A novel dual-monomer magnetic molecularly imprinted polymer (D/L-MMIP) was developed for the selective extraction of luteolin (Lut) by synergistically combining covalent and non-covalent imprinting strategies. The D/L-MMIP was synthesized via free radical polymerization using phenylboronic acid-functionalized Fe 3 O 4 nanoparticles (covalent monomer) and acrylamide (non-covalent monomer). Comprehensive characterization (FT-IR, XRD, SEM, VSM) verified the successful fabrication of D/L-MMIP, confirming its molecularly imprinted structure, spherical morphology, and strong magnetic responsiveness, with Fe 3 O 4 enabling rapid separation (<30 s). The adsorption kinetics reached equilibrium within 120 min at room temperature. Adsorption studies demonstrated pseudo-second-order kinetics (R 2 > 0.99) and Langmuir isotherm behavior (R 2 > 0.99), yielding a maximum adsorption capacity (q max ) of 15.9 mg·g −1 , which was significantly higher than those of non-imprinted (D/L-MNIP, 7.3 mg·g −1 ) and single-monomer MIPs (S/L-MMIP: 4.2 mg·g −1 ; S/L-CMMIP: 7.4 mg·g −1 ). The D/L-MMIP exhibited exceptional selectivity for Lut (imprinting factor = 4.76) over structurally similar flavonoids (quercetin, rutin, hyperoside), with selectivity coefficients (α) of 6.15, 2.64, and 1.53, respectively. The pH-responsive boronic ester bond facilitated efficient template release under acidic conditions (pH ∼ 2.1), while the material maintained 83.5 % of its adsorption capacity after five regeneration cycles. When applied to Lonicera japonica Thunb. extracts, D/L-MMIP enrichment improved Lut purity from 36 % to 60 % (as determined by HPCE), demonstrating its practical applicability. This study provides a sustainable and efficient strategy for the selective separation of bioactive compounds, leveraging the complementary advantages of covalent and non-covalent interactions for enhanced molecular recognition.

Luteolin (LUT) is a natural dietary ingredient with a wide range of biological functions and has been used in skin improvement. Skin lipid oversecreting is an exceedingly common disorder of the pilosebaceous glands and popular in adolescents. This study evaluated the effect of LUT on ameliorating oleic acid (OA) induced skin lipid accumulation in C57BL/6 mice and SZ95 cells. The level of major lipids and some key lipid metabolism markers were assessed, including triglyceride (TG), non-esterified fatty acid (NEFA), cholesterol, sterol regulatory-binding protein 1 and peroxisome proliferator-activated receptors. In vivo data showed decreasing significantly cholesterol levels and in vitro data demonstrated the remarkable influence in the reduction of TG and NEFA levels. Besides, LUT affected the diversity and structure of gut bacteria, an increase in the relative abundance of Akkermansia, Bacteroides and Alistipes , which were associated with the anti-oxidative and anti-inflammatory process. Additionally, our study presented that LUT significantly reduced the level of malondialdehyde (MDA, a lipid peroxidation marker), inhibited the expression of pro-inflammatory factor (TNF-α, IL-6) as well as up-regulated the mRNA expression level of SIRT1, indicating LUT alleviating inflammation and enhancing antioxidation may be associate with the activation of SIRT1 pathway.

Background Glioblastoma multiforme (GBM) is the most common and worst-prognosed primary malignant tumor in the central nervous system. Epithelial mesenchymal transition (EMT) is an important cause of postoperative invasion and recurrence in GBM. Due to the presence of the blood-brain barrier, local therapy may be the preferred route for treatment of GBM after surgery. Methods Our objective is to develop an anti- EMT functional hydrogel for post-GBM surgery tamponade. We fabricated self-assembled luteolin nanoparticles (LU NPs) via the solvent evaporation method, and characterized their morphology and particle size using scanning electron microscopy (SEM), transmission electron microscopy (TEM), and dynamic light scattering (DLS). Furthermore, biological evaluations were conducted through CCK-8 assays, EdU staining, flow cytometry for cell cycle analysis, scratch assays, transwell assays and western blot. Ultimately, we constructed a polyvinyl alcohol (PVA) hydrogel loaded with luteolin nanoparticles and validated its therapeutic efficacy in vivo experiments. Results We successfully synthesized LU NPs, which significantly inhibited GBM cell proliferation, as well as GBM cell invasion and migration in vitro. Furthermore, LU NPs downregulated the EMT signaling pathway. We discovered that the observed anti-tumor effects of LU NPs were dependent on the function of β-catenin. Additionally, we successfully constructed a PVA hydrogel loaded with LU NPs (LU@gel). Finally, in the postoperative model of intracranial GBM xenograft in mice, LU@gel effectively suppressed GBM proliferation and EMT, significantly prolonging the survival time of the mice. Conclusions In summary, we have demonstrated that LU@gel exhibits potent anti-GBM effects, primarily attributed to the inhibition of β-catenin-mediated cell proliferation and EMT.

This study aimed to examine the potential drug-drug interaction (DDI) between vandetanib and luteolin in vivo and in vitro, with the objective of establishing a scientific foundation for their appropriate utilization in clinical settings. Sprague-Dawley (SD) rats were randomly divided into two groups: a control group (vandetanib administered by gavage alone) and an experimental group (vandetanib and luteolin administered together). A series of blood samples were collected at different time intervals. The plasma concentrations of vandetanib and its metabolite N-demethyl vandetanib in rats were determined using an ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS). Incubation systems were set up with rat liver microsomes (RLM) and human liver microsomes (HLM) to measure the Michaelis-Menten constant (Km) and half-maximum inhibitory concentration (IC50) values. Additionally, the inhibitory mechanism of luteolin on vandetanib was also investigated. Ultimately, the molecular mechanism of inhibition was examined through the utilization of molecular docking techniques. In vivo animal experiment results showed that compared with the control group, the AUC(0-t) and Cmax of vandetanib in the experimental group were significantly increased. The findings from the in vitro experiments revealed that luteolin exhibited a moderate inhibitory effect on the metabolism of vandetanib. The IC50 values for RLM and HLM were determined to be 8.56 μM and 15.84 μM, respectively. The identified inhibition mechanism was classified as mixed. This study utilized molecular docking analysis to provide additional evidence supporting the competitive inhibition of luteolin on vandetanib in CYP3A4. The data presented in our study indicated a potential interaction between vandetanib and luteolin, which may necessitate the need for dose adjustment during their co-administration in clinical settings.

Mixed metal oxides derived from a metal–organic framework (MOF) sacrificial template take advantage of the original morphology of MOFs, excellent electrocatalytic properties, mesoporous structure, and more active sites and transport channels, which is favorable to construct a high-performance electrochemical sensor. Here, polyhedral CoZn-MOFs are synthesized by precipitation at room temperature, and then they are etched using Ni2+ to obtain hollow polyhedral NiCoZn hydroxide (NiCoZn-OH) nanocages. After self-assembly with graphene oxide (GO) nanosheets and annealing, the hollow polyhedral NiCoZn mixed metal oxide/reduced graphene oxide (NiCoZnOX/rGO) mixture with a size of ∼700 nm is obtained and used for electrochemically detecting luteolin (LU), which shows a high sensitivity (7.16 μA·μM–1·cm–2 in 0–60 μM) and a low limit of detection (64 nM, S/N = 3). Moreover, it has excellent selectivity in the presence of ascorbic acid and is successfully used to detect LU in diluted human urine, indicating a great potential in clinical applications.

Estrogen receptor (ER)‑negative breast tumors are associated with low survival rates, which is related to their ability to grow and metastasize into distal organs. The aryl hydrocarbon receptor (AhR), a ligand‑activated transcription factor that is involved in several biological processes, is a promising anti‑metastatic target. Luteolin, a non‑toxic naturally occurring plant flavonoid with diverse biological activities, has been demonstrated to be effective against certain types of cancer, and has also been described as a ligand of AhR. In the present study, various cancer cell lines were first investigated following treatment with luteolin, and luteolin exhibited the lowest IC50 in MDA‑MB‑231 cells. Then, the efficiency of luteolin in suppressing the metastasis of ER‑negative breast cancer in vitro was assessed. MDA‑MB‑231 cells were treated with luteolin in vitro. Subsequently, MTT assay and flow cytometry were used to detect cell viability, the cell cycle and apoptosis, and a Transwell assay was used to evaluate cell invasion. In addition, reverse transcription‑semi‑quantitative PCR and western blot were performed to detect the mRNA and protein expression levels of matrix metalloproteinase (MMP)‑2 and MMP‑9. In addition, the number of surface tumor nodules was measured in vivo, in mice bearing B16‑F10 tumors, following treatment with luteolin. Luteolin inhibited the viability and induced the apoptosis of MDA‑MB‑231 cells, which was accompanied by cell cycle arrest. This was associated with a decrease in the expression of the pro‑metastatic markers C‑X‑C chemokine receptor type 4 (CXCR4), MMP‑2 and MMP‑9, which was reversed by AhR inhibition. Furthermore, it was identified that luteolin could inhibit the metastasis in a B16F10 mouse xenograft model, and the levels of MMP‑9, MMP‑2 and CXCR4 were significantly decreased in the lung tissues isolated from tumor‑bearing nude mice following luteolin treatment. In conclusion, luteolin is a potential molecule for inhibiting breast cancer invasion and metastasis, which could have promising clinical applications.

Luteolin is a flavonoid compound with a variety of pharmacological effects. In this study, we explored the effects of luteolin on monocrotaline (MCT) induced rat pulmonary arterial hypertension (PAH) and underlying mechanisms. A rat PAH model was generated through MCT injection. In this model, luteolin improved pulmonary vascular remodeling and right ventricular hypertrophy, meanwhile, luteolin could inhibit the proliferation and migration of pulmonary artery smooth muscle cells induced by platelet-derived growth factor-BB (PDGF-BB) in a dose-dependent manner. Moreover, our results showed that luteolin could downregulate the expression of LATS1 and YAP, decrease YAP nuclear localization, reduce the expression of PI3K, and thereby restrain the phosphorylation of AKT induced by PDGF-BB. In conclusion, luteolin ameliorated experimental PAH, which was at least partly mediated through suppressing HIPPO-YAP/PI3K/AKT signaling pathway. Therefore, luteolin might become a promising candidate for treatment of PAH.

Background Chemical liver injury is one of the main causes of acute liver failure and death. To date, however, treatment strategies for acute liver injury have been limited. Therefore, there is an urgent need to find new therapeutic targets and effective drugs. NOD-like receptor pyrin domain containing 3 (NLRP3) inflammasome is a complex of multiple proteins that has been shown to induce cell death under inflammatory and stress pathologic conditions and is thought to provide new targets for the treatment of a variety of diseases. Purpose The purpose of this study was to investigate whether luteolin has a protective effect on the liver and further elucidate whether it is realized through the thioredoxin interacting protein (TXNIP)-NLRP3 axis. Study design Acute hepatic injury in mice caused by intraperitoneal injection of lipopolysaccharide (LPS) was treated with or without luteolin. Methods Male C57BL/6 mice and mouse primary hepatocytes were selected. TXNIP protein knockdown was achieved by siRNA , qPCR and Western blot were performed to explore the mechanism of luteolin in alleviating acute liver injury. Results The results indicated that luteolin had a markedly protective effect on acute liver injury induced by LPS in mice by inhibiting the TXNIP-NLRP3 axis. Luteolin inhibits NLRP3 inflammasome activation by suppressing TXNIP, apoptosis associated speck-like protein containing a CARD domain (ASC), caspase-1, interleukin-1β (IL-1β) and IL-18 to reduce liver injury. In addition, luteolin inhibits LPS-induced liver inflammation by inhibiting the production of inflammation-related gene tumor necrosis factor-α (TNF-α), IL-10, and IL-6. What's more, luteolin alleviated LPS-induced hepatocyte injury by inhibiting oxidative stress and regulating MDA , SOD , and GSH levels. However, the protective effect of luteolin on acute LPS-induced liver injury in mice was blocked by si-TXNIP in vitro . Conclusions These combined data showed that luteolin may alleviate LPS-induced liver injury through the TXNIP-NLPR3 axis, providing new therapeutic targets and therapeutic drugs for subsequent studies.

A poly (methylene blue)/copper nanowires modified nano-carbon paste electrode (PMB/CuNWs@nano-CPE) was developed for ultra-sensitive detection of luteolin. The morphology of copper nanowires was characterized by transmission electron microscopy (TEM). Cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) were utilized to investigate the electrochemical properties of the fabricated electrodes. In comparison with bare nano-carbon paste electrodes (nano-CPE), the PMB/CuNWs@nano-CPE presented better electrocatalytic activity towards luteolin, including reduced oxidation peak potential and enhanced oxidation peak current. The influencing factors on sensitivity were discussed, such as instrumental parameters, concentration of methylene blue, electropolymerization cycles, the amount of copper nanowires, pH value, accumulation potential and accumulation time. Under optimal conditions, two linear calibration ranges of 0.5 nM−0.01 μM and 0.01 μM−1 μM and the detection limit (LOD) of 0.1 nM (S/N = 3) were obtained for luteolin. The satisfying results were achieved for the determination of luteolin in Chinese medicine capsule samples.

A novel molecularly imprinted polymer (MIP) electrochemical sensor was prepared and applied for highly selective and sensitive electrochemical determination of luteolin . The MIP film was electrodeposited on the indium-tin oxide (ITO) glass electrode surface through the electropolymerization of β-cyclodextrin (β-CD) as the functional monomer and luteolin as the template molecule. The MIP sensor displays a rapid electrochemical response, high sensitivity and specific binding of the template luteolin. Under the optimized experimental conditions, the linear response range for luteolin concentrations by the sensor is 5.0 × 10 −8 mol/L to 3.0 × 10 −5 mol/L and the detection limit is 2.4 × 10 −8 mol/L (S/N = 3). The analytical performance of the proposed MIP sensor is evaluated for the detection of luteolin in the traditional medicine as a real sample with good accuracy and precision.

Scope The study aimed to investigate the regioselectivity of methylation of luteolin (3′,4′,5,7-tetrahydroxyflavone) in human in vitro and in vivo. Methods and results Recombinant human catechol- O -methyltransferase (COMT) and human liver S9 were utilized to study the kinetics of meta (3′)- and para (4′)- methylation of luteolin, and urine samples from volunteers after giving a luteolin-containing formulation were collected to determine the ratio of para-/meta-production. The results showed luteolin favored a para-methylation, with a ratio of of para-/meta-production in CL int (1.43 in recombinant human COMT and 1.47 in human liver S9), which was contrary to the known substrates of COMT. However, the result of urine sample assay showed a preference of meta-methylation with a ratio of of para-/meta-production (0.460 ± 0.126). To elucidate the mechanism for different preference of methylation of luteolin in vitro and in vivo, metabolism stability of the meta- and para-methylated luteolin was evaluated in human liver microsomes and recombinant human CYP450s, which revealed that para-methylated luteolin was more easily demethylated by human CYP1A2 and CYP3A4/5 than meta-methylated luteolin. Conclusion Luteolin was a rare substrate of human COMT favoring a para-methylation, but further demethylation by human CYP1A2 and CYP3A4/5 caused a preference of accumulation in meta-methylated luteolin in vivo.

In this study, luteolin-oxidized lotus root starch (OLRS) nanoparticles (NPs) were developed to improve the stability and antioxidant activity of luteolin . Results showed that a stable luteolin-OLRS NPs was formed using luteolin and OLRS (oxidation degree, 15%) in the weight ratio of 3:1, as well as anti-solvent and solvent in the volume ratio of 10:1. Under this condition, the particle size, polydispersity index and zeta-potential of luteolin-OLRS NPs was 305 nm, 0.173 and −20.8 mV, respectively. The analysis of transmission electron microscopy , X-ray diffractometer and Fourier transform infrared spectroscopy demonstrated that the luteolin was successfully encapsulated in OLRS NPs, giving an encapsulation efficiency of 87.2%. The release characteristic and antioxidant activity of encapsulated luteolin were further investigated. Results exhibited that the OLRS NPs enabled luteolin to be stable in simulated gastric fluid and sustained release in simulated intestinal fluid, leading to the enhancement of antioxidant activity of luteolin.
