Alzheimer's disease (AD) is the most common neurodegenerative disease that causes dementia in the elderly. Total tau in the blood is a potentially useful biomarker for predicting early AD. However, during detection, the non-specific adsorption of high-abundance biomolecules in the sample will block the detection signal and reduce the detection sensitivity. Based on this, this work described an electrochemical biosensor with high antifouling performance based on loach mucus-inspired guanosine-based hydrogel for the specific detection of tau protein. The exploited electrochemical biosensor shows high sensitivity and selectivity for tau protein detection, with a wide linear response range from 1.00 × 10 −2 to 1.00 × 10 2 ng/mL and a low limit of detection (LOD) of 1.31 × 10 −3 ng/mL. This LOD is lower than the critical tau protein concentration (0.2 ng/mL) that distinguishes AD cases, indicating that this simple tau protein immunoassay has the potential for clinical application. Furthermore, due to the facile fabrication, good biocompatibility , facile biofunctionalization, and antifouling property of this hydrogel film, hydrogel-based biosensors also have the potential to detect other biomarkers in complex biological samples.

Dipeptidyl peptidase-IV (DPP-IV) inhibitors play a critical role in the treatment of diabetes and metabolic diseases. This study combines computational simulations with experimental validation to identify peptides with potential DPP-IV inhibitory activity from wheat proteins. A peptide database was constructed through trypsin digestion simulation, and screening was performed using the ConPLex deep learning algorithm, leading to the identification of four promising peptides: TENEWK (Thr-Glu-Asn-Glu-Trp-Lys), NFVSER (Asn-Phe-Val-Ser-Glu-Arg), LDLPSK (Leu-Asp-Leu-Pro-Ser-Lys) and QHEQR (Gln-His-Glu-Gln-Arg). Experimental results showed that the IC50 values of these four peptides were 4.96 mM, 3.07 mM, 2.89 mM, and 4.18 mM, respectively, and they were all competitive DPP-IV inhibitors with significant inhibitory activity. Molecular dynamics (MD) simulations revealed the inhibitory mechanism by which inhibitory peptides led to the disappearance of the α-helix of the DPP-IV active center. Further tau-random accelerated molecular dynamics (tau-RaMD) simulations were used to calculate the binding residence time of each peptide, providing insights into their binding stability and key interacting residues. The combination of computational and experimental approaches significantly improved the accuracy and efficiency of screening peptides with DPP-IV inhibitory potential, providing a promising research foundation for developing peptide-enriched health foods.

Curcumin, a molecule of labile natural product, has multiplex pharmacological effects, including antioxidant, antimicrobial, and anticancer effects as well as anti-Alzheimer’s disease (AD) properties. However, the drawbacks of curcumin such as poor stability and bioavailability seriously limited its application in drug development. Numerous literature studies suggested the conversion of curcumin to metal complexes and explored the clearance of β-amyloid fibrils by curcumin conjugates. However, the research about the inhibition effect of curcumin complex on τ protein aggregation is progressing slowly, and its interactive orientation with the related proteins from the steric effect has been ignored. Inspired by the perspective on interactive orientation, we conjugated curcumin molecule on the positive-charged scaffold of Ru(II) complexes (bipyridine for Ru1, phenanthroline for Ru2) to construct an octahedral structure. After being conjugated with a ruthenium octahedral framework, curcumin exhibited a stronger ability to not only inhibit aggregation of R3, the τ peptide corresponding to the third repeat unit of the microtubule binding domain (residues 306–336 in full τ) (Ru1 with an IC50 of 0.5 μM and Ru2 with an IC50 of less than 3 μM) but also disaggregate R3 fibers with dose-dependence through an exothermic and dynamic process. Autodock simulation was applied to get a hairpin-docking mode in the inhibition of R3 aggregation. Herein, our results provide a rational strategy emphasizing the interactive orientation from an octahedral steric structure to design the anti-AD drugs with natural compound.

Alzheimer’s disease (AD) is a widely distributed neurodegenerative disease characterized clinically by cognitive deficits and pathologically by formation of amyloid-β (Aβ) plaque and neurofibrillary tangles (NFTs) in the brain. Vanadium is a biological trace element that has a function to mimic insulin for diabetes. Bis(ethylmaltolato) oxidovanadium (IV) (BEOV) has been reported to have a hypoglycemic property, but its effect on AD remains unclear. In this study, BEOV was supplemented at doses of 0.2 and 1.0 mmol/L to the AD model mice APPSwe/PS1dE9 for 3 months. The results showed that BEOV substantially ameliorated glucose metabolic disorder as well as synaptic and behavioral deficits of the AD mice. Further investigation revealed that BEOV significantly reduced Aβ generation by increasing the expression of peroxisome proliferator-activated receptor gamma and insulin-degrading enzyme and by decreasing β-secretase 1 in the hippocampus and cortex of AD mice. BEOV also reduced tau hyperphosphorylation by inhibiting protein tyrosine phosphatase-1B and regulating the pathway of insulin receptor/insulin receptor substrate-1/protein kinase B/glycogen synthase kinase 3 beta. Furthermore, BEOV could enhance autophagolysosomal fusion and restore autophagic flux to increase the clearance of Aβ deposits and phosphorylated tau in the brains of AD mice. Collectively, the present study provides solid data for revealing the function and mechanism of BEOV on AD pathology.

The advancement of Alzheimer’s disease (AD) is directly correlated to the phosphorylation damage of Tau protein (Tau-441), which is considered the most reliable indicator for early detection of AD. Nevertheless, the concentration of Tau-441 in human serum remains considerably low. To address this issue, a paper-based fluorescence immunosensor based on internal filter effect (IFE) was developed for the detection Tau-441. The microfluidic paper-based channel was prepared and modified with a gold layer to attach primary antibodies and increase the sensitivity of the sensor. All at once, Au@CeO 2 nanorods was employed as a fluorescence probe to hydrolyze p-nitrophenyl phosphate ( pNPP ), which generates hydrolyzed product p-nitrophenol (PNP) can reduce or quench the fluorescence intensity of N-doped graphene quantum dots (NGQDs). The immunosensor was designed to detect an increase in the amount of Tau-441 collected as the fluorescence intensity of NGQDs decreased. The developed sensor demonstrated satisfactory results in clinical serum samples, offering a promising concept for an immunoassay tagged with nanozymes for early detection of AD. Overall, this microfluidic paper-based fluorescent immunosensor has the potential to improve early diagnosis of AD.

Alzheimer's disease (AD) incidence is rising with no effective treatments. Eleutheroside B (EB) has neuroprotective potential, but its therapeutic effects and mechanisms in AD are unclear. This study investigated EB's effects on AD pathologies and elucidated its underlying mechanisms using a transgenic Caenorhabditis elegans ( C. elegans ) model. We found that EB could ameliorate cognitive impairment, locomotor dysfunction, and shortened lifespan induced by amyloid-β(Aβ) and Tau aggregation in C. elegans , and reduce the relative expression levels of Aβ, Tau, and phosphorylated Tau (p-Tau) proteins. EB significantly improved the health status and anti-aging capacity of the worms, as evidenced by prolonged healthspan, elevated pharyngeal pumping rate and body bend frequency, and increased body length and width. Notably, EB exhibited no observable toxicity even at high-dose administration. Furthermore, EB activated key antioxidant transcription factors skn-1 , daf-16 , and hsf-1 ; it also increased the expression levels of antioxidant enzymes superoxide dismutase-3(SOD-3) and glutathione S-transferase 4(GST-4), decreased reactive oxygen species (ROS) levels, and elevated the expression of heat shock proteins Hsp-4 and Hsp-6. EB also reduced the accumulation of p62/SQST-1 protein, promoted the colocalization of lgg-1::GFP with lysosomes, and increased the relative mRNA expression of autophagy-related genes aak-2 , unc-51 , bec-1 , vps-34 , and lgg-1 . After targeted knockdown of these genes, the protective effects of EB against AD-related pathologies were abolished. Our findings demonstrate that EB exhibits anti-aging properties and potential for ameliorating AD, which is likely mediated via antioxidative activities and autophagy induction.

Blood-based detection of Tau presents a promising strategy for early screening of Alzheimer's disease (AD). This study introduces a novel surface plasmon resonance imaging (SPRi) platform that employs an array of low-cost monochromatic LEDs as excitation light sources, effectively replacing conventional spectrometer-based systems to achieve cost reduction and enhanced operational stability. We developed an adaptive fitting algorithm that automatically identifies optimal spectral bands for analysis, enabling high-precision SPR measurements across an extended dynamic range. The system demonstrates exceptional performance with a RI resolution of 1.96 × 10 −6 RIU and a dynamic range of 0.0136 RIU. To further enhance sensitivity, we implemented a signal amplification strategy based on localized sample heating, which promotes molecular enrichment at both the center and periphery of evaporating droplets. This approach increases local target concentrations and enhances interaction probability with surface-immobilized capture probes. The label-free amplification method eliminates the requirement for secondary signal enhancement, substantially simplifying the detection workflow. Using this integrated approach, we achieved ultrasensitive Tau detection with a limit of detection (LOD) of 437 fg/mL (7.95 fM) and established a linear detection range spanning from 1 pg/mL to 10 ng/mL. This technique provides a robust platform for AD-related Tau quantification and demonstrates significant potential for facilitating early AD diagnosis.

The progression of Alzheimer’s disease (AD) is positively correlated with the phosphorylation damage of Tau-441 protein, which is the marker with the most potential for the early detection of AD. The low content of Tau-441 in human serum is a major difficulty for the realization of content detection. Herein, we prepared an electrochemical immunosensor modified with Poly(3,4-ethylene-dioxythiophene)-poly (styrene sulfonate) (PEDOT: PSS)/Carboxylated multi-walled carbon nanotube (MWCNTs-COOH) nanocomposites based on electrochemical immunoassay technology for the low-concentration detection of Tau-441. The immunosensor based on the nanocomposite can take advantage of the characteristics of conductive polymers to achieve electrical signal amplification and use MWCNTs-COOH to increase the contact area of the active site and bond with the Tau-441 antibodies on the electrode. The physicochemical and electrical properties of PEDOT: PSS/MWCNTs-COOH were studied by in situ characterization techniques and electrochemical characterization methods, indicating that the immunosensor has high selectivity and sensitivity to the Tau-441 immune reaction. Under optimized optimal conditions, the electrochemical immunosensor detected a range of concentrations of Tau-441 to obtain a low detection of limit (0.0074 ng mL−1) and demonstrated good detection performance through actual human serum sample testing experiments. Therefore, the study provides an effective reference value for the early diagnosis of AD.Keywords:PEDOT: PSS;MWCNTs-COOH;nanocomposites;electrochemical immunosensor;Tau-441

Glutamate (Glu) excitotoxicity and amyloid-β (Aβ) deposition are significant factors in the occurrence and development of Alzheimer’s disease (AD). Dauricine and daurisoline are two alkaloid components of Menispermum dauricum DC . that have a protective effect on the nervous system. The protection of dauricine and daurisoline on Glu-injured PC12 cells and the protection dauricine on Aβ 25-35 -injured PC12 cells were investigated in this study. The results of the study demonstrated that on PC12 cells damaged by Glu (20 mM), dauricine and daurisoline (3 and 10 μM) increased the cell viability, reduced cell apoptosis, and enhanced mitochondrial membrane potential (MMP) levels. Dauricine and daurisoline can also reduce the levels of intracellular ROS and free Ca 2+ , and suppression the expression of CaM, p-CaMKII, and p-Tau in Glu-damaged PC12 cells. In addition, on PC12 cells damaged by Aβ 25-35 (30 μM), dauricine (3 and 10 μM) can also significantly increase the cell viability and MMP levels, inhibit cell apoptosis, reduce intracellular ROS and free Ca 2+ levels, and down-regulate protein expression of CaM, p-CaMKII, and p-Tau. This study demonstrates for the first time that dauricine and daurisoline may inhibit the excessive phosphorylation of Tau protein and subsequent cell apoptosis by suppressing the Ca 2+ -CaM/CaMKII pathway, thereby protecting PC12 cells damaged Glu or Aβ 25-35 in vitro . Dauricine and daurisoline have the potential to treat AD effectively and have further research value.

Alzheimer's disease (AD) is characterized by extracellular amyloid plaques composed of β-amyloid (Aβ) and intracellular neurofibrillary tangles containing hyperphosphorylated tau protein. No effective therapy is available for this disease. In this study, we investigated the potential therapeutic effects of dauricine (DAU), a benzyl tetrahydroisoquinoline alkaloid, on AD, and found that DAU administration significantly improved cognitive impairments in 3xTg-AD mice by decreasing Aβ plaques and hyperphosphorylated tau and increasing the hippocampal ATP level. Proteomic and western blot analyses revealed that DAU treatment mainly modified the expression of proteins involved in mitochondrial energy metabolism, such as Aco2, Ndufs1, Cox5a, and SDHB, and that of synapse-related proteins such as Syn1 and Syn2. Pathway analysis revealed that DAU modulated the tricarboxylic acid cycle, synaptic vesicle cycle, glycolysis, and gluconeogenesis in 3xTg-AD mice. Our study suggests that DAU may be a potential drug for the treatment of AD.

A two-step electropolymerization strategy was constructed to establish an electrochemical sensor method for sensitive and selective detection of betamethasone. First, a poly-(2-aminoterephthalic acid) (P(ATA)) layer, which can significantly enhance electronic conductivity, was synthesized on the surface of the bare electrode using electropolymerization. Subsequently, taurine (Tau), which is rich in sulphonate groups, was electropolymerized and introduced into the P(ATA) layer to obtain P(Tau)/P(ATA)/GCE. Then, the electropolymerization parameters and test conditions were optimized, including the concentration of polymerization solution, the scanning rate, the polymerization cycles, buffer pH and accumulation time. Characterization of the obtained electrodes done using scanning electron microscopy and several electrochemical characterization methods proved the successful preparation of the P(Tau)/P(ATA)/GCE. Electrochemical tests of betamethasone at different concentrations (0.2-25μg·mL-1) under optimal measurement conditions exhibited excellent linearity, good sensitivity. Compared with commonly employed HPLC method, the proposed sensor method was much fast, simple, environmentally friendly, and cost-effective. Compared with other sensor methods, P(Tau)/P(ATA)/GCE sensor exhibited good sensitivity and excellent stability. Moreover, the P(Tau)/P(ATA)/GCE sensor was successfully applied to quantitatively detect betamethasone in cosmetics, which provides a new method for the monitoring of illegal addition of glucocorticoids in cosmetics.
