Immune checkpoint inhibitors (ICIs) are effective agents for tumor immunotherapy. However, their clinical effectiveness is unsatisfactory due to off-target effects and a suppressive immune microenvironment. This study developed a nanodrug delivery system for bladder cancer (BCa) using PCL-MPEG and PCL-PEG-CHO to synthesize internal hydrophobic and external hydrophilic micelles (PP) that encapsulated water-insoluble astragaloside IV (PPA). The aldehyde group on the surface of PPA reacted with the amino group of aPD-L1, allowing the decoration of this antibody on the surface of the micelles. The resultingPPA@aPD-L1effectively piggybacked astragaloside IV and aPD-L1 antibody. These findings suggest that PPA@aPD-L1 is relatively stable in circulation and efficiently binds to BCa cells with the aid of aPD-L1. Additionally, this strategy prolongs the drug’s retention time in tumors. Compared to PBS, PP, and PPA with PPA + aPD-L1 groups, PPA@aPD-L1significantly prolonged the survival of mice with BCa and reduced tumor volume. Mechanistic studies showed that PPA inhibited the NF-κB and STAT3 signaling pathways in tumor cells. Additionally, PPA@aPD-L1increased IFN-γ and decreased IL-10 expression in bladder tumors, affecting the number and type of intratumorally infiltrating T cells. Our study presents a simple and effective drug delivery system that combines herbal monomers with ICIs. It has demonstrated a potent ability to suppress tumor growth and holds potential for future applications.

Current smart cling films for pork preservation often face two key challenges: either they lack real-time freshness monitoring capabilities, or their antibacterial components rely on visible light—limiting penetration depth and easily inducing lipid oxidation in pork. To address these gaps, this work fabricated an electrospun bilayer smart cling film with integrated preservation and monitoring functions. The film comprises a hydrophilic outer layer (polycaprolactone [PCL]/polyvinyl alcohol [PVA]/chitosan [CS]/anthocyanin [ACN], for pH-responsive color indication) and a hydrophobic inner layer (PCL as the matrix, loaded with aluminum phthalocyanine [AlPc], for near-infrared-driven antibacterial activity). We systematically investigated the effect of AlPc loading (5 %–15 %) on film performance. Results showed that the film with 15 % AlPc exhibited the highest level of comprehensive properties among the tested groups: its antibacterial and antioxidant capacities were significantly enhanced, alongside improved physical characteristics—hydrophobicity of the inner antibacterial layer increased by 17 % (in terms of water contact angle), and water vapor transmission (WVT) was reduced by 19 %. Importantly, the film displayed distinct color changes (light red to dark red) in pH 3–11 buffer solutions and during pork storage, enabling real-time freshness monitoring. Moreover, it extended the freshness period of pork to 5 days. These findings demonstrate that the prepared bilayer film effectively addresses the limitations of existing cling films, offering significant potential for simultaneous pork shelf-life extension and freshness monitoring.

The preparation of polyesteramides (PEAs) with regular block structure has long been a challenge, owing to the occurrence of the ester-amide exchange during the polymerization at high tempereture. In this work, we report a simple and green method to prepare polyamide 6 (PA6) based triblock PEA whose structure is poly(ε-caprolactone)-block-PA6-block-poly(ε-caprolactone) (PCL- b -PA6- b -PCL) using tetraphenyl tin as catalyst without solvent. The first segment PA6 was synthesized via cationic ring-opening polymerization (ROP) of ε-caprolactam (CPL), later, ROP of ε-caprolactone (CL) was involved to extend polycaprolactone (PCL) segments by using tetraphenyl tin as catalyst. By varying the proportion of CPL and CL, series PA6-based PEAs was prepared and characterized by gel permeation chromatography, 1 H, DOSY, and 13 C NMR. The results showed that PEA containing more proportion of CL presented a higher molecular weight with narrow polydisperse index. In particular, the lower water absorption and lower oligomers content infers that the PEA could have a prospective application in processing precision electronic devices.

Polyesteramides have attracted much attention due to their intriguing properties. However, the synthesis of di- or triblock polyesteramides based on polyamide 6 (PA6) remains challenging owing to their harsh reaction conditions. In this work, we introduced a new strategy to synthesize poly(ε-caprolactone)-block-PA6-block-poly(ε-caprolactone) (PCL-b-PA6-b-PCL) in a mild way. A series of well-designed copolymers were characterized by gel permeation chromatography, 1H, DOSY, and 13C NMR, and their crystallization and melting behavior, degradation rate, and compatibilization effect on PA6/PCL blends were found to be strongly composition dependent. Interestingly, the copolymers exhibited diverse spherulite morphologies, including curving stems and banded spherulites, which also correlated with the different relative content of PA6. Besides, PA6 nanodomains could act as physical cross-linking points to improve the tensile strength and Young’s modulus of PCL. This work fills the synthesis gap of well-defined PA6-based triblock polyesteramides, providing new materials for material modification and elucidation of some basic scientific questions.

A novel biodegradable 1,2,3-triazolium-functionalized PEG- b -PCL copolymer (TAPEC) was synthesized by the “click” coupling of methoxypolyethylene glycol azide and α-propargyl-ω-hydroxyl-poly(ε-caprolactone), followed by the quaternization of the 1,2,3-triazole moiety with iodomethane. All the intermediates and TAPEC were characterized by 1 H NMR, FT-IR, and gel permeation chromatography (GPC). Taking advantage of the characteristics of ionic liquid and block copolymer, this ion-containing diblock copolymer is expected to be used as a novel compatibilizer in mixed biopolyester for regulating the interface and crystallization behaviors. Hence, the TAPEC was evaluated as a compatibilizer and an interface emulsifier in the blends of polylactic acid (PLA) and poly(ε-caprolactone) (PCL). Non-isothermal crystallization experimental results showed that the TAPEC with the higher amount of ε-caprolactone units induces a plasticization and nucleate effect that increased the crystallization ability of the PLA phase; meanwhile, in the PCL phase, the agminated ionic cluster acting as a nucleating agent significantly increased the crystalline of PCL.

This study investigates the design of polyrotaxane-based solid polymer electrolytes (SPEs) by grafting polycaprolactone (PCL) onto hydroxypropylated polyrotaxane (HyPR) and examining the synergistic effects of cross-linking and polymer grafting on key material properties. Incorporation of long-chain PEG crosslinker resulted in SPEs with high ionic conductivity, achieving 5.6 × 10 −6 S/cm at 30 °C and 1.5 × 10 −4 S/cm at 60 °C. Comparative analysis of short-chain and long-chain crosslinkers revealed the trade-off between enhancing segmental mobility for ionic transport and maintaining sufficient thermal and mechanical stability, underscoring the need for further optimization to achieve a well-balanced network structure. Overall, this work demonstrates the potential of polyrotaxane-based cross-linked networks as a versatile platform for high-performance SPEs and offers insights for the rational design of next-generation solid-state electrolyte materials.

Resveratrol which is a natural polyphenol with good biological activity is mainly used in medicine, but it has poor photostability in vitro and short plasma half-life in vivo. Multi-armed poly( ε -caprolactone) (PCL) has attracted great attention because the increased number of terminal group broadens the polymeric flexibility in biomedical application. In the work, six-armed PCL was coupled with resveratrol through the hydrolysable ester bond to solve its disadvantage and realize protection for more resveratrol. The synthesized product was characterized with 1 H NMR, GPC and DSC. The molecular weight of the designed polymer was less than the experimental, and the polymeric distribution of molecular weight was narrow. In addition, resveratrol changed the physical form of six-armed PCL. The resveratrol coupling with six-armed PCL presented better photostability compared to original resveratrol. The concentration curve of the coupled resveratrol extended to the plain terrain after 80 min of irradiation, while the original resveratrol continued to decline. They were 72.10% and 52.35% at 2 h, respectively. Moreover, six-armed PCL could effectively protect resveratrol in vivo, and buy time for reaching the target site. The in vitro released amount of the original resveratrol was 52.89% within 3 h, while the coupled resveratrol was only 6.28%. Then this study provides a promising strategy for efficient usage of resveratrol.

The crystallization behavior of poly(e-caprolactone)/poly(ethylene glycol) (PCL/PEG) blend was investigated by differential scanning calorimetry (DSC) and polarized microscopy (POM). Individual phase transition peaks in the DSC curves for both PEG and PCL in all the polymer blends with different PCL contents were observed. The crystallization and melting peak temperatures of PEG were at 41 and 65°C, respectively; while the crystallization and melting temperatures of PCL located at 28 and 56°C, respectively. In-situ POM results demonstrated that spherulites crystalline morphology was formed for both PCL and PEG homopolymers. In PEG/PCL blend, however, both the phase separation morphology and spherulitic morphology can be observed. In blends with 30 or 50 wt % PCL, the PCL component formed dispersed phase and crystallized at lower temperature. However, in blends with 70% PCL, the phase inversion behavior occurred. The continuous PCL phase crystallized at 35°C, while the PEG dispersed phase crystallized at a lower temperature. Fractional crystallization behavior of PEG and PCL was controlled by temperature. The spherulites growth rate of PEG was greatly influenced by temperature, instead of the content of PCL component in the PCL/PEG blends.

Crystallization of polymer chains confined on a surface greatly influences surface properties. We have grafted comb-like copolymer, consisting of poly(2-hydroxyethyl methacrylate) (PHEMA) backbone and semicrystalline poly(ε-caprolactone) (PCL) side chains, on silicon surface and investigated the crystallization of such confined PCL chains upon solvent evaporation by using atomic force microscopy (AFM), grazing incidence wide-angle X-ray scattering (GI-WAXS), and polarized optical microscope (POM). Our studies reveal that the PCL chains align and form “flat-on” lamellae at a low PCL chain density. As the chain density increases, the comb-like polymer (PHEMA-g-PCL) chains undergo pancake-to-mushroom-to-brush transition, and the lamellae turn from “flat-on” to “edge-on” in orientation. Further increasing PCL chain density leads the “edge-on” lamellae to change from a quasi-two-dimensional (quasi-2D) to quasi-three-dimensional (quasi-3D). Because of the confinements of polymer chains, we can observe the evolution of spherulites at different stages during the mushroom-to-brush transition of PHEMA-g-PCL chains. The confinements also result in knobbly substructures in the edge-on lamellae.

Background The main challenge of polymeric micelles as drug delivery systems is that the actual delivery efficiency is not as high as expected, which is closely related with the interactions with the complex biological environments such as blood components, phagocytosis, and biodistribution. Herein, we expect to understand these concerns for the clinically relevant micelles that composed of methoxypolyethylene glycol (MPEG) with identical chain length And poly(ε-caprolactone) (PCL) with tunable chain length (PCL n -MPEG) (n=20, 30, and 40) wherein doxorubicin was encapsulated as a model drug. Methods The doxorubicin-loaded PCL n -MPEG micelles were prepared by a dialysis method and characterized by dynamic light scattering and transmission electron microscopy. The surface PEG density and chain conformation were investigated by dissipative particle dynamics simulation. The stability of the micelles was detected by nanoparticle tracking analysis. The effects of PCL chain length on the blood components, phagocytosis, and biodistribution were assayed in vitro and in vivo. Results The micelles exhibited spherical morphology with a diameter about 30nm. The PEG chain conformation from "mushroom-like" to "brush-like" was evident. The micelles have no remarkable effect on the red blood cells, blood coagulation, and platelet activation. Interestingly, the protein adsorption was affected and dependent on the chain conformation, with lowest adsorption for PCL 30 -MPEG, which also has the loWest phagocytosis. The stability of the micelles was in the order of PCL 40 -MPEG>PCL 30 -MPEG>PCL 20 -MPEG which was dependent on the PCL chain length. The micelles mainly accumulated in liver, with the order consistent with their stability, indicating that, besides the phagocytosis, the stability of the micelle plays an important role in biodistribution as well. The related mechanisms were proposed and discussed. Conclusion Manipulating the PEG/PCL ratio of the micelle is an effective approach to modulate the protein adsorption, phagocytosis, and biodistribution, which may be a prerequisite for clinical applications.

In this paper, the environment-friendly, water-soluble carbon dots (CDs) with stable photoluminescence (PL) have been prepared via the one-step pyrolysis of lotus leaf. Then the as-prepared CDs containing abundant hydroxylic and carboxylic groups were employed as cocatalyst with tartaric acid (TA) in ring-opening polymerization (ROP) of ε-caprolactone (ε-CL). The low-toxic organic acid TA, as main catalyst, was used to catalyze the ROP of ε-CL efficiently. The fluorescent CDs@PCL hybrids were obviously hydrophobic and they exhibited an excellent biocompatibility, and biodegradability due to the existence of PCL. Therefore the hydrophobic, biodegradable and multi-color fluorescent CDs@PCL hybrids may have potential applications in biomedicine, photocatalyst, bioimaging, and environmental analysis. Furthermore the application of CDs in catalyzing and initiating polymerization reaction will exemplify the versatility of CDs in the most unexpected fields.

Mechanical injury is the primary initiating cause of iatrogenic tracheal stenosis (TS), while bacterial infection and persistent inflammatory responses are crucial exacerbating factors driving fibrotic progression. In this study, we constructed a Tanshinone IIA / PCL nanofiber-covered airway stent (PCL@Tan-IIA) using electrospinning technology. Characterizations revealed that the PCL@Tan-IIA stent possessed hydrophobic surface characteristics, favorable mechanical strength, and desirable drug-release kinetics, coupled with potent antibacterial efficacy against planktonic methicillin-resistant Staphylococcus aureus (MRSA). Mechanistically, the stent exerted anti-inflammatory activity by downregulating the secretion of pro-inflammatory cytokine IL-6 and upregulating the expression of anti-inflammatory cytokine IL-10 in macrophages. Concurrently, it exhibited anti-fibrotic effects through inhibiting the migration and activation of human pulmonary fibroblasts. Moreover, the PCL@Tan-IIA stent implanted in New Zealand rabbits inhibited local airway bacterial proliferation, alleviated inflammation and reduced excessive fibrous tissue hyperplasia. This work provides a promising strategy to overcome stent-induced restenosis and has potential for clinical translation in the management of TS.
