This study is aimed at exploring the release of bound polyphenols (BP) from insoluble dietary fiber (IDF) and its mechanism by solid-state fermentation (SSF) via Trichoderma viride. The results indicated that BP released by SSF (5.55 mg GAE/g DW) was significantly higher than by alkaline hydrolysis. In addition, 39 polyphenols and catabolites were detected, and the related biotransformation pathways were speculated. Quantitative analysis showed that SSF released more ferulic acid, p-coumaric acid, and organic acids, which led to advances in antioxidant, α-amylase, and α-glucosidase inhibitory activities. Furthermore, structural characteristics (scanning electron microscopy, X-ray diffraction, thermos gravimetric analysis, and Fourier transform infrared spectroscopy) and dynamic changes of carbohydrate-hydrolyzing enzymes indicated that the destruction of hemicellulose and the secretion of xylanase were vital for releasing BP. Overall, this study demonstrated that SSF was beneficial to release BP from IDF, which could provide insight into utilizing agricultural byproducts in a more natural and economical way.

The postharvest losses of litchi caused by litchi downy blight are considerably high. We identified a natural antifungal volatile pyrone, 6-pentyl-2H-pyran-2-one (6PP), synthesized by Trichoderma erinaceum LS019-2 and investigated as biocontrol for litchi downy blight and preservation. 6PP significantly inhibited the growth and sporangial germination of Peronophythora litchii, the causal agent of litchi downy blight, and caused severe cellular and intracellular destructions, as evidenced by electron microscopic analysis. Furthermore, in the treatment, the fruit kept better color, higher weight, and antioxidant activity, so it can maintain freshness and prolong shelf life. Metabolome analysis confirmed the decline of lipids and the accumulation of organic acids in litchi fruits in response to 6PP treatment. These effects from 6PP could alleviate disease effects and prolong the shelf life of litchi fruits. These findings suggested that 6PP could be a useful natural product to control downy blight disease and a new preservative of litchi fruits.

Navel orange peel is a potential source of soluble dietary fiber. In order to make full use of this resource and improve its physiological activities, the effects of single and mixed strains fermentation modifications were compared in this study. The structural, physicochemical, and functional properties of soluble dietary fiber fermented by Trichoderma viride and/or Aspergillus niger were investigated. The results showed that mixed fermentation resulted in significantly higher activity of complex cellulase than single fermentation. On the second day, when T. viride and A. niger were inoculated at a ratio of 3:1, the activities of carboxymethyl cellulase , β -glucosidase, and filter paper lyase were 1.54, 1.61, and 1.30 times those of single fermentation. Mixed fermentation increased the yield of soluble dietary fiber from 6.63% to 15.73%. Scanning electron microscopy and Fourier transform infrared spectroscopic analyses suggested that soluble dietary fiber produced by mixed fermentation had smaller particle size, lower crystallinity, and more disrupted structure. Moreover, the functional properties of soluble dietary fiber were remarkably improved. After mixed fermentation, the glucose and cholesterol adsorption amounts, and α -amylase inhibition activity increased by 1.51, 1.63, and 1.76 times, respectively. In summary, mixed fermentation was an efficient and environmentally friendly modification method. Modified SDF can be utilized in the food processing industry, expanding the range of by-product applications.

Streptomyces lydicus A01 and Trichoderma harzianum P1 are potential biocontrol agents of fungal diseases in plants. S. lydicus A01 produces natamycin to bind the ergosterol of the fungal cell membrane and inhibits the growth of Botrytis cinerea. T. harzianum P1, on the other hand, features high chitinase activity and decomposes the chitin in the cell wall of B. cinerea . To obtain the synergistic biocontrol effects of chitinase and natamycin on Botrytis cinerea , this study transformed the chit42 gene from T. harzianum P1 to S. lydicus A01. The conjugal transformant (CT) of S. lydicus A01 with the chit42 gene was detected using polymerase chain reaction (PCR). Associated chitinase activity and natamycin production were examined using the 3, 5-dinitrosalicylic acid (DNS) method and ultraviolet spectrophotometry, respectively. The S. lydicus A01- chit42 CT showed substantially higher chitinase activity and natamycin production than its wild type strain (WT). Consequently, the biocontrol effects of S. lydicus A01- chit42 CT on B. cinerea , including inhibition to spore germination and mycelial growth, were highly improved compared with those of the WT. Our research indicates that the biocontrol effect of Streptomyces can be highly improved by transforming the exogenous resistance gene, i.e. chit42 from Trichoderma , which not only enhances the production of antibiotics, but also provides a supplementary function by degrading the cell walls of the pathogens.

Dietary fiber (DF) is a carrier of abundant bound polyphenols (BP). The present work was to investigate the release characteristic and mechanism of BP from the insoluble dietary fiber of navel orange peel (NOP-IDF) by mixed solid-state fermentation (SSF) with Trichoderma reesei and Aspergillus niger . The optimized fermentation conditions were set as follows: inoculation ratio of T. reesei to A. niger 3:1, delayed inoculation time 28 h and initial pH 6.5. The compositions of BP were identified and quantified by UPLC-ESI-QTOF-MS/MS and HPLC-ESI-QQQ-MS/MS, then the biotransformation pathways were partially speculated. Moreover, cellulase, xylanase and β -glucosidase activities were strongly correlated with the BP content. The SEM showed the cell wall and the crystalline structure of the NOP-IDF was effectively decomposed. Furthermore, mixed SSF significantly enhanced in vitro antioxidant activities of BP. In summary, our study demonstrated that mixed SSF was a feasible method for the BP release.
