Anticancer effects of morin-7-sulphate sodium, a flavonoid derivative, in mouse melanoma cells

Hua-Wen Li,Tang-Bin Zou,Qing Jia,En-Qin Xia,Wen-Jun Cao,Wen Liu,Tai-Ping He,Qin Wang

Background Increasing evidence supports the anticancer effects of morin in vitro and in vivo . However, the role of morin-7-sulphate sodium (NaMoS), a water-soluble flavonoid derivative synthesized from morin remains unclear. The present study investigated the tumor suppression by NaMoS in mouse melanoma cells. Materials and methods We synthesized the flavonoid derivative morin-7-sulphate sodium according to the method described for quercetin-sulphate derivative, and further isolated, purified and identified the compound. Cell proliferation in vitro was assessed using a CCK-8 assay. The wound healing assay was performed to evaluate cell motility , and flow cytometry was used to detect cellular apoptosis . Protein levels of vimentin , matrix metalloproteinase 9 (MMP9), phosphorylation of Akt1/2/3 (p-Akt1/2/3), extracellular signal-regulated kinase 1/2 (p-ERK1/2) and Caspase3 in B16F10 cells were detected by immunohistochemistry and Western blot . Results The results suggest that cell proliferation was markedly decreased in NaMoS-treated groups (1, 10, 25, 50, 100, 500, 1000   μM) in a dose-dependent manner compared with the Control group and the IC 50 was 221.67   μM at 48   h. NaMoS at 200   μM concentration significantly inhibited the invasion and promoted apoptosis of B16F10 cells. Moreover, protein level of Caspase3 increased significantly in B16F10 cells treated by NaMoS. Immunohistochemistry and Western blot further confirmed that NaMoS decreased the expression of vimentin, MMP9, p-Akt1/2/3 and p-ERK1/2 in B16F10 cells. Conclusions This study provides robust evidence that NaMoS, a water-soluble flavonoid, manifests anticancer properties and may act as a signal transduction inhibitor in melanoma cells.

Highly efficient bioconversion of flavonoid glycosides from citrus-processing wastes in solvent-buffer systems

Yucong Zou,Xuan Xin,Haixia Xu,Hongwei Yuan,Xiaofeng Li,Yigang Yu,Guanglei Zhao

An efficient whole-cell biocatalytic method was developed for the cascade hydrolysis of flavonoid glycosides to their valuable hydrolyzates with high substrate conversions. Cells of Aspergillus niger showed both α-L-rhamnosidase and β-D-glucosidase activities, catalyzing the hydrolysis of hesperidin (HES) through two pathways in one pot: (1) α-L-rhamnosidase catalyzed HES hydrolysis to hesperetin-7-O-glucoside (HG) and subsequently β-D-glucosidase catalyzed HG hydrolysis to hesperetin, and (2) β-D-glucosidase catalyzed direct hydrolysis of HES to hesperetin. Both the HES conversion and product composition were controllable by adjusting the key reaction conditions and addition of different sugars. Remarkably, high conversions (above 90%) of HES were achieved by using green solvents in place of traditional organic solvents for dissolving polar substrates. The type and concentration of natural deep eutectic solvents (NADESs) in buffer solutions evidently influenced both the catalytic activity and selectivity of the cells. Inverted fluorescence microscopy and OD260/OD280 analysis suggested that NADESs can enhance the cellular membrane permeability, but high concentration of NADESs may damage cellular membranes and reduce the activity of enzymes. Furthermore, the whole-cell catalysts showed good scaled-up capability and stability. The whole cells also showed a relatively broad substrate spectrum towards various citrus flavonoid glycosides and their derivatives, with substrate conversions of 58.1%–98.2% and monoglycoside percentages of 73.3%–98.9%. This demonstration of whole-cell catalysis for one-pot cascade hydrolysis of flavonoid glycosides offers an eco-friendly, scalable and flexible platform method for the production of useful and valuable flavonoid monoglycosides and aglycones from inexpensive substances.

Design, Synthesis and Biological Evaluation of Ligustrazine-Flavonoid Derivatives as Potential Anti-Tumor Agents

Hui Wang,Wenxi Zhang,Yatao Cheng,Xinyu Zhang,Nannan Xue,Gaorong Wu,Meng Chen,Kang Fang,Wenbo Guo,Fei Zhou,Herong Cui,Tao Ma,Penglong Wang,Haimin Lei

In the clinic some anti-tumor drugs have shown damage to normal blood vessels, which could lead to vascular diseases. Therefore, it is necessary to evaluate the effects of anti-tumor drugs on normal blood vessels at the beginning of the drug design process. In this study, ligustrazine (TMP) and flavonoids were selected as raw materials. Sixteen novel TMP-flavonoid derivatives were designed and synthesized. Interestingly, compounds14and16were obtained by hydrolysis of a dihydroflavone to a chalcone under alkaline conditions. The cytotoxicity of the TMP-flavonoid derivatives was evaluated on five human tumor cell lines and one classical type of normal endothelial cell lines (HUVEC-12) by an MTT assay. Part of the derivatives showed better anti-tumor activities than the corresponding raw materials. Among them, compound14exhibited the closest activity to the positive control against the Bel-7402 cell line (IC50= 10.74 ± 1.12 μM; DDP IC50= 6.73 ± 0.37 μM) and had no toxicity on HUVEC-12 (IC50> 40 μM). Subsequently, fluorescence staining and flow cytometry analysis indicated that compound14could induce apoptosis of Bel-7402 cell lines. Moreover, the structure-activity relationships of these derivatives were briefly discussed.Keywords:TMP-flavonoid derivatives;anti-tumor;human tumor cell lines;HUVEC-12;fluorescence staining;flow cytometry

Abelmoschi Corolla non-flavonoid components altered the pharmacokinetic profile of its flavonoids in rat

Linling Lu,Dawei Qian,Jianming Guo,Yefei Qian,Boyi Xu,Mei Sha,Jinao Duan

Aim Abelmoschi Corolla is a well-known herbal medicine used for the treatment of chronic renal disease . Flavonoids are the major bioactive ingredients of Abelmoschi Corolla, but some non-flavonoid components also exist in this herb. In order to clarify the influences of non-flavonoid components on the pharmacokinetics profile of the flavonoid fraction from Abelmoschi Corolla (FFA), an investigation was carried out to compare the pharmacokinetic parameters of seven flavonoid components after administration of FFA and after administration of FFA combined with different non-flavonoid fractions. Materials and methods A selective and sensitive UPLC–MS/MS method was established to determine the plasma concentrations of the seven compounds. Sprague-Dawley rats were allocated to four groups which orally administered FFA, FFA combined with macromolecular fraction (FFA–MF), FFA combined with small molecule fraction (FFA–SF) and FFA combined with MF–SF (FFA–MF–SF) with approximately the same dose of FFA. At different time points, the concentration of rutin (1), hyperoside (2), isoquercitrin (3), hibifolin (4), myricetin (5), quercetin-3′- O -glucose (6), quercetin (7) in rat plasma were determined and main pharmacokinetic parameters including T 1/2 , T max , AUC and C max were calculated using the DAS 2.0 software package. The statistical analysis was performed using the Student's t -test with P <0.05 as the level of significance. Results Flavonoids almost had similar pharmacokinetics profile that were rapidly absorbed, reached the peak concentration at 30–60   min in group A, but the pharmacokinetic profiles and parameters of these flavonoids changed when co-administered with non-flavonoid components. It was found that AUC of five flavonoids but not hibifolin and quercetin in group FFA–SF and group FFA–MF–SF increased ( P <0.05) in comparison with group FFA while the tendency was not observed in group FFA–MF. Moreover, seven flavonoids had varying degrees of differences in the pharmacokinetics parameters such as C max , T max and T 1/2 ( P <0.05) in group FFA–MF, FFA–SF and FFA–MF–SF by comparison with group FFA. Conclusion These results indicate that non-flavonoid components could improve the bioavailability and delay the elimination of some flavonoids in rat.

Self-assembly of Hyaluronic Acid-Cu-Quercetin flavonoid nanoparticles: synergistic chemotherapy to target tumors

Hanxun Yue,Xuan Zhao,Qin Yong,Min Shi,Xiaofeng Jiang,Yating Zhang,Xian Yu

Background In this study, a natural compound quercetin (Qu) was investigated for its various antitumor effects. However, due to its poor water solubility and low bioavailability, its clinical application is limited. To overcome this constraint, a modification was to Qu, which resulted in the creation of novel flavonoid self-assembling nanoparticles (HCQ NPs). Methods HCQ NPs were synthesized by a self-assembly method and characterized using transmission electron microscopy, the Malvern Zetasizer instrument, X-ray photoelectron spectroscopy (XPS), the ultraviolet-visible spectrophotometric method (UV-vis), Fourier transform infrared (FITR) and inductively coupled plasma mass spectrometry. Extracellular, methylene blue spectrophotometric analysis was used to determine the ability of HCQ NPs to react with different concentrations of H 2 O 2 to form hydroxyl radicals ( • OH). Intracellular, DCFH-DA staining was used to detect the ability of HCQ NPs to react with H 2 O 2 to generate reactive oxygen species. Flow cytometry was used to detect the uptake of HCQ NPs by MDA-MB-231 cells at different time points. The biocompatibility of HCQ NPs was evaluated using the Cell Counting Kit-8 (CCK-8) assay. Calcein AM/PI double staining and the CCK-8 assay were used to evaluate the synergistic antitumor effect of HCQ NPs and H 2 O 2 . Results HCQ NPs showed uniformly sized analogous spherical shapes with a hydrodynamic diameter of 55.36 ± 0.27 nm. XPS revealed that Cu was mainly present as Cu 2+ in the HCQ NPs. UV−vis absorption spectrum of the characteristic peak of HCQ NPs was located at 296 nm. Similarly, FTIR spectroscopy revealed a complex formation of Qu and Cu 2+ that substantially changed the wavenumber of the 4-position C = O characteristic absorption peak. Based on the proportion of Qu and Cu 2+ (1:2), the total drug loading of Qu and Cu 2+ in the HCQ NPs for therapeutic purposes was calculated to be 9%. Methylene blue spectrophotometric analysis of • OH indicated that Cu can lead to the generation of • OH by triggering Fenton-like reactions. HCQ NPs rapidly accumulated in MDA-MB-231 cells with the extension of time, and the maximum accumulation concentration was reached at about 0.5 h. Calcein AM/PI double staining and CCK-8 revealed synergistic antitumor effects of HCQ NPs including the chemotherapeutic effect of Qu and chemodynamic therapy by Cu 2+ in a simulated tumor microenvironment. HCQ NPs demonstrated very low toxicity in LO2 cells in the biocompatibility experiment. Conclusion This study show cases a new method of creating self-assembled flavonoid HCQ NPs that show great for fighting cancer.

Integrated Functional Omics Analysis of Flavonoid-Related Metabolism in AtMYB12 Transcript Factor Overexpressed Tomato

Xuanlin Meng,Xingchen Zhao,Xiangyu Ding,Yang Li,Guodong Cao,Zhaohui Chu,Xiuli Su,Yuanchen Liu,Xiangfeng Chen,Jinggong Guo,Zongwei Cai,Xinhua Ding

Genetic engineering (GE) technology is widely used in plant modification. However, the results of modification may not exactly meet the expectations. Herein, we propose a new multi-omics method for GE plant evaluation based on the optimized use of the metID algorithm. Using this method, we found that flavonoid accumulation was at the expense of the great sacrifice of l-phenylalanine in GE tomatoes for the first time. Meanwhile, the ceramide series of sphingolipid is synthesized de novo from l-serine, and ceramides are the primary source of vesicles coated with flavonoids and secreted from the endoplasmic reticulum. Therefore, the accumulation of the ceramide series of sphingolipid changed the cell component of intracellular organelles. Furthermore, the improvement of the method allows us to identify more metabolites related to dysregulated pathways.

Studies on the Flavonoid Composition of Pleioblastus amarus Leaves and Shoots Based on Targeted Metabolomics

Yongmei Chen,Qingwen Wei,Jing Wang,Chuanyao Huang,Zhenghao Wang,Yuchun Liu

Background:Pleioblastus amarus(Keng) Keng f. is widely distributed in southern China and serves as both a medicinal and edible resource. Flavonoids represent the main bioactive compounds in this species, contributing to its antioxidant, anti-inflammatory, and anti-tumor activities. Methods:In this study, a targeted metabolomics approach based on liquid chromatography–tandem mass spectrometry (LC–MS/MS) was applied to analyze 35 flavonoid compounds in dry leaf and shoot samples ofP. amarus. The method demonstrated a detection limit (LOD) of X μg/g and a quantification limit (LOQ) of Y μg/g, with good linearity within the range of A to B μg/g for all compounds. Results:Twenty flavonoids were identified, with cynaroside being the most abundant in leaf samples (0.013 mg/g) and isovitexin being the most abundant in shoot samples (0.001 mg/g). The total flavonoid content in dry leaf (0.19 mg/g) was significantly higher than in dry shoot (0.022 mg/g). Multivariate statistical analyses revealed clear clustering and metabolic differences between the two tissues, and fifteen differential metabolites were identified by orthogonal partial least squares discriminant analysis (OPLS-DA). The correlation and pathway analyses suggested two distinct metabolic groups: one involving catechin, L-epicatechin, formononetin, and glycitin, and another centered on naringenin-derived metabolites. Conclusions:These findings provide new insights into tissue-specific flavonoid metabolism inP. amarus, highlighting distinct patterns in the leaves and shoots. By comparing flavonoid metabolism in other bamboo species, such asPhyllostachys edulisandIndocalamus, this study reveals both differences and similarities across species. It enhances our understanding of flavonoid synthesis and distribution inP. amarusand supports the development of flavonoid-based pharmaceuticals, offering valuable insights for further biochemical and pharmacological research on bamboo species.

Hydroxyl substitution patterns affect flavonoid–HSA binding: Mechanistic insights from multispectral spectroscopy and molecular simulations

Ziang Zhao,Xinyi Fang,Yanbing Gao,Qiaomei Sun

Flavonoids, diverse plant-derived compounds, show distinct binding behaviors with human serum albumin (HSA), influencing their pharmacokinetics. We examined three flavonoids—chrysin, baicalin, glycitein—using multispectral analysis and molecular simulations. Fluorescence (time-resolved, synchronous, steady-state), circular dichroism, dynamic light scattering and 3D fluorescence confirmed static quenching, ground-state complex formation, increased α-helix content, reduced protein size, and enhanced hydrophobic exposure. Thermodynamic results (ΔG < 0) showed spontaneous, Hydrogen bonds and van der Waals forces driven binding exhibits strong, moderate, and weak affinity, respectively(Kb ≈ 10 3 –10 5  M −1 ); chrysin had the strongest affinity due to optimal 5,7-dihydroxyl substitution, while glycitein's isoflavone structure decreased its affinity. Docking pinpointed Sudlow site III as the binding site. Molecular dynamics revealed increased helicity and decreased HSA flexibility, implying conformational tightening. These findings link flavonoid structural features—hydroxyl pattern and ring connectivity—to binding strength, site specificity, and HSA stability, offering mechanistic insight for designing flavonoid-based therapeutics and improving pharmacokinetic predictions.

Selenomethionine regulates flavonoid biosynthesis through the BroMYB34–BroCHSs module in kale (Brassica oleracea var. acephala)

Xin Fu,Yuxiang Liu,Lixia Wang,Xiaolong Li,Rui Shu,Tiantian Yao,Jian Jiao,Xiaoyan Zhao,Jingjuan Li,Yihui Zhang,Jianwei Gao,Fengde Wang

Kale is a super vegetable that is rich in a variety of nutrients and has antioxidant and anti-cancer properties. Selenomethionine (SeMet) is a type of organic selenium compound that enhances fruit quality; however, there is a lack of research on its application in kale. The study identified 10 mg·L−1 as the optimal SeMet concentration for kale, and selenium gradually accumulated after SeMet treatment, which significantly improved the antioxidant capacity of kale. Metabolome and transcriptome analysis of exogenous SeMet in kale revealed flavonoids as the main differential metabolites, and differential genes were most significantly enriched in flavonoid-related pathways. In addition, SeMet significantly induced the expression of MYB-type transcription factor BroMYB34. BroMYB34 binds to the BroCHSs promoter, a key gene for flavonoid synthesis, to activate its transcription. Transient overexpression of BroMYB34 could significantly increase the expression of BroT26.C02gCHS, BroT26.C03gCHS3, and BroT26. C09gCHS3 stimulates flavonoid synthesis to enhance the antioxidant capacity of kale. Overall, the findings of this study preliminarily reveal the molecular mechanism of SeMet promoting flavonoid biosynthesis of kale seedlings.

Improved flavonoid content in mulberry leaves by solid-state fermentation: Metabolic profile, activity, and mechanism

Qiannan Zhao,Xiaohai Yan,Yuan Yue,Tianli Yue,Yahong Yuan

This study was aimed at investigating the mechanisms of release and transformation of total flavonoids (TFs) from mulberry leaves (MLs) by solid-state fermentation (SSF) via Aspergillus cristatus . An intelligent model based on a back-propagation artificial neural network (BP–ANN) was established to predict the TFs in fungi co-fermented MLs. After prediction and experimental validation, the TFs released by SSF (72.55 mg rutin equivalents/g dry weight) were significantly higher than that of unfermented (24.42 mg rutin equivalents/g dry weight). In addition, the biotransformation mechanism of flavonoids during SSF was proposed by analyzing the untargeted metabolome , including the flavonoid and phenylpropanoid biosynthetic pathways . Pearson's correlation coefficient analysis indicated that the 29 up-regulated catabolites (especially loureirin D and arachidoside) after SSF contributed to increased ABTS (2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) radical scavenging activities and α-glucosidase inhibitory activities. Furthermore, structural characteristics (by scanning electron microscopy, thermogravimetric analysis , and FTIR spectroscopy) and analyses of dynamic changes in carbohydrate-hydrolyzing enzymes revealed that the destruction of hemicellulose was essential to releasing TFs by SFF. Industrial relevance In this study, a back-propagation artificial neural network (BP–ANN) was successfully used to establish a co-fermentation with Aspergillus cristatus for releasing and transforming flavonoids of mulberry leaves (MLs) through solid-state fermentation (SSF). The present study has immeasurable importance toward making maximum use of MLs. Moreover, the findings of this study also provide new insights into understanding the release and biotransformation mechanism of flavonoids from MLs by SSF.

Fermented orange peel mitigates biogenic amines in Huangjiu: Insights into flavonoid-histidine/tyrosine decarboxylase interactions

Xinjie Wang,Jinzhou Lu,Dan Yu,Shiyin Tao,Yang Zhang,Jiayou Li,Jianxing Yu

This study aims to evaluate the inhibitory effect and molecular mechanism of orange peel (OP) and its fermented product (FOP) by Rhizopus stolonifer JP1118 on the accumulation of biogenic amines (BAs) in Chinese rice wine (Huangjiu). UHPLC-Q-TOF-MS/MS technology was utilized to analyze the differences in metabolites between OP and FOP. Huangjiu fermentation experiments were conducted to compare the effects of different OP/FOP concentrations on BAs formation, and molecular docking was used to verify the interaction between flavonoid components and BAs synthesis-related decarboxylases. The results indicated that the content of 13 flavonoid compounds in FOP was more than double that in OP. The 4 g/L FOP treatment group demonstrated the most inhibition of BAs formation, and the inhibition on histamine was particularly significant. Molecular docking results confirmed that 11 flavonoids in FOP could stably bind to histidine decarboxylase (HDC), and 10 flavonoids could stably bind to tyrosine decarboxylase (TDC). They occupy the active sites of the enzymes via hydrogen bonding and hydrophobic interactions, thereby impeding substrate binding and inhibiting the synthesis of histamine and tyramine. This research offers a technical solution for low-amine Huangjiu, references for BAs control in other fermented foods and agricultural by-product recycling, with academic and industrial significance.

Isolation, purification, and bioactivity studies of flavonoid compounds from the fruiting bodies of Sanghuangporus vaninii

Zhang Lihong,Zhao Shuang,Lin Guo,Zheng Tian,Li Jingyi,Wang Qian,Liu Jingxin

In this study, flavonoids (SVF) extracted from Sanghuangporus vaninii were purified using AB-8 macroporous resin. After ultrasonication-assisted extraction, the crude flavonoid content was determined to be 149.15 mg/g; subsequent optimization of the SVF purification process resulted in an increase in the total flavonoid content up to 724.36 mg/g, corresponding to a 4.83-fold enhancement. Furthermore, Liquid Chromatography-Mass Spectrometry (LC-MS) analysis identified 25 flavonoid components, with isoquercitrin, catechin, and rutin as the major constituents. The purified flavonoids (PSVF) exhibited significantly enhanced antioxidant capacity and exerted potent inhibitory effects on α-glucosidase, α-amylase, and pancreatic lipase. Compared with crude SVF, PSVF also showed stronger antibacterial activity against Escherichia coli (E. coli ) and Staphylococcus aureus (S. aureus) . Additionally, in RAW264.7 macrophages, PSVF concentration-dependently suppressed the Lipopolysaccharide (LPS)-induced release of NO, TNF-α, IL-1β, and IL-6. Collectively, these findings indicate that AB-8 macroporous resin purification effectively improves both the purity and bioactivity of SVF, thereby supporting the potential application of PSVF in functional foods and pharmaceutical products.
