Ratiometric Fluorescent Mapping of pH and Glutathione Dictates Intracellular Transport Pathways of Micellar Nanoparticles

Zexuan Ding,Guhuan Liu,Jinming Hu

Visualization of intracellular transport pathways is crucial to investigate the internalization mechanism and understand the intracellular behavior of nanomaterials. Herein, we rationalized the design of micellar nanoparticles (NPs) for ratiometric fluorescent mapping of intracellular pH and glutathione (GSH), two essential parameters for maintaining normal cellular functions. Specifically, pH-sensitive naphthalimide-based probe (NPI) and pH-inert rhodamine B (RhB) were covalently labeled to double hydrophilic block copolymers (DHBCs) using the thiolactone chemistry, enabling the covalent attachment of NPI and RhB to one molecule with a redox-responsive disulfide linkage. The dually labeled DHBCs exhibited blue/orange dual emissions in acidic pH, which was further converted into green/orange dual emissions in neutral pH because of the deprotonation of NPI moieties and the sole green emission in the presence of GSH at neutral pH because of the decreased Förster resonance energy transfer efficiency between an NPI donor and an RhB acceptor as a result of GSH-mediated cleavage of disulfide bonds. These remarkable ratiometric fluorescence changes allowed for not only the simultaneous mapping of the intracellular pH and GSH but also the intracellular transport pathways of internalized NPs.

Three-Dimensional Mapping of Retrograde Multi-Labeled Motor Neuron Columns in the Spinal Cord

Jianyi Xu,Xiaofeng Yin,Yisong Qi,Bo Chen,Yusha Li,Peng Wan,Yingtao Yao,Dan Zhu,Baoguo Jiang,Tingting Yu

The quantification and distribution characteristics of spinal motor neurons play important roles in the study of spinal cord and peripheral nerve injury and repair. In most research, the sole retrograde labeling of each nerve or muscle could not simultaneously obtain the distributions of different motor neuron subpopulations. Therefore, it did not allow mapping of spatial relationships of different motor neuron columns for disclosing the functional relationship of different nerve branches. Here, we combined the multiple retrograde labeling, optical clearing, and imaging for three-dimensional (3D) visualization of motor neurons of multiple brachial plexus branches. After screening fluorescent tracers by the labeling feasibility of motor neurons and fluorescence compatibility with optical clearing, we performed mapping and quantification of the motor neurons of ulnar, median, and radial nerves in the spinal cord, then disclosed the relative spatial distribution among different neuronal subpopulations. This work will provide valuable mapping data for the understanding of the functional relationships among brachial plexus branches, hopefully facilitating the study of regeneration of axons and remodeling of motor neurons in peripheral nerve repair.

Mapping Potential Engineering Sites of Mycobacterium smegmatis porin A (MspA) to Form a Nanoreactor

Jinyue Zhang,Jiao Cao,Wendong Jia,Shanyu Zhang,Shuanghong Yan,Yuqin Wang,Panke Zhang,Hong-Yuan Chen,Wenfei Li,Shuo Huang

Protein nanopores can be engineered as nanoreactors to investigate single-molecule chemical reactions. Recent studies have demonstrated that Mycobacterium smegmatis porin A (MspA) nanopore is a superior engineering template acknowledging its geometrical advantages. However, reported engineering of MspA to form a nanoreactor has focused only on site 91 and mapping of other engineering sites have never been performed before. By taking tetrachloraurate(III) ([AuCl4]−) as a model reactant, potential engineering sites within the pore constriction of MspA have been thoroughly investigated. It is discovered that the produced event amplitude is inversely correlated to the cross-sectional diameter of the pore constriction size at the engineering site, providing evidence that site 91 is actually already the optimum place to introduce the chemical reactivity. Other unavailable engineering sites, which either significantly interfere with the pore assembly or produce reactive sites facing to the pore’s exterior instead of to the pore lumen, were also spotted and discussed. All results demonstrated above have provided a complete map of engineering sites within the constriction area of MspA and may be beneficial as a reference in future engineering of corresponding nanoreactors.

Mechanistic Mapping of Additive Cracking for Hybrid SEI Construction in Aqueous Zinc-Ion Batteries

Boyuan Liu,Heng Zhou,Shiqiang Wei,Shuangming Chen,Hong Wang,Hengjie Liu,Yixiu Wang,Jialin Shi,Zijun Zhang,Qian Zhou,Zhandong Wang,Li Song

Electrolyte additives represent a promising strategy for regulating the solid-electrolyte interphase (SEI) in aqueous zinc-ion batteries. However, the underlying molecular mechanisms by which these additives mediate SEI formation remain insufficiently understood. Here, we employ 1-acetyl-2-pyrrolidone (APD) as a model organic additive to mechanistically map molecular cracking pathways involved in constructing a robust SEI on zinc anodes. Using in situ synchrotron radiation vacuum ultraviolet photolysis mass spectrometry (SR-VUPM), for the first time, we reveal the dynamic cleavage process of the additive to form a hybrid SEI. The in situ synchrotron radiation X-ray diffraction and Fourier transform infrared spectroscopy further indicate that the resulting hybrid SEI serves as a hydrophobic barrier and regulates the uniform deposition of zinc. The functional SEI plays a crucial role in optimizing interfacial reaction kinetics and deposition/stripping reversibility. As a result, the Zn/Zn symmetric cells deliver over 1500 h at 5.0 mA cm −2 and 5.0 mA h cm −2 . The cycle life of the Zn/I 2 @AC full battery is extended to 6000 cycles, while the optimized pouch cell also displays excellent cyclic stability. These findings provide mechanistic insight into additive-derived SEI formation for developing durable metal anodes.

Sensitivity-tunable flexible ion–electron synergistic pressure sensor for high-resolution occlusal force mapping

Yueyang Chen,Xin Yi,Zhihao Wang,Chen Shao,Rongdong Zeng,Shaohui Zhang

A bilayer ion–electron design harmonizes high sensitivity with rapid dynamic response. • Tunable sensitivity is achieved by tailoring MXene doping from 0.02 to 0.10 mg/mL. • 2 wt% CNT loading optimizes percolation, avoiding rigid filler agglomeration. • Robust durability is validated under saliva wetting, shear, and bionic occlusion. High-resolution, wide-range occlusal force monitoring is crucial for diagnosing temporomandibular disorders and optimizing dental prosthetics. However, current clinical tools lack sufficient flexibility, spatial resolution, and dynamic response range. This work develops a flexible, sensitivity-tunable pressure sensor tailored for quantitative full-arch occlusal force mapping. An ion–electron synergistic piezoresistive sensor was fabricated in a bilayer heterogeneous structure, integrating an ionically conductive, MXene-doped poly(AMPS-co-AA) hydrogel sensory layer with an electronically conductive carbon nanotube/thermoplastic polyurethane (CNT/TPU) transduction film. Interfacial adhesion was optimized using O₂ plasma activation, PEDOT modification, and hot-press crosslinking. This synergistic design harmonizes high sensitivity with rapid dynamic response (58 μs for the electronic layer and 3 ms for the ionic layer). The fabricated device demonstrated a wide dynamic response range up to 30 N and outstanding mechanical durability over 1000 cycles. Systematic screening revealed that a 2 wt% CNT loading forms an optimal percolation network that minimizes interfacial charge-transfer resistance, whereas a 3 wt% loading induces microscale contact loss and signal fluctuations. Evaluations using a 3D-printed bionic oral model across full-arch dental positions showed good agreement with the FEA-predicted stress gradients. This sensing platform offers a practical, high-fidelity approach for digitized occlusal force mapping and oral biomechanical assessment. A Dual-Readout Near-Infrared Fluorescent Fiber Probe for High Spatiotemporal Resolution Neurotransmitter Mapping

Shin Seyoung,Kim Yeji,Yoo Jeongeun,Cho Jeonghyeon,Song Youngho,Lee Seungju,Park Seongjun,Cho Soo-Yeon

Neuromodulators such as dopamine (DA) operate through volume transmission in the brain, creating spatially distributed fields that shape circuit excitability. However, conventional sensing modalities, including voltammetry and fiber photometry, inherently collapse these multidimensional dynamics into one-dimensional, single-point signals, thereby obscuring spatial heterogeneity and propagation behavior. Here, we introduce the Neural Remote Imaging Fiber Interface (NeuRIFI), a dual-readout near-infrared (NIR) sensing platform that bridges localized neurochemical detection and field-level spatial interpretation. By functionalizing implantable optical fibers with DNA-functionalized SWCNT nanosensors, this platform leverages NIR fluorescence emission within the tissue-transparent optical window to enable synchronous fiber-guided photometry and remote NIR imaging without genetic labeling. In this configuration, the photometric channel provides a spatially anchored validation reference, while the remote imaging channel preserves the spatial information on neurochemical signals. We validated the interface across tissue phantoms and ex vivo mouse brains, demonstrating reliable deep-brain signal acquisition at a 3 mm depth. Furthermore, by computing frame-to-frame intensity derivatives, we resolved transient DA flux at a temporal resolution of 20 ms, a spatial resolution of ∼1.5 μm per pixel, and a concentration sensitivity of 90 nM, enabling high spatiotemporal resolution mapping of neurochemical transport across the sensing surface. Collectively, this dual-readout architecture provides a framework linking point-validated sensing to remotely observable spatial structures, offering spatial accessibility for studying complex neurochemical landscapes in deep-brain structures and extending neurochemical field mapping toward in vivo settings.

Underway mapping of coastal seawater pH using an automated shipboard analyzer with spectrophotometric detection

Hangqian Li,Shu Wang,Shulu Zheng,Tengyue Fang,Huilin Shu,Yi Xu,Xianghui Guo,Eric P. Achterberg,Liyang Zhan,Jian Ma

The development of field-deployable methods and instruments for the measurement of pH and other carbonate parameters is important for the assessment of the marine carbon cycle , ocean acidification and marine carbon dioxide removal techniques. In this study, a high-precision fully automated integrated syringe-pump-based environmental-water analyzer for pH ( i SEA-pH) was developed. The pH is determined spectrophotometrically using purified indicator dye with a high precision (better than ±0.001) and high frequency (3.5 min/sample). For the short-term analysis, the measurement frequency was 18 h −1 , which revealed pH = 7.8148 ± 0.0005 (n = 104) for aged surface seawater (S = 35) from the western Pacific. For long-term analysis, the measurement frequency was 2 h −1 for 4 days, and the results showed that pH = 7.8148 ± 0.0010 (n = 200). Three commonly used pH indicators (meta-cresol purple, thymol blue and phenol red) were purified with improved flash chromatography procedures. The autonomous i SEA-pH can automatically correct for the influence of temperature, salinity and other factors on pH measurements to achieve rapid and accurate on-site measurements, which meet the “climate” goal of the Global Ocean Acidification Observing Network (uncertainty is ±0.003). Three identical i SEA-pH systems were developed and successfully applied in mesocosm experiments and several coastal and open ocean cruises with excellent in field performance.

Mutual Inductance-Based Array Sensor for Continuous Monitoring and Mapping of Localized Brain Deformations

Ziwei Liu,Jianzheng Li,Yapeng Fu,Sijia Yu,Wenjun Li,Yiqing Yang,Jiajia Wang,Sihui Yu,Peiwen Yu,Kailiang Xu,Yajie Qin,Songlin Zhang,Huisheng Peng,Xuemei Sun

The brain undergoes constant dynamic deformations, which are intricately linked to its intrinsic properties and pathological states. However, continuous monitoring of these deformations presents significant challenges due to the limited temporal and spatial resolutions of conventional imaging techniques. Here, it is developed an implantable deformation array sensor capable of real-time, continuous monitoring and mapping of brain deformations. The sensor features microcoil arrays embedded in ultrathin films and operates based on the principle of mutual inductance, achieving exceptional temporal resolution (<100 µs) and distance resolution (<5 µm). It is implanted the sensor between the skull and the cortex and successfully tracked continuous brain deformations across four cortical sites in rat models. These experiments are conducted under scenarios of isoflurane inhalation, mechanical impacts, and intracranial hemorrhage, where such detailed monitoring and mapping has not been achieved previously. The results reveal that brain deformations dynamically vary over time in response to both physiological fluctuations and pathological events, with deformation amplitudes decreasing as the distance from the primary site increases. This innovative approach provides a novel platform for exploring brain dynamics and their associations with neurological disorders and disease progression.

A Cyclodextrin-Hosted Ir(III) Complex for Ratiometric Mapping of Tumor Hypoxia In Vivo

Peng Xiao,Chunyan Liu,Tiancong Ma,Xiuhong Lu,Lihong Jing,Yi Hou,Peisen Zhang,Gang Huang,Mingyuan Gao

Hypoxia is considered as a key microenvironmental feature of solid tumors. Luminescent transition metal complexes particularly those based on iridium and ruthenium have shown remarkable potentials for constructing sensitive oxygen-sensing probes due to their unique oxygen quenching pathway. However, the low aqueous solubility of these complexes largely retards their sensing applications in biological media. Moreover, it remains difficult so far to use the existing complexes typically possessing only one luminescent domain to quantitatively detect the intratumoral hypoxia degree. Herein, an Ir(III) complex showing red emissions is designed and synthesized, and innovatively encapsulated within a hydrophobic pocket of Cyanine7-modified cyclodextrin. The Ir(III) complex enables the oxygen detection, while the cyclodextrin is used not only for improving the water solubility and suppressing the luminescence quenching effect of the surrounding aqueous media, but also for carrying Cyanine7 to establish a ratiometric oxygen fluorescence probe. 2D nuclear magnetic resonance is carried out to confirm the host–guest structure. The oxygen-responsive ability of the resulting ratiometric probe is evaluated through in vitro cell and multicellular experiments. Further animal studies about tumor oxygen level mapping demonstrate that the probe can be successfully used for quantitatively visualizing tumor hypoxia in vivo.

High-Resolution Mapping of Aerosol Optical Depth and Ground Aerosol Coefficients for Mainland China

Lianfa Li

Aerosols play an important role in climate change, and ground aerosols (e.g., fine particulate matter, abbreviated as PM2.5) are associated with a variety of health problems. Due to clouds and high reflectance conditions, satellite-derived aerosol optical depth (AOD) products usually have large percentages of missing values (e.g., on average greater than 60% for mainland China), which limits their applicability. In this study, we generated grid maps of high-resolution, daily complete AOD and ground aerosol coefficients for the large study area of mainland China from 2015 to 2018. Based on the AOD retrieved using the recent Multi-Angle Implementation of Atmospheric Correction advanced algorithm, we added a geographic zoning factor to account for variability in meteorology, and developed an adaptive method based on the improved full residual deep network (with attention layers) to impute extensively missing AOD in the whole study area consistently and reliably. Furthermore, we generated high-resolution grid maps of complete AOD and ground aerosol coefficients. Overall, compared with the original residual model, in the independent test of 20% samples, our daily models achieved an average test R2of 0.90 (an improvement of approximately 5%) with a range of 0.75–0.97 (average test root mean square error: 0.075). This high test performance shows the validity of AOD imputation. In the evaluation using the ground AOD data from six Aerosol Robotic Network monitoring stations, our method obtained an R2of 0.78, which further illustrated the reliability of the dataset. In addition, ground aerosol coefficients were generated to provide an improved correlation with PM2.5. With the complete AOD data and ground coefficients, we presented and interpreted their spatiotemporal variations in mainland China. This study has important implications for using satellite-derived AOD to estimate aerosol air pollutants.

Mapping the relationship between atopic dermatitis and gut microbiota: a bibliometric analysis, 2014–2023

Yilin Wang,Bingkun Wang,Shiyou Sun,Zhongzhi Wang

Background: Atopic dermatitis (AD) is a chronic inflammatory skin condition affecting a significant portion of the population, with prevalence rates of 25% in children and 7–10% in adults. AD not only poses physical challenges but also profoundly impacts patients’ mental well-being and quality of life. The stability of gut microbiota is crucial for overall health and can influence AD progression by modulating immune function, skin barrier integrity, and neuroendocrine signaling, which may be an effective target for the prevention and treatment of AD. Thus, exploring the interactions between AD and gut microbiota, particularly in infants, can provide insights into potential preventive and therapeutic strategies. This study aimed to explore the correlation between AD and gut microbiota while providing an overview of current research trends and emerging areas of interest in this field. Methods: A comprehensive search was conducted on the Web of Science Core Collection (WOSCC) for relevant publications from January 1, 2014, to December 31, 2023. English-language articles and reviews were included. Two investigators independently screened the publications, and visual analysis was performed using CiteSpace, VOSviewer, Scimago Graphica, and Microsoft Excel software. Results: A total of 804 articles were included, showing a significant increase in publications over the past decade. The United States, Wageningen University, and University Ulsan (represented by Hong SJ) had the highest number of published papers. Nutrients was the journal with the most publications, while the Journal of Allergy and Clinical Immunology had the highest number of citations and centrality among co-cited journals.

SynClear: A One-Step Synchronous Clearing and Labeling Strategy for Multiscale 3D Brain Mapping

Xiao Xiao,Canyu Ma,Yashuo Wang,Jiazhu Zhu,Jiajia Chen,Simei Li,Yongjie Wang,Wei Gong,Ke Si

High-resolution three-dimensional imaging is essential for resolving the multiscale organization of biological tissues. However, conventional workflows treat tissue clearing and molecular labeling as separate steps, leading to a kinetic mismatch between reagent transport and probe binding that limits imaging depth, labeling uniformity, and throughput. Here, we introduce SynClear, a one-step strategy that synchronizes nuclear labeling with tissue clearing by embedding fluorescent probes within a chemically engineered clearing medium. This integrated formulation enables rapid and uniform labeling across millimeter-scale samples while preserving endogenous fluorescence and remaining compatible with multiplexed immunostaining. We demonstrate the general applicability of SynClear across diverse tissue types, including mouse brain, peripheral organs, and post-mortem human cortex. In mouse brain sections, SynClear supports accurate 3D atlas registration and quantitative mapping of cytoarchitecture. In glioblastoma models, it resolves pathological features across scales, from tumor boundaries to immune microenvironments. In human cortex, it enables laminar-resolved structural analysis and neuronal subtype mapping. By coupling labeling and clearing within a single chemical framework, SynClear provides a robust and scalable platform for volumetric tissue imaging, with potential applications in both basic neuroscience and translational pathology.
