Wound healing is a complicated process that involves hemostasis, antibacterial defense, and tissue regeneration. Conventional treatment methods, such as surgical suturing, have inherent limitations, necessitating the exploration of new ones. Hydrogels can create a moist environment that facilitates wound healing, making them an ideal material for wound healing. In this study, a procoagulant polysaccharide mixture (carboxymethyl chitosan/sodium alginate/fucoidan; CAF) was designed. Hydrogels were prepared using CAF and an oxidized starch/tannic acid blend (OT) at different ratios. Through comprehensive evaluations, such as gelation time, swelling capacity, and antibacterial efficacy, an optimal hydrogel (COT) was identified. This COT hydrogel was formed by mixing 3% CAF and OT solutions at a ratio of 2:1 (v/v). The associated gelation process occurred rapidly within 13 s. COT hydrogel exhibited self-healing properties, and a high swelling rate (~3109 ± 74%). It also demonstrated high antibacterial activity, facilitating enhanced protection against infection. Additionally, COT hydrogel exhibited biocompatibility and biosafety. COT hydrogel demonstrated low cytotoxicity on mice fibroblast cells (L929) and good hemocompatibility in vitro. Moreover, in vivo evaluations revealed that it did not cause skin irritation or allergic reactions. Importantly, COT hydrogel significantly outperformed the commercially available hydrogel with its hemostatic and wound healing performance (p< 0.001,p< 0.01).

Fucoidan (GKP) alleviates OVX-induced bone loss by suppressing osteoclastogenesis. • GKP improves systemic metabolism and identifies betaine as a key metabolite effector. • Betaine attenuates osteoclastogenesis by regulating the OPG/RANK/RANKL signaling. Postmenopausal osteoporosis (PMOP), characterized by excessive bone resorption and impaired bone remodeling, remains a significant global health challenge. This study investigated the protective effects of fucoidan from giant kelp (GKP) against PMOP and its underlying mechanisms in ovariectomized mice. GKP supplementation attenuated bone loss by preserving trabecular microarchitecture, normalizing systemic mineral homeostasis, and suppressing osteoclastogenesis, as evidenced by the downregulation of osteoclast-related genes. 16S rRNA gene sequencing revealed that GKP reshaped the gut microbiota by decreasing the Bacteroidota/Firmicutes ratio and enriching beneficial taxa, which promoted short-chain fatty acid production. Targeted metabolomics revealed that GKP induced profound systemic metabolic reprogramming, particularly amino acid metabolism, and identified betaine as a key systemic metabolite effector upregulated following GKP treatment. In vitro assays demonstrated that betaine suppressed receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation and mitigated inflammation. In vivo validation demonstrated that betaine intervention recapitulated the osteoprotective phenotype observed in GKP-treated mice, which was associated with regulation of the osteoprotegerin/RANKL/RANK signaling axis. These results indicate that GKP exerts anti-osteoporotic effects in association with changes in gut microbiota and systemic metabolism, while betaine-mediated regulation of the OPG/RANKL/RANK signaling may contribute to its osteoprotective activity. BACKGROUND Chitosan (CTS) and chitosan oligosaccharides (COS) are widely used in the food industry due to their bioactive properties. However, their cationic nature enables interaction with whey protein isolate (WPI), affecting protein digestibility. Fucoidan (FUC), a naturally occurring anionic sulfated polysaccharide from brown algae, can form electrostatic complexes with CTS and COS to partially alleviate this effect; however, this process requires optimization. RESULTS Chitosan–fucoidan (CTS–FUC) and chitosan oligosaccharide–fucoidan (COS–FUC) complexes were successfully prepared and characterized using Fourier transform infrared (FTIR) spectroscopy, scanning electron microscopy, and other analytical instruments, confirming that complexation occurred via electrostatic interactions between amino groups of CTS and COS and the sulfate groups of FUC. Subsequently, the impact of these carbohydrates on WPI's digestibility was assessed during simulated gastrointestinal digestion. The CTS–FUC and COS–FUC complexes mitigated the inhibition of protein digestion caused by CTS and by COS alone. Specifically, the addition of the CTS–FUC complex increased the degree of hydrolysis of WPI by 24.6% compared with the corresponding mixed group, whereas the COS–FUC complex increased it by 22.6% compared with its corresponding mixed group. Furthermore, compared with the group containing CTS + FUC or COS + FUC, both preformed complexes increased protein concentration to 1.08-fold. The free trypsin concentration in the CTS–FUC + WPI group was 1.11 times that in the CTS + FUC + WPI group, whereas the concentration in the COS–FUC + WPI group was 1.06 times that in the COS + FUC + WPI group. Spectroscopic and chemical methods further revealed that the complexes reduced the binding of CTS and COS to protein and trypsin. CONCLUSION Overall, this study demonstrates that CTS–FUC and COS–FUC complexes effectively alleviated the negative impact of CTS and COS on protein digestion, offering new insights into mitigating their adverse nutritional effects in food systems. © 2026 Society of Chemical Industry.

TEMPO/NaClO/NaBr is applied to controlled degradation of fucoidan. • TEMPO/NaClO/NaBr enables efficient LMWF production from fucoidan in 0.5 h. • Galactose changes are consistent with possible C6 oxidation under TEMPO mediation • The TEMPO-free system caused 18.97% fucose loss and 6.13% sulfate loss. Low-molecular-weight fucoidan (LMWF) is highly desirable for food and pharmaceutical applications, yet conventional degradation methods often compromise its bioactive sulfate content. In this study, we employed a TEMPO/NaClO/NaBr system to degrade fucoidan under mild alkaline conditions and systematically investigated the degradation kinetics, structural changes, and anticoagulant activity of the resulting LMWF products. The degradation process was monitored by NaOH consumption and molecular weight distribution analysis. The optimal reaction time was determined to be 0.5 h, during which the molecular weight decreased from 337.4 kDa to 24.3 kDa, yielding LMWF with a relatively narrow molecular weight distribution (PDI = 1.08). Structural characterization by monosaccharide composition analysis, FT-IR, and 1 H NMR suggested that TEMPO-mediated oxidation favored oxidation at C6 primary hydroxyl groups, as indicated by changes in galactose and galacturonic acid contents, while no significant loss of total sulfate content was observed. In contrast, the non-selective NaClO/NaBr system caused substantial losses of fucose (18.97%) and total sulfate content (6.13%). Notably, the TEMPO-derived LMWF retained potent anticoagulant activity in the APTT assay while exerting a minimal effect on the PT pathway, suggesting a potentially lower bleeding risk. This study provides a promising strategy for the controlled preparation of LMWF with minimal loss of total sulfate content and a potentially favorable safety profile. Fucoidan-coated pH sensitive liposomes were designed for targeted delivery of gemcitabine (FU-GEM PSL) to treat pancreatic cancer (PC). FU-GEM PSL had a particle size of 175.3 ± 4.9 nm, zeta potential of −19.0 ± 3.7 mV, encapsulation efficiency (EE) of 74.05 ± 0.17 %, and drug loading (DL) of 21.27 ± 0.05 %. Cell experiments in vitro showed that FU-GEM PSL could increase the release of GEM and drug concentration, and could inhibit tumor cell proliferation by affecting the cell cycle. FU-GEM PSL entered cells through macropinocytosis and caveolin-mediated endocytosis to exert effects. Meanwhile, the expression of P-selectin was detected in human tissues, demonstrating the feasibility of targeting FU. Moreover, combined with animal experiments in vivo, FU-GEM PSL could inhibit the development of PC. Furthermore, anti-tumor experiments in vivo carried on BALB/c mice indicated that FU-GEM PSL had tumor suppression abilities and safety. Therefore, FU-GEM PSL is a promising formulation for PC therapy.

Cyclophosphamide (cy) is a widely used cancer drug. Many researchers have focused on the prevention and alleviation of its side effects, particularly damage to the intestinal mucosal barrier. In this study, we examined the effects of fucoidan, isolated from Acaudina molpadioides, on mice with intestinal mucosal damage induced by cyclophosphamide. Our results showed that fucoidan intervention could relieve injury such as decreasing inflammation and increasing the expression of tight junction proteins, and 50 kDa fucoidan significantly increased the abundance of short chain fatty acid (SCFA) producer Coprococcus, Rikenella, and Butyricicoccus (p < 0.05, p < 0.001, and p < 0.05, respectively) species within the intestinal mucosa compared with the cyclophosphamide group, as determined by 16S rDNA gene high-throughput sequencing. In addition, SCFAs, particularly propionate, butyrate, and total SCFAs, were increased in the feces, and SCFA receptors were upregulated in the small intestine. The protective effects of fucoidan on cyclophosphamide treatment may be associated with gut microflora, and 50 kDa fucoidan had superior effects. Therefore, fucoidan may have applications as an effective supplement to protect against intestinal mucosal barrier damage during chemotherapy.

Mesoporous silica nanoparticles (MSN) are effective drug delivery carriers because of their adjustable large pore size and high porosity. In this study, complex nanoparticles containing disulfide bonds (SS) were designed and prepared as curcumin (Cur) carriers by using fucoidan (FUC) and MSN as the polymer matrix. The product was characterized using scanning electron microscopy, transmission electron microscopy, dynamic light scattering, Fourier-transform infrared spectroscopy, and an N 2 adsorption and desorption test. When the mass ratio of MSN to FUC was 2:1, the nanospheres particle size was the smallest (295.6 ± 0.98 nm, −35.2 ± 0.8 mV). Furthermore, the curcumin encapsulation rate by MSN-Cur-SS-FUC was over 90%, and the cumulative release rate in 24 h was over 80% due to the combined effect of weak acidity and high glutathione concentration in the tumor site microenvironment. When the Cur concentration was 50 μg/mL, the cell viability of free Cur was 63.8%, the cell viability of MSN-Cur-SS-FUC was 14.5%, and the cell viability of MSN-SS-FUC at the same concentration remained above 74.6%. MSN-SS-FUC composite nanoparticles showed a good delivery of Cur, a lipid-soluble active compound, and provides a new delivery route for other lipid-soluble and poorly bioavailable active compounds.

Background: Humans with hypertensive heart disease are more likely to experience heart failure, arrhythmia, myocardial infarction, and sudden death, and it is crucial to treat this condition. Fucoidan (FO) is a natural substance derived from marine algae that has antioxidant and immunomodulatory activities. FO has also been shown to regulate apoptosis. However, whether FO can protect against cardiac hypertrophy is unknown. Methods: We investigated the effect of FO in hypertrophic models in vivo and in vitro. C57BL/6 mice were given an oral gavage of FO (300 mg/kg/day) or PBS (internal control) the day before surgery, followed by a 14-day infusion of Ang II or saline. AC-16 cells were treated with si-USP22 for 4 h and then treated with Ang II (100 nM) for 24 h. Systolic blood pressure (SBP) was recorded, echocardiography was used to assess cardiac function, and pathological changes in heart tissues were assessed by histological staining. Apoptosis levels were detected by TUNEL assays. The mRNA level of genes was assessed by qPCR. Protein expression was detected by immunoblotting. Results: Our data showed that USP22 expression was lowered in Ang II-infused animals and cells, which could promote cardiac dysfunction and remodeling. However, treatment with FO significantly upregulated the expression of USP22 and reduced the incidence of cardiac hypertrophy, fibrosis, inflammation, and oxidative responses. Additionally, FO treatment lowered p53 expression and apoptosis while increasing Sirt 1 and Bcl-2 expression. Conclusion: By reducing the level of Ang II-induced apoptosis through the regulation of USP22/Sirt 1 expression, FO treatment might improve cardiac function. According to this study, FO might be potential targeted approach for treating heart failure.

Wound healing is a largely unmet medical issue in trauma, burn, and diabetes. In this study, a pullulan-based and nanoparticle-loaded smart microneedle patch is designed to release drugs differentially based on the needs of wound healing. Chitosan and fucoidan are first used to prepare moxifloxacin (MOX)-loaded nanoparticles (MOXNPs) with a diameter of 258.0 ± 10.86 nm, PDI 0.19 ± 0.06, and surface charge 45.1 ± 3.9 mV. MOXNPs, lidocaine (LH), and thrombin (TH) are then incorporated to a 30 % ( w /w) pullulan-based microneedle patch (TH + LH + [email protected] ). TH + LH + [email protected] possesses uniform and cone-shaped microneedles with a length of 725 μm, demonstrating good biocompatibility, sufficient strength for skin penetration, fast skin dissolution within 55 ± 5 min, rapid release of TH and LH within 1 h, and sustained release of MOX for 24 h. TH + LH + [email protected] heals mice skin wounds completely within 7 days and restores collagen deposition with accelerated cell proliferation , granulation, and reduced pro-inflammatory cytokines. In conclusion, this study utilizes combined polysaccharides to develop a smart multifunctional microneedle platform that achieves rapid hemostasis/analgesia and sustained bactericidal action. The smart and combined therapy is a potential strategy for high-quality wound healing.

Due to the malignant proliferation and severe tumor fibrosis of pancreatic cancer (PC), the efficacy of chemotherapy has been largely limited. Peptidyl-prolyl cis -trans isomerase NIMA-interacting 1 (Pin1) promotes tumor progression by promoting collagen synthesis and tumor cell proliferation . In this study, we developed nano-liposomes (FU-GEM/Pin1 siRNA@LIP) that act simultaneously on pancreatic tumor cells. FU-GEM/Pin1 siRNA@LIP were designed with suitable particle size and potential, and effectively loaded with Pin1 siRNA and fucoidan (FU) while demonstrating good release stability and biocompatibility . Our study showed that FU-GEM/Pin1 siRNA@LIP effectively inhibited the proliferation of Panc01 and Panc02 tumor cells and significantly enhanced the uptake of these two types of cells in vitro . FU-GEM/Pin1 siRNA@LIP might cause tumor damage, promote apoptosis and reduce collagen formation in vivo . Molecular and histological studies demonstrated that FU-GEM/Pin1 siRNA@LIP regulated the expression of CDH11 and α-SMA, and also reducing collagen synthesis of peripheral matrix. In summary, our experiments highlight that FU-GEM/Pin1 siRNA@LIP is an effective combination delivery system. It can inhibit tumor cell proliferation in vitro , promote tumor cell apoptosis and reduce collagen synthesis in vivo . This system could provide a promising therapeutic option for PC.

Parkinson's disease is a neurodegenerative disease that is characterized by the loss of dopaminergic neurons. Fucoidan, which has emerged as a neuroprotective agent, is a marine-origin sulfated polysaccharide enriched in brown algae and sea cucumbers. However, variations in structural characteristics exist among fucoidans derived from different sources, resulting in a wide spectrum of biological effects. It is urgent to find the fucoidan with the strongest neuroprotective effect, and the mechanism needs to be further explored. We isolated and purified four different fucoidan species with different chemical structures and found that Type II fucoidan from Fucus vesiculosus (FvF) significantly improved mitochondrial dysfunction, prevented neuronal apoptosis, reduced dopaminergic neuron loss, and improved motor deficits in an 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced PD mouse model. Further mechanistic investigation revealed that the ATP5F1a protein is a key target responsible for alleviating mitochondrial dysfunction of FvF to exert neuroprotective effects. This study highlights the favorable properties of FvF for neuroprotection, making FvF a promising candidate for the treatment of PD.

Acute wound healing still faces numerous challenges, such as excessive inflammatory response, prolonged hemostasis time, and insufficient tissue regeneration, which often lead to delayed wound healing and scar formation. To address these issues, this study developed a multifunctional “physical-ionic” dual-network hydrogel (PPF hydrogel). This hydrogel combines an elastic polyvinyl alcohol (PVA) scaffold with a rigid ionic network composed of γ-polyglutamic acid (γ-PGA) and fucoidan, cross-linked via Ca 2+ . The resulting hydrogel possesses a highly interconnected porous structure, exhibiting excellent mechanical strength and moderate tissue adhesion. In vitro and in vivo evaluations showed that the PPF hydrogel has good blood and cell compatibility, rapidly achieving hemostasis and promoting the proliferation and migration of L929 fibroblasts. In a mouse full-thickness skin resection model, the PPF significantly accelerated wound closure while reducing epidermal hyperplasia and promoting orderly collagen deposition. Mechanistically, the PPF effectively modulates the immune microenvironment by downregulating pro-inflammatory cytokines (TNF-α, IL-6) and upregulating anti-inflammatory factors (IL-10) and VEGF. This regulation promotes M2 macrophage polarization and angiogenesis, thereby accelerating the transition from the inflammatory phase to the remodeling phase. This dual-network hydrogel offers a promising therapeutic strategy for high-quality acute wound repair in regenerative medicine.
