en This link goes to a English section zh This link goes to a Chinese section Diabetic wound infections are a common complication of diabetes, which severely impact patients' quality of life. Effective treatment of diabetic wound infections remains one major challenge in the clinic, partially due to the formation of bacterial biofilm and antibiotic resistance. It is imperative to develop non-antibiotic-dependent strategies to efficiently eradicate biofilm infections in diabetic wounds. In this work, an innovative nanospray (CDs-HM) was successfully prepared by linking one acoustic sensitizer (hematoporphyrin monomethyl ether, HMME) to CDs fabricated with hemin, nickel(II) chloride, and polymer ethylene imine. CDs-HM exhibited catalase-enhanced sonodynamic properties and photothermal-enhanced chemodynamic properties. In vitro experiments and transcriptomic analysis demonstrated that CDs-HM successfully killed the bacteria and destroyed bacterial biofilm by disrupting bacterial cell membrane integrity, inducing oxidative stress, and inhibiting ATP production through synergistic photothermal therapy/chemodynamic therapy/sonodynamic therapy (PTT/CDT/SDT) effects. In the diabetic wound infection mouse model, CDs-HM remarkably eradicated MRSA biofilm, reduced inflammation levels, and promoted angiogenesis/collagen deposition so as to accelerate wound healing. It is noteworthy that CDs-HM displayed superior bacteria killing and wound healing effects compared with conventional vancomycin and nanosilver dressing interventions. The practicality and efficiency of CDs-HM endowed it with broad clinical translation prospects. It paved the way for developing novel strategies for combating diabetic wound infections. 摘要 en This link goes to a English section zh This link goes to a Chinese section 糖尿病伤口感染是糖尿病的常见并发症,严重影响患者的生活质量。

Black phosphorus quantum dots (BPQDs) with excellent biocompatibility, outstanding photothermal and photodynamic efficacies have attracted significant attention in cancer therapy. However, the low environmental stability and poor dispersity of BPQDs limit their practical applications. In the present work, biocompatible anionic waterborne polyurethane (WPU) nanoparticles were synthesized from castor oil to encapsulate the BPQDs. The WPU–BPQDs with a BPQDs loading capacity of about 13.8% (w/w) exhibited significantly improved dispersion and environmental stability without affecting the photothermal efficiency of BPQDs. Intriguingly, it was found that WPU encapsulation led to significant enhancement in the reactive oxygen species (ROS) generation of BPQDs, which indicated the enhanced photodynamic efficacy of the encapsulated BPQDs as compared to the bare BPQDs. The effect of solution pH on the ROS generation efficiency of BPQDs and the pH variation caused by BPQDs degradation was then investigated to explore the possible mechanism. In acidic solution, ROS generation was suppressed, while BPQDs degradation led to the acidification of the solution. Fortunately, after being encapsulated inside the WPU nanoparticles, the degradation rate of BPQDs became slower, while the acidic environment around BPQDs was favorably regulated by WPU nanoparticles having a special electrochemical double layer consisting of interior COO− and exterior NH(Et3)+, thus endowing the WPU–BPQDs-boosted production of ROS as compared to the bare BPQDs. Considering the undesired acidic tumor environment, this unique pH regulation effect of WPU–BPQDs would be beneficial for in vivo photodynamic efficacy. Both in vitro and in vivo experiments showed that WPU–BPQDs could effectively improve photodynamic therapy (PDT) and maintain outstanding photothermal therapy (PTT) effects. Together with the excellent dispersity, biocompatibility, and easy biodegradability, WPU–BPQDs can be a promising agent for PDT/PTT cancer treatments.

Phototherapy as a promising cancer diagnostic and therapeutic strategy has aroused extensive attention. However, single-wavelength near-infrared (NIR) light–triggered combinational treatment of photothermal therapy (PTT) and photodynamic therapy (PDT) is still a great challenge. Herein, a multifunctional micelle activated by a single-wavelength laser for simultaneous PTT and PDT as well as fluorescence imaging is developed. Briefly, new indocyanine green (IR820) is conjugated to d -α-tocopheryl polyethylene glycol 1000 succinate (TPGS) via the linker 6-aminocaproic acid, and then, chlorin e6 (Ce6) is encapsulated into the micelles formed by TPGS–IR820 conjugates to fabricate TPGS–IR820/Ce6 micelles. As the well-designed TPGS–IR820 conjugate shares a similar peak absorption wavelength with Ce6, this micelle can be applied with a single NIR laser (660 nm). The stable micelles exhibit excellent photothermal conversion efficiency in vitro and in vivo as well as high singlet oxygen generation capacity in tumor cells. After efficient cellular internalization, the as-prepared micelles display outstanding anticancer activity upon single NIR laser irradiation in vitro and in vivo. Furthermore, TPGS–IR820/Ce6 micelles show negligible systemic toxicity. The highly safe and effective TPGS–IR820/Ce6 micelles can offer an innovative strategy to construct single NIR light–induced PTT and PDT combined phototherapy nanoplatforms via suitable modification of organic phototherapeutic agents.

Given the increase in public safety incidents due to bacterial infections, sensitive detection and real-time inactivation of pathogenic bacteria have garnered increasing attention in the field of food safety, clinical diagnosis, and environmental monitoring. In this study, we constructed a “two-in-one” PB@DPA-Ce-GMP@Van (PCV) nanoplatform possessing enzyme-like properties and photothermal therapeutic (PTT) and photodynamic therapeutic (PDT) abilities for the detection and inactivation of pathogenic bacteria. PCV was captured on the surface of S. aureus to form a PCV/S. aureus complex. After centrifugation, PCV suspensions could catalyze non-fluorescent Amplex Red (AR) into fluorescent substrates and fluorescent scopolamine (SC) into non-fluorescent substrates. Hence, a ratiometric fluorescence probe with SC/AR reading was constructed for the sensitive detection of bacteria. The detection range reached 10 1 –10 7 colony-forming unit (CFU)/mL, and the detection limit was as low as 5.0 CFU/mL. Furthermore, approximately 99.7 % of free S. aureus can be inactivated by the PTT and PDT abilities of PCV. Thus, this nanoplatform exhibits a novel advantage in detecting and inactivating bacteria. Moreover, the bacterial infection wound model indicated that PVC exerts good disinfection ability. This study is of immense significance in the application of targeted sensing, eliminating bacterial infection, and even clinical therapy.

Nanocatalytic therapeutic agents with triple synergistic treatment modes of chemodynamic therapy (CDT), photothermal therapy (PTT) and photodynamic therapy (PDT) are emerging nanomaterials in malignancy treatment. Nevertheless, the passive diffusion and transport of nanomaterials result in poor permeability in tumor lesions, severely affecting the effectiveness of synergistic therapy. Herein, a dual-source-driven parachute-like Au 2 Pt@PMO@ICG Janus nanomotor (APIJNS) is prepared by an interfacial energy-mediated anisotropic growth strategy for PTT-mediated CDT/PDT triple synergistic cancer therapy. Such nanomotor can realize self-thermophoresis drive under the trigger of near-infrared light, which can effectively enhance the active permeability and uptake of the APIJNS in the tumor tissue, thus achieving efficient PTT/PDT/CDT synergistic therapeutic effect. In addition, parachute-like APIJNS exhibits high-efficiency catalase (CAT)-like activity and can catalyze the overexpressed H 2 O 2 in the tumor microenvironment (TME) to generate oxygen (O 2 ), not only alleviating the hypoxia in the tumor lesion, but also converting it into singlet oxygen ( 1 O 2 ) to activate the photosensitizer ICG, achieving PDT. This work provides new ideas for enhancing triple synergistic cancer therapy by improving tumor permeability with dual-drive Janus nanomotors based on dual noble metal nanozymes.

Successful synthesis, colloidal stability, broad optical absorption, efficient photothermal conversion and ROS generation under 808 nm laser were confirmed. In vitro assays were conducted using 5637 bladder cancer cells, and a murine bladder tumor model was established to evaluate the anticancer efficacy. Biocompatibility assessments were also carried out. Results The composite was constructed by integrating MnFe2O4 nanoparticles onto TiVNbMoC3 MXene nanosheets, which formed a layer - on - layer Schottky junction. Then, cRGD peptides were conjugated to the surface for tumor targeting. Comprehensive characterization confirmed successful synthesis, favorable colloidal stability, broad optical absorption, and efficient photothermal conversion and reactive oxygen species (ROS) generation under 808 nm laser irradiation. In vitro, the cRGD-targeted nanocomposite was efficiently internalized by 5637 bladder cancer cells and showed significantly higher anticancer efficacy than doxorubicin (DOX), including inhibition of proliferation, migration, colony formation, and induction of apoptosis and immunogenic cell death. In a murine bladder tumor model, the nanocomposite achieved the strongest tumor growth suppression and reduced final tumor volume more effectively than DOX. Biocompatibility assessments showed minimal hemolysis and no major organ damage at therapeutic doses. Conclusion This MXene-based Schottky junction nanocomposite represents a promising targeted platform for dual-modal photothermal/photodynamic therapy combined with immune activation in bladder cancer, warranting further translational investigation.

Large scale bone defects after bone tumor resection are difficult to reconstruct and repair, and there is also the possibility of tumor recurrence. Photothermal therapy (PTT) has the function of inhibiting tumor cells, but the risk of damage to normal cells is the main factor limiting the clinical application of PTT drugs, and most of them have a weak effect on regeneration for bone defects. Therefore, specific biomaterials that simultaneously eliminate bone tumors, have low toxicity, and promote osteogenesis have attracted considerable attention. In this paper, we successfully fabricated bioactive bredigite scaffolds (MS-BRT) functionalized with MoSe2 nanocrystals using a combination of 3D printing and hydrothermal methods. MS-BRT scaffolds not only have low toxicity and good osteogenic ability, but also have the ability to kill bone tumors by photothermal therapy. Using scanning electron microscopy (SEM), X-ray photoelectron spectroscopy (XPS), and an infrared thermal camera, MoSe2 nanocrystals were demonstrated to be successfully modified on the surface of BRT scaffolds. The MoSe2 nanocrystals endow the scaffolds with excellent photothermal properties, which can be well controlled by varying the hydrothermal reaction time and laser power density. Furthermore, the MS-BRT scaffolds can effectively kill MG-63 and HeLa cells and promote the adhesion and proliferation of osteoblasts. The performance of osteoblastic activity was assessed by alkaline phosphatase staining and alizarin red S staining, which results suggest that both MS-BRT and BRT have favorable osteogenic properties. This study combines the photothermal properties of semiconducting MoSe2 nanocrystals with the osteogenic activity of bioceramic scaffolds for the first time, providing a broader perspective for the development of novel biomaterials with dual functions of bone tumor treatment and bone regeneration.

As a controllable, simple method with few side effects, near-infrared (NIR) light-based photothermal therapy (PTT) has been proven an effective cancer therapeutic approach. However, PTT-induced inflammation is a potential negative factor. And the overexpressed heat shock proteins (HSPs) by cancer cells can protect them from hyperthermia during PTT. In this work, small-size Ti 3 C 2 T x MXene nanosheets with high photothermal conversion efficiency in the region of NIR, high cargo loading capability and good free radical scavenging capability were chosen for cancer PTT and anti-inflammation. And (-)-epigallocatechin gallate (EGCG) was applied to form EGCG/Fe metal-polyphenol nanodots on the nanosheets. EGCG being released in acid cancer cells could reduce the expression of HSPs and could be used for anti-inflammation. As a result, the complex nanosheets named MXene@EGCG could achieve enhanced cancer PTT and be anti-inflammatory. Both in vitro and in vivo studies proved the good photothermal ability of MXene@EGCG and demonstrated that it could inhibit the expression of HSPs in tumor cells and relieve PTT-induced inflammation. Therefore, the nanosheets show good results in tumor ablation with a low level of inflammation, which provides another possibility for cancer therapy. Statement of significance Photothermal therapy (PTT)-induced inflammation plays an essential role in some important stages of tumor development and is unfavorable for cancer treatment. And hyperthermia leads to the overexpression of heat shock proteins (HSPs) in cancer cells, which limits the therapeutic effect of PTT. Therefore, we coated small-size Ti 3 C 2 T x MXene nanosheets with (-)-epigallocatechin gallate (EGCG)/Fe metal-polyphenol nanodots and named them as MXene@EGCG. This system shows a good photothermal conversion efficiency at 808 nm. And it can release EGCG in cancer cells to inhibit the expression of HSPs, thus achieving an enhanced cancer PTT. Both MXene and EGCG can also diminish the PTT-trigged inflammation. Both in vitro and in vivo studies prove the good anti-cancer PTT effect and anti-inflammation capability of MXene@EGCG.

Chemodynamic therapy (CDT) with selective therapeutic and minimal side effects has attracted increasing attention in recent years. Nevertheless, the limited H2O2 and acid contents in the tumor microenvironment suppress the efficiency of the Fenton reaction. To overcome these issues, photothermal therapy (PTT) is employed to amplify the CDT effect and realize synergistic PTT/CDT. Herein, MoS2 sheets are functionalized with copper (Cu2O–MoS2), which integrates enhanced CDT and PTT to combat tumor cells. The 808 nm light activated Cu2O–MoS2 nanoplatform appears to be a potential candidate to improve the synergistic effect on the enhanced Fenton-like reaction and photothermal performance.

Currently, it is still a challenge to construct an integrated multi-functional nanoplatform with autofluorescence-free bioimaging and temperature sensing and photothermal therapy (PTT) for tumors located at 10 mm below the surface of local tissue (deep tissue) using gold nanorods (GNRs). Herein, through the self-assembly and surface modification of persistent luminescent nanoparticles (PLNPs) and GNRs using hexadecyl trimethyl ammonium bromide (CTAB) and phosphotungstic acid (PW 12 ) molecules, biocompatible PLNP-GNR composite nanoplatforms were constructed to simultaneously monitor autofluorescence-free bioimaging and temperature change during in vitro and in vivo PTT process using a single wavelength (635 nm) light excitation strategy. Especially, the nanoplatforms demonstrated that the local-tissue temperature can increase by 26 °C under 635 nm excitation (0.8 W·cm −2 ) and the photothermal conversion efficiency is ~37%. Among them, PLNPs were acted as optical probes for NIR bioimaging and ratiometric nanothermometer and the temperature sensitivity reaches to ~0.064 K −1 . This study is helpful for reducing photodamage to normal tissue through the accurate control of excitation light power during deep-tissue PTT process.

With the trend of sustainable development and the complex medical environment, there is a strong demand for multimodal antibacterial cellulose wound dressing (MACD) with photothermal therapy (PTT). Herein, a novel MACD fabrication strategy with PTT was proposed and implemented through graft polymerization of an imidazolium ionic liquid monomer containing iron complex anion structure. The fabricated hydrogels exhibited excellent antibacterial properties because of the efficient photothermal conversion ability (68.67 %) of ionic liquids and the intrinsic structural characteristic of quaternary ammonium salts. The antibacterial ratio of cellulosic hydrogel dressings to S. aureus and E. coli could reach 99.57 % and 99.16 %, respectively. Additionally, the fabricated hydrogels demonstrated extremely low hemolysis rates (<5 %) and excellent cell viability (~>85 %). Furthermore, in vivo antibacterial experimental results proved that the fabricated antibacterial dressings could significantly accelerate wound healing. Therefore, the proposed strategy would provide a new method of designing and preparing high-performance cellulose wound dressings.

Photothermal therapy (PTT), as a noninvasive and local treatment, has emerged as a promising anti-tumor strategy with minimal damage to normal tissue under spatiotemporally controllable irradiation. However, the necrosis of cancer cells during PTT will induce an inflammatory reaction, which may motivate tumor regeneration and resistance to therapy. In this study, polyoxometalates and a chloroquine diphosphate (CQ) co-loaded metal–organic framework nanoplatform with hyaluronic acid coating was constructed for efficient ovarian cancer therapy and anti-inflammation. Our results demonstrated that this nanoplatform not only displayed considerable photothermal therapeutic capacity under 808 nm near-infrared laser, but also had an impressive anti-inflammatory capacity by scavenging reactive oxygen species in the tumor microenvironment. CQ with pH dependence was used for the deacidification of lysosomes and the inhibition of autophagy, cutting off a self-protection pathway induced by cell necrosis–autophagy, and achieving the synergistic treatment of tumors. Therefore, we combined the excellent properties of these materials to synthesize a nanoplatform and explored its therapeutic effects in various aspects. This work provides a promising novel prospect for PTT/anti-inflammation/anti-autophagy combinations for efficient ovarian cancer treatment through the fine tuning of material design.Keywords:polyoxometalate;PTT;anti-autophagy;anti-inflammatory;cancer therapy
