Pectin-coated liposomes for pectinase-triggered thymol release: A novel on-demand sterilization approach for orange preservation

Zikai Li,Yuhe Dong,Tong Wu,Wanying Zhu,Haohan Jing,Tao Jiang,Yao Liu,Xi Yu,Ying Xiao,Tian Zhong

To reduce food contamination caused by foodborne pathogens, improve encapsulation stability and targeting efficacy of antimicrobial agents, and minimize the required dosage, we developed a pectin-coated liposome containing thymol (Thy@Lip@Pec). The pectin shell of this formulation can be enzymatically degraded by pectinase secreted by fungi, thereby triggering the on-demand release of thymol. Under the action of pectinase at 1.0 mg/mL, this formulation released 76.45 % of thymol within 6 h. Thy@Lip@Pec treatment significantly inhibited Aspergillus niger and Penicillium spp. colonies on agar plates. Moreover, while maintaining quality indicators of oranges, Thy@Lip@Pec significantly suppressed postharvest diseases caused by A. niger and Penicillium spp., reducing the decay incidences of A. niger from 80.00 % (control) to 6.67 %, and that of Penicillium spp. from 93.33 % (control) to 33.34 % in artificially inoculated oranges. This on-demand release antimicrobial material system represents an advanced and efficient strategy for food preservation.

Physicochemical and antimicrobial properties of soy protein isolate films incorporating high internal phase emulsion loaded with thymol

Yuan Zhao,Linfan Shi,Zhongyang Ren,Qun Liu,Yucang Zhang,Wuyin Weng

Oil-in-water (O/W) high internal phase (HIP) emulsion was prepared to investigate its effects on the physicochemical properties and antimicrobial properties of soy protein isolate (SPI)-based films. The particle size and migration degree of oil droplets in the SPI film-forming solution with HIP emulsion and the films were lower than those with conventional O/W emulsion or oil. The SPI-based emulsion films with HIP emulsion containing 30 % oil had the lowest water vapor permeability (1.15 × 10 −10 g·m −1 ·s −1 ·Pa −1 ), glass transition temperature (40.93 °C) and tensile strength (4.47 MPa), and the highest transparency value (12.87) and elongation at break (160.83 %). The antimicrobial test of the SPI-based emulsion films loaded with thymol showed that the thymol encapsulation efficiency, sustained release effect, and growth inhibition effect on microbes were higher for the films with HIP emulsion than those for the films with O/W emulsion or oil.

In-situ electrospinning of thymol-loaded polyurethane fibrous membranes for waterproof, breathable, and antibacterial wound dressing application

Yunpeng Yue,Xiaobao Gong,Wenling Jiao,Yang Li,Xia Yin,Yang Si,Jianyong Yu,Bin Ding

Skin-like flexible membrane with excellent water resistance and moisture permeability is an urgent need in the wound dressing field to provide comfort and protection for the wound site. Despite efforts that have been made in the development of waterproof and breathable (W&B) membranes, the in-situ electrospinning of W&B membranes suitable for irregular wound surfaces as wound dressings still faces huge challenges. In the current work, a portable electrospinning device with multi-functions, including adjustable perfusion speed for a large range from 0.05 mL/h to 10 mL/h and high voltage up to 11 kV, was designed. The thymol-loaded ethanol-soluble polyurethane (EPU) skin-like W&B nanofibrous membranes with antibacterial activity were fabricated via the custom-designed device. Ultimately, the resultant nanofibrous membranes composed of EPU, fluorinated polyurethane (FPU), and thymol presented uniform structure, robust waterproofness with the hydrostatic pressure of 17.6 cm H 2 O, excellent breathability of 3.56 kg m −2 d −1 , the high tensile stress of 1.83 MPa and tensile strain of 453%, as well as high antibacterial activity. These results demonstrate that the new-type device has potential as a portable electrospinning apparatus for the fabrication of antibacterial membranes directly on the wound surface and puts a new way for the development of portable electrospinning devices.

A comparative study on stabilizing and releasing thymol by pre-formed V-type starch and β-cyclodextrin

Qing Gao,Jiabao Zheng,Paul Van der Meeren,Bin Zhang,Xiong Fu,Qiang Huang

Cyclodextrins (CD) are well known to form host-guest assemblies with hydrophobic compounds. As a structural homologue, starch-guest inclusion complexes (ICs) have become promising in the food, agriculture , and pharmaceutical industries due to their low cost and versatility. This study compared the stabilizing and releasing abilities for thymol of three kinds of pre-formed V-type starch (Water-V, Salt-V, and Alkali-V) and β-CD. The loading efficiency for thymol decreased in the order of Water-V > Alkali-V > CD > Salt-V when removing the free thymol. Water-V, β-CD, and their ICs had better thermal stability with maximum thermal decomposition temperatures over 320 °C. Under non-cold chain daily storage conditions for 4 d, the percentage of thymol released from starch/CD-thymol ICs was 14% (Water-V), 35% (Salt-V), 34% (Alkali-V), and 43% (CD). The Water-V made ICs had good sustained release properties and long-term antibacterial activity , which kept strawberries fresh for 6 d without decay.

Identification of CYP isoforms involved in the metabolism of thymol and carvacrol in human liver microsomes (HLMs)

Dong Rui-Hua,Fang Zhong-Ze,Zhu Liang-Liang,Ge Guang-Bo,Cao Yun-Feng,Li Xiao-Bao,Hu Cui-Min,Yang Ling,Liu Ze-Yuan

Carvacrol and thymol are phenolic compounds with similar structures isolated from many aromatic plants, and have been demonstrated to exert multiple pharmacological effects. The metabolic and pharmacokinetic behaviour of thymol and carvacrol has received much attention. Carvacrol and thymol have been demonstrated to undergo phase I metabolism such as hydroxylation reaction. However, drug-metabolizing enzymes involved in this process remain unclear. Given that cytochrome P450 s (CYPs) are involved in most phase I metabolism, the aim of the present study was to investigate the role of CYPs in the metabolism of thymol and carvacrol. After incubation with human liver microsomes (HLMs) in the presence of NADPH, a new metabolite and two metabolites were detected for thymol and carvacrol, respectively. A combination of chemical inhibition studies and assays with recombinant CYP isoforms demonstrated that CYP2A6 was the predominant drug-metabolizing enzyme involved in the metabolism of thymol and carvacrol. All these results remind the researchers that special attention should be paid on pharmacokinetic and clinical outcomes when thymol or carvacrol was co-administrated with other compounds mainly undergoing CYP2A6-mediated metabolism.

Effect of potassium salts on the structure of γ-cyclodextrin MOF and the encapsulation properties with thymol

Xiaodan Pan,Shahid Ahmed Junejo,Chin Ping Tan,Bin Zhang,Xiong Fu,Qiang Huang

BACKGROUND Thymol is a natural essential oil with strong volatility, low solubility, poor dispersion, strong irritation, and an unpleasant smell, which often requires appropriate porous materials to encapsulate thymol during the application process. However, the encapsulation efficiency of thymol in inclusion complexes is low, and new methods of encapsulation need to be developed. In the present study, the encapsulation capacity, storage stability, and antibacterial activity of thymol were investigated using γ-cyclodextrin (γ-CD) metal–organic frameworks (MOFs) by cocrystallization and high-temperature adsorption methods. The effect of different potassium salts (i.e. KOH, KCl, and KAc) on the structure and complexation of γ-CD-MOFs was also analyzed. RESULTS Compared with γ-CD, the thymol encapsulation capacity of γ-CD-MOFs was increased by two- to three-fold, with the encapsulation content following the order: KAc-γ-CD-MOF (293.8 mg g –1 ) > KOH-γ-CD-MOF (287.7 mg g –1 ) > KCl-γ-CD-MOF (249.3 mg g –1 ). The anions in the solution participate in the coordination and influence the symmetry relationship between atoms and ions. This explains the differences in both the three-dimensional γ-CD-MOF structure and the thymol encapsulation amount, as well as the high storage stability of thymol. CONCLUSION The in vitro release kinetics and antibacterial experiments showed that the inclusion complexes prepared by γ-CD-MOFs had higher stability, sustainability, and antibacterial activity, which suggests that it is an excellent complex material for industrial and agricultural applications. © 2022 Society of Chemical Industry.

Preparation and Enhanced Antimicrobial Activity of Thymol Immobilized on Different Silica Nanoparticles with Application in Apple Juice

Yuhao Liu,Xutao Li,Jie Sheng,Yuyang Lu,Huimin Sun,Qixiang Xu,Yongheng Zhu,Yishan Song

In order to diminish the application limitations of essential oils (EOs) as natural antimicrobial components in the food industry, novel antimicrobial materials were designed and prepared by immobilization of thymol derivatives on silica particles with different morphologies (hollow mesoporous silica nanoparticles, MCM-41, amorphous silica). The structural characteristics of antimicrobial materials were estimated by FESEM, FT-IR, TGA, N2 adsorption-desorption, and small-angle XRD, and the results revealed that both mesoporous silica nanoparticles maintained the orderly structures and had good immobilization yield. Furthermore, the antibacterial performance tests showed that mesoporous silica nanoparticles greatly enhanced the antimicrobial activity of thymol against two representative foodborne bacteria (Escherichia coliandStaphylococcus aureus), and the application of the antimicrobial support was tested in apple juices inoculated withE. coli. The MBC of functionalized mesoporous silica supports was established to be below 0.1 mg/mL againstE. coliandS. aureus, which is much lower than that of free thymol (0.3 mg/mL and 0.5 mg/mL againstE. coliandS. aureus, respectively). In addition, at a range from 0.05 mg/mL to 0.2 mg/mL, immobilized hollow mesoporous silica nanoparticles (HMSNs) can inhibit the growth ofE. coliin apple juice and maintain good sensory properties during 7 days of storage. Keywords:silica nanoparticles;mesoporous structure;antimicrobial activity;immobilization;thymol

Activity of the botanical compound thymol against kiwifruit rot caused by Fusarium tricinctum and the underlying mechanisms

Weizhen Wang,Linan He,Zhuzhu Zhang,Wenzhi Li,Jia Chen,Tingting Chen,Youhua Long

BACKGROUND Kiwifruit rot is an important disease caused by different fungal pathogens, which can lead to huge economic loss in the kiwifruit industry. The aims of this study were to discover an effective botanical compound that significantly inhibits the pathogens causing kiwifruit rot, evaluate its control efficacy against this disease, and reveal the underlying mechanisms. RESULTS A strain of Fusarium tricinctum (GF-1), isolated from diseased kiwifruit, could cause fruit rot in both Actinidia chinensis var. chinensis and Actinidia chinensis var. deliciosa . Different botanical chemicals were used for antifungal activity test against GF-1 and thymol was the most effective one with a 50% effective concentration (EC 50 ) of 30.98 mg L −1 . The minimal inhibitory concentration (MIC) of thymol against GF-1 was 90 mg L −1 . Control efficacy of thymol against kiwifruit rot was evaluated and the results indicated that thymol could effectively decrease the occurrence and spread of kiwifruit rot. The mechanisms underlying the antifungal activity of thymol against F. tricinctum were investigated, and it showed that thymol could significantly damage the ultrastructure, destroy the plasma membrane integrity, and instantaneously increase energy metabolisms of F. tricinctum . Further investigations indicated that thymol could extend shelf life of kiwifruit by increasing their storability. CONCLUSION Thymol can effectively inhibit F. tricinctum that is one of the causal agents of kiwifruit rot. Multiple modes of action are involved in the antifungal activity. The results of this study indicate that thymol can be a promising botanical fungicide to control kiwifruit rot and provide useful references for thymol application in agriculture system. © 2023 Society of Chemical Industry.

Inhibitory Activity and Mechanism of Action with Thymol against the Blueberry Pathogenic Fungi Caused by Neopestalotiopsis clavispora

Shengjie Ye,Liang Shuai,Donglan Luo,Liangjie Ba

Decay caused byNeopestalotiopsis clavisporais an important postharvest disease of blueberries that seriously affects the commercial value of blueberry fruit. In this paper, we studied the inhibitory activity and mode of action of thymol against the pathogenic fungus of blueberries caused byNeopestalotiopsis clavispora. The results demonstrated that thymol administration could limit mycelial growth in vitro; the inhibitory effect was positively connected with thymol mass concentrations, and the minimal inhibitory concentration (MIC) was 100 mg/L. Further investigations revealed that MIC thymol treatment dramatically reduced the germination of pathogenic spores and led to an increase in the conductivity of the pathogen, leakage of contents, and a decrease in pH. Propidium iodide (PI) staining experiments demonstrated that MIC thymol caused damage to mycelial cell membranes. Additionally, MIC thymol treatment promoted mycelium malondialdehyde content accumulation, inhibited superoxide dismutase (SOD) and catalase (CAT) enzyme activities, decreased adenosine triphosphate (ATP), adenosine diphosphate (ADP), and adenosine monophosphate (AMP) content and energy charge levels, and the fluorescence intensity of mycelium caused by MIC thymol treatment was significantly increased by the 2,7-Dichlorodi-hydrofluorescein diacetate (DCFH-DA) assay. The results of this study indicate that thymol suppresses the proliferation ofNeopestalotiopsis clavisporaby compromising the integrity of their cell membranes, promoting the accumulation of cellular reactive oxygen species (ROS), and interfering with energy metabolism. Keywords:Neopestalotiopsis clavispora;thymol;blueberry;postharvest disease;antifungal activity

Thymol and trans-cinnamaldehyde induce apoptosis through a metacaspase-dependent mitochondrial pathway in food-spoilage yeast Zygosaccharomyces rouxii

Huxuan Wang,Zhonghua Peng,Cong Wang,Yanan Zhu,Fei Xia,Hongmin Sun

This work aimed for the first time to provide detailed insights into thymol and trans -cinnamaldehyde's mechanisms of action on the food-spoilage yeast Zygosaccharomyces rouxii and offers evidence in favor of the activation of an apoptosis-like phenotype. The action mechanisms of thymol and trans -cinnamaldehyde were investigated by the measurement of a series of typical apoptotic features using flow cytometer or microplate reader. Moreover, quantitative reverse transcription PCR (QRT-PCR) was performed to investigate the effects of thymol and trans -cinnamaldehyde on the transcription of key regulators of apoptosis in Z. rouxii . The results indicated that the treatment of Z. rouxii with thymol or trans -cinnamaldehyde (minimum inhibitory and subinhibitory concentrations) triggered reactive oxygen species (ROS) accumulation, elevated intracellular Ca 2+ level, and depolarized mitochondrial membrane potential (MMP) coupled with hallmarks of apoptosis including mitochondrial cytochrome c (cyt c) release, metacaspase activation, phosphatidylserine (PS) exposure, and DNA fragmentation. Moreover, thymol or trans -cinnamaldehyde treatment upregulated the transcription of proapoptotic regulators including Yca 1, Dnm 1, Nuc 1, Ndi 1, and Mmi 1 and downregulated the transcription of antiapoptotic regulators of Fis 1 and Cdc 48, further confirming the apoptosis induced by thymol and trans -cinnamaldehyde in Z. rouxii . In summary, thymol and trans -cinnamaldehyde probably induced apoptosis through a metacaspase-dependent mitochondrial pathway in Z. roxuii . These findings provide theoretical support for the development of safe and efficient agents used in food preservation. Practical Application The results will provide a new idea for the systematic analysis of the antifungal mechanisms of thymol and trans -cinnamaldehyde and also provide a theoretical support for the development and application of natural food preservatives, which is of positive significance for the effective control of the spoilage caused by Z. rouxii in food processing and storage and the protection of consumers’ health.

Fabrication and characterization of chitosan nanoemulsions loading thymol or thyme essential oil for the preservation of refrigerated pork

Ting Liu,Liu Liu

The purposes of this study were to develop stable thyme essential oil chitosan nanoemulsions (TEO-CSs) and thymol chitosan nanoemulsions (T-CSs), and to evaluate their characterization and antibacterial properties. The effects of the TEO-CSs and T-CSs on preservation of refrigerated pork at 4 °C were also assessed. The results indicated that the mean particle size values of the TEO-CSs and T-CSs were 139.47 and 123.30 nm, and the encapsulation efficiencies were 80.99% and 70.72%. The absolute zeta potential values of the TEO-CSs and T-CSs were 30.87 and 32.42 mV respectively, indicating that the stable TEO-CSs and T-CSs were fabricated. The information on FTIR, XRD and TGA inferred that chitosan was physically mixed with thymol and TEO without chemical reaction, and the antibacterial properties of thymol and TEO still remained. Furthermore, the antibacterial assays verified that the TEO-CSs and T-CSs strongly inhibited Staphylococcus aureus ( S. aureus ) and Escherichia coli ( E. coli ) planktonic bacteria and biofilms. Fresh pork treated with the TEO-CSs and T-CSs had significantly improved color parameters (a ⁎ , b ⁎ , L ⁎ and ΔE values) during storage at 4 °C. The TEO-CSs and T-CSs effectively extended the shelf life of refrigerated pork to more than 6 days by measuring the pH values and total viable count (TVC) of bacteria. Our results highlighted that the TEO-CSs and T-CSs, as natural novel antibacterial packaging materials, had outstanding antibacterial activities and effectively preserved food.

Thymol contributes to preserving the postharvest freshness of blueberries by enhancing antioxidant and ascorbate-glutathione (AsA-GSH) cycle

Ba Liangjie,Luo Chenglin,Wang Xiaogang,Ye Shengjie,Luo Donglan,Cao Sen

Blueberries are prone to softening and rotting shortly after harvest, which not only complicates post-harvest preservation but also significantly impairs their quality. In this study, the regulatory mechanism of thymol pre-harvest treatment on the quality and ascorbic acid-glutathione (AsA-GSH) cycle of blueberries during storage was investigated. The results demonstrated that thymol treatment impeded the decline of total flavonoids, total phenols, reducing sugars, and starch content in blueberry fruit. In addition, thymol treatment suppressed the activities of α-amylase, β-amylase, and total amylase. In addition, thymol increased the activities of ascorbate peroxidase (APX), glutathione reductase (GR), monodehydroascorbate reductase (MDHAR), and dehydroascorbate reductase (DHAR) and maintained high AsA and GSH contents and AsA/(AsA + DHA) and GSH/(GSH + oxidized glutathione (GSSG)) ratios in blueberries. In conclusion, thymol improved the overall postharvest antioxidant capacity of blueberries and maintained their quality by delaying the reduction of antioxidants, inducing antioxidant enzyme activities, and enhancing the metabolic activities of the AsA-GSH cycle. Therefore, thymol pre-harvest treatment can serve as an efficient and natural pre-harvest treatment agent for preserving postharvest blueberry quality, effectively maintaining the post-harvest quality of blueberries and extending their storage life.
