Integrating spectroscopic techniques and molecular dynamics simulations to probe the interaction between hesperidin and soy protein isolate and the consequent functional improvements

Chaoyang Ma,Deao Song,Jinghui Chen

Natural polyphenols, when combined with proteins, can endow proteins with unique functional properties. This research synthesized soybean protein isolate (SPI)-hesperidin (HP) complexes and systematically examined the contact between their structure and functional characteristics. The experimental findings demonstrated that the conjugation of SPI with HP resulted in a significant increase in particle size from 279.2 ± 1.4 nm to 377.6 ± 19.2 nm, accompanied by a marginal elevation of ζ-potential from −24.6 ± 1.3 mV to −22.0 ± 0.6 mV. The spectroscopic analyses, employing multiple techniques, showed that reduction in the α-helix and marked fluorescence quenching of SPI and brought about alterations in its secondary structure, characterized by a reduction in the α-helix and a corresponding increase in the β-sheet. Moreover, molecular docking and dynamics simulations provided compelling evidence that SPI formed complexes with HP via hydrogen bonding and hydrophobic interactions. In addition, Functional characterization studies revealed that at an HP concentration of 1.25 mg/mL, the complex exhibited enhanced protein flexibility (57.60%), surface hydrophobicity (41.72%), foaming activity (4.70%), and emulsifying activity (28.16%), whereas foam stability and emulsifying stability decreased by 5.79% and 12.91%, respectively. This research further illuminated the intricate interaction mechanism between proteins and polyphenols, shedding new light on the applications of SPI and HP in food.

Yang Weiqi,Wang Wenhao,Hu Yuanjia,Zheng Jinkun,Wang Yanhui,Chen Guodong,Li Deqiang,Li Pengfei,Qian Zhengming

This study presents a rapid and eco-friendly analytical method for quantifying hesperidin and naringin in mandarin peel, orange peel, and pomelo peel, thereby promoting resource utilization and sustainable practices. The method employed straightforward grinding with diatomite for 2 min, followed by extraction using a mixture of N,N-dimethylformamide and methanol (40:60, v/v ) with ultrasonic assistance for 2 min, which facilitates the efficient release of bioactive flavonoids. The analysis was performed using high-performance liquid chromatography with a Agilent InfinityLab Poroshell 120 EC-C 18 column at 30 °C, employing a gradient elution of 0.1% formic acid aqueous solution and acetonitrile, achieving separation within 5 min. Detection was carried out at 283 nm. Method validation confirmed that this approach exhibits high specificity, precision, and stability. Compared to traditional methods, this approach reduces total analysis time to 9 min, decreases both solvent and energy consumption, and lowers costs. The method’s Greenness Evaluation Metric for Analytical Methods score of 5.691, outperforming other published approaches. The method was applied to analyze 36 batches of citrus peels (12 batches each of mandarin peel, orange peel, and pomelo peel). Hesperidin content in mandarin peel samples ranged from 3.29% to 7.15%, in orange peel samples from 2.06% to 6.03%, and naringin in pomelo peel from 0.07% to 12.40%, demonstrating high analytical accuracy and strong practicality. In conclusion, the established rapid, simple, efficient, and environmentally friendly analytical method effectively supports quality control of citrus peel materials, promotes resource utilization of citrus processing by-products, enabling their transformation into high-value ingredients for the functional food, nutraceutical, cosmetic, and pharmaceutical industries. It aligns with the concept of the green circular economy, and possesses significant potential for widespread application.

Hesperidin alleviates kidney injury induced by nano-copper through regulating endoplasmic reticulum stress and autophagy via AMPK-AKT/mTOR pathway in broiler chicken

Lijiao Zhao,Heyun Zhu,Haoye Li,Shuaihao Guo,Jingchun Wang,Zhaoxin Tang,Jianzhao Liao,Jianying Guo

Nano-copper (nanoCu), a nanoscale form of copper (Cu), has been widely applied in the animal husbandry industry. However, it is of great concern that the Cu exposure to humans and animals can cause toxic damage to multiple organs. Hesperidin (Hes), a flavonoid found in citrus fruits, has shown mitigating effects on various organ damages. In our study, we aimed to investigate the toxic effects of nanoCu exposure on the kidneys of chickens and the therapeutic efficacy of Hes in nanoCu-induced renal injury. Our results indicated that Hes could alleviate endoplasmic reticulum stress (ERS) caused by nanoCu exposure. Intriguingly, nanoCu significantly disrupted endoplasmic reticulum (ER) homeostasis, which manifested as increased levels of key proteins such as GRP78, PERK, and CHOP, as well as elevated H 2 O 2 levels. But Hes treatment exhibited an opposite trend. Furthermore, nanoCu significantly induced autophagy in renal cells, which was accompanied by the activation of the AMPK-AKT/mTOR pathway and a concurrent increase in the protein levels of ATG5, Beclin 1, LC3 Ⅱ/LC3 Ⅰ, and ULK1, along with a decrease in the level of p62. However, it can be reversed by Hes treatment. These findings suggest that Hes could mitigate ER homeostasis disruption and autophagy caused by nanoCu exposure, providing new insights into the pharmacological treatment of Hes in related health issues.

Platycodon grandiflorus polysaccharides combined with hesperidin exerted the synergistic effect of relieving ulcerative colitis in mice by modulating PI3K/AKT and JAK2/STAT3 signaling pathways

Yang Liu,Quanwei Sun,Xuefei Xu,Mengmeng Li,Wenheng Gao,Yunlong Li,Ye Yang,Dengke Yin

Ulcerative colitis (UC) is a chronic inflammatory disorder with a complex etiology, characterized by intestinal inflammation and barrier dysfunction. Platycodon grandiflorus polysaccharides (PGP), the primary component of Platycodon grandiflorus , and hesperidin (Hesp), a prominent active component in Citrus aurantium L. (CAL), have both demonstrated anti-inflammatory properties. This study aims to elucidate the underlying mechanism of the synergistic effect of PGP combined with Hesp on UC, focusing on the coordinated interaction between the phosphatidylinositol 3-kinase (PI3K)/protein kinase B (AKT) and Janus kinase 2 (JAK2)/signal transducer and activator of transcription 3 (STAT3) signaling pathways. A mouse model of UC induced by dextran sulfate sodium (DSS) and a cell model using lipopolysaccharide (LPS)-induced RAW264.7/IEC6 cells were employed to investigate the in vitro and in vivo anti-inflammatory effects of PGP combined with Hesp on UC and its potential mechanism of action. The results indicated that compared to the effects of either drug alone, the combination of PGP and Hesp significantly modulated inflammatory factor levels, inhibited oxidative stress, regulated colonic mucosal immunity, suppressed apoptosis, and restored intestinal barrier function in vitro and in vivo . Further in vitro studies revealed that PGP significantly inhibited the PI3K/AKT signaling pathway, while Hesp significantly inhibited the JAK2/STAT3 signaling pathway. The use of inhibitors and activators targeting both pathways validated the synergistic effects of PGP combined with Hesp on the PI3K/AKT and JAK2/STAT3 signaling pathways. These findings suggest that PGP combined with Hesp exhibits a synergistic effect on DSS-induced colitis, potentially mediated through the phosphatase and tensin homolog (PTEN)/PI3K/AKT and interleukin-6 (IL-6)/JAK2/STAT3 signaling pathways.

Hesperidin PLGA nanoparticles potentiate the efficacy of aPD-1 in treating triple negative breast cancer by regulating CCL2 and ADPN expression in cancer-associated adipocytes

Ningchao Luo,Lisha Ma,Ninghui Ma,Jiale Wei,Hongyan Zhang,Wanyu Jin,Yujie Li,Jingbin Shi,Yang Xiong

Triple negative breast cancer (TNBC) represents a heterogeneous subtype of breast cancer characterized by an unfavorable prognosis due to its aggressive biology. Cancer-associated adipocytes (CAAs) play an active role in tumor development, invasion and metastasis, and response to treatment by secreting various cytokines. CAAs secrete CCL2 and ADPN which significantly affect the efficacy of aPD-1 in treating breast cancer. Our recent research has demonstrated that Hesperidin, a natural phenolic compound, significantly inhibits CCL2, elevates ADPN secreted by CAAs in vitro and in vivo, remodels the immune microenvironment, and potentiates the efficacy of aPD-1 in triple-negative breast cancer. We used Oil red staining, Bodipy 493/503 staining and quantitative real-time PCR to verify the formation of CAAs. ELISA was used to detect levels of CCL2, ADPN secreted by CAAs. Changes in the number of immune cells in mouse tumor tissues were detected using flow cytometry and immunofluorescence. Our data suggest that Hesperidin PLGA nanoparticles significantly reduced CCL2 and increased ADPN secreted by CAAs, which concurrently decreased the recruitment of M2 macrophages, Tregs and MDSCs while increased the infiltration of CD8 + T cells, M1 macrophages and DCs into tumor, thus significantly potentiated the efficacy of aPD-1 in vivo. This study provides a new combined strategy for the clinical treatment of triple-negative breast cancer by interfering with CCL2, ADPN secreted by CAAs to enhance the efficacy of immunotherapy.
