As a recently developed detection technique, photoelectrochemical (PEC) measurement has received extensive attention. However, owing to the lack of a comprehensive approach to engineer the photo-generated carriers, the performance is still limited. As an example, a significant use of PEC measurement might be in understanding the roles of glutathione in physiological and pathological processes. Here, we developed a new approach for engineering the photo-generated carriers with the aid of the synergic effect of self-doping, surface plasmon resonance, electrical field amplification effect, etc. Thus, a highly sensitive multicomponent PEC platform has been developed, in which Au nanorods are decorated on the surface of self-doped TiO2 nanowires, followed by surface receptor functionalization with hemin for capturing glutathione. The synergic effect effectively increases generation, separation and transfer kinetics of the photo-generated carriers, which can be further increased by using a mixture of Au nanorods with different aspect ratios to tune the absorption wavelength to the entire UV-visible region. As a result, this system exhibits a broad linear range from 10 nM to 17.5 μM and low detection limit down to 8.6 nM for detecting glutathione, about 1–2 orders of magnitude lower than most existing PEC sensors.

Simultaneously eliminating tumor cells and filling the bone defects after tumor resection is still highly challenging for clinical bone tumor treatment. Three-dimensional (3D) printing technology , which provides scaffolds with patient-specific accurate geometry and desirable feature, has gained great attention for supplying a promising candidate for therapy of bone tumors. In this work, we constructed multifunctional scaffolds by coating hemin and doxorubicin (DOX) on the strut surface of 3D-printed bioactive glass ceramics (BGC-HM-DOX). Owing to the surficial hemin particles, BGC-HM-DOX scaffolds showed excellent photothermal response under near-infrared laser irradiation. Besides, BGC-HM-DOX scaffolds could effectively kill osteosarcoma cells in vitro and suppress tumor growth in vivo via the combination of photothermal therapy and chemotherapy. Therefore, this scaffold may provide a high clinical potential for precision medicine in tumor-induced bone defect management.

The advancement of novel detection strategies, especially those activatable by near-infrared (NIR) light, is of great importance for advancing biomedical sensing. Herein, we report a rationally designed, NIR-excitable nanoprobe, UCNP‑hemin@F127, for the sensitive detection and imaging of peroxynitrite (ONOO - ). The central design feature is the dual role of the biological molecule hemin, which acts simultaneously as a luminescence resonance energy transfer (LRET) acceptor to quench the emission of core‑shell upconversion nanoparticles (UCNP) and as a specific recognition unit for ONOO - . Co‑encapsulated with the UCNP donor within an amphiphilic F127 matrix, hemin enables a highly efficient “turn‑on” sensing mechanism. Upon interaction with ONOO - , the porphyrin ring of hemin is cleaved, disrupting the LRET process and restoring the fluorescence. This nanoprobe allows rapid, selective, and sensitive detection of ONOO - in solution, achieving a detection limit of 37.41 nM and a signal enhancement of up to 7.7‑fold. It demonstrates good biocompatibility and has been successfully applied for visualizing both exogenous and endogenous ONOO - in live cells under 980 nm excitation. This integrated quencher‑sensor strategy, employing a native biomolecule as a key functional module, provides a reliable tool for monitoring ONOO - and offers a versatile design platform for the development of advanced LRET‑based bio-probes.

The traditional G-quadruplex/hemin horseradish peroxidase has poor stability and a strong background signal and is not suitable for the detection of heavy metals such as mercury ions. In this work, we used the previous DNA strand for mercury ion detection and optimized it. Hemin is covalently bound to the 5′ and 3′ ends of the DNA strand. In the covalent binding state of double Hemin, the G-quadruplex has a strong catalytic activity and can catalyze the oxidation of hydrogen peroxide by ABTS. The formation of double chains with the addition of the S2 chain provides a rigid structure for detecting mercury ions. At the same time, after the addition of mercury ions, the formation of T-Hg2+-T structure further hinders the formation of G-quadruplex and reduces the absorbance value. The detection range in ultrapure water was 0–1000 nM, and the limit of detection concentration was 0.61 pM. In the human serum, the detection range is 0–300 nM and the LOD is 0.1 nM. In addition, the biosensor has excellent selective specificity and is expected to be used to detect mercury ions in organic biological environments and industrial environments contaminated by heavy metals.

In the present work, a highly sensitive and specific fluorescent biosensor for blood glucose monitoring is developed based on hemin-functionalized graphene quantum dots (GQDs) and glucose oxidase (GOx) system. The GQDs which are simply prepared by pyrolyzing citric acid exhibit strong fluorescence and good water-solubility. Due to the noncovalent assembly between hemin and GQDs, the addition of hemin can make hydrogen peroxide (H 2 O 2 ) to destroy the passivated surface of GQDs, leading to significant fluorescence quenching of GQDs. Based on this effect, a novel fluorescent platform is proposed for the sensing of glucose. Under the optimized conditions, the linear range of glucose is from 9 to 300 μM, and the limit of detection is 0.1 μM. As unique properties of GQDs, the proposed biosensor is green, simple, cost-efficient, and it is successfully applied to the determination of glucose in human serum. In addition, the proposed method provides a new pathway to further design the biosensors based on the assembly of GQDs with hemin for detection of biomolecules.

Hemin-functionalized TiO2 nanoparticles were prepared and assembled on glassy carbon electrode through the electrostatic attraction between positively charged poly(diallyldimethylammonium chloride) and negatively charged hemin and TiO2. The scanning electron microscopy and X-ray diffraction characterization showed that the assembled hemin-TiO2 film displayed a mesoporous structure comprising plenty of TiO2 nanoparticles in rutile phase. The Fourier transform infrared spectra and UV-visible diffuse reflectance spectra indicated that hemin was successfully incorporated into TiO2 film and significantly enhanced the absorption of TiO2 film to visible light. The hemin-TiO2 film exhibited a pair of well-defined redox peaks of hemin by cyclic voltammetry, which could effectively catalyze the electroreduction of H2O2. Thus, the hemin-TiO2 modified electrode was employed as an electrochemical sensor for H2O2 determination, showing a sensitive response linearly proportional to the concentration of H2O2 from 3.0 × 10−7 to 4.7 × 10−4 mol·L−1. At the same time, the photoelectrocatalytic activity of TiO2 under visible light illumination was dramatically promoted by hemin. The hemin-TiO2 modified electrode produced a photoelectrochemical response linearly proportional to hydroquinone in the concentration range from 4.0 × 10−7 to 3.0 × 10−5 mol·L−1.

The encapsulation of catalytic active centers into polymer hydrogels represents a simple and effective strategy for enzyme mimicry. Current methods only enable the preparation of macroscopic enzyme carriers, which are far from natural enzymes in terms of their sizes, local chemical and physical properties, and catalytic activities. In this work, a nanogel strategy to immobilize hemin, a mimic of the active center for horseradish peroxidase (HRP), into a polyacrylamide (PAAm) matrix is demonstrated. Hemin was modified using N-(3-aminopropyl) methacrylamide hydrochloride (APMA) and the modified hemin together with excess APMA were covalently incorporated into PAAm nanogels. The resulting Hem@Gel shows enhanced catalytic activity and environmental tolerance compared with hemin and unmodified nanogel Hem/Gel, which has been demonstrated via the catalytic oxidation of various organic dyes. Further characterization and molecular simulations reveal the Fe–N coordination between the pendant groups and the enrichment of dye molecules and H2O2 in PAAm nanogels, which together enhance the local reaction rate and realize an overall improvement in catalytic efficiency.

The electrochemical inactivation of microorganisms by a hemin/graphite felt (GF) composite electrode was investigated, and Escherichia coli was treated as the testing species. The composite electrode was constructed by chemically bonding hemin molecules onto an amino-mineralized GF (AGF) surface. Then, the electrode was characterized systematically by electrochemical methods, and the kinetic parameters of the modified electrode were investigated. The hemin molecules on the surface of the composite electrode have high activity for the reduction of O 2 . When the composite electrode was applied with negative potentials, the dissolved oxygen was electrochemically reduced to reactive oxygen species (ROS, such as H 2 O 2 and OH) at the cathode surface. The ROS can cause biological damage and can eventually result in the death of bacteria. A sterilizing rate up to 99.9% could be obtained after 60 min of inactivation. Thus, this composite electrode could be applied to disinfect drinking water efficiently at a low potential (−0.6 V vs. SCE) without any addition of chloride.

An electrochemical aptasensor is described for the detection of prostate specific antigen (PSA). The aptasensor is based on the use of hemin-functionalized graphene-conjugated palladium nanoparticles (H-Gr/PdNPs) deposited on a glassy carbon electrode. The nanocomposites integrate the high electrical conductivity of graphene with the easily functionalized surface chemistry of PdNPs and their excellent catalytic property. The hemin placed on graphene acts as both a protective agent and an in-situ redox probe. The PdNPs provide numerous binding sites for the immobilization of DNA-biotin via coordinative binding between Pd and amino groups. A sensitive and specific PSA assay was attained by immobilizing the PSA aptamer via biotin-streptavidin interaction. The resulting aptasensor has a linear response that covers the PSA concentration range from 0.025 to 205 ng·mL −1 , with a 8 pg·mL −1 lower detection limit (at −0.362 V, scan rate: 0.1 mV·s −1 , S/ N = 3). The method was applied to the quantitation of PSA in spiked serum samples, giving recoveries ranging from 95.0 to 100.3%.
A signal amplified and approving electrochemical aptasensor was constructed for the determination of prostate specific antigen (PSA) based on the use of hemin-functionalized graphene conjugated to palladium nanoparticles (H-Gr/PdNPs). The sensor has a wide linear range, a relatively low detection limit, satisfying stability and high specificity.

In this work, an ultrasensitive chemiluminescence (CL) aptasensor was prepared for indirect hemin detection based on hemin aptamer modified magnetic graphene oxide polymers (MGO@H-Atp@Co-PP). First, magnetic graphene oxide (MGO) was prepared, and MGO has a large specific surface area and easy separation characteristics. GO and MGO were characterized using scanning electron microscopy (SEM), X-ray powder diffraction (XRD) and Fourier transform infrared spectroscopy (FTIR). Then, the hemin aptamer (H-Atp) was immobilized on the surface of the MGO, followed by cobalt porphyrin (Co-PP) being modified on its surface by the charge attracting effect between H-Atp and Co-PP. The immobilization properties of MGO to H-Atp and the adsorption properties of MGO@H-Atp to Co-PP were researched through the curves of kinetics and the curves of thermodynamics. When hemin existed in solution, Co-PP was removed from the surface of MGO@H-Atp@Co-PP due to the specific recognition ability between hemin and H-Atp while MGO@H-Atp@hemin was separated by a magnet, and Co-PP caused changes to the CL signal. So under optimized CL conditions, hemin could be assayed indirectly within the linear concentration range from 8.0 × 10−11–4.8 × 10−8 mol L−1, and the detection limit was 3.9 × 10−12 mol L−1 (3δ) with the relative standard deviation (RSD) of 3.6%. The CL aptasensor was finally used for the determination of hemin in practical serum samples, and recoveries ranged from 97% to 105%. The satisfactory results revealed the potential applications of the MGO@H-Atp@Co-PP-CL aptasensor for hemin detection in monitoring of drugs and diagnosis of diseases.

Poly(aniline-luminol-hemin) nanocomposites are prepared on an electrode surface through electropolymerization, and a highly sensitive electrochemiluminescence (ECL) biosensor for choline is developed based on the poly(aniline-luminol-hemin) nanocomposites and an enzyme catalyzed reaction of choline oxidase (CHOD). The obtained nanocomposites are characterized by scanning electron microscopy (SEM), atomic absorption spectrometry (AAS) and ECL. The results indicate that hemin can be incorporated into the poly(aniline-luminol) nanocomposites using the facile electropolymerization method, and the poly(aniline-luminol-hemin) nanocomposites are rod shaped porous nanostructure. Moreover, the poly(aniline-luminol-hemin) nanocomposites exhibit higher ECL intensity than poly(aniline-luminol) nanocomposites in alkaline media due to the catalytic effect of hemin on the ECL of the polymerized luminol and the electron transfer ability of hemin in the nanocomposites. CHOD is immobilized on the surface of the poly(aniline-luminol-hemin) nanocomposites modified electrode with glutaraldehyde, and the ECL biosensor based on poly(aniline-luminol-hemin)/CHOD exhibits a wider linear range for the choline detection. The enhanced ECL signals are linear with the logarithm of concentration of choline over the range of 1.0×10 −11 ∼1.0×10 −7 mol L −1 with a low detection limit of 1.2×10 −12 mol L −1 . Moreover, the proposed biosensor is successfully applied to the detection of choline in milk.

Synergistic therapy is considered to be an efficient method for cancer therapy and bacterial elimination with excellent efficacy and high controllability. A new graphene-based nanozyme family member, Hemin–rGO@Au NFs, was synthesized and demonstrated synergistic antibacterial capability. The composite benefits from a high peroxidase-like catalytic activity, excellent photothermal conversion efficiency in the NIR region and fast electron transfer capability. The synthetic procedure of the nanozyme composite is simple but it shows an impressive synergistic antibacterial effect. We expect that these findings will lead to new and wide applications of rGO–Au composites in the biological field.
