Preparation, in vitro and in vivo evaluation of phloretin-loaded TPGS/Pluronic F68 modified mixed micelles with enhanced bioavailability and anti-ageing activity

Jiaying Li,Tingyuan Li,Mingjie Gong,Xiaowen Wang,Qinyang Hua,Xia Jiang,Qilong Wang,Elmurat Toreniyazov,Jiangnan Yu,Xia Cao,Michael Adu-Frimpong,Ximing Xu

Phloretin exhibits strong antioxidant and anti-ageing properties by inhibiting mitochondrial oxidation of glutamate, succinic acid, and ascorbic acid. However, its clinical application is limited by poor aqueous solubility and low oral bioavailability. To enhance its bioavailability and efficacy, we incorporated phloretin into nano-micelles (phloretin-MM) using the thin film dispersion method. Characterisation revealed that the optimal formulation had TPGS and Pluronic F68 in a 4:1 ratio as the excipients, which resulted in spherical micelles with an average particle size of 33.28 nm and an encapsulation efficiency of 71.2 ± 0.48%. The in vitro release profile showed that the phloretin-MM showed significantly higher cumulative release rates than free phloretin across various pH conditions, while the pharmaceutical analysis in rats indicated that phloretin-MM significantly improved the oral bioavailability of phloretin (about 5 folds) in circulation. Additionally, through the analysis of the staining of zebrafish under light microscopy and the average gray value, it can be concluded that phloretin has anti-ageing drug effect, and phloretin-MM is better than free phloretin. These findings suggest that TPGS/Pluronic F68-modified phloretin-MM could serve as an excellent nano-drug carrier system, potentially enhancing the solubility, bioavailability, and anti-ageing effects of phloretin for broader clinical applications.

Anti-Ageing Effect of Physalis alkekengi Ethyl Acetate Layer on a d-galactose-Induced Mouse Model through the Reduction of Cellular Senescence and Oxidative Stress

Kaiyue Sun,Yingting Sun,Heyang Li,Dongyao Han,Yuting Bai,Rong Zhao,Zijiao Guo

We aimed to study the effects of an ethyl acetate fraction ofPhysalis alkekengi(PAE) ond-galactose (d-gal)-induced senescence and the underlying mechanism. Firstly, analysis of the phytochemical composition revealed total flavonoids, total phenolics, total saponins, rutin, and luteolin contents of 71.72 ± 2.99 mg rutin equivalents/g, 40.19 ± 0.47 mg gallic acid equivalents/g, 128.13 ± 1.04 mg oleanolic acid equivalents/g, 1.67 ± 0.07 mg/g and 1.61 ± 0.01 mg/g, respectively. The mice were treated withd-gal for six weeks, and from the fifth week, the mice were administered with PAE by gavage once a day for five weeks. We found significantd-gal-induced ageing-related changes, such as learning and memory impairment in novel object recognition and Y-maze, fatigue in weight-loaded forced swimming, reduced thymus coefficient, and histopathological injury of the liver, spleen, and hippocampus. The PAE effectively protected from such changes. Further evaluation showed that PAE decreased the senescence-associatedβ-galactosidase of the liver, spleen, and hippocampus, as well as the oxidative stress of the liver, plasma, and brain. The abundance of flavonoids, phenols, and saponins in PAE may have contributed to the above results. Overall, this study showed the potential application of PAE for the prevention or treatment of ageing-associated disorders. Keywords:Physalis alkekengi;learning and memory;exercise endurance;cellular senescence;oxidative stress

Roles of free radical on the formation of acetaldehyde in model wine solutions under different ultrasound parameters: A key bridge-link compound for red wine coloration during ageing

Zhen-Dan Xue,Qing-An Zhang,Hong-Rong Zheng

In order to explore the effects of ultrasound on the formation of acetaldehyde and its mechanism in model wine solutions, ultrasound conditions and free radicals were investigated by response surface methodology and electron paramagnetic resonance spectroscopy (EPR), respectively. The results indicate that ultrasound does induce the production of acetaldehyde with the maximum amount under the conditions of ultrasound power density 0.2 W/cm 2 , 48 min and 32 °C. The hydroxyl radicals and the 1-hydroxyethyl free radicals are the main initiator and precursor for acetaldehyde, respectively. Furthermore, the stronger the 1-hydroxyethyl free radicals captured by EPR, the lower the formation of acetaldehyde. In addition, the content of Fe 2+ and ethanol also exerted a certain influence on the acetaldehyde formation. In conclusion, ultrasound does promote the production of acetaldehyde in the model wine solutions, which is beneficial for well understanding the mechanism of ultrasound in modifying the wine color and accelerating ageing.

Construction of layered double hydroxide composites derived from tungsten tailing particles for simultaneously enhancing fire protection and anti-ageing properties of intumescent fire-resistant coatings applied in wood surface

Feiyue Wang,Hui Liu,Long Yan

Tungsten tailing is a great deal of solid waste produced in the production process of tungsten metal, for which there is no simple and effective treatment method in the world. The success synthesis of layered double hydroxide composites (TTF-LDH) by co-precipitation method from tungsten tailing is a feasible way to solve this problem. The structure and properties of TTF-LDH were confirmed by X-ray diffraction (XRD), scanning electron microscopy (SEM) and Fourier transform infrared spectroscopy (FTIR). The obtained TTF-LDH was in combination with intumescent flame retardants (IFR) to achieve a series of intumescent fire resistance coatings. The effects of TTF-LDH on the fire protection, smoke suppression and anti-ageing performances of coatings were evaluated by fire resistance test , smoke density test, cone calorimeter test and accelerated ageing test, while the flame-retardant mechanism of TTF-LDH in coatings was revealed. The results illustrate that an appropriate amount of TTF-LDH can remarkably enhance smoke suppression and fire-resistant abilities of coatings, among which the coating containing 3 wt % TTF-LDH (IFRC-LDH 3 ) exhibits the optimal synergistic effect , revealing a smoke density rating value of 17.3%, a total heat release of 7.0 MJ/m 2 and an equilibrium backside temperature of 146.2 °C at 900 s. Meanwhile, the addition of TTF-LDH can strengthen the char formation and thermal stability abilities of the coatings, and the residual weight of IFRC-LDH 3 rises to 35.7% at 800 °C, as supported by thermogravimetric (TG) analysis. The results of accelerated ageing test demonstrate that the presence of TTF-LDH can strengthen structural stability of coating, decrease migration and decomposition of IFR, thus giving the coating better integrity and durability of fire-resistant and smoke suppression properties.
