Unraveling the inhibitory mechanism of composite allelochemicals on Microcystis aeruginosa: Integrated transcriptomic and metabolomic insights

Jie Li,Liu Shao,Anglu Shen,Peimin He

Harmful cyanobacterial blooms (HCBs), driven by eutrophication, pose critical threats to aquatic ecosystems and public health. Current studies focused on inhibitory effect of single-component allelochemical on HCBs, yet cyanobacteria in natural environments are simultaneously exposed to multiple allelochemicals. Thus, it is more meaningful to study composite allelochemicals formulation for HCBs control. Our previous study found that the composite allelochemicals agent of 4-tert-butylcatechol and l -lysine (CAA-TL) can synergistically inhibit the growth of Microcystis aeruginosa . In this study, we investigated the inhibitory mechanism of CAA-TL against M. aeruginosa , integrating physiological, transcriptomic, and metabolomic approaches. CAA-TL reduced the cell density of M. aeruginosa by more than 90 % within four days, and significantly decreased the Chl- a content and chlorophyll fluorescence parameters. The integrated transcriptomic and metabolomic results showed that: (1) 471 upregulated and 312 downregulated differential expressed genes (DEGs) and 433 upregulated and 215 downregulated differential metabolites (DMs) were identified; (2) CAA-TL markedly impacted the expression of photosynthesis related genes (e.g., PsbA , PsbL , PsaB and PsaK ) indicating the inhibition of photosynthetic activity; (3) CAA-TL impacted DNA and protein synthesis through downregulation of nucleotide metabolism; (4) CAA-TL disrupted energy supply via the dysregulation of energy metabolism pathways. These findings reveal the inhibitory mechanism of CAA-TL via an integrated omics approach and provide theoretical support for their eco-friendly application in HCBs control.

Effects of Prometryn Exposure Scenarios on Microcystis aeruginosa Growth and Water Qualities in Incubator Experiments

Zhenjiang Yang,Suiliang Huang,Xiaowen Huang,Xiaofu Liang,Waseem Akram,Zobia Khatoon

Although multiple herbicide exposures are more prospective to occur in water, many previous studies were carried out as single herbicide exposure. To investigate the toxic effect of prometryn on cyanobacteria and water qualities, single and double prometryn exposures (at different growth phases) onMicrocystis aeruginosagrowth and concentrations of nutrients were compared after a 44-day experiment. Results indicated that under single exposure, maximum inhibition rates were 4.7–12.0% higher than those under double exposures. Correspondingly, the maximumMicrocystis aeruginosadensities and growth rates under single exposure were 10.3–21.1% and 19.5–37.7% lower than those under double exposures (p< 0.05), respectively. These findings revealed that repeated prometryn exposures resulted in a reduction in biological effects, because the time of application and the concentration injected during the first application were both significant factors in the biological effects of prometryn. Prometryn exposure scenarios did not have a significant effect on nutrient or nutrient consumption concentrations (p> 0.05). In general, the pattern of nutrient limitation showed a shift from phosphorus to nitrogen limitation. The quantified relationships betweenMicrocystis aeruginosagrowth rates and consumed nutrients were studied. Based on the above findings, we believe that a high-dose and single prometryn exposure is a more effective exposure pattern for limiting cyanobacteria growth.

Effects of PET and Gallic Acid on Growth, Photosynthesis, and Oxidative Stress of Microcystis aeruginosa

Wang Tiantian,Liu Haicheng

Utilizing the allelopathic effect of plants to inhibit algae growth has gained significant attention from researchers in recent years. However, there are large amounts of microplastics in the aquatic environment, which may influence the inhibitory effects of allelochemicals on algae. Consequently, this research aimed to examine the individual and combined toxicity of gallic acid (GA) and polyethylene glycol terephthalate (PET) microplastics on Microcystis aeruginosa. At a concentration of 50 mg/L, PET significantly enhanced algal cell growth and chlorophyll a synthesis, while inducing oxidative stress without damaging the cell membrane. On the other hand, low concentrations of GA promote algal cell growth. However, high concentrations of GA not only inhibit growth and photosynthesis but also result in cellular abnormalities and severe oxidative damage, as well as membrane disruption. Moreover, it was observed that the exposure of PET diminished the toxicity of GA to M. aeruginosa . The combined exposure to GA and PET resulted in a slight increase in chlorophyll a content, as well as phycocyanin (PC), alloxyphycocyanin (APC), and phycoerythrin (PE) content, along with maximum photochemical efficiency (Fv/Fm), compared to GA exposure alone. In the combined system of GA and PET, the contents of superoxide dismutase (SOD) and malondialdehyde (MDA) were lower than those in the single GA system. Additionally, both systems showed a degree of improvement compared to the control group. Scanning electron microscopy (SEM) observed that the presence of PET resulted in reduced damage to algal cells compared to exposure to GA alone. This research establishes a foundation for understanding the synergistic impacts of allelochemicals and microplastics on M. aeruginosa , thereby providing a reference for managing cyanobacteria in water blooms.

Effects of fish feed addition scenarios with prometryn on Microcystis aeruginosa growth and water qualities

Zhenjiang Yang,Suiliang Huang,Wenwen Kong,Liang Chen,Priakhina Ekaterina,Zobia Khatoon,Muhammad Nabil Ashraf,Waseem Akram

The number of undesirable environmental impacts of fish feed has been reported widely. Although repeated fish feed exposures are more prospective to occur in water, previous studies were mostly conducted as a single exposure of fish feed. In order to fill these gaps, a 40 days incubator experiment was conducted to explore the effects of fish feed addition scenarios during the lag phase with prometryn on both Microcystis aeruginosa growth and concentrations of nutrients. The maximum algae densities in groups of single exposure were 6.0–26.2% and 8.8–74.4% higher than those in groups of double and triple exposures, respectively ( P  < 0.05). At the beginning of the experiment, concentrations of nutrients in groups with different feed exposure scenarios were significantly different. The pattern of nutrient limitation showed a transformation from phosphorus limitation to nitrogen limitation generally. Furthermore, the average inhibition rates of algae by prometryn in the case of a single fish feed exposure were 4.6–9.4% lower than those under double exposures, and 22.0–26.8% lower than those under triple exposures ( P  < 0.05). In addition, algae growth rates have been developed as a function of concentrations of consumed nutrients ( R 2 = 0.410–0.932). Based on the above results, we concluded that in terms of limiting algae growth multiple low-dosage additions of fish feed were considered as a better addition pattern. By optimizing feed addition scenarios, there is considerable potential to increase the environmental sustainability of aquaculture.

Simultaneous photoinactivation of Microcystis aeruginosa and photocatalytic degradation of microcystin-LR by erythrosine-based photosensitizer

Nuzahat Habibul,Xiayida Mamatimin,Wei Song,Mihebai Yilimulati

The simultaneous control of toxic cyanobacterial blooms and degradation of microcystins (MCs) remains a critical challenge for environmental risk reduction. Herein, we demonstrate the novel application of erythrosine (ERY), an FDA-certified food additive, as a potent photoactive agent for concurrent control of cyanobacterial blooms and removal of microcystin-LR (MC-LR) contaminants. ERY treatment achieved effective growth inhibition of Microcystis aeruginosa . Mechanistic studies revealed that ERY-generated including reactive oxygen species (ROS) severely damaged cellular structures and photosynthesis systems. Notably, ERY-treated samples showed a significantly greater reduction in extracellular MC-LR content compared to conventional H 2 O 2 treatment, confirming its superior microcystin degradation efficiency. The degradation products of MC-LR were elucidated through LC-MS/MS analysis, and degradation pathways and reactive sites were verified by theoretical calculations. Furthermore, ecotoxicological evaluation using Lemna minor and rapid photodegradation of ERY (95.6% within 4 days) confirmed its minimal phytotoxicity and the risk from the parent MC‑LR is eliminated, but the potential hazard of transient intermediates requires further study. These findings position ERY as a promising and environmentally benign photosensitizer for harmful algal bloom mitigation.

Palmatine chloride inhibits Microcystis aeruginosa via photosynthetic impairment, oxidative stress, and altered cellular energy status

Xuemei Chen,Xueying Fan,Ping Zhang,Caihong Qin,Kai Li,Bingbing Qin,Junlei Wang,Hongqing Li,Yan Xiao

Harmful cyanobacterial blooms threaten aquatic ecosystems and public health through cyanotoxin release and oxygen depletion. This study evaluated the inhibitory activity of palmatine chloride (PC) against Microcystis aeruginosa under different light and temperature conditions and investigated the associated physiological, biochemical, ultrastructural, and transcriptional responses. At 25°C, PC maintained concentration-dependent inhibition across 50–125 μmol·m −2 ·s −1 , achieving 48–52%, 92–96%, and 90–98% inhibition at 4, 8, and 12 mg/L, respectively, within seven days. At 35°C, 12 mg/L PC produced near-complete inhibition within 48 h. Specifically, PC markedly impaired photosynthetic performance, increased reactive oxygen species and malondialdehyde levels, altered antioxidant responses, and induced membrane disruption, thylakoid disorganization, and cytoplasmic leakage. N -acetylcysteine co-treatment did not restore growth or photosynthetic parameters, indicating that PC inhibition was not solely attributable to an NAC-reversible ROS-dependent process. Transcriptomic profiling and ATP measurements further indicated substantial disruption of cellular energy homeostasis, characterized by downregulation of genes involved in photosynthesis, respiration, and central carbon metabolism, together with an 84% decrease in intracellular ATP. Preliminary toxicity tests suggested relatively low acute toxicity to Daphnia magna and Vibrio fischeri under the tested conditions. Besides, PC had limited effects on Spirulina platensis but modestly inhibited Chlorella vulgaris , while no sustained accumulation of extracellular microcystin equivalents was detected. Collectively, PC strongly inhibited Microcystis aeruginosa , accompanied by photosynthetic impairment, oxidative stress, structural damage, and disruption of cellular energy homeostasis. These findings support further evaluation of PC as a candidate botanical algicide.

Differential effects of environmentally relevant norfloxacin on Microcystis aeruginosa and Chlorella vulgaris: Growth and metabolism

Mihebai Yilimulati,Dongyuan Zhang,Jieyu Wang,Zaimire Abudushalamu,Wentao Zhang,Nuzahat Habibul

The ecological effects of antibiotics at environmentally relevant low concentrations have attracted considerable attention, but how they regulate algal growth and influence bloom risks through selective, dose-dependent, and algae-specific mechanisms remains poorly understood. Moreover, most conventional toxicological studies have used high concentrations at the mg/L level, which are orders of magnitude higher than actual environmental concentrations (ranging from ng/L to μg/L). To address this gap, the present study systematically compared the selective effects and metabolic mechanisms of norfloxacin on the cyanobacterium Microcystis aeruginosa and the green alga Chlorella vulgaris under environmentally relevant concentrations. Low NOR concentrations (0.01–0.5 μg/L) stimulated the growth of Microcystis aeruginosa , increased its photosynthetic pigment content, enhanced photosystem activity, and activated metabolic pathways related to nucleotide and amino acid synthesis. Interestingly, higher NOR concentrations (5–100 μg/L) suppressed photosynthesis, induced oxidative stress, promoted the release of extracellular organic matter, and markedly disrupted central energy metabolism, including carbon fixation, glycolysis, the pentose phosphate pathway, and the tricarboxylic acid cycle. In contrast, Chlorella vulgaris exhibited greater tolerance, as evidenced by maintained photosynthetic homeostasis, moderate metabolic reorganization, and no significant fluctuation in cell abundance. These findings indicate that environmentally relevant concentrations of norfloxacin may selectively stimulate the growth of the cyanobacterium Microcystis aeruginosa through a “low-dose promotion, high-dose inhibition” pattern mediated by key metabolic pathways. The differential algae-specific responses suggest that norfloxacin pollution could potentially increase the risk of cyanobacterial blooms, although this possibility remains to be validated through co-culture or field studies.

Visible-light-drivenself-floating Ag2MoO4/TACN@LF photocatalyst inactivation of Microcystis aeruginosa: Performance and mechanisms

Gongduan Fan,Chenjian Cai,Zhuoyi Chen,Jing Luo,Banghao Du,Shangwu Yang,Jiaxin Wu

The poor light transmittance , low recycling rate and potential secondary pollution problems of powder photocatalysts limit the practical application of photocatalytic technology. Therefore, there is an urgent need to develop stable floating photocatalysts to alleviate the problem of harmful algal blooms (HABs). In this work, a floating Ag 2 MoO 4 /TACN@LF composite was synthesized for the inactivation of Microcystis aeruginosa ( M. aeruginosa ) by employing a simple oscillatory impregnation method. The Ag 2 MoO 4 /TACN@LF floating photocatalyst exhibited stable algae removal performance under various conditions, and 100% chlorophyll a removal rate could be achieved within 4 h of visible light irradiation under the optimal photocatalyst loading of 6 g/L. The effects of Ag 2 MoO 4 /TACN@LF floating catalyst on algal cell membrane permeability, phycobiliprotein, intracellular antioxidant enzyme activities and algal organic matter were further studied. Moreover, the scavenging experiments demonstrated that •OH was the predominant reactive oxygen species (ROSs). Ag0 and 3 g-C 3 N 4 * also made a non-negligible contribution to the inactivation of algal cells. What's more, the Ag 2 MoO 4 /TACN@LF still exhibited 92% inactivation efficiency for M. aeruginosa after 5 cycles reused experiments, indicating its promising potential for practical application of alleviating HABs.

Activated carbon fiber loaded nano zero-valent iron for Microcystis aeruginosa removal: Performance and mechanisms

Guoming Zeng,Dong Liang,Xuanhao Fan,Yu He,Rui Zhang,Xiaoling Lei,Haoxuan Wei,Da Sun

Cyanobacterial blooms caused by Microcystis aeruginosa threaten environmental safety and daily life. In this study, an activated carbon fiber-supported nano zero-valent iron composite (ACF-nZVI) was developed to remove Microcystis aeruginosa . The results showed that nZVI was evenly distributed on the activated carbon fibers, preventing aggregation and oxidation. ACF-nZVI achieved a removal efficiency of more than 90 % within a pH range of 3–7. During the reaction, H 2 O 2 , which was generated by Fe 0 , was activated to form · OH and · O – 2 , which dismantled antioxidant enzymes and induced lipid peroxidation. Additionally, ACF-nZVI destroyed the cell wall and membrane, resulting in protein and humus leakage and causing 92.34 % cell damage and death. In this study, an environmentally friendly and stable nanomaterial was developed, offering a novel approach for the safe, cost-effective, and efficient removal of cyanobacteria.

Exudates of Microcystis aeruginosa on oxidative stress and inflammatory responses in gills of Sinocyclocheilus grahami

Jun Li,Xuexiu Chang,Sen Zhao,Yuanwei Zhang,Qi Pu,Yuting Wang,Jiaojiao Li

Early cyanobacterial blooms studies observed that exposure to blue-green algae led to fish gills impairment. The objective of this work was to evaluate the toxic mechanisms of exudates of Microcystis aeruginosa (MaE) on fish gills. In this study, the toxic mechanism of MaE (2×10 6 cells/mL) and one of its main components phytosphingosine (PHS) with two concentrations 2.9 ng/mL and 145 ng/mL were conducted by integrating histopathology, biochemical biomarkers, and transcriptomics techniques in Sinocyclocheilus grahami ( S. grahami ) for 96  h exposure. Damaged gill tissue with epithelial hyperplasia and hypertrophy, remarkable Na + /K + -ATPase (NKA) enzyme activity, disrupted the redox homeostats including lipid peroxidation and inflammatory responses were observed in the fish of MaE exposure group. Compare to MaE exposure, two concentrations of PHS exposure appeared to be a trend of lower degree of tissue damage, NKA activity and oxidative stress, but induced obviously lipid metabolism disorder with higher triglycerides, total cholesterol and total bile acid, which might be responsible for inflammation responses in fish gill. By transcriptome analysis, MaE exposure were primarily enriched in pathways related to gill function and immune response. PHS exposure, with higher number of differentially expressed genes (DEGs), were enriched in Toll-like receptor (TLR), Mitogen-Activated Protein Kinase (MAPK) and NOD-like receptor protein 3 (NLRP3) pathways. We concluded that MaE and PHS were induced the inflammatory responses, with oxidative stress-induced inflammation for MaE exposure but lipid metabolism disorder-induced inflammation for PHS exposure. The present study provided two toxin-induced gill inflammation response pathways under cyanobacterial blooms, which could be a scientific basis for the ecological and health risk assessment in the aquatic environment.

Inhibitory effect of malonic acid on Microcystis aeruginosa: Cell morphology, oxidative stress and gene expression

Chunyang Tao,Xiaojun Niu,Dongqing Zhang,Yuejin Liu,Lingling Zhou,Yunhao Han,Yujing Mo,Kaixin Wu,Yu Lin,Zhang Lin

The utilization of allelochemicals , which are regarded as eco-friendly algaecides for mitigating harmful cyanobacterial blooms (HCB), has attracted increasing attention, owing to their negligible toxicity, high selectivity and excellent degradability. In the present study, the inhibitory effects of malonic acid on Microcystis aeruginosa were investigated in terms of growth indicators, cell morphology, antioxidant enzymes activities and gene expression. The results indicated that the exposure to 0.5 mmol L −1 malonic acid for 7 d caused a high microalgae inhibition rate of 86.88 %. Furthermore, the analysis of scanning electron microscopy (SEM) and transmission electron microscope (TEM) showed that the presence of malonic acid resulted in the disruption of cell integrity and the changes in cell morphology. Specifically, malonic acid was observed to cleave microalgae cells, while concurrently oxidizing the microalgae organic matter. In addition, under the exposure of 0.5 mmol L −1 malonic acid, the levels of malondialdehyde (MDA) were 3.28 times as the control group on d-5, while the activities of superoxide dismutase (SOD) and catalase (CAT) were first increased by 5.96 and 11.73 fold compare to the control group on d-5, and then decreased, due to oxidative stress induced by malonic acid. Transcriptome-based gene expression analysis revealed that the presence of malonic acid significantly inhibited the transcription of genes ( rbcL , psbA1 ) associated with photosynthesis , and the expression of DNA repair gene ( recA ), stress response gene ( ftsH ) and microcystin synthesis genes ( mcyD , mcyH ) were up-regulated. These findings provide novel insights into the inhibitory effect of malonic acid on M. aeruginosa , suggesting that malonic acid has the potential to control M. aeruginosa blooms.

Efficient photocatalytic inactivation of Microcystis aeruginosa via self-floating Ag3VO4/BiVO4 hydrogel under visible light

Gongduan Fan,Xia Li,Jiuhong Lin,Xinyi Wu,Ling Zhang,Jiaxin Wu,Yingmu Wang

In this work, a facile separation of self-floating Ag 3 VO 4 /BiVO 4 hydrogel (ABVH) photocatalyst was constructed via the freeze-thaw method for the inactivation of Microcystis aeruginosa . The as-synthesized photocatalyst exhibited excellent photocatalytic performance, resulting in the degradation efficiency of chlorophyll a reaching approximately 99.6% within 4 h under visible light. The cell morphology, intracellular organic matter (IOM), extracellular organic matter (EOM), and antioxidant system were severely disrupted during the photocatalytic process. The inactivation mechanism could be ascribed to the ·OH and h + generated by hydrogel photocatalyst under visible light irradiation. Furthermore, five repetitive experiments and cyclic compressive stress-strain tests demonstrated that the as-prepared hydrogel possessed excellent reusability and mechanical strength properties. Besides, the hydrogel photocatalyst showed excellent potential for the removal of M. aeruginosa in the presence of natural organic matter (NOM). Overall, the synthesized self-floating hydrogel photocatalyst may offer a novel insight into the control of harmful cyanobacterial blooms.
