CW-MFC for Efficient Degradation of Ofloxacin-Containing Wastewater and Dynamic Analysis of Microbial Communities

Li Yanhong,Zhao Zikang,Zhao Yao,Tang Shen,Zhang Lihao,Zhang Jie,Peng Yuqing

The overuse of antibiotics poses critical threats to environmental and human health. This study developed a constructed wetland-microbial fuel cell (CW-MFC) system integrated with pre-acclimated Shewanella oneidensis MR-1 to enhance ofloxacin (OFL) removal from wastewater. The CW-MFC demonstrated exceptional removal efficiency (>96%) for OFL across tested concentration gradients, significantly outperforming conventional constructed wetlands (CW) in both antibiotic elimination and conventional pollutant reduction. Microbial community analysis revealed that S. oneidensis MR-1 inoculation enhanced microbial diversity under OFL stress while promoting enrichment of electroactive Firmicutes phylum members compared to non-inoculated systems. The synergistic integration of bioelectrochemical processes and microbial consortia modification in CW-MFCs effectively addressed antibiotic contamination challenges. These findings establish CW-MFC technology combined with electroactive bacteria acclimation as a sustainable solution for pharmaceutical wastewater treatment, providing critical insights for optimizing antibiotic removal strategies in engineered ecosystems.

Cr(VI) bioreduction enhanced by the electron transfer between flavin reductase and persistent free radicals

Cheng Yu,Yassine Riahi,Qian Wang,Mengyang Feng,Abdelkader Mohamed,Ke Dai,Peng Cai,Qiaoyun Huang

Persistent free radicals (PFRs) in biochar are an important electron shuttle for mediating electron transfer, which has significant impact on the biogeochemical redox reactions. Although the influence of biochar on the extracellular electron transfer (EET) for redox cycle has been extensively studied, the molecular mechanism for promoting the EET with PFRs remains poorly understood. This study investigated the oxygen-centered PFRs-mediated Cr(VI) reduction by Shewanella oneidensis MR-1 (MR-1) and exhibited the molecular mechanism of electron transfer between flavin substances and PFRs. Results showed that the Cr(VI) bioreduction rate by MR-1 increased from 31% to 70% with the addition of biochar. Electrochemical results illustrated that biochar increased biocurrent generation in the Cr(VI) bioreduction process. 3D-EEM and LC/MS spectra indicated that MR-1 secreted the flavin mononucleotide (FMN) reductase that relied on the [H] to provide the electrons. Electron paramagnetic resonance spectra illustrated that PFRs in biochar accepted the electrons from FMN reductase and stored those bioelectrons. Because of the oxidation of FMN, the electron transfer from FMN reductase to PFRs would increase the intracellular reactive oxygen species, which further produced the extracellular ·O 2 − . The reduced PFRs released the bioelectrons, accelerating the Cr(VI) reduction by ·O 2 − . Together with the results of the mutant strains experiment, it was found that the EET by c -cytochrome and free radicals contributed to the Cr(VI) bioreduction by 7.1% and 92.9%, respectively. These findings revealed that the PFRs could participate in the EET process and promote the redox reactions, providing a new approach for enhancing the remediation of heavy metal pollution by microorganisms and suggesting the important role of PFRs in the electron transfer process.

Low-concentration resveratrol attenuates the spoilage potential of Shewanella putrefaciens in fish fillets

Hongniu Zhou,Qingyun Wang,Li Ni,Wen Zhang

8 μg/mL resveratrol suppresses spoilage potential without affecting growth. • Resveratrol causes apparent flagellar damage in S. putrefaciens JR-6. • Anti-spoilage effects persist after 30 serial passages with resveratrol. • Resveratrol-passaged JR-6 reduces TVB-N and spoilage volatiles in fish fillets. • Transcriptomics and docking suggest multi-target effects of resveratrol. Shewanella putrefaciens is a specific spoilage organism with strong spoilage potential in aquatic products. At a subinhibitory concentration of 8 μg/mL (1/8 MIC), resveratrol significantly reduced adhesion, biofilm formation, swimming motility, and extracellular protease production ability in S. putrefaciens JR-6 without affecting bacterial growth or membrane integrity. These inhibitory effects persisted after 30 serial passages. Transmission electron microscopy revealed shortened and apparently broken flagella in resveratrol-exposed cells. In fish fillet assays, inoculation with resveratrol-passaged strains delayed TVB-N accumulation and markedly reduced the semi-quantitative levels of three characteristic spoilage-associated volatile compounds. Transcriptomic analysis identified 76 differentially expressed genes associated with amino acid and nitrogen metabolism, oxidative phosphorylation, flagellar assembly, biofilm formation, and two-component systems. RT-qPCR confirmed significant downregulation of genes involved in histidine metabolism ( hisB and hisC ), nitrogen metabolism ( gltD and gltB ), oxidative phosphorylation ( coxA and coxC ), two-component systems ( dctM and dctP ), and quorum sensing ( gene3325 ). Molecular docking predicted interactions between resveratrol and the corresponding proteins through hydrogen bonding and hydrophobic contacts. These findings provide insight into the multi-target modulation of spoilage-associated phenotypes by low-concentration resveratrol and support its further evaluation as a natural preservative for aquatic products.

EDDS enhanced Shewanella putrefaciens CN32 and α-FeOOH reductive dechlorination of carbon tetrachloride

Li Yang Zhou,Shuai Chen,Hui Li,Shu Guo,Yong Di Liu,Jie Yang

S,S-ethylenediamine-N,N-disuccinic acid (EDDS) enhanced reductive dissolution of α-FeOOH by Shewanella putrefaciens CN32 (CN32), resulting in formation of surface-bound Fe(II) species ( Fe II EDDS) to improve reductive dechlorination of carbon tetrachloride (CT). The pseudo-first-order rate constants for bio-reduction extents of α-FeOOH by CN32 in the presence of 1.36 mM EDDS was 0.023 ± 0.0003 d −1 which was higher than without EDDS. The enhancement mechanism of bio-reduction was attributed to the strong complexation ability of EDDS to formed Fe III EDDS, which could be better utilized by CN32. The dechlorination kinetic of CT by Fe II EDDS (2.016 h −1 ) in the presence of 1.36 mM EDDS was 24 times faster than without EDDS. Chloroform were detected as main products for the degradation of CT. The chemical analyses and morphological observation showed that combination between EDDS and Fe 2+ produced Fe II EDDS complex, which had a reductive potential of −0.375 V and significantly enhanced CT dechlorination. The results showed that EDDS played an important role in enhancing the bio-reduction of α-FeOOH to accelerate reductive dechlorination of CT.

Label‐free proteomics study on Shewanella putrefaciens regulated by ε‐poly‐lysine treatment

Hou W.,Zhang Y.,Zhang Y.,Yue Q.,Wang L.,Min T.,Wang H.

AimsThe purpose of this study was to investigate the regulatory mechanism of ε‐PL on Shewanella putrefaciens. Methods and ResultsProteomics analysis of inhibitory effect of ε‐PL against S. putrefaciens was performed by label‐free quantitative assay based on high‐resolution mass spectrometry (MS). Quantification of 2206 proteins was obtained with high confidence, and a total of 36 differentially expressed proteins (DEPs), with 10 and 26 proteins showing upregulation and downregulation, respectively, were identified. Upon Go functional enrichment, 11, 5 and 8 specific Go terms in biological processes, molecular functions and cellular components were identified, respectively. Six KEGG pathways, including ‘ribosome’, were significantly enriched. Among the ribosome pathway, there were seven DEPs and all of them were distributed on large and small subunits of ribosome.ConclusionsThe significant downregulation of proteins, large subunits of ribosomal proteins RP‐L18, L30 and L27, small subunits ribosomal proteins S16 and S20, and RNA polymerase β' subunit protein rpoC were the critical action sites of ε‐PL to inhibit S. putrefaciens growth.Significance and Impact of the StudyShewanella putrefaciens is one of the representative fish‐spoilage bacteria regardless of fish type, and poses significant problems for the fish brewery. A better understanding of the antibacterial mechanism of ε‐PL on S. putrefaciens could make important contributions to development of biological control strategies of these economically important pathogens.

Impact of Fe(II) oxidation in the presence of iron-reducing bacteria on subsequent Fe(III) bio-reduction

Rong Chen,Hui Liu,Man Tong,Lei Zhao,Peng Zhang,Deng Liu,Songhu Yuan

The interplay of Fe(II) oxidation and Fe(III) bio-reduction occurs widely in both natural and engineered redox-dynamic systems. This study aimed to unravel the impact of Fe(II) oxidation by O 2 in the presence of iron-reducing bacteria on subsequent Fe(III) bio-reduction. Mixed solutions of Fe 2+ (0.1–0.5 mM) and Shewanella oneidensis strain MR-1 (MR-1, 2.0 × 10 7  CFU/mL) at neutral pH were first exposed to laboratory air for Fe(II) oxidation and bacterial inactivation, and then the resultant Fe(III) suspensions were switched to anoxic conditions for bio-reduction by the surviving bacteria. In the oxidation step, the coexisting MR-1 was inactivated by 0.8–1.71 orders of magnitude within 60 min. In the subsequent bio-reduction step, the resultant Fe(III) was bio-reduced by the surviving MR-1. Bio-reduction of the resultant Fe(III) by the surviving MR-1 was 1.8–2.5 times faster than that of the Fe(III) that was produced from Fe 2+ oxidation without MR-1 by fresh MR-1 cells at 2.0 × 10 7  CFU/mL. Although MR-1 inactivation during Fe(II) oxidation may inhibit Fe(III) bio-reduction, the increase in bio-availability of the resultant Fe(III) and the residual reactivity of dead cells led to net enhancement of bio-reduction under the tested conditions. Lepidocrocite was the sole Fe(III) mineral that was produced from Fe 2+ oxidation without MR-1, while 19% ferrihydrite was produced from Fe 2+ oxidation in the presence of MR-1. The formation of low-crystallinity ferrihydrite accounts for the increase in bio-availability of the Fe(III) minerals. The findings of this study highlight an important but overlooked impact underlying the interplay of Fe(II) oxidation and Fe(III) bio-reduction.

Intra/extracellular transmembrane electron transfer for bioelectric enhancement mediated by in-situ bio-FeS and polypyrrole coating

Dan Yuan,Shungang Wan,Yong Xiao,Lei Sun

This study involves the construction of an “inside-out” electronic network channel with in-situ formation of biogenic FeS (bio-FeS) nanoparticles in cells of Shewanella oneidensis MR-1 ( S . oneidensis MR-1) and a coating of conductive polypyrrole (PPy) on the outer surface of entire cells. The microbial fuel cell driven by bio-FeS and PPy co-modified S . oneidensis MR-1 (FeS/PPy@MR-1) has a maximum power density 3.6 times higher than that of the native strain. Electrochemical analysis demonstrates that the modifications of bio-FeS and PPy markedly enhance the direct electron transfer and c -type cytochrome-bound flavin-mediated single-electron reaction pathways and expand the electron output flux. The electron-donating ability of FeS/PPy@MR-1 is 7 times greater than that of the native strain. Bio-FeS and PPy can mediate novel intra/extracellular transmembrane electron transport pathways independent of the traditional respiratory chain, bridge spatially discrete redox environments, and optimize electron transfer pathways to intracellular to extracellular transfer electrons rapidly and maximally. These findings provide insights into conductive nanomaterial-mediated intra/extracellular transmembrane electron transfer, which is critical for the environment and bioenergy fields.

Iron Cycle Tuned by Outer-Membrane Cytochromes of Dissimilatory Metal-Reducing Bacteria: Interfacial Dynamics and Mechanisms In Vitro

Sheng-Song Yu,Jie-Jie Chen,Rui-Fen Cheng,Yuan Min,Han-Qing Yu

The biogeochemical cycle of iron is of great importance to living organisms on Earth, and dissimilatory metal-reducing bacteria (DMRB) with the capability of reducing hematite (α-Fe2O3) by outer-membrane (OM) cytochromes play a great role in the iron cycle. However, the dynamic binding of cytochromes to α-Fe2O3 at the molecular level and the resulting impact on the photon-to-electron conversion of α-Fe2O3 for the iron cycle are not fully understood. To address these issues, two-dimensional IR correlation analysis coupled with molecular dynamics (MD) simulations was conducted for an OmcA–Fe2O3 system as OmcA bonds stronger with hematite in a typical DMRB,Shewanella. The photoelectric response of α-Fe2O3 with the OmcA coating was evaluated at three different potentials. Specifically, the binding groups from OmcA to α-Fe2O3 were in the sequence of carboxyl groups, amide II, and amide I. Further MD analysis reveals that both electrostatic interactions and hydrogen bonds played essential roles in the binding process, leading to the structural changes of OmcA to facilitate iron reduction. Moreover, the OmcA coating could store the photogenerated electrons from α-Fe2O3 like a capacitor and utilize the stored electrons for α-Fe2O3 reduction in dark and anoxic environments, further driving the biogeochemical cycle of iron. These investigations give the dynamic information on the OM protein/hematite interaction and provide fundamental insights into the biogeochemical cycle of iron by taking the photon-induced redox chemistry of iron oxide into consideration.

Metabolic profiling, antibacterial effects, and preservation mechanisms of Paenibacillus provencensis metabolites in refrigerated Sea Bass (Perca fluviatilis)

Li Liu,Yakun Guo,Weijia Zhang,Fan Bai,Jinlin Wang,Ruichang Gao,Xiaoming Jiang,Yuanhui Zhao,Xinxing Xu

This study proposed to investigate the inhibition mechanism of the Paenibacillus provencensis (PP-2) metabolites on the dominant spoilage bacteria of sea bass and the application in preservation. Shewanella (71.11 %) and Pseudomonas (13.54 %) were the main genera. The Shewanella putrefaciens (L-1) and Pseudomonas fragi (L-4) were isolated and purified based on spoilage factors as dominant spoilage bacteria. PP-2 supernatant metabolites at minimum inhibitory concentration (MIC, 150 mg/mL) inhibited L-1 and L-4 by more than 95 % with significant antioxidant properties. The PP-2 metabolites were detected by LC-MS mainly including organic acids and derivatives, lipids molecules, organic heterocyclic compounds, and organic oxides. According to the transmission electron microscope (TEM) results, Additionally, the metabolites demonstrated antioxidant properties and disrupted bacterial cell membranes, highlighting their potential for controlling seafood spoilage. PP-2 metabolites significantly lowered pH ( p  < 0.05), TBARS values (0.33 ± 0.01 μmol/g), and TVB-N levels (24.06 ± 2.25 mg/100 g) compared to Nisin-treated and control groups after 6–8 d. After 8 d, colony counts in the PP-2 group (7.90 ± 0.01 log CFU/g) remained below the spoilage threshold. PP-2 metabolites also reduced protein oxidation, with carbonyl content at 0.33 ± 0.01 μmol/g and sulfhydryl content at 5.26 ± 0.18 μmol/g. The metabolites preserved textural properties, showing slower declines in hardness, elasticity, and chewiness, and lower juice leakage (9.37 ± 1.27 %). Metabolomic analysis revealed that PP-2 metabolites disrupted several key bacterial metabolic pathways, leading to bacterial death. These results suggest that PP-2 metabolites enhance seafood preservation by targeting bacterial metabolic processes.

Enhancing the extracellular electron transfer ability via Polydopamine@S. oneidensis MR-1 for Cr(VI) reduction

Boyu Jia,Tianbao Liu,Juanjuan Wan,Andrei Ivanets,Yujia Xiang,Lijuan Zhang,Xintai Su

Microbial reduction of hexavalent chromium (Cr (VI)) shows better efficiency and cost-effectiveness. However, immobilization of Cr (III) remains a challenge as there is a limited supply of electron donors. A greener and cleaner option for donating external electrons was using bioelectrochemical systems to perform the microbial reduction of Cr(VI). In this system, we constructed a polydopamine (PDA) decorated Shewanella oneidensis MR-1 ( S. oneidensis MR-1) bioelectrode with bidirectional electron transport, abbreviated as PDA@ S. oneidensis MR-1. The conjugated PDA distributed on the intracellular and extracellular of individual S. oneidensis MR-1 has been shown to accelerate electron transfer by outer membrane C-type cytochromes and flavin-bound MtrC/OmcA pathway by various electrochemical analyses. As expected, the PDA@ S. oneidensis MR-1 biofilm achieved 88.1% Cr (VI) removal efficiency (RE) and 58.1% Cr (III) immobilization efficiency (IE) within 24 h under the autotrophic conditions at the optimal voltage (−150 mV) compared with the control potential (0 mV). The PDA@ S. oneidensis MR-1 biofilm showed increased RE activity was attributed to the shortening of the distance between individual bacteria by PDA. This research provides a viable strategy for in situ bioremediation of Cr(VI) polluted aquatic environment.

Enhanced Cr(VI) bioreduction by biochar: Insight into the persistent free radicals mediated extracellular electron transfer

Cheng Yu,Xiaoxi Zhu,Abdelkader Mohamed,Ke Dai,Peng Cai,Shilin Liu,Qiaoyun Huang,Baoshan Xing

Biochar can act as a shuttle to accelerate the extracellular electron transfer (EET) by exoelectrogens. However, it is poorly understood how the persistent free radicals (PFRs) in biochar affected EET and the redox reaction. Herein, the effects of the biochar and chitosan modified biochar (CBC) on the Cr(VI) bioreduction by Shewanella oneidensis MR-1 (MR-1) was investigated. Kinetic study indicated that the Cr(VI) bioreduction rate constant by MR-1 was increased by 1.8–33.7 folds in the presence of biochar, and by 2.7–60.2 folds in the presence of CBC, respectively. Moreover, Cr(VI) bioreduction rates increased with the decreasing pH. Results suggested that the electrostatic attraction between Cr(VI) and redox-active particles could accelerate the EET by c -cytochrome due to the promotion of the Cr(VI) migration from aqueous phase to biochar or CBC. Electron paramagnetic resonance analysis suggested that the PFRs affected the electron transfer from the ·O 2 - generated by MR-1 to Cr(VI) and accelerate the Cr(VI) bioreduction. Remarkably, in the presence of PFRs, this electron shuttling process was dependent on the non-metal-reducing respiratory pathway. Our results offer new insights that free radicals may be widely involved in the EET and strongly impact on the redox reaction in the environment.

Shewanella-Augmented Cerium Oxide Nanoclusters With Superior Antioxidant Activity for Enhanced Radioprotection of Intestine

Haidong Li,Yuanyuan Shi,Weihao An,QiFeng Gao,Yaxin Hao,Zhencun Cui,Feifei Zhang,Yina Liu,Xiaolei Chen,Qiang Wu,Hui Wang,Wangsuo Wu,Longlong Tian

Developing oral radioprotectants for radiation-induced gastrointestinal syndrome (RIGS) is hindered by the non-renewable catalytic capacity of conventional antioxidants and the harsh gastrointestinal biological barriers. Herein, we developed a self-regenerating bio-hybrid nanozyme, SW@CeO 2 , by in situ growth of cerium oxide on electroactive Shewanella oneidensis (SW). Uniquely, this system exploits bacterial extracellular electron transfer (EET) to continuously fuel the reversible Ce 3+ /Ce 4+ redox cycle, thereby sustaining the catalytic scavenging of radiation-induced reactive oxygen species (ROS). Following oral administration, SW@CeO 2 demonstrates exceptional gastrointestinal stability, favorable biodistribution, and efficient intestinal colonization. In a lethal irradiation mouse model, the bio-hybrid effectively mitigates oxidative stress, preserves intestinal barrier integrity, and reshapes the gut microbiota by enriching beneficial taxa such as Lactobacillaceae . Remarkably, SW@CeO 2 increases the survival rate from 0% to 80%. This study presents a novel paradigm of harnessing microbial electron transport to sustain inorganic nanozyme activity, providing a robust and translatable solution for radioprotection.
