Understanding effect of molecular structure of imidazole-based ionic liquids on catalytic performance for biomass inulin hydrolysis

Peng Lu,Zhi-Ping Zhao,Xing-Ya Wang,Gong-Jia Lan,Xiao-Lan Wang

Developing a green hydrolysis process of biomass for the production of the value-added compounds has been widely concerned. This article aimed at elucidating the structure-property relationship of ionic liquids (ILs) as catalysts, i.e., effect of the chemical structures of anion and cation of ILs on their acidity and catalytic properties of inulin hydrolysis. Twenty kinds of imidazole-based ILs and one kind of quaternary ammonium ILs were synthesized. Inulin hydrolysis for the reducing sugar production was investigated with a series of functionalized ILs under moderate temperature (75   °C) and atmospheric pressure. The chemical structure, acidity and catalytic property of ILs were experimentally characterized and theoretically analyzed. The possible catalytic hydrolysis mechanisms of polysaccharide or lignocellulose by ILs were detailedly analyzed. This work demonstrated that the catalytic performance of catalysts depends not only on its acidity, but also on the chemical structure. It was revealed that imidazole-based ILs showed better catalytic performance than quaternary ammonium ILs in the inulin hydrolysis system, and SO 3 H-functionalized sulfonate ILs obtained more efficient catalytic property because the hydrolysis reaction was catalyzed by the synergy of anion and cation, compared with the sulfuric acid with stronger acidity, which can cause serious equipment corrosion in biomass hydrolysis process. In all these tested ILs, (1-(4-sulfonic acid)-butyl-3-ethylimidazolium hydrogen sulfate) ([C 2 MIM-PS][HSO 4 − ]) exhibited the best catalytic performance, and the yield of reducing sugar was 100%. Understanding the structure-property relationship of ILs will be helpful to design the covalent immobilization strategy of ILs to improve its reusability and eliminate the potential harmfulness to the environment.

The effect of Maillard reaction on the lactose crystallization and flavor release in lactose/WPI/inulin encapsulation

Zhuofan Huang,Kaixin Li,Lingjun Ma,Fang Chen,Xiaosong Hu,Song Miao,Junfu Ji

The crystallization of lactose usually causes the structural collapse and core material escape of flavor encapsulations. The objective of this study was to investigate the effects of different grafting degrees of WPI-inulin Maillard reaction products on the lactose crystallization and the subsequent release behaviors. Ethyl acetate was chosen as the model volatile flavor and the encapsulations were prepared by freeze-drying. The results found that the encapsulation efficiency was significantly increased from 30% to over 80% by using MRPs as wall materials. Those microparticles showed the greater flavor retention and lower moisture adsorption. In addition, the encapsulations produced by the proper Maillard reaction times (e.g., 48 h and 72 h) could effectively delay the lactose crystallization and thus improve the structural stability of the matrix. This innovation finding aims to use the Maillard reaction to control the crystallization behaviors and enhance the usefulness of high-lactose containing products in encapsulation systems.

Intervention time modified the effect of inulin on high-fat diet-induced obesity and gut microbial disorders

Xiaofang Li,Ping Chen,Yingying Zhang,Jiaming Zhang,Shanshan Shen,Kaikai Li

Various epidemiological studies support that inulin improves the intestinal environment and decreases the risk of obesity. The same as host feeding rhythms, gut microbiota itself displays daily fluctuations in composition and function. The purpose of this study was to investigate the influences of intervention time of inulin to alleviate high-fat diet (HFD)-induced obesity based on chrono-nutrition. Male C57BL/6J mice were given HFD and gavaged inulin at light-on time (HFM) or light-off time (HFN) daily. Energy expenditure (EE), 16S rRNA sequencing in colonic contents, and short-chain fatty acids (SCFAs) analysis were carried out after 12 weeks. Both in HFM and HFN group, the HFD-induced weight gain was significantly reduced by 20.91% and 23.32%, and the mice in HFN group showed no significant difference in weight gain with the control group. Besides, mice in HFN group showed a tendency to increase EE and fecal SCFAs contents in addition to the relative content of colonic beneficial bacteria. As the light-off period of mice is their active period, these results suggested that inulin intake at active period exhibited relatively stronger effects on ameliorating obesity and improving the gut microbial environment.

3D-printed dysphagia-friendly gels with tailored inulin-konjac glucomannan mixtures: Swallowing safety enhancement and controlled lycopene release

Qian Chen,Yueqi Wang,Hong Lin,Huan Xiang,Yanyan Wu

This study proposed an innovative integration of dynamic digestion model with additive manufacturing technology to investigate how inulin (IN)/konjac glucomannan (KGM) ratios modulate myofibrillar protein (MP) gel networks for dysphagia management. The optimized IN/KGM fomulation (P8–1:2, IN/KGM = 1:2 ( w /w)) promoted the formation of densely network structures. The P8–1:2 gel exhibited highest lycopene encapsulation efficiency (88.07 %) and excellent rheological properties while mantianing swallowing safety and 3D printability. During gastric digestion, KGM delayed matrix disintegration through carboxylic group-mediated pH buffering, whereas swelling barrier effect of IN impeded pepsin infiltration, suppressing lycopene burst release. The P8–1:2 gel showed the highest bioavailability (94.83 %) of lycopene at 120 min of gastrointestinal digestion. The IN/KGM synergistic strategy enabled mediate molecular interplay networks, achieving coordinated regulation of gel “swallowing performance - printing precision and digestive responsiveness”, which provided novel insights for personalized dysphagia diet design and precision nutrition.

Enhancement of environmental and gastrointestinal digestive stability of probiotic-loaded liposomes: Study of inulin gelation and prebiotic effects

Jing Liu,Nianwen Zhang,Chaofan Guo,Ruilin Luo,Xingwei Wang,Xiaoyu Yin,Jianxin Cao,Xianchao Feng,Xuejiao Wang

This study develops a liposome–inulin gel delivery system to enhance the environmental and gastrointestinal stability of Lactobacillus rhamnosus . As the inulin gel ratio increases from 1:1 to 1:3, the encapsulation efficiency of probiotic-loaded system (L-Lip@gel) significantly increases, reaching ~89% ( P  < 0.05), accompanied by an increase in particle size, reduced zeta potential (−6.2 mV) and a higher polydispersity index (0.47). Microstructural characterisation indicates that the inulin gel coating interacts with the hydrophilic core of liposomes via hydrogen bonding and steric effects, forming a stable composite structure. During 28 days of storage, viable probiotic counts remain >6 log cfu/mL and exceed to 7 log cfu/mL under acidic conditions, oxidative stress and simulated gastrointestinal digestion. The release behaviour follows both the zero-order and Korsmeyer–Peppas kinetic models with significantly reduced membrane permeability ( P  < 0.05). Overall, the proposed system effectively enhances probiotic stability and bioavailability.

The gut microbiota mediates the antidepressant effects of imipramine: Evidence from fecal microbiota transplantation and prebiotic inulin intervention

Chen Yonglan,Lan Yuxin,Yi Jingyan,Qi Xiaoyi,Liu Li,Liu Sha,Xu Xinxin,Liu Huan,He Yufeng,He Bing,Ma Xiaochi,Lü Muhan,Liang Sicheng

Background Many antidepressants exert their effects by modulating the gut microbiota, but it is unknown if this mechanism extends to the tricyclic antidepressant imipramine. Objective We probed the extent to which specific gut bacterial taxa mediate the antidepressant effects of imipramine. Material and methods Using a chronic unpredictable mild stress (CUMS) mouse model, we administered imipramine, the prebiotic inulin, or their combination, and systematically evaluated depressive-like phenotypes. Gut microbial dynamics were profiled by 16S rRNA sequencing, and fecal microbiota transplantation (FMT) was performed to determine the transmissibility of microbiota-derived antidepressant effects. Immunofluorescence staining was used to quantify brain-derived neurotrophic factor (BDNF) expression and to assess intestinal barrier integrity via ZO-1 and occludin. Results Imipramine treatment significantly increased the relative abundances of putative A. muciniphila and L. reuteri , both of which showed strong correlations with behavioral improvements. Furthermore, FMT from imipramine-treated donors effectively alleviated CUMS-induced depressive-like behaviors, upregulated BDNF expression and restored gut barrier integrity in recipient mice. Co-administration of imipramine and inulin produced synergistic antidepressant effects. Conclusions Imipramine partially exerts its antidepressant activity through microbiota modulation, with putative A. muciniphila and L. reuteri emerging as important mediators. The superior efficacy of the imipramine-inulin combination highlights a promising microbiota-targeted strategy for advancing antidepressant therapy.

Structural Reconfiguration of Antarctic Krill Protein by Ultrasound-Assisted Inulin Glycosylation for Improved Resveratrol Incorporation and Emulsion Stability

Na Li,Zisheng Zhai,Yao Qin,Xusong Wang,Hai Chi,Fawen Yin,Dayong Zhou,Deyang Li

Abstract Antarctic krill protein (AKP) is a high-value marine protein limited by aggregation, low solubility, and poor compatibility with hydrophobic bioactives. Here, we applied ultrasound-assisted inulin glycosylation to prepare AKP–inulin conjugates and AKP–inulin–resveratrol ternary complexes, systematically examining their structure–function relationships. Ultrasound enhanced glycosylation, reaching the highest grafting degree at 480 W, and improved colloidal stability, as indicated by reduced particle size and increased absolute zeta potential. Structural analyses revealed α-helix reduction, increased β-sheet and disordered structures, molecular unfolding, and enhanced cross-linking. The 480 W ternary complex exhibited superior emulsifying activity, high interfacial protein adsorption, improved 10-day storage stability, and enhanced antioxidant capacity, with DPPH scavenging reaching 63.41%. These results demonstrate that ultrasound-assisted glycosylation reconfigures AKP into a resveratrol-containing, interfacially active matrix with improved colloidal and emulsion stability, providing mechanistic insight into protein–polysaccharide interactions and supporting the development of AKP-based ternary complexes for functional food applications.

Development and characterization of tilapia skin collagen-inulin oleogel as the potential fat substitute in beef patty formulations

Yingying Gu,Weiwei Xu,Yanjie Guo,Yongfang Gao,Jie Zhu

The effects of inulin addition, olive oil content, and ultrasonic treatment on the rheological, texture, and structural properties of collagen-based oleogels were investigated in this study. Furthermore, the fat substitution ability of the oleogel in low-fat beef patties was evaluated. Initially, a uniform and dense network cross-linked structure was found when the ratio of collagen to inulin complex was 1:5. The oleogel sample exhibited good stability and oil binding ability with an additional amount of 50 % olive oil. Ultrasonic treatment improved the stability of the oleogel structure in all samples. Additionally, the addition of inulin reduced cooking loss in beef patties. Beef patties prepared at a 50 % fat substitution level showed physical properties that were the least different from those of pure adipose tissue (control group), which could significantly reduce the content of saturated fatty acids and improve the storage stability of beef patties. This study provided guidance for the application of collagen-inulin oleogel in food processing.

Inulin and metal-phenolic network single-cell encapsulation: Boosting the tolerance and intestinal colonization capability of Bifidobacterium lactis

Mengjun Wang,Shuo Zhang,Siyan Liu,Linyu Nian,Suilou Wang,Chongjiang Cao

Probiotics play a vital role in human health, but maintaining their high viability during processing and oral administration remains challenging. In this study, we utilized inulin and tannic acid (TA)/Ca 2+ metal-phenolic networks (MPNs) as coating materials to encapsulate anaerobic Bifidobacterium lactis (BL) using a layer-by-layer self-assembly strategy to improve their tolerance to the adverse gastrointestinal microenvironment and colonization in the intestinal tract. The results demonstrated that the three-dimensional gel coating formed by inulin could protect the MPNs coating from degradation by gastric acid and improve the ability of BL to resist gastric acid and bile salts, significantly increasing the survival rate of probiotics. Additionally, MPNs could effectively scavenge free radicals and resist cell damage caused by oxidative stress. Importantly, TA in MPNs could form strong interactions (hydrogen bonds) with mucin in the intestine, greatly prolonging the residence time of BL (more than 96 h). Moreover, the raw materials used to prepare BL@TA@Inulin are food-grade, thus ensuring high biosafety. In summary, BL@TA@Inulin represents a novel probiotic single-cell encapsulation approach to achieve highly active and adhesive delivery of probiotics, with potential applications in developing healthy foods and treating gastrointestinal tract-related diseases.

Synergistic Regulation of Ferroptosis via an Oral Solid Lipid Nanocomplex-Embedded Inulin Gel for Targeted Ulcerative Colitis Therapy

Luolan Ouyang,Jia Ke,Shanshan Xiang,Jing Ding,Shufen Zhang,Zixu Wang,Si Chen,Tingting Meng,Hong Yuan,Fangying Yu,Fuqiang Hu

Ulcerative colitis (UC), a chronic inflammatory disorder of the colon, remains challenging to treat due to limited efficacy with current therapies. Ferroptosis, a newly identified form of programmed cell death, contributes significantly to UC pathogenesis and might offer promising therapeutic potential. Herein, we developed an oral synergistic regulation of ferroptosis using a solid lipid nanocomplex-embedded inulin gel (SLN/Que&OA@Gel) for colon-targeted UC therapy; solid lipid was utilized to encapsulate oleic acid (OA) and quercetin (Que) for better intestinal bioavailability, which was further embedded in an inulin gel to realize colon targeting. The system combines OA─which modulates lipid composition to reduce ferroptosis susceptibility─with Que, an antioxidant that scavenges reactive oxygen species (ROS) and suppresses oxidative stress to limit ferroptosis progression. Simultaneously, the incorporation of the inulin gel matrix ensures gastrointestinal tract protection, mucoadhesion, prolonged retention time at the lesion site, and sustained drug release, which promise a better curative effect. In vivo studies demonstrate that SLN/Que&OA@Gel effectively remodels the UC-associated pathological microenvironment by alleviating intestinal damage, restoring intestinal barrier integrity, and reducing inflammation. This strategy of synergistic regulation of ferroptosis and microenvironment remodeling presents a promising therapeutic avenue for UC management.

Insights into ultrasound-modified soy protein isolate-inulin hydrogels: Focus on binding mechanisms, stability, and riboflavin controlled-release behavior

Sai Yang,Ziteng Lian,Yifei Chi,Yaqi Tang,Mengmeng Wang,Jiexu Zhang,Xiaohong Tong,Huan Wang

The loose structure of natural soy protein isolate (SPI) gels limits their application in food nutrient delivery systems. This study investigated the effects of varying inulin (INU) concentrations combined with ultrasound treatment on the structural characteristics, stability, and riboflavin digestion and release of SPI-based hydrogels. Increasing INU concentration enhanced the textural properties and water-holding capacity (WHC) of the composite gels, converting free water into immobilized water. The addition of 2 % INU with ultrasound treatment (SU-2) markedly improved rheological properties, yielding a smoother, denser microstructure and increasing protein disorder. Molecular docking and molecular dynamics simulations indicated that the SPI-INU gel structure was stabilized primarily through hydrogen bonding and hydrophobic interactions. The modified gels improved riboflavin loading and stability, with SU-2 exhibiting the highest bioaccessibility (85.82 %) and chemical stability (76.01 %). This study contributes to the development of natural protein/polysaccharide gel carriers for targeted slow release of hydrophilic bioactives.

Industrial additive and dietary fiber inulin induces tumor-selective necrotic death via mitochondrial lipid peroxidation involving MYC and YAP

Ming Li,Aidana Kablan,Gulziya Toktau,Ashfaq Ahmad,Lun Meng,Ziye Li,Zhaohe Zuo,Songgula Gemingbai,Aknazar Otebay,Arailym Satiyeva,Yingqiu Xie

Inulin as a prebiotic fiber and an industrial additive, has been shown to regulate gut and small intestinal microbiome for health, but non-microbiota-dependent cellular pathways remain unclear. Here we aim to determine how the natural polysaccharide inulin reshapes metabolic stress responses especially in colon cancer cells. In tumor cells treated by inulin, oxidized lipids accumulated in the perimitochondrial region and were accompanied by mitochondrial fragmentation, Golgi dispersion, endoplasmic reticulum (ER) disorganization, lysosomal remodeling, increased mitochondrially encoded cytochrome c oxidase II (MTCO2), and nuclear p21, indicating organelle stress coupled to metabolic reprogramming. Non-malignant cells showed elevated lipid peroxidation but limited necrotic death, consistent with greater metabolic tolerance. YAP/MYC inhibition increased inulin sensitivity. Importantly, ataxia telangiectasia and Rad3-related (ATR) supported stress adaptation, particularly in normal cells. Additionally, inulin decreased nuclear active MYC and YAP and enhanced nuclear androgen receptor (AR). Network pharmacology identified inulin against colon cancer by AR-related hub gene PTGS2 (prostaglandin G/H synthase-2) and validated by a synergistic combination with progesterone. Thus, inulin triggers tumor-selective metabolic stress and disrupts adaptive MYC/YAP, providing a rationale for metabolism-based precision cancer therapy.
