Metformin-loaded β-TCP/CTS/SBA-15 composite scaffolds promote alveolar bone regeneration in a rat model of periodontitis

Xu Wanghan,Tan Wei,Li Chan,Wu Keke,Zeng Xinyi,Xiao Liwei

Periodontitis is a progressive infectious inflammatory disease, which leads to alveolar bone resorption and loss of periodontal attachment. It is imperative for us to develop a therapeutic scaffold to repair the alveolar bone defect of periodontitis. In this study, we designed a new composite scaffold loading metformin (MET) by using the freeze-drying method, which was composed of β-tricalcium phosphate (β-TCP), chitosan (CTS) and the mesoporous silica (SBA-15). The scaffolds were expected to combine the excellent biocompatibility of CTS, the good bioactivity of β-TCP, and the anti-inflammatory properties of MET. The MET-loaded β-TCP/CTS/SBA-15 scaffolds showed improved cell adhesion, appropriate porosity and good biocompatibility in vitro. This MET composite scaffold was implanted in the alveolar bone defects area of rats with periodontitis. After 12 weeks, Micro-CT and histological analysis were performed to evaluate different degrees of healing and mineralization. Results showed that the MET-loaded β-TCP/CTS/SBA-15 scaffolds promoted alveolar bone regeneration in a rat model of periodontitis. To our knowledge, this is the first report that MET-loaded β-TCP/CTS/SBA-15 scaffolds have a positive effect on alveolar bone regeneration in periodontitis. Our findings might provide a new and promising strategy for repairing alveolar bone defects under the condition of periodontitis.

Injectable Alginate/β-TCP Composite Hydrogel Incorporating P34HB/MgO+PEG Coaxial Electrospun Microfibers for Minimally Invasive Treatment of Osteonecrosis

Yu Hu Zhou,Zi Hao Zou,Jian Xiang Teng,Zhan Yu Wu,Si Wei Luo,Xu Ning,Chuan Ye,Long Yang,Wei Seong Toh

Avascular necrosis of the femoral head (ANFH) is a debilitating musculoskeletal disorder that is typically caused by impaired blood supply to the hip joint. In treating irregular bone defects that resulted from ANFH, injectable hydrogels are particularly attractive as they can be administered in a minimally invasive manner and conform to the variable shape of a bone defect. However, they often lack the biochemical and mechanical properties for effective bone repair. To address these issues, an injectable composite hydrogel, SA/β-TCP@PMP, composed of sodium alginate (SA) and β-tricalcium phosphate (β-TCP) crosslinked with glucono-delta-lactone (GDL), and enforced with varying concentrations of poly(3-hydroxybutyrate- co -4-hydroxybutyrate)/magnesium oxide and polyethylene glycol (P34HB/MgO+PEG; PMP) coaxial electrospun microfibers are formulated. By varying the concentrations (0%, 1%, 2%, 4%) of PMP microfibers, the physical properties of the composite hydrogel, including its injectability, surface morphology, swelling, degradation, and mechanical properties, as well as magnesium ions (Mg 2+ ) release can be finely tuned. Among all, SA/β-TCP@2%PMP composite hydrogel demonstrates superior angiogenic and osteogenic properties, and promotes robust new bone formation in a rabbit model of steroid-induced ANFH. Overall, this novel composite hydrogel offers significant application potential for minimally invasive treatment of irregular bone lesions in ANFH.

Bioactive ion engineered 3D printed TCP scaffolds promote vascularized bone regeneration in diabetic bone defects by restoring mitochondrial homeostasis

Yue He,Man Shu,Zhimin Li,Yun Liu,Ziyang Xu,Kelie Wang,Yu Zhang,Teliang Lu

Diabetes mellitus (DM)-associated hyperglycemia disrupts vascularized bone regeneration through mitochondrial dysfunction in repair-related cells and hindering scaffold-mediated bone defect repair. To overcome this challenge, we developed an innovative 3D-printed β-tricalcium phosphate scaffold co-doped with zinc and silicon ions (Zn/Si-TCP). In vitro evaluations revealed that Zn/Si-TCP significantly enhanced cellular viability of both endothelial cells and osteoblasts under hyperglycemic conditions, achieving this through dual mechanisms: (1) effective reactive oxygen species (ROS) scavenging (35 % reduction), and (2) restoration of mitochondrial homeostasis, as demonstrated by 1.5-fold upregulation of mitochondrial fusion protein Mfn1 and 0.6-fold downregulation of fission protein Drp1. The scaffold notably improved mitochondrial function, enhancing membrane potential by 1.4-fold and boosting ATP generation by 1.2-fold. These synergistic effects translated to superior biological performance, with 1.5-fold enhancement in angiogenic capacity (tube formation assay) and 1.9-fold increase in osteogenic activity (ALP assay). In vivo studies in a diabetic rat calvarial defect model demonstrated the remarkable efficacy of Zn/Si-TCP scaffolds, which achieved a 4.6-fold increase in vascular density and a 2.0-fold improvement in bone volume fraction (BV/TV) compared to undoped TCP controls at 8 weeks post-implantation, indicating successful vascularized bone reconstruction facilitated by Zn/Si-TCP scaffolds. This study establishes a new paradigm in diabetic bone repair by developing a dual-functional scaffold that coordinately promotes angiogenesis and osteogenesis through mitochondrial homeostasis regulation.

A novel ternary polymer/β-TCP composite microspheres with polydopamine-encapsulated nano zinc oxide coating for bone tissue engineering

Zhen Zhang,Geng Chen,Jun Wang,Xin Hu,Peijie Hou,Chengdong Xiong,Lifang Zhang

Addressing the prevalent clinical issue of bone defects, this study successfully developed a novel ternary copolymer material (PLCD) based on polylactic acid (PLA), polycaprolactone (PCL), and polydioxanone (PPDO). This ternary material combines the strength of PLA, the toughness of PCL, and the controllable rapid degradation properties of PPDO. making it a promising candidate for biomedical applications. By using an optimized emulsion method combined with finite element analysis, microspheres with a particle size range of 500–700 μm were prepared and subsequently fabricated into microsphere scaffolds using the solvent sintering method. To enhance osteoinductive and osseointegration capabilities, various proportions of β-tricalcium phosphate (β-TCP) were uniformly dispersed within the microspheres, creating composite scaffolds. Additionally, nano zinc oxide were coated on the microsphere scaffold surface using the in situ polymerization properties of dopamine, imparting antibacterial properties and enhancing functionality. Physical and biological evaluations of the microsphere scaffolds revealed excellent overall performance, including an interconnected porous structure, porosity and mechanical properties matching those of cancellous bone, suitable degradation behavior, and in vitro mineralization ability. Furthermore, scaffolds coated with polydopamine and loaded with nano zinc oxide exhibited strong initial antibacterial effects, and promoted osteogenic differentiation. These results indicate that the developed composite microsphere scaffolds have significant potential in bone tissue engineering applications, offering a promising solution for effective bone regeneration.

Fabrication and characterization of a bioactive composite scaffold based on polymeric collagen/gelatin/nano β-TCP for alveolar bone regeneration

Chao Yao,Prisana Pripatnanont,Junbiao Zhang,Srisurang Suttapreyasri

One strategy to correct alveolar bone defects is use of bioactive bone substitutes to maintain the structure of defect site and facilitate cells and vessels' ingrowth. This study aimed to fabricate and characterize the freeze-dried bone regeneration scaffolds composed of polymeric Type I collagen, nano Beta-tricalcium phosphate (β-TCP), and gelatin. The stable structures of scaffolds were obtained by thermal crosslinking and EDC/NHS ((1-ethyl-3-(3-dimethylaminopropyl) carbodiimide)/(N-hydroxysuccinimide)) chemical crosslinking processes. Subsequently, the physicochemical and biological properties of the scaffolds were characterized and assessed. The results indicated the bioactive composite scaffolds containing 10% and 20% (w/v) nano β-TCP exhibited suitable porosity (84.45 ± 25.43 nm, and 94.51 ± 14.69 nm respectively), a rapid swelling property (reaching the maximum swelling rate at 1 h), excellent degradation resistance (residual mass percentage of scaffolds higher than 80% on day 90 in PBS and Type I collagenase solution respectively), and sustained calcium release capabilities. Moreover, they displayed outstanding biological properties, including superior cell viability, cell adhesion, and cell proliferation. Additionally, the scaffolds containing 10% and 20% (w/v) nano β-TCP could promote the osteogenic differentiation of MC3T3-E1. Therefore, the bioactive composite scaffolds containing 10% and 20% (w/v) nano β-TCP could be further studied for being used to treat alveolar bone defects in vivo.

Development of Strong and Tough β-TCP/PCL Composite Scaffolds with Interconnected Porosity by Digital Light Processing and Partial Infiltration

Yanlong Wu,Ruomeng Chen,Xu Chen,Yongqiang Yang,Jian Qiao,Yaxiong Liu

Strong and tough β-TCP/PCL composite scaffolds with interconnected porosity were developed by combining digital light processing and vacuum infiltration. The composite scaffolds were comprised of pure β-TCP, β-TCP matrix composite and PCL matrix composite. The porous β-TCP/PCL composite scaffolds showed remarkable mechanical advantages compared with ceramic scaffolds with the same macroscopic pore structure (dense scaffolds). The composite scaffolds exhibited a significant increase in strain energy density and fracture energy density, though with similar compressive and flexural strengths. Moreover, the composite scaffolds had a much higher Weibull modulus and longer fatigue life than the dense scaffolds. It was revealed that the composite scaffolds with interconnected porosity possess comprehensive mechanical properties (high strength, excellent toughness, significant reliability and fatigue resistance), which suggests that they could replace the pure ceramic scaffolds for degradable bone substitutes, especially in complex stress environments. Keywords:porous composite scaffolds;mechanical properties;digital light processing;infiltration;gradient structure

Doping polyvinyl alcohol can improve the injectability of biological ceramics in 3D printing and influence the adhesion of cells to the scaffolds after sintering

ZhiDong Lin,XuLin Hu,LiLun Zhong,DeQiang Peng,JunLin Lu,Jian He,Xiaofei Shen,ChengDong Xiong,Tao Xu,Wei Niu

β tricalcium phosphate (β-TCP) is a common biological ceramic in bone tissue engineering due to its excellent biocompatibility and biodegradability. However, owing to its inherent properties of poor injectability and forming ability, the application of TCP in extrusion 3D printing is quite limited. To solve this problem, we innovatively printed gelatin and PVA (Polyvinyl alcohol) binary systems by taking advantage of the ability of gelatin to solidify rapidly at low temperatures and the ability of PVA to effectively improve phase separation during extrusion. Here, we fabricated a series of novel Gel-PVA-TCP scaffolds by incorporating different concentrations of PVA (5%, 10%, 15%) through 3D printing. Collectively, the addition of PVA made differences in three ways. First, compared to the PVA-free system, the novel printing system significantly improved the printability of TCP paste. The printing temperature was decreased to 35–45 °C by doping different concentrations of PVA. Next, the novel printing system maintained the excellent forming ability and printing precision of the scaffolds. Compared to the macropores of scaffolds without doping PVA (456.1 ± 11.2 μm), the macropores of Gel-PVA-TCP were 486.1 ± 26.5, 446.1 ± 15.2, and 443.7 ± 26.3 μm. Last, after sintering, the scaffolds with different concentrations of PVA exhibited similar compressive strength (3.82 ± 0.22, 3.34 ± 0.23, 3.74 ± 0.38, 3.47 ± 0.48 Mpa) and distinct micropores (4.44 ± 0.7, 1.54 ± 0.3, 2.59 ± 0.6, 3.59 ± 1.0 μm) on the surfaces of the scaffolds. Moreover, it was found that differences in the microstructure significantly facilitate the adhesion of bone marrow mesenchymal stem cells to scaffolds. Hence, the methods we provided hold a great promise for application in 3D printing techniques.

Solar-driven scalable hygroscopic gel for recycling water from passive plant transpiration and soil evaporation

Zou Hao,Yang Xinge,Zhu Jingling,Wang Fan,Zeng Ziya,Xiang Chengjie,Huang Danfeng,Li Jun,Wang Ruzhu

In the face of escalating global water scarcity, intensified by population growth and agricultural demands, there is an urgent need to improve water efficiency in greenhouse cultivation. The rapid development of atmospheric water harvesting technology offers us an opportunity to address this issue. Here a novel, solar-driven, scalable hygroscopic gel, termed TCP-Li, that demonstrates exceptional water uptake capacity (3.38 g water  g sorbent −1 ) and rapid solar thermal water release was reported. As a proof-of-concept application, we utilize this material in a transpiration and evaporation adsorption device (TEAD), which leverages the properties of TCP-Li to efficiently harvest water vapour from plant transpiration and soil evaporation. TEAD operates passively, absorbing moisture at high relative humidity during the night and releasing water for irrigation during the day using natural sunlight. Greenhouse experiments confirmed the efficacy of TEAD in ensuring normal plant growth while providing additional irrigation water (87.1 g per plant and 1,890.6 g m −2 ), thereby achieving an average water-saving effect of 44.9%. This study introduces a groundbreaking solution aimed at enhancing water efficiency in greenhouse cultivation, offering substantial potential for commercial and large-scale agricultural applications and significantly contributing to mitigating the global water crisis and optimizing water use in modern agriculture.

Dual-sized macro-micro porous biodegradable Zn-Mg-β-TCP scaffolds for bone defect repair: In vitro and in vivo evaluation

Kang Wang,Zexin Liu,Lin Liu,Tiantian Wang,Kun Chen,Runhua Yao,Ruiqiang Hang,Xiaohong Yao

Zinc-based materials have been extensively researched as biodegradable bone repair materials in recent years, but their low degradation rate and limited bioactivity need to be further improved. In this study, a new-type dual-sized macro-micro porous Zinc-Magnesium-β-tricalcium phosphate (DMMP Zn-Mg-β-TCP) scaffolds were designed and fabricated by vacuum heating-press sintering. The mechanical properties, degradation behaviors, in vitro biocompatibility, in vivo osteogenesis, and biological safety of the scaffolds were systematically evaluated. The results show that DMMP Zn-Mg-β-TCP has suitable mechanical properties, degradation properties, and excellent osteogenic properties. The compressive yield strength range of the scaffolds is 12.90–25.46 MPa, and the elastic modulus range is 0.31–0.44 GPa, meeting the mechanical properties required for implant materials in cancellous bone. In vitro degradation tests showed that the corrosion rate of the scaffolds ranges from 2.25 to 2.61 mm/y. Furthermore, the scaffolds exhibited excellent in vitro cell compatibility. After a 24-week animal experiment, the scaffold demonstrated excellent osteogenic capacity during bone healing, while maintaining biomechanical stability throughout its degradation process. In summary, the DMMP Zn-Mg-β-TCP has the potential to become a novel biodegradable bone implant material.

Biodegradation of CP/TCP by a constructed microbial consortium after comparative bacterial community analysis of long-term CP domesticated activated sludge

Xiaowen Sun,Ling Chen,Cheng Liu,Yin Xu,Wei Ma,Hong Ni

The objective of this study was to construct a microbial consortium for effective biodegradation of chlorpyrifos (CP) and its hydrolysis product, 3,5,6-Trichloro-2-pyridinol (TCP). A activated sludge from an organophosphorus pesticide factory was domesticated under long-term (20 weeks) CP stress, and the dynamic change in bacterial communities was analyzed by high-throughput sequencing. Then, a microbial consortium MC-BSPK was constructed of Bacillus sp. MC-B, Serratia sp. MC-S, Pseudomonas sp. MC-P, and Klebsiella sp. MC-K, which were significantly enriched during the domestication process. The biodegradation capacities of the microbial consortium MC-BSPK reached 100% for CP within 9 days and 88.61% for TCP within 15 days under the optimized degradation conditions (pH 8.0 and 31 °C). High-performance liquid chromatography (HPLC) revealed that CP could be degraded by the microbial consortium MC-BSPK into TCP, probably through hydrolysis of the P–O ester bond, and further degraded into other small molecules. A bioassay revealed that the virulence of CP toward Drosophila melanogaster W1118 was clearly reduced by the microbial consortium MC-BSPK biodegradation. Thus, the easily constructed microbial consortium MC-BSPK with high CP/TCP degradation capacities has the potential for application in pesticide-contaminated bioremediation.

Assessment of artificial bone materials with different structural pore sizes obtained from 3D printed polycaprolactone/β-tricalcium phosphate (3D PCL/β-TCP)

Zhao Qianjuan,Shan Rong,Liu Shengxi,Liu Xuanhao,Liu Bin,Song Fuxiang

Artificial bone is the alternative candidate for the bone defect treatment under the circumstance that there exits enormous challenge to remedy the bone defect caused by attributes like trauma and tumors. However, the impact of pore size discrepancy for regulating new bone generation is still ambiguous. Using direct 3D printing technology, customized 3D polycaprolactone/β-tricalcium phosphate (PCL/β-TCP) artificial bones with different structural pore sizes (1.8, 2.0, 2.3, 2.5, and 2.8 mm) were successfully prepared, abbreviated as the 3D PCL/β-TCP. 3D PCL/β-TCP exhibited a 3D porous structure morphology similar to natural bone and possessed outstanding mechanical properties. Computational fluid dynamics analysis indicated that as the structural pore size increased from 1.8 to 2.8 mm, both velocity difference (from 4.64 × 10−5 to 7.23 × 10−6 m s−1) and depressurization (from 7.17 × 10−2 to 2.25 × 10−2 Pa) decreased as the medium passed through. In vitro biomimetic mineralization experiments confirmed that 3D PCL/β-TCP artificial bones could induce calcium–phosphate complex generation within 4 weeks. Moreover, CCK-8 and Calcein AM live cell staining experiments demonstrated that 3D PCL/β-TCP artificial bones with different structural pore sizes exhibited advantageous cell compatibility, promoting MC3T3-E1 cell proliferation and adhesion. In vivo experiments in rats further indicated that 3D PCL/β-TCP artificial bones with different structural pore sizes promoted new bone formation, with the 2.5 mm group showing the most significant effect. In conclusion, 3D PCL/β-TCP artificial bone with different structural pore sizes could promote new bone formation and 2.5 mm group was the recommended for the bone defect repair.

Osteogenesis enhancement of silk fibroin/ α-TCP cement by N-acetyl cysteine through Wnt/β-catenin signaling pathway in vivo and vitro

Tao Feng,Junjie Niu,Bin Pi,Yingjie Lu,Jinning Wang,Wen Zhang,Bin Li,Huilin Yang,Xuesong Zhu

High brittleness and lack osteogenesis are two major limitations of calcium phosphate cement (CPC) in application in bone defect reconstruction. Here we prepared a composite calcium phosphate cement by mixing N-acetyl cysteine loaded silk fibroin solution with α-tricalcium phosphate. In vitro cytology experiment revealed that SF-NAC/α-TCP could significantly increase the activity of exocrine ALP and up-regulated expression of bone-related genes. However, NAC up-regulated gene expression could be significantly suppressed by DKK1. We propose that NAC functioning as osteogenic factor by activating the Wnt/β-catenin signaling pathway may be the possible mechanism of up-regulation of osteogenic genes. Bone regeneration in vivo shown in a rat femur defect was enhanced by the addition of NAC in SF/α-TCP. In addition, the combination intensity of cement-bone interface was improved. The combination SF-NAC/α-TCP might be developed into a promising tool for bone tissue repair in the clinic.
