Beneficial effects of biomimetic nano-sized hydroxyapatite/antibiotic gentamicin enriched chitosan–glycerophosphate hydrogel on the performance of injectable polymethylmethacrylate

Yue Sa,Man Wang,Hongbing Deng,Yining Wang,Tao Jiang

Injectable polymethylmethacrylate (PMMA) bone cement is a widely used bone substitute in cemented arthroplasty, vertebroplasty and osteoporosis fractures. However, due to the inappropriate stiffness, poor bioactivity and high polymerization temperature of PMMA, aseptic loosening of the implanted cement at the bone–cement interface still could be observed in a high rate of patients. To improve the performance of PMMA, artificial extracellular matrices like chitosan–glycerophosphate (CS–GP) thermosensitive hydrogel was introduced into PMMA acting as a pore forming agent and osteoconductive nano-sized hydroxyapatite (nano-HA)/antibiotic gentamicin (GM) as a carrier. It is shown that CS–GP thermosensitive hydrogel can effectively create open pores at the surface of the PMMA cement, which is believed to facilitate bone tissue ingrowth and improve the cement anchorage at the bone–cement interface in future clinical applications. Meanwhile, such a hydrogel effectively decreases the maximum polymerization temperature to below 30 °C, prolongs the working time to more than 720 s and produces cement with a proper modulus of elasticity and a compressive yield strength ranging from 402 to 584 MPa and from 3.1 to 5.9 MPa, respectively. Furthermore, the incorporated nano-HA particles sufficiently increase the mineralization capacity of the cement without compromising its mechanical properties and the incorporated GM remarkably enhances the anti-bacterial activity of the cement. More importantly, nano-HA and GM enriched CS–GP thermosensitive hydrogel effectively improve the overall performance of PMMA cement without influencing the cell survival, suggesting the injectable p-PMMA/CS–GP/nano-HA/GM cement will hold strong promise for future bone reconstruction applications.

Preparation and performance of porous hydroxyapatite/poly(lactic-co-glycolic acid) drug-loaded microsphere scaffolds for gentamicin sulfate delivery

Cao Xing,Dai Lingjun,Sun Shichang,Ma Rui,Liu Xiangli

The prevention/treatment of osteomyelitis infection by combining the local antibiotic delivery system with the bone regeneration scaffold can effectively overcome the drawbacks of systemic antibiotic administration and realize the full cycle control of drug release. Herein, by introducing sodium carboxymethyl cellulose (SCMC) cross-linking agent to improve the binding force of poly(lactic-co-glycolic acid) (PLGA) microspheres and porous hydroxyapatite (HAp) bone scaffold. The porous HAp/PLGA drug-loaded microsphere bone scaffold for gentamicin sulfate (GS) delivery was successfully prepared. The optimal preparation parameters, drug release characteristics, SCMC enhancement mechanism, antibacterial properties and bone cell activity of porous HAp/PLGA drug-loaded microsphere bone scaffolds were comprehensively investigated. The results showed that the 0.1% SCMC-modified porous HAp/PLGA drug-loaded microsphere bone scaffold has a cumulative drug release of 45.0 ± 0.90% on the first day, which is about 20% lower than that of pure PLGA drug-loaded microspheres. Moreover, its drug release can be sustained and stably released for more than 17 d, which is attributed to the enhancement of the binding force between the microspheres and HAp by SCMC (combination for more than 3 weeks). Meanwhile, the diameter of the antibacterial ring expanded from the initial 10 ± 0.50 to 28 ± 1.2 mm after 14 d, which also indicated the sustained and stable release of GS. Alamar Blue analysis results showed that 0.1% SCMC-modified composite bone scaffold is beneficial to the proliferation activity of bone cells, and its 14 day activity increased by 20%. The above results indicate that the SCMC-modified composite bone scaffold has the potential to treat/prevent osteomyelitis.

Synergistically enhancing the antibacterial and antibiofilm activities of anion exchange membrane by chemically assembling gentamicin and N-chloramine layers

Yuyang Yao,Yan Zhang,Yundi Dong,Keqin Ruan,Yifei Shao,Geting Xu,Junbin Liao,Zhenlu Shen,Jiangnan Shen

Biofouling is one of significant obstacles decaying the high separation performance and long-term stability of polymer membranes during the application. To explore the desirable anti-biofouling anion exchange membrane (AEM), in this work, a series of modified AEMs having surface antibacterial functional layers on surface has been prepared by using gentamicin and N-chloramine for electrodialysis (ED). The antibacterial functional layer on surface was constructed via chemical assembly. By using CuSO 4 and H 2 O 2 as the trigger, l -dopa was oxidized on surface of commercial AEM (Fuji type II) to form a carboxyl layer. Thereafter, gentamicin was chemically assembled via the amide condensation reaction . It was chlorinated with diluted NaClO solution to endow it with synergistic antibacterial activity by using gentamicin. Investigation demonstrates that the optimized membrane of P( l -Dopa)@GM-Cl has high antibacterial activity against Staphylococcus aureus (S. aureus) and Escherichia coli (E. coli). Among them, flow cytometry shows that the antibacterial rate of P( l -Dopa)@GM-Cl against E. coli is more than 99%, which is much higher than original membrane (28.4%). P( l -Dopa)@GM-Cl exhibits the enhanced performance in inhibiting biofilm observed by scanning electron microscopy analysis . This work may provide a reference for the construction of anti-biofouling ion exchange membrane.

Production of high affinity monoclonal antibody and development of indirect competitive chemiluminescence enzyme immunoassay for gentamicin residue in animal tissues

Peng Dai,Yan Zhang,Yanping Hong,Jianhua Xiong,Huaying Du,Luying Duan,Dan Wang,Yubo Wang,Weijie Deng,Bruce D. Hammock,Wuying Yang

To determine gentamicin residues in animal tissues, a monoclonal antibody (Mab) was produced and a sensitive indirect competitive chemiluminescent enzyme immunoassay (icCLEIA) was developed. At first, gentamicin was conjugated with bovine serum albumin as immunogens which were used to immunize BALB/c mice. Then, an anti-gentamicin Mab was prepared by hybridoma technology. Finally, a sensitive icCLEIA was developed with an 50% inhibition concentration (IC 50 ) of 0.067 ng/mL for gentamicin. The limit of detection of the icCLEIA was 0.002 ng/mL. The cross reactivity of the Mab with structural analogues were<0.01%. The recoveries of gentamicin ranged from 80 to 101% and coefficient of variation was <6.4% in pork and fish samples. Samples were detected by UPLC-MS/MS for evaluating reliability of the icCLEIA. The results suggested that the prepared anti-gentamicin Mab can be used for rapid and convenient immunoassays to detect gentamicin residues in animal tissues.

Cucurbit[8]uril-mediated fluorescent indicator displacement: a novel “turn-on” fluorescent probe for gentamicin detection in water and food samples

Qinghong Bai,Yu Xia,Yukun Kang,Ying Yang,Yahui Yang,Li Zeng,Chun Liu,Xin Xiao

Gentamicin (GEN), an aminoglycoside antibiotic, is widely used in treating infections within the livestock industry but readily causes food residues and environmental pollution. This study developed a novel water-soluble supramolecular fluorescent probe DBTPy 2+ @Q[8], through host–guest interactions between the methylpyridine-modified benzothiophene cationic fluorophore (DBTPy 2+ ) and cucurbit[8]uril (Q[8]). This enabled specific GEN recognition and detection via the fluorescence indicator displacement (FID) mechanism. The DBTPy 2+ -Q[8] assembly induced significant fluorescence quenching, while GEN addition rapidly restored fluorescence. The restored intensity exhibited a good linear relationship with GEN concentration in the range of 0.1–10 μM (R 2  = 0.996), with a limit of detection (LOD) of 0.206 μM. The probe exhibited good selectivity and anti-interference capability, achieving 92.0%–104.1% spiked recoveries in water and food samples, and enabled specific GEN fluorescence imaging in living cells. This work establishes a novel supramolecular host–guest strategy for fluorescent probes enabling rapid, quantitative, specific GEN detection.

Fabrication of cost-effective antifouling sensors with conjugation of hyaluronic acid and phase-transited BSA for the rapid detection of gentamicin

Yinqiang Xia,Jin Zhao,Zheying Gao,Yifei Ruan,Siqi Wang,Liangbin Hu,Tianli Yue

Matrix interference poses persistent and significant challenges against rapidly detecting gentamicin residue in foods. Here, we fabricated synergistic antifouling surfaces using the phase transition of bovine serum albumin (PTB) and hyaluronic acid (HA) on substrates. The hybrid PTB + HA coatings showed ultralow adsorption of BSA and lysozyme, requiring 10 times less PTB dosage. With PTB's assistance, the modification of HA proved to be much more cost-effective than the sulfhydrated HA. The adsorption of E. coli on the hybrid coatings could be reduced by 92.6 %, illustrating good properties of antibacterial adsorption. Furthermore, the hybrid surfaces demonstrated good stability despite being rinsed with strong acid and alkaline solutions. Based on the immune-inhibition assay, the sensitive antifouling sensors were developed with a low detection limit (1.8 ng/mL for gentamicin) and satisfactory recovery rate (94.0 %–104.0 %) in goat milk. All tests could be completed within 10 min, confirming the sensor system's feasibility and efficiency.

UiO-66-NH2-Deposited Gold Nanoparticles Enable Enhanced Interference-Resistant Immunochromatographic Assay for Rapid Detection of Gentamicin in Animal-Derived Foods

Yimeng Pang,Zehao Yang,Xiaohua Liu,Xing Shen,Hongtao Lei,Xiangmei Li

Gentamicin (GEN) is a broad-spectrum antibiotic widely used in livestock production, and its excessive residues in animal-derived foods pose potential health risks to consumers. However, conventional colloidal gold immunochromatographic assays (AuNPs-ICA) often suffer from low sensitivity and poor tolerance to sample matrices. Herein, a UiO-66-NH2framework decorated with gold nanoparticle (UiO-66-NH2@Au)-based ICA was employed to construct an ICA platform for GEN detection, combining the optical advantages of AuNPs with the protective and stable octahedral framework of the Metal-organic framework (MOF) to enhance antibody stability under extreme conditions. The method achieved limits of detection for GEN of 0.1 µg/kg in four tested matrices, with recoveries of 80.1–112.0% and coefficients of variation below 11.7%. Compared to traditional AuNPs-ICA, the sensitivity was improved by up to 30-fold, the pH tolerance range was expanded from 6–8 to 4–10, and the organic solvent tolerance to organic solvents expanded up to 40%. Verification with 40 real samples demonstrated excellent correlation (R2> 0.99) with results from commercial ELISA kits. This UiO-66-NH2@Au-ICA platform offers a new technical solution with high sensitivity, strong good anti-interference performance, and robustness for rapid field detection of GEN residues in products of animal origin and holds significant practical importance for advancing rapid food safety detection technologies.

Dual-Responsive Nanogels with Cascaded Gentamicin Release and Lysosomal Escape to Combat Intracellular Small Colony Variants for Peritonitis and Sepsis Therapies

Shuang Xie,Yu Li,Wenxiong Cao,Jiawen Peng,Kun Huang,Jie Meng,Xiaohong Li

Intracellular bacteria are the major cause of serious infections including sepsis and peritonitis, but face great challenges in fighting against the stubborn intracellular small colony variants (SCVs). Herein, the authors have developed nanogels (NGs) to destroy both planktonic bacteria and SCVs and eliminate excessive inflammations for peritonitis and sepsis therapies. Free gentamicin (GEN) and hydroxyapatite nanoparticles (NPs) with GEN loading and mannose grafts (mHA G ) are inoculated into ε-polylysine NGs to obtain NG@G1-mHA G2 through crosslinking with phenylboronic acid and tannic acid. The H 2 O 2 consumption after reaction with phenylboronic esters and the elimination of free radicals by tannic acid alleviates the escalated inflammatory status to promote sepsis therapy. After mannose-mediated uptake into macrophages, the acid-triggered degradation of mHA G NPs generates Ca 2+ to destabilize lysosomes and the efficient lysosomal escape leads to reversion of hypometabolic SCVs into normal phenotype and their sensitivity to GEN. In a peritonitis mouse model, NG@G1-mHA G2 treatment provides strong and persistent bactericidal effects against both extracellular bacteria and intracellular SCVs and extends survival of peritonitis mice without apparent hepatomegaly, splenomegaly, pulmonary edema, and inflammatory cell infiltration. Thus, this study demonstrates a concise and versatile strategy to eliminate SCVs and relieve inflammatory storms for peritonitis and sepsis therapies without infection recurrence.

Protein-reduced gold nanoparticles mixed with gentamicin sulfate and loaded into konjac/gelatin sponge heal wounds and kill drug-resistant bacteria

Yini Zou,Ruiqi Xie,Enling Hu,Ping Qian,Bitao Lu,Guangqian Lan,Fei Lu

Timely antibacterial treatment of wounds reduces the probability of wound infection and promotes wound healing. However, the materials used to treat wounds often fail to provide both sterilization (especially for super bacteria) and moisture, and some may even cause secondary injury to the wound. In this study, gold nanoparticles (Au NPs) of average grain diameter of 3 ± 1 nm were prepared using egg white as the reductant. These particles showed no aggregation and pink fluorescence. Au NPs were mixed with gentamicin sulfate (GS) and loaded into a mixture of konjac glucomannan (KGM) and gelatin as wound dressing (KGM/ [email protected] NPs/GS). Antibacterial experiments showed that the Au NPs amplified the antibacterial activity of GS; Au NPs/GS efficiently eliminated bacteria, especially super bacteria. Cytotoxicity tests indicated that KGM/ [email protected] NPs/GS showed basically no cytotoxicity to L929 cells. In addition, KGM/ [email protected] NPs/GS possesses good water absorption, water retention, and enhanced mechanical properties, which can provide a moist environment for wounds and promote healing. In conclusion, our study showed that the antibacterial activity of KGM/ [email protected] NPs/GS is better than that of only GS and that it efficiently eliminated super bacteria. Therefore, KGM/ [email protected] NPs/GS can be used for killing superbugs, inhibiting bacterial growth, and promoting wound healing.

The Brown Sugar Mediated Carbon Quantum Dots as a Novel Fluorescence Sensor for Sensitive Detection of Gentamicin and Its Application in Foods

Xinran Guo,Yanxin Guo,Xinyue Chen

In this work, a novel fluorescence sensing strategy was proposed for the detection of gentamicin based on fluorescent carbon quantum dots (CQDs) and gold nanoparticles (AuNPs). Herein, the CQDs were green-synthesized for the first time via a one-step hydrothermal method utilizing brown sugar as the precursor. In the presence of citrate-stabilized AuNPs, the fluorescence of CQDs was quenched efficiently. Gentamicin, on the other hand, had a higher affinity for AuNPs and was able to compete with CQDs for a preferential binding to AuNPs, which ultimately led to the aggregation of AuNPs and freeing of CQDs in solution, causing the fluorescence recovery of CQDs. Based on the above phenomenon, the concentrations of gentamicin could be ascertained by detecting the variations in fluorescence intensity of CQDs. This sensing strategy exhibited excellent selectivity in various antibiotics. At the same time, the method displayed outstanding sensitivity for gentamicin, which was successfully applied to real samples detection. Keywords:carbon quantum dots;gold nanoparticles;green synthesis;fluorescence probe;gentamicin

Exogenous cystine increases susceptibility of drug-resistant Salmonella to gentamicin by promoting oxidation of glutathione metabolism and imbalance of intracellular redox levels

Junyuan Du,Zhiyi Wu,Chunyang Zhu,Heng Yang,Feike Zhao,Binghu Fang

Introduction: Antibiotic overuse has caused the development of bacterial resistance, which is a major threat to public health. Intracellular metabolic processes are essential for maintaining the normal physiological activities of bacteria, and an increasing body of research has demonstrated a significant association between metabolic alterations and the development of drug resistance. Numerous studies have demonstrated that the addition of adjuvants can counteract bacterial antibiotic resistance.Method: Cystine treatment was verified in vitro to promote the lethal effect of gentamicin on Salmonella using in vitro bactericidal counting methods. The metabolic differences in Salmonella enterica Typhimurium standard strain ATCC 14028 with or without the addition of cystine were analyzed via untargeted metabolomics. The multifunctional electronic enzyme marker was used to determine intracellular reduced glutathione/oxidized glutathione (GSH/GSSG), ferrous iron on (Fe2+), and reactive oxygen species (ROS) levels. The expression of glutathione and stress genes was determined using real-time quantitative PCR.Result: We confirmed that exogenous cystine increased the lethal effect of gentamicin against strain S. enterica Typhimurium (ATCC 14028) and other clinically resistant Salmonella serotypes. Exogenous cystine stimulated the metabolism of the cell and activated the glutathione pathway while altering the GSH/GSSG ratio, which placed bacteria in a state of redox imbalance with increased Fe2+ and ROS levels. Our results suggest that when bacterial redox levels are reprogrammed, bacterial susceptibility to antibiotics can also change.Discussion: This study confirms that cystine enhances the antimicrobial efficacy of gentamicin against drug-resistant Salmonella. Through the application of metabolomics, the underlying metabolic mechanisms by which cystine exerts its effects on Salmonella have been elucidated, offering a novel perspective in the domain of metabolic reprogramming aimed at counteracting drug resistance. Furthermore, these findings reinforce the potential role of small-molecule metabolites as effective adjuvants to enhance antibiotic action.

Chitosan-gentamicin conjugate attenuates heat stress-induced intestinal barrier injury via the TLR4/STAT6/MYLK signaling pathway: In vitro and in vivo studies

Xueting Niu,Canying Hu,Shengwei Chen,Jiaying Wen,Xiaoxi Liu,Yanhong Yong,Zhichao Yu,Xingbin Ma,Chengpeng Li,Mohamad Warda,A.M. Abd El-Aty,Ravi Gooneratne,Xianghong Ju

Heat stress (HS) has a negative impact on animal health. A modified chitosan-gentamicin conjugate (CS-GT) was prepared to investigate its potential protective effects and mechanism of action on heat stress-induced intestinal mucosa injury in IPEC-J2 cells and mouse 3D intestinal organs in a mouse model. CS-GT significantly ( P  < 0.01) reversed the decline in transmembrane resistance and increased the FITC-dextran permeability of the IPEC-J2 monolayer fusion epithelium caused by heat stress. Heat stress decreased the expression of the tight binding proteins occludin , claudin1, and claudin2. However, pretreatment with CS-GT significantly increased ( P  < 0.01) the expression of these tight binding proteins. Mechanistically, CS-GT inhibited the activation of the TLR4/STAT6/MYLK signaling pathway induced by heat stress. Molecular docking showed that CS-GT can bind effectively with TLR4 . In conclusion, CS-GT alleviates heat stress-induced intestinal mucosal damage both in vitro and in vivo . This effect is mediated, at least partly, by the inhibition of the TLR4/STAT6/MYLK signaling pathway and upregulation of tight junction proteins. These findings suggest that CS-GT may have therapeutic potential in the prevention and treatment of heat stress-related intestinal injury.
