellent mechanical strength and flexibility. Inspired by the hierarchical structure and mechanical robustness of nacre, in this work, we choose konjac glucomannan (KGM) as the mortar and MXene as the brick to prepare an ultrathin, flexible and environmentally friendly MXene/KGM composite film via a facile vacuum-assisted filtration method. MXene nanosheets are introduced into KGM matrix to assemble a nacre-like structure through strong hydrogen bonding interactions between the rich surface-terminating groups of MXene and the hydroxyl groups of KGM. As expected, the as-prepared MXene/KGM composite film with 40 wt% MXene content exhibits optimal mechanical properties, including a tensile strength of 145.8 MPa and an elongation at break of 8.1 %. Moreover, this composite film also achieves an EMI shielding effectiveness of 24 dB, demonstrating a balance between mechanical properties and EMI shielding effectiveness. The resulting environmentally friendly MXene/KGM composite film demonstrates significant potential for application in next-generation flexible electronic devices.

The development of green and environmentally friendly flame retardant materials has attracted widespread attention in modern architecture. In this work, a kind of novel aerogel was prepared by the incorporation of Al(OH) 3 coated hollow glass microspheres (HGM) into konjac glucomannan (KGM) matrix and then cross-linking by CaCl 2 (abbreviated as KGM-HGM-Al(OH) 3 ). The prepared KGM-HGM-Al(OH) 3 aerogel exhibits a porous structure with a high porosity of 93.93%, endowing it with excellent thermal insulation performance. In addition, it exhibits excellent thermal stability, and the mass loss of KGM-HGM-Al(OH) 3 aerogels is 37.6% at 800°C. More importantly, the aerogel shows excellent flame retardancy, with the peak heat release rate (pHRR) and total heat release (THR) of 21.43 kW m −2 and 1.895 kJ m −2 , respectively. Furthermore, the limiting oxygen index (LOI) is 54.5%; this superior performance is primarily attributed to the synergistic effect of HGM-Al(OH) 3 . The hollow structure of the hollow glass microspheres effectively delays the internal heat transfer, Al(OH) 3 decomposes endothermically at high temperatures to form an Al 2 O 3 barrier layer, while releasing water vapor to dilute combustible gas, so that the KGM-HGM-Al(OH) 3 aerogel has excellent flame retardancy. The aerogel material has the advantages of low cost and a simple preparation method, and has extensive application value in the fields of building fire resistance and battery fire resistance.

The objective of this study was to enhance the water barrier properties and antifungal efficacy of KGM film by incorporating (attapulgites-Ag nanoparticles, APTs-AgNPs) composites. The X-ray photoelectron spectroscopy (XPS), Transmission electron microscopy (TEM), and Energy dispersive X-ray (EDS) analyses indicated that the successful fabrication of APTs-AgNPs composites. Water vapour permeability (WVP), moisture content (MC), water solubility (WS) and water absorption (WA) values decreased by 19.15%, 12.50%, 74.96% and 41.74% respectively, while the water contact angle (WCA) increased by 134.74%. These results demonstrated that the water barrier property of KGM/APTs-AgNPs films was superior to that of pure KGM films. The Rheological properties, fourier transform infrared (FTIR) spectrum, X-ray diffraction (XRD) patterns and Scanning electron microscopy (SEM) images results indicated that hydrogen bonds formed between homogeneous APTs-AgNPs (1.5 wt%) and KGM, which enhanced the water barrier properties of the KGM/APTs-AgNPs films. The KGM/APTs-AgNPs composite films possessed excellent oxygen permeability, Mechanical properties, UV barrier properties, opacity properties and thermostability. Moreover, KGM/APTs-AgNPs films slowly released Ag + and exhibited long-acting antifungal efficacy against Aspergillus niger , as the inhibition circle diameter was 20.70 ± 0.32 mm for 10 days. Therefore, KGM/APTs-AgNPs films could be a potential alternative to petroleum-based films.

A novel drug-polysaccharide conjugate with konjac glucomannan (KGM) as a drug carrier was fabricated through the esterification of ibuprofen (IBU), an anti-inflammatory drug, with KGM. The influences of the reaction conditions, such as the amount of ibuprofen acryl chloride, reaction time, reaction temperature, and the amount of catalyst, on the degree of substitution were investigated. KGM ibuprofen ester (KGM-IBU) was characterized by Fourier transform infrared spectrometry (FTIR), X-ray diffraction (XRD), solid-state 13C NMR, scanning electron microscopy (SEM), transmission electron microscopy (TEM), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC), and dynamic mechanical analysis (DMA). The hydrophobic structure of IBU in KGM-IBU was proven by the fluorescence emission spectra of pyrene. In addition, by using commercially available ibuprofen sustained-release capsules (IBU-SRC) as a control, the in vitro controlled release performance of KGM-IBU was evaluated. The cumulative release of IBU-SRC within 36 h was 94%, while that of KGM-IBU within 36 h was 77%. The results showed that KGM-IBU had better sustained-release performance without a burst release effect. The obtained products could be used as a potential biocompatible sustained-release drug delivery system.

All-polysaccharide GT/KGM emulsion gels serve as multifunctional dysphagia carriers. • Increasing GT strengthened the network, immobilized water, and improved lubrication. • IDDSI classification shifted from Level 4 to Level 6 with increasing GT concentration. • GT/KGM gels significantly protected β-carotene against time, heat, light, and pH stress. Texture-modified foods for dysphagia should provide bolus safety, oral comfort, structural stability, and nutritional value, but many current systems rely mainly on thickening or pureeing and have limited capacity to protect lipophilic nutrients. In this study, gum tragacanth/konjac glucomannan (GT/KGM) emulsion gels were developed for dysphagia. The gels were prepared with a fixed KGM concentration of 0.2% (w/v) and GT concentrations of 0.1%, 0.3%, 0.5%, 0.7%, and 0.9% (w/v), corresponding to KG1-KG5. Then, the resulting gels were evaluated for stability, structure, water distribution, rheological and tribological behavior, IDDSI classification, and β-carotene retention. Experimental results showed that increasing GT concentration improved droplet dispersion and shifted part of the relatively mobile water toward network-immobilized water. All samples exhibited solid-like viscoelastic behavior and shear-thinning flow, indicating deformable but structured gel networks. Tribological analysis showed Stribeck-like lubrication, with GT-rich formulations exhibiting lower friction at intermediate and high sliding speeds. TPA combined with IDDSI testing revealed a GT-driven transition from Level 4 (KG1) to Level 5 (KG2-KG3) and Level 6 (KG4-KG5). Both KG4 and KG5 met IDDSI Level 6. KG4 had lower hardness, gumminess, chewiness, and springiness than KG5. β-Carotene-loaded gels maintained high retention under heating, light exposure, and different pH conditions. These results show that all-polysaccharide emulsion gels can control texture for swallowing and protect lipophilic nutrients. They may be used to develop foods for people with dysphagia in the future. Download: Download high-res image (137KB) Download: Download full-size image GT/KGM emulsion gels for dysphagia-oriented texture control, oral lubrication, and β-carotene protection. GT and KGM were used to construct all-polysaccharide emulsion gels with corn oil as the lipid phase. Increasing GT concentration reinforced the gel network, improved water retention and reduced friction during simulated oral processing, supporting IDDSI-based swallowing suitability. The structured lipid phase further served as a protective carrier for β-carotene under environmental stresses. These results suggest that GT/KGM emulsion gels can integrate swallowing safety, oral comfort and lipophilic nutrient protection for dysphagia-oriented food design.

Fabricated SA/KGM/DMY edible films via hydrogen-bond-mediated supramolecular assembly. • Optimized 8% DMY loading for balanced mechanical, barrier and bioactive properties. • Achieved simultaneous browning inhibition and microbial suppression in fresh-cut apples. Food packaging is crucial for freshness preservation, with edible films gaining widespread attention. This study prepared sodium alginate (SA)/konjac glucomannan (KGM) composite films containing dihydromyricetin (DMY), with 8% DMY as optimal. Fourier transform infrared (FT-IR) spectroscopy, molecular docking, and isothermal titration calorimetry (ITC) confirmed hydrogen bonding, stabilizing DMY. The film exhibited potent antioxidant activity (88.9% DPPH scavenging), antimicrobial efficacy against both Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus (92.05% and 95.20% inhibition, respectively; log₁₀ reduction >2.50 at 12% DMY loading). Fresh-cut apple trials showed it delayed quality loss, inhibited browning (lower PPO activity), and suppressed microbial proliferation (3.97 log CFU/g vs. 5.28 log CFU/g control). The key innovations are: (i) elucidating DMY stabilization in a polysaccharide matrix via hydrogen bonding through docking and ITC; (ii) integrating antioxidant, antimicrobial, and barrier functions in a single edible film; and (iii) demonstrating dual efficacy against enzymatic browning and microbial spoilage in fresh-cut produce. Water-in-water (W/W) emulsions as innovative transport for hydrophilic functional ingredients inherently lack stability due to the ultralow interfacial tension and thick interfacial layer. In this work, we first proposed a simple strategy to construct stable dextran (DEX)-in-poly (ethylene glycol) (PEG) W/W emulsions by utilizing the unique phase preference of konjac glucomannan/tragacanth gum (KT) hydrogels formed based on the cooling-induced intermolecular hydrogen bonds. KT hydrogels were demonstrated to present selective partition to PEG phase and could form physically crosslinking network, thus significantly improving the rheological behavior and stability of DEX-in-PEG (D/P) emulsions with no flocculation after 31 d of storage at 25 °C. Meanwhile, the average droplet size of emulsions decreased from 15.37 to 8.48 μm with TG concentration tuning from 0.2 % to 0.8 %. When encapsulating riboflavin (RIB) in D/P emulsions stabilized by KT-4 hydrogel, the storage and lighting stability of RIB were enhanced with retention rate of 60.94 % and 21.89 %, respectively. In addition, the release of RIB in the simulate gastrointestinal environment substantially reduced and the antioxidant activity of RIB was maximumly protected due to the unique pH-responsiveness of KT hydrogel network. This work provides a feasible strategy for designing stable W/W emulsions stabilized by natural polysaccharide hydrogels, and biocompatible carriers for hydrophilic functional ingredients.

Antibacterial hydrogels have emerged as a promising approach for effective wound treatment. However, despite extensive research on the fabrication of antibacterial hydrogels, it remains challenging to develop injectable, biocompatible, transparent, and mass-producible hydrogels with antibacterial properties. In this study, we successfully fabricated an antibacterial drug-loaded composite hydrogel, named CC45/OKG40/HS, through a Schiff base reaction between carboxymethyl chitosan (CC) and oxidized konjac glucomannan (OKG), followed by the encapsulation of stevioside-stabilized honokiol (HS) micelles. The CC45/OKG40/HS hydrogel exhibited several favorable properties, including a short gel time (<10 min), high water content (>92%), injectability, good adhesiveness, self-healing ability, and high transparency. In vitro experiments confirmed its excellent antibacterial properties, antioxidant activities, and high biocompatibility (no cytotoxicity, hemolysis ratio <5%). Furthermore, in vivo evaluation demonstrated that the CC45/OKG40/HS0.5 hydrogel accelerated wound healing by relieving inflammatory responses and enhancing re-epithelization. Given its feasibility for mass production, the findings showed that the CC45/OKG40/HS hydrogel has the potential as an advanced antibacterial wound dressing for commercial use.
