Biogenic PdAu nanoparticles (bio-PdAu NPs) synthesized by microorganisms exhibit remarkable catalytic activity relevant to environmental remediation. This study investigates the biosynthetic pathway of bio-PdAu NPs synthesized by Shewanella oneidensis MR-1, revealing that the bioreduction of Pd(II) and Au(III) follows first-order kinetics. Inhibition of NADH dehydrogenase (rotenone, Rot-PdAu), c-Cyts (diuron, Diu-PdAu), and hydrogenase (Cu2+, Cu-PdAu) decreased total metal-ion reduction efficiencies and Pd/Au composition, in which hydrogenases contributed most, followed by NADH dehydrogenase and c-Cyts. Notably, the presence of inhibitors affected the Au(III) reduction efficiency, altering particle size and composition. Statistical analysis indicated average particle sizes of 45.45, 50.15, and 13.16 nm for Rot-PdAu, Diu-PdAu, and Cu-PdAu, respectively, with a positive correlation to kinetic constants (R2 = 0.82). During biosynthesis, intracellular and extracellular pathways contributed 76.55 and 23.44%, respectively. For electrocatalytic hydrogen evolution reaction, the maximum current density (jmax) of bio-PdAu NPs exhibits substantially superior electrocatalytic performance compared to monometallic counterparts, with current densities approximately 1.8–3.2 times higher than individual bio-Pd or bio-Au systems. These findings deepen the understanding of the controlled synthesis pathways of bio-PdAu NPs and offer insights for developing biobimetallic NPs for environmental applications.

Mo 2 C has recently been introduced into microbial cathode electrocatalysis due to its well-known catalytic activity for hydrogen evolution reaction (HER), but a low electrode potential (i.e., a high energy input) is required usually. However, in the range of potential where HER cannot occur, its effect on microorganism/cathode interfacial kinetics remains unknown. Herein, an active Mo 2 C interface is constructed on conventional carbon cloth electrode to investigate its impact on electric-driven fumarate reduction by a typical electroactive microorganism ( Shewanella oneidensis MR-1) at a potential of −0.36 V (vs . standard hydrogen electrode) that cannot meet the HER requirement. The Mo 2 C-functionalized electrode achieves a greatly increased current consumption density of ∼0.041 mA cm −2 , about three times that of the control, indicating a faster fumarate reduction rate. This substantially proves that the enhancement of microbial electrocatalytic kinetics at Mo 2 C-functionalized interface is not confined to the induced hydrogen evolution. By analyzing its interaction with extracellular electron transfer mediators (e.g., c -type cytochromes and riboflavin), a robust inward extracellular electron transfer process based on the rapid electrochemical reaction of riboflavin at the Mo 2 C-functionalized interface is elaborated. This work provides a new understanding of the mechanism by which the nanostructured Mo 2 C interface enhances microbial cathode electrocatalysis.

In natural or engineered anaerobic environments, iron oxidation-driven microbial denitrification plays a critical role in the water or wastewater treatment. Herein, we report a previously unidentified metallic iron (Fe 0 )-dependent denitrification mode driven by the electro-syntrophic interaction between electroactive microorganism and denitrifier. In a model denitrifying consortium of Shewanella oneidensis and Pseudomonas aeruginosa , we find that P. aeruginosa can accept electrons for nitrate reduction via the constructed electron transfer system of Fe 0 – S. oneidensis – P. aeruginosa . In the electro-syntrophic consortium, the membrane-bound CymA–OmcA–MtrC protein complexes of S. oneidensis drive the generation, transfer and consumption of electrons, thus enabling modulation of microbial metabolic activity. Specially, using Fe 0 as the sole electron donor, S. oneidensis can act as a bio-engine to harvest electrons and conserve energy from Fe 0 biocorrosion. Electrons released by S. oneidensis are utilized by P. aeruginosa for accomplishing microbial denitrification. Metatranscriptomics analysis demonstrated that the direct electron cross-feeding process facilitates the expression of genes encoding for denitrification enzymes, intracellular electron transfer proteins, and quorum sensing of P. aeruginosa . The Fe 0 -dependent electronic syntrophy in this work could provide a metabolic window for the growth of denitrifiers that is a new insight into nitrate removal or global nitrogen cycle.

The co-contamination of hexavalent chromium Cr(VI) and chlorpyrifos (CPF) poses a severe challenge to traditional remediation technologies due to synergistic toxicity. This study constructed a bio-material synergistic system (FeO/C@ S. oneidensis MR-1) for their simultaneous removal. Results demonstrated that under optimal conditions (0.08 g/L FeO/C, 10% amount of inoculation, pH 7.0), the removal rates of Cr(VI) and CPF reached 75.5% and 93.7%, respectively, maintaining robust degradation efficiency even under a severe organic shock load of 50 mg/L CPF. Importantly, comprehensive multi-scale characterizations (SEM, CLSM, EDS, XRD, FTIR, XPS, 3D-EEM), revealed a bidirectional bio-material synergy: FeO/C acted as an electron shuttle and play a role of bio-driven electron allocation, preferentially driving the Fe(III)/Fe(II) cycle to detoxify Cr(VI), while dynamically secreted extracellular polymeric substances (EPS) acted as an active interface to enrich CPF ensuring the efficient biomineralization of CPF and complex heavy metals. This dual action effectively alleviated acute oxidative stress, enabling cell viability to recover to 80%. For real-world applications, a clear takeaway is that this self-sustaining system entirely eliminates the need for external chemical electron donors. It offers a scalable, bio-driven electron allocation green engineering strategy for the in-situ bioremediation of complex heavy metal-organic wastewater.

This study elucidates the mechanism by which hexavalent chromium [Cr(VI)] synergistically promotes malathion biodegradation in Shewanella oneidensis MR-1. Exposure to 5 mg/L Cr(VI) significantly enhanced the malathion degradation rate by 2.3-fold, achieving 96.79 % removal within 72 h under optimized conditions (5 % inoculum, pH 6). Analysis of cell membrane parameters combined with microscopic observation techniques indicated that Cr(VI) induced biofilm densification and membrane lipid peroxidation. Metabolomic profiling identified three critical Cr(VI)-activated pathways: (1)tryptophan-mediated redox homeostasis, (2)sulfur assimilation for antioxidant substances biosynthesis, and (3)cofactor biosynthesis for electron transport optimization. These metabolic adaptations increased membrane permeability and accelerated reactive oxygen species diffusion. The findings highlight Cr(VI) as a metabolic modulator that reprograms microbial catabolism via redox signaling, providing a mechanistic basis for developing biocatalysis strategies in multi-pollutant environments.

Dissimilatory iron-reducing bacteria (DIRB) offer an eco-friendly approach to address nanoscale zero-valent iron (nZVI) surface passivation problem, yet their efficacy is limited by the slow direct extracellular electron transfer (EET) between DIRB and Fe(III) oxide passivation layers (Fe( III )OPL). This study demonstrates that natural dissolved humic substances (DHS)-mediated electron shuttling unlocking nZVI reactivity in Shewanella oneidensis MR-1 (MR-1) system for enhance Cr(VI) removal. Seven DHSs from diverse sources were evaluated, revealing that Cr(VI) removal rate constants ( k obs ) correlate strongly with DHS's quinone content (C=O) and electron transfer capacity (ETC), quantified as k obs = 1.66 C C = O − 9.39 ( P < 0.0007) and k obs = 19.37 ETC + 0.74 ( P < 0.0005). DHS-mediated systems achieve 2.0–12.1-fold higher k obs than nZVI/MR-1 alone. The enhancement mechanism of DHS-mediated Cr(VI) removal by nZVI/MR-1 is that DHS effectively facilitates Fe( III )OPL dissolution on the nZVI surface via electron shuttling, maintaining nZVI reactivity and amplifying Fe(II) generation by 1.65-fold. Moreover, DHS stimulates MR-1 to secrete more extracellular polymeric substances (EPS), enhancing Cr(VI) adsorption and reduction. This work elucidates DHS's dual role in maintaining nZVI activity and amplifying Cr(VI) removal by nZVI/MR-1, while establishing a predictive framework linking DHS molecular traits parameters to Cr(VI) removal kinetic, offering sustainable strategies for heavy metal-contaminated water bodies remediation by integrating biogeochemical and material engineering principles.

Microbially mediated iron redox processes are of great significance in the biogeochemical cycles of elements, which are often coupled with soil organic matter (SOM) in the environment. Although the influences of SOM fractions on individual reduction or oxidation processes have been studied extensively, a comprehensive understanding is still lacking. Here, using ferrihydrite, Shewanella oneidensis MR-1, and operationally defined SOM components including fulvic acid (FA), humic acid (HA), and humin (HM) extracted from black soil and peat, we explored the SOM-mediated microbial iron reduction and hydroxyl radical (•OH) production processes. The results showed that the addition of SOM inhibited the transformation of ferrihydrite to highly crystalline iron oxides. Although FA and HA increased Fe(II) production over four times on average due to complexation and their high electron exchange capacities, HA inhibited 30–43% of the •OH yield, while FA had no significant influence on it. Superoxide (O2•–) was the predominant intermediate in •OH production in the FA-containing system, while one- and two-electron transfer processes were concurrent in HA- and HM-containing systems. These findings provide deep insights into the multiple mechanisms of SOM in regulating microbially mediated iron redox processes and •OH production.

With the death and decomposition of widely distributed photosynthetic organisms, free natural pigments are often detected in surface water, sediment and soil. Whether free pigments can act as photosensitizers to drive biophotoelectrochemical metabolism in nonphotosynthetic microorganisms has not been reported. In this work, we provide direct evidence for the photoelectrophic relationship between extracellular chlorophyll a (Chl a ) and nonphotosynthetic microorganisms. The results show that 10 μg of Chl a can produce significant photoelectrons (∼0.34 A/cm 2 ) upon irradiation to drive nitrate reduction in Shewanella oneidensis . Chl a undergoes structural changes during the photoelectric process, thus the ability of Chl a to generate a photocurrent decreases gradually with increasing illumination time. These changes are greater in the presence of microorganisms than in the absence of microorganisms. Photoelectron transport from Chl a to S. oneidensis occurs through a direct pathway involving the cytochromes MtrA, MtrB, MtrC and CymA but not through an indirect pathway involving riboflavin. These findings reveal a novel photoelectrotrophic linkage between natural photosynthetic pigments and nonphototrophic microorganisms, which has important implications for the biogeochemical cycle of nitrogen in various natural environments where Chl a is distributed.

Exogenous electron mediators (EMs) can facilitate extracellular electron transfer (EET) via electron shuttling processes, but it is still unclear whether and how biofilm formation is affected by the presence of EMs. Here, the impacts of EMs on EET and biofilm formation were investigated in bioelectrochemical systems (BESs) with Shewanella oneidensis MR-1, and the results showed that the presence of five different EMs led to high density current production. All the EMs substantially promoted biofilm formation with 15–36 times higher total biofilm DNA with EMs than without EMs, and they also increased the production of extracellular polymeric substances, which was favorable for biofilm formation. The current decreased substantially after removing EMs from the medium or by replacing electrodes without biofilm, suggesting that both biofilm and EMs are required for high density current production. EET-related gene expression was upregulated with EMs, resulting in the high flux of cell electron output. A synergistic mechanism was proposed: EMs in suspension were quickly reduced by the cells and reoxidized rapidly by the electrode, resulting in a microenvironment with sufficient oxidized EMs for biofilm formation, and thus, besides the well-known electron shuttling process, the EM-induced high biofilm formation and high Mtr gene expression could jointly contribute to the EET and subsequently produce a high density current. This study provides a new insight into EM-enhanced current production via regulating the biofilm formation and EET-related gene expression.

Microbial fuel cells with mixed bacteria always show better power output capacity than those with single bacteria due to synergetic effects between species. Here, a rational-designed synthetic three-species alliance system based on Shewanella oneidensis MR-1 (the model exoelectrogen), Pseudomonas aeruginosa, and Lactobacillus plantarum was constructed upon process optimization, the three-species alliance system in MFC delivered a maximum power density of ∼ 207 mW/m 2 by using the glucose as the substrate, which was 2 ∼ 58 times higher than that of MFC inoculated with single species. In addition, the synthetic alliance system could generate electricity from broad substrate spectrum (lactose, saccharose, maltose, arabinose, cellobiose) with enhanced performance. Electrochemical and metabolism analysis indicated inter-species synergetic enhancement on charge transfer, substrate utilization, and biofilm formation was the underlying mechanism for performance improvement in MFC by this synthetic alliance system. In this system, L. plantarum uses glucose to produce lactic acid as the electrogenic substrate and electron donor of S. oneidensis MR-1, and P. aeruginosa uses glucose to produce another electron shuttle phenazine, which can also promote the growth of S. oneidensis MR-1 biofilm and help the alliance system to transfer electrons better. This work demonstrated a new strategy to enhance the MFC performance with a rational designed synthetic bacterial community, which would further renew the toolbox for MFC optimization.

Large quantities of low-concentration uranium-contaminated water from the nuclear industry poses a critical environmental challenge due to the limitations of current methods on removal efficiency and adsorption selectivity. This study developed a novel biofilm cathode for enhanced uranium adsorption by leveraging the quorum sensing (QS) system of Shewanella oneidensis MR-1. Biofilm formation was induced using exogenous acyl-homoserine lactones (AHLs), with C4-HSL (10 μmol/L) identified as the most efficient signal molecule. It significantly improved biofilm properties, increasing thickness by 107.7 % versus the control without AHLs, boosting extracellular protein content, and increasing the proportion of living cells. Microstructure analysis (CLSM) revealed a “base-polysaccharide-gel” three-dimensional structure, where proteins, lipids, and β-polysaccharides form a functional gel layer that provides the primary functional matrix for uranium binding. The C4-HSL-induced biofilm exhibited a 43.4 % higher reduction peak current and a 33.3 % decrease in electron transfer resistance, confirming improved electron transfer efficiency. Furthermore, coupling CLSM, SEM-EDS, FT-IR and XPS analysis indicated uranium capture was primarily dominated by complexation/coordination with functional groups on extracellular polymeric substances (EPS), supplemented by electrochemical reduction of 25.91 % soluble U(VI) to insoluble U(IV). Accordingly, a possible mechanism model of QS-driven biofilm cathode is proposed for optimizing biofilm structure and enhancing uranium capture. Finally, verification tests in actual uranium-contaminated groundwater demonstrated the biofilm cathode exhibited exceptional performance with a 99.4 % uranium recovery rate and high selectivity (K d,U at 34.57 L/g). These findings highlight the significant potential of AHLs-triggered QS as a powerful strategy to optimize bioelectrochemical properties for highly-efficient uranium remediation and resource recovery.

Probing of the single-cell level extracellular electron transfer highlights the maximum output current for microbial fuel cells (MFCs) at hundreds of femtoampere per cell, which is difficult to achieve by existing devices. Past studies focus on the external factors for boosting charge-extraction efficiency from bacteria. Here, we elucidate the intracellular factors that determine this output limit by monitoring the respiratory-driven shrinking kinetics of a single magnetite nanoprobe immobilized on a single Shewanella oneidensis MR-1 cell with plasmonic imaging. Quantified dissolving of nanoprobes unveils a previously undescribed bio-current fluctuation between 0 and 2.7 fA on a ∼40 min cycle. Simultaneously tracing of endogenous oscillations indicates that the bio-current waves are correlated with the periodic cellular electrokinesis. The unsynchronized electron transfer capability in the cell population results in the mean current of 0.24 fA per cell, significantly smaller than in single cells. It explains why the averaged output current of MFCs cannot reach the measured single-cell currents. This work offers a different perspective to improve the power output by extending the active episodes of the bio-current waves.
