Evaluating the bacterial activity effectively is critical to addressing the challenges posed by bacterial infections. Electrochemistry offers significant advantages in accuracy and cost efficiency compared with methods that are more time-consuming or require expensive instrumentation. This study initially established an electrochemical method for detecting bacterial activity using heat treatment as the pretreatment step. Subsequent optimization of the heat treatment and detection conditions further enhanced the method efficiency. The detection results of Escherichia coli (E. coli) and Staphylococcus aureus (S. aureus) under different heating conditions showed that the peak current values of E. coli and S. aureus were the highest after heating at 50 °C for 15 min. The dynamic changes in the electrochemical signals from the heat-treated bacteria with alteration in heating conditions were indicated to be related to the effects of heating on the external structure and purine metabolism of the bacterial cells. Although heat-treated E. coli and S. aureus exhibited similar peak potentials, different substances were observed to have contributed to these potentials. A positive linear correlation was observed between the peak current and bacterial concentration. Compared to the plate counting method, the electrochemical method detected the activity changes in E. coli and S. aureus from the logarithmic to the stationary phase earlier. The findings support the broad application of electrochemical methods in microbial detection and provide valuable insights into the purine metabolism of heat-stressed bacteria.

Statement of problem Staphylococcus aureus ( S aureus ) and Streptococcus mutans ( S mutans ) are pathogenic bacteria associated with denture-related stomatitis. Zinc (Zn), a widely used metallic antibacterial agent, can induce the production of reactive oxygen species (ROS) in cells, thereby inhibiting microbial growth and proliferation; studies, however, are lacking. Purpose The purpose of this in vitro study was to determine the antibacterial and antibiofilm function of Zn 2+ against S aureus and S mutans and assess gene expression changes to clarify its inhibitory mechanisms. Material and methods Zn 2+ at one-fourth minimum inhibitory concentration (MIC), one-half MIC, and MIC was co-cultured with the bacteria. Membrane integrity was evaluated by alkaline phosphatase (AKP) activity and live-dead staining. The oxidative stress mechanism was assessed by intracellular reactive oxygen species (ROS) and lipid peroxidation levels. Deoxyribonucleic acid (DNA) integrity was evaluated using agarose gel electrophoresis. Ribonucleic acid (RNA) sequencing combined with gene ontology (GO) and the Kyoto encyclopedia of genes and genomes (KEGG) bioinformatics analyses was performed to identify Zn²⁺-affected molecular pathways. The effect of Zn 2+ on key gene expression in S aureus and S mutans was evaluated by quantitative reverse transcription polymerase chain reaction (qRT-PCR). Results In S aureus , Zn 2+ upregulated ROS and malondialdehyde (MDA) levels and disrupted oxidative phosphorylation, which indicated the induction of oxidative stress and caused bacterial death. In S mutans , Zn 2+ interfered with metabolic pathways by targeting 2-component systems (TCS), weakening acid resistance, impairing biofilm adhesion, and disrupting cell wall peptidoglycan synthesis, ultimately resulting in bacterial death. The qRT-PCR results were consistent with those of GO and KEGG analyses. Conclusions Differentially expressed genes (DEGs) in S aureus were primarily enriched in pathways related to DNA replication, mismatch repair, and oxidative phosphorylation, suggesting that Zn 2+ co-culture significantly affected genetic stability and redox-associated cellular functions. In contrast, DEGs in S mutans were predominantly involved in microbial metabolism, molecular systems, and bioadhesion pathways, with no significant changes in oxidative stress markers.

Staphylococcus aureus is a major foodborne pathogen characterized by strong virulence and biofilm-forming ability, posing a persistent threat to food safety. Aquilaria sinensis (Lour.) Gilg, a medicinal and edible plant found in tropical regions such as Hainan, has rarely been investigated for its quorum-sensing inhibitory (QSI) potential. In this study, four types of A. sinensis essential oils were systematically evaluated for their ability to inhibit quorum sensing and related virulence in S. aureus . Among them, the Tong-Ti-Xiang type supercritical essential oil (TTC) exhibited the strongest activity. At a sub-inhibitory concentration of 125 μg/mL (1/4 MIC), TTC reduced biofilm formation by up to 78% without affecting bacterial growth. Confocal laser scanning Microscopy and scanning electron microscopy analyses further confirmed that TTC compromised the density and structural integrity of the biofilms. Moreover, TTC suppressed α-hemolysin activity and significantly downregulated quorum-sensing-regulated genes involved in biofilm formation and adhesion, including icaA , icaD , clfA , clfB , and agrA . Gas chromatography mass spectrometry analysis revealed that TTC is rich in chromone derivatives, among which 2-phenethyl-4H-chromen-4-one showed strong binding affinity toward the adhesion-associated IcaA protein and the QS-associated AgrA protein in molecular docking analysis, suggesting it as a key active compound responsible for the observed QSI effect. Overall, these findings highlight TTC as a promising natural anti-biofilm and anti-virulence agent for controlling foodborne pathogens.

YM155 is a clinically evaluated anticancer with a fused naphthoquinone-imidazolium scaffold. In this study, we demonstrated that based on weak or cryptic antibacterial activity of YM155 against methicillin-resistant Staphylococcus aureus (MRSA) (MIC of 50 μg/ml), some congeneric compounds with short alkyl chains (e.g., c5 with a hexyl chain) at the N3 position of the scaffold, displayed more potent antibacterial activity against MRSA (MIC of 3.13 μg/ml), which is in a clinically achievable range. Their antibacterial activity was evident against Gram-negative bacteria, only in the presence of the outer membrane-permeabilizing agent, polymyxin B. The antibacterial efficacy of c5 was confirmed using the Drosophila systemic infection model. We also characterized five spontaneous c5-resistant MRSA mutants that carry mutations in the ubiE gene, for quinone metabolism and respiratory electron transfer, and subsequently exhibited reduced respiration activity. The antibacterial activity of c5 was compromised either by an antioxidant, N-acetylcysteine, or in an anaerobic condition. These suggest that the antibacterial mechanism of c5 involves the generation of reactive oxygen species (ROS), presumably during respiratory electron transport. This study provides an insight into “drug redirecting,” through a chemical modification, based on an ROS-generating pharmacophore.

A novel antimicrobial peptide named as phe 11 -protonectin was designed and synthesized by replacing the eleventh amino acid of protonectin with phenylalanine. Compared with broad-spectrum protonectin, this antimicrobial peptide showed greatly improved selectivity against Gram-positive bacteria. It was adopted to analyze Gram-positive bacteria utilizing its strong binding to cellular membrane. Using Staphylococcus aureus ( S. aureus ) as a model bacterium, a dual-site recognition-based chemiluminescent method was successfully established. Immunoglobulin G (IgG) was adopted as the capture agent for S. aureus , while alkaline phosphatase-conjugated phe 11 -protonectin was adopted as the tracer agent for S. aureus . Thus an enzyme-conjugated sandwich complex of S. aureus was formed, which allowed selective and sensitive analysis of this pathogen. Utilizing the dual-site recognition mechanism, the method showed a wide linear range of 1.0 × 10 3 - 1.0 × 10 7 CFU mL −1 and a low detection limit of 2.9 × 10 2 CFU mL -1 (3σ). The whole detection process can be accomplished within 130 min. It was successfully applied to analyze S. aureus in various sample matrixes including physiological saline injection, human urine and milk. The results demonstrated the application potential of the antimicrobial peptide in bacterial analysis.

Purpose Staphylococcus aureus (S. aureus) is an important bacterial pathogen, which creates infective inflammation to human being and animals. Mangiferin (MG) is one of the natural flavonoids with anti-inflammatory, anti-bacterial, and anti-oxidative properties. However, the anti-apoptosis and anti-autophagy of MG are unknown. Hence, this study was aimed to research the inhibition of MG on S. aureus-induced apoptosis and autophagy in RAW264.7 cells. Methods The RAW264.7 cells were pretreated with MG, or pretreated with SP600125 or anisomycin synchronously, and then infected with S. aureus (MOI=100:1). The viability and proliferation status of RAW264.7 cells were detected by MTT and EdU assay. The relative expression of TNF-α, IL-6 and IL-10 protein was tested with ELISA. The levels of Bax, Bcl-2, caspase-3, c-Jun N-terminal kinase (JNK), extracellular-regulated protein kinase (ERK), p38, LC3, Beclin-1, p62, phosphorylated JNK, phosphorylated p38 and phosphorylated ERK in cells were detected by Western blotting. The apoptosis rate of RAW264.7 cells was analyzed by flow cytometric assay. Results The study showed that MG significantly attenuated RAW264.7 cells apoptosis and autophagy caused by S. aureus. MG alleviated S. aureus-induced apoptosis by down-regulating the protein level of active caspase-3 and Bax and up-regulating the level of Bcl-2. MG also inhibited S. aureus-induced autophagy via decreasing the protein level of LC3-II/LC3-I and Beclin-1 or increasing the protein expression of p62. This protective role was dependent on the up-regulation of JNK signal pathway, which was confirmed by using JNK agonist and inhibitor. Conclusion Our results demonstrated that MG might protect RAW264.7 cells from S. aureus-induced apoptosis and autophagy via inhibiting JNK/Bax-dependent signal pathway. Therefore, MG may be a potential agent against pathological cell damage induced by S. aureus infection.

Staphylococcus aureus poses a severe public health problem as one of the vital causative agents of healthcare- and community-acquired infections. There is a globally urgent need for new drugs with a novel mode of action (MoA) to combat S. aureus biofilms and persisters that tolerate antibiotic treatment. We demonstrate that a benzonaphthopyranone glycoside, chrysomycin A (ChryA), is a rapid bactericide that is highly active against S. aureus persisters, robustly eradicates biofilms in vitro, and shows a sustainable killing efficacy in vivo. ChryA was suggested to target multiple critical cellular processes. A wide range of genetic and biochemical approaches showed that ChryA directly binds to GlmU and DapD, involved in the biosynthetic pathways for the cell wall peptidoglycan and lysine precursors, respectively, and inhibits the acetyltransferase activities by competition with their mutual substrate acetyl-CoA. Our study provides an effective antimicrobial strategy combining multiple MoAs onto a single small molecule for treatments of S. aureus persistent infections.

Treatment of acute osteomyelitis (OM) poses a significant challenge because of the difficulty in removing bacteria and the aberrant innate immune responses at the infection site. Topical ozone therapy has demonstrated favorable outcomes in treating OM, but its high reactivity and short half-life severely limit its clinical application. To address this issue, a suitable ozone delivery system that can promote the therapeutic effects of topical ozone therapy in OM is needed. In this study, we developed ozone-loaded fluorinated hyaluronic acid nanoparticles embedded in a thermoresponsive hydroxypropyl chitin (HPCH) hydrogel (Ozone@Gel) for treating acute OM. The ozone-delivering nanocomposite hydrogel could release ozone continuously and stably at the injection site for over 15 days. The ozone loaded in this ozone-delivering nanocomposite hydrogel was about 100 mg/L. In vitro antibacterial experiments demonstrated that the Ozone@Gel effectively attenuated Staphylococcus aureus proliferation and reduced biofilm formation by inhibiting the expression of bacterial polysaccharide-related genes (agr, atlE, aap, icaA, and icaBC). Further, animal experiments showed that Ozone@Gel effectively reduced bacterial load at the site of infection by enhancing neutrophil antimicrobial activity, thereby alleviating the progression of acute OM. Thus, this novel therapeutic strategy based on topical ozone therapy is a promising and feasible intervention for treating acute S. aureus OM.

Gut microbiota plays a crucial role in resisting the invasion of pathogens, particularly multidrug-resistant (MDR) bacteria, which pose a significant threat to public health. While exercise offers numerous health benefits, its impact on host colonization resistance remains largely unclear. In this study, we demonstrate that moderate exercise significantly reduces gut colonization by methicillin-resistant Staphylococcus aureus (MRSA), a clinically important MDR pathogen. Moreover, we identify an understudied strain of the intestinal probiotic Dubosiella newyorkensis (L8) as a critical factor in mediating exercise-induced colonization resistance against MRSA. Mechanistically, L8 enhances the deprivation of fucose, a crucial carbon source essential for MRSA growth and pathogenicity. This process relies on the high binding affinity of pyruvate to the ILE257 site of the lactate dehydrogenase in L8. Overall, our work highlights the importance of moderate exercise in maintaining host colonization resistance and demonstrates L8 as a probiotic in protecting against MRSA colonization.

Optical fiber bacterial sensors have attracted attention in many fields due to their unique advantages. However, it is still difficult to promote its practicality which is mainly subject to their low sensitivity, inferior stability, and reliability. This work reported here an optical fiber surface plasmon resonance (SPR) biosensor enhanced by gold nanoparticles (AuNPs)/Au film coupling for label-free detection the Staphylococcus aureus ( S. aureus ), while both the gold substrates were successfully biofunctionalized with goat anti-human immunoglobulin G (IgG) as the probe to capture and detect the bacteria. In the experiment, the refractive index(RI) sensitivity of the Multimode-Singlemode-Multimode optical fiber (MSMOF) coated with Au film was improved by 19 % after the AuNPs were homogeneously decorated on the Au film surface. To detect S. aureus by the sensor , the SPR wavelength variation in the transmission spectrum was recorded, and its limit of detection (LOD) was demonstrated experimentally to be about 50 CFU/mL and was obtained theoretically to be 1.18 CFU/mL during its concentration range of 50–10 8 CFU/mL. Also, it shows great linearity in the range of 10 2 -10 8 CFU/mL and the sensor exhibits excellent specificity and stability. This kind of SPR-enhanced optical fiber biosensor is fully promising for its applications in biomedicine, food safety, pharmaceuticals, etc.

Featured ApplicationDuring food processing, the surface of food and processing equipment is easily contaminated by microorganisms, which will cause food spoilage or the spread of disease, threatening food safety. Plasma-activated solution can be used as a novel food disinfectant, enhancing food quality. Plasma-activated solution will not leave chemical residues or cause environmental pollution. It is a green and environmentally friendly disinfectant and promises to replace traditional chemical disinfectants.AbstractPlasma-activated solution has attracted more attention in the food industry due to no chemical residue and good bacteriostatic properties. This study aimed to evaluate the effects of plasma-activated hydrogen peroxide solution (PAH) on the morphophysiology ofStaphylococcus aureusbiofilms. PAH was prepared using dielectric-barrier-discharge plasma and incubated withS. aureusbiofilms for 0–40 min. Changes in biofilm morphophysiology were evaluated with laser scanning confocal microscopy, electron microscopic images, reactive oxygen species (ROS) content, metabolic capacity, and 1% agarose gel. Results indicated that the population ofS. aureusin the biofilms was reduced by 4.04-log after incubation with PAH for 30 min. The thickness and metabolic capacity of biofilms were decreased, the ROS content and DNA fragments of bacteria increased after PAH treatments. Data suggested that PAH treatments significantly destroyed the morphophysiology ofS. aureus(ATCC 6538) biofilms and could be considered as a valuable anti-biofilm technology to reduce foodborne pathogens on food and/or in food facilities. Keywords:foodborne pathogen;cold plasma;nonthermal antimicrobial treatment;metabolic capacity;DNA damage
