Enhanced stripping voltammetric response of Hg2+, Cu2+, Pb2+ and Cd2+ by ZIF-8 and its electrochemical analytical application

Yaru Chu,Feng Gao,Fei Gao,Qingxiang Wang

The zeolitic imidazolate framework ZIF-8 was synthesized and mixed with chitosan (CS) solution to obtain a homogeneous dispersion. Then the dispersion of ZIF-8 and CS (ZIF-8-CS) modified glassy carbon electrode is fabricated and utilized for stripping voltammetric analysis of Hg 2+ , Cu 2+ , Pb 2+ and Cd 2+ for the first time. The large surface area effect and strong enriching effect of the nanosized ZIF-8 to the four heavy metal ions contribute to significantly enhance analytical signals. Under the optimum conditions, the stripping peak current densities linearly increased with Hg 2+ concentration in the range from 1.0   μM to 80.0   μM, and with Cu 2+ , Pb 2+ and Cd 2+ concentrations in the range from 1.0   μM to 100.0   μM. The analytical sensitivity for the four ions are estimated to be 14.20   μA·μM −1 ·cm −2 (Hg 2+ ), 2.797   μA·μM −1 ·cm −2 (Cu 2+ ), 5.203   μA·μM −1 ·cm −2 (Pb 2+ ) and 1.390   μA·μM −1 ·cm −2 (Cd 2+ ), respectively. The limits of detection for the four ions were estimated to be 0.0293   μM (Hg 2+ ), 0.108   μM (Cu 2+ ), 0.0619   μM (Pb 2+ ) and 0.135   μM (Cd 2+ ), respectively. The ZIF-8 based modified electrode also presented excellent anti-interference ability, and good reliability for monitoring the four ions in real water samples.

Photothermal Soft Nanoballs Developed by Loading Plasmonic Cu2–xSe Nanocrystals into Liposomes for Photothermal Immunoassay of Aflatoxin B1

Xue Li,Lin Yang,Chen Men,Yi Fen Xie,Jia Jun Liu,Hong Yan Zou,Yuan Fang Li,Lei Zhan,Cheng Zhi Huang

Photothermal effects (PTEs) have been greatly concerned with the fast development of new photothermal nanomaterials. Herein we propose a photothermal immunoassay (PTIA) by taking mycotoxins (AFB1) as an example based on the PTEs of plasmonic Cu2–xSe nanocrystals (NCs). By loading plasmonic Cu2–xSe NCs into liposomes to form photothermal soft nanoballs (ptSNBs), on which aptamer of AFB1 previously assembled, a sandwich structure of AFB1 could be formed with the aptamer on ptSNBs and capture antibody. The heat released from the ptSNBs under NIR irradiation, owing to the plasmonic photothermal light-to-heat conversion through photon–electron–phonon coupling, makes the temperature of substrate solution increased, and the increased temperature has a linear relationship with the AFB1 content. Owing to the large amounts of plasmonic Cu2–xSe NCs in the ptSNBs, the PTEs get amplified, making AFB1 higher than 1 ng/mL detectable in food even if with a rough homemade immunothermometer. The proposal of PTIA opens a new field of immunoassay including developing photothermal nanostructures, new thermometers, PTIA theory, and so on.

A novel phthalohydrazide derivative as a multi-targeted fluorescent probe for monitoring trace Co2+ and Cu2+ in water

Luyue Wang,Yuzhu Chen,Zhengyang Xing,Jie Ma

Real-time monitoring of metal ion species and concentrations in water is a highly active research field. In the paper, a novel fluorescent molecule, (E)-N'-((5-(9-phenyl-9H-carbazol-3-yl) thiophen-2-yl) methylene) terephthalohydrazide (PCTMT), is synthesized via Suzuki reaction and Schiff-base reaction in turn. In the DMF-H 2 O (70 % v/v) solvent system, the PCTMT presents a good aggregation-induced enhancement (AIE) fluorescent effect and good selectivity and anti-interference for monitoring Co 2+ and Cu 2+ in water with a wide pH range. The probe's detection limit (LOD) reached 28.2 nM and 38.7 nM for Co 2+ and Cu 2+ , respectively. The binding constants between PCTMT and Co 2+ and Cu 2+ are calculated as 1.8 ×10 17 M -2 and 2.3 ×10 16 M -2 , at a 2:1 mol ratio. In the probe system, Co 2+ and Cu 2+ can be distinguished by oxalate titration. The detection mechanism is discussed through testing analyses with the theoretical simulation calculation via the density functional theory (DFT) and time-dependent density functional theory (TD-DFT). The probe also indicates good detection capacities in real water samples. This study may promote the development of coordinated-type ion probes and innovation in water quality detection technology.

A Double Network Composite Hydrogel with Self-Regulating Cu2+/Luteolin Release and Mechanical Modulation for Enhanced Wound Healing

Yue Li,Yunpeng Wang,Yuanyuan Ding,Xi Fan,Liansong Ye,Qingqing Pan,Bowen Zhang,Peng Li,Kui Luo,Bing Hu,Bin He,Yuji Pu

Designing adaptive and smart hydrogel wound dressings to meet specific needs across different stages of wound healing is crucial. Here, we present a composite hydrogel, GSC/PBE@Lut, that offers self-regulating release of cupric ions and luteolin and modulates mechanical properties to promote chronic wound healing. The double network hydrogel, GSC, is fabricated through photo-cross-linking of gelatin methacrylate, followed by Cu2+-alginate coordination cross-linking. On one hand, GSC allows for rapid Cu2+ release to eliminate bacteria in the acidic pH environment during inflammation and reduces the hydrogel’s mechanical strength to minimize tissue trauma during early dressing changes. On the other hand, GSC enables slow Cu2+ release during the proliferation stage, promoting angiogenesis and biocompatibility. Furthermore, the inclusion of pH- and reactive oxygen species (ROS)-responsive luteolin nanoparticles (PBE@Lut) in the hydrogel matrix allows for controlled release of luteolin, offering antioxidant and anti-inflammatory effects and promoting anti-inflammatory macrophage polarization. In a murine model of Staphylococcus aureus infected wounds, GSC/PBE@Lut demonstrates exceptional therapeutic benefits in antibacterial, anti-inflammatory, angiogenic, and tissue regeneration. Overall, our results suggest that smart hydrogels with controlled bioactive agent release and mechanical modulation present a promising solution for treating chronic wounds.

Application of HMN-Cu2+ gel with anticoagulant function in promoting rapid vascular endothelial repair in blood fluid environment

Yi'an Chen,Mengxue Zhou,Jingyue Wang,Yunji Fei,Xiaoling Yan,Xin Hao,Ansha Zhao

Atherosclerotic plaques can be eliminated using approaches such as endarterectomy. However, intimal damage resulting from surgery is frequently challenging to address. Traditional surgical techniques often incorporate foreign substances such as modified stents or drugs to counter the consequences of intimal defects. Such treatment makes it difficult to repair vascular defects in endothelial segments. It is prone to cause problems such as hyperplasia and abnormal blood supply due to vascular endothelial defects. In this study, hyaluronic acid (HA), a natural extracellular mechanism component that is not prone to actively induce coagulation, was employed as a biomimetic material for intimal repair. Through chemical grafting modification, the gel was endowed with rapid photocuring performance within 3 s, and the scientific attributes of the gel-series materials were enhanced. The additional incorporation of Cu 2+ into the gel-prepared HMN-Cu 2+ hydrogel augmented the anticoagulant effect of the material while enhancing blood cell compatibility. The results of material science experiments indicate that HMN gel can adhere stably to various biological interfaces in a fluid environment mimicking blood flow, and its swelling and degradation properties are appropriate. Cell experiments and blood evaluation demonstrated that the gel exhibited favorable erythrocyte compatibility and anticoagulant and endothelial spreading capabilities during intimal repair. The gel has the potential to substitute the intima barrier, improve the intima repair environment, and facilitate subsequent intima repair. It can effectively resolve the problem of intima layer irreparability during novel low-damage vascular surgeries such as intima stripping and plaque rotation.

Biofilm-camouflaged Prussian blue synergistic mitochondrial mass enhancement for Alzheimer's disease based on Cu2+ chelation and photothermal therapy

Lianxin Li,Yu Xiong,Yuewen Zhang,Yujiao Yan,Ruixin Zhao,Fengmei Yang,Meng Xie

Alzheimer's disease (AD) is one of the most common neurodegenerative diseases characterized by cognitive and memory impairment. Metal ion imbalance and Mitochondrial dysfunction, leading to abnormal aggregation of β-amyloid protein (Aβ), are key factors in the pathogenesis of AD. Therefore, we designed a composite nanometer system of red blood cell (RBC) membranes-encapsulated Prussian blue nanoparticles (PB/RBC). Prussian blue nanoparticles (PBNPs) can chelate Cu 2+ and reduce reactive oxygen species (ROS). The RBC membranes are a kind of natural long-lasting circulating carrier. At the same time, through NIR irradiation, the excellent photothermal ability of PBNPs can also temporarily open the blood-brain barrier (BBB), enhance the transmission efficiency of PB/RBC across the BBB, and depolymerize the formed Aβ deposits, thereby achieving the optimal therapeutic effect. In vitro and in vivo studies demonstrated that PB/RBC could inhibit Cu 2+ -induced Aβ monomers aggregation, eliminate the deposition of Aβ plaques, improve the quality of mitochondria, restore the phagocytic function of microglia, alleviate neuroinflammation in APP/PS1 mice, and repair memory damage. In conclusion, our biofilm-camouflaged nano-delivery system provides significant neuroprotection by inhibiting Cu 2+ -induced Aβ monomers aggregation, photothermally depolymerizing Aβ fibrils and reducing the level of ROS, thus effectively ameliorating and treating AD.

An Ion Imprinted Polymers Grafted Paper-based Fluorescent Sensor Based on Quantum Dots for Detection of Cu2+ Ions

Xin-Ran WANG,Bo-Wei LI,Hui-Yan YOU,Ling-Xin CHEN

A novel highly selective fluorescent sensor was fabricated based on the microfluidic paper-based analytical device for the determination of Cu 2+ in aqueous samples. The glass fiber paper was activated using HCl solution and incubated in CdTe quantum dots (QDs) solution to obtain a fluorescent substrate. The Cu 2+ imprinted polymers (Cu-IIP) was synthesized using 3-aminopropyl triethoxysilane (APTES) as functional monomer and TEOS as cross-linker on the surface of the glass fiber in paper at room temperature. This fluorescent sensor was successfully applied to determine the amount of Cu 2+ in aqueous samples that mainly depended on the electron transition between the CdTe quantum dots and the Cu 2+ for specific recognition Cu 2+ . A good linearity was presented in the range of 0.032–3.2 mg L −1 , and the limit of detection was achieved at 0.012 mg L −1 . Satisfactory recoveries ranging from 99.9% to 122.5% were attained for spiked seawater and lake water samples with four concentration levels of Cu 2+ . The paper@QDs@Cu-IIP showed a good performance and significant application perspectives for rapid detection of Cu 2+ by comparing with the results of inductively coupled plasma-mass spectrometry (ICP-MS).

Multifunctional Theranostic Agent of Cu2(OH)PO4 Quantum Dots for Photoacoustic Image-Guided Photothermal/Photodynamic Combination Cancer Therapy

Wei Guo,Zhenyu Qiu,Chongshen Guo,Dandan Ding,Tianchan Li,Fei Wang,Jianzhe Sun,Nannan Zheng,Shaoqin Liu

Image-guided phototherapy is considered to be a prospective technique for cancer treatment because it can provide both oncotherapy and bioimaging, thus achieving an optimized therapeutic efficacy and higher treatment accuracy. Compared to complicated systems with multiple components, using a single material for this multifunctional purpose is preferable. In this work, we strategically fabricated poly(acrylic acid)- (PAA-) coated Cu2(OH)PO4 quantum dots [denoted as Cu2(OH)PO4@PAA QDs], which exhibit a strong near-infrared photoabsorption ability. As a result, an excellent photothermal conversion ability and the photoactivated formation of reactive oxygen species could be realized upon NIR irradiation, concurrently meeting the basic requirements for photothermal and photodynamic therapies. Moreover, phototherapeutic investigations on both cervical cancer cells in vitro and solid tumors of an in vivo mice model illustrated the effective antitumor effects of Cu2(OH)PO4@PAA upon 1064-nm laser irradiation, with no detectable lesions in major organs during treatment. Meanwhile, Cu2(OH)PO4@PAA is also an exogenous contrast for photoacoustic tomography (PAT) imaging to depict tumors under NIR irradiation. In brief, the Cu2(OH)PO4@PAA QDs prepared in this work are expected to serve as a multifunctional theranostic platform.

Nonstoichiometric Cu2−xSe nanocrystals in situ produced on the surface of carbon nanotubes for ablation of tumor cells

Qiang Wang,Wen Long Li,Hong Yan Zou,Hui Liu,Cheng Zhi Huang

Carbon nanotubes (CNTs) are well known as photo-thermal agents for the ablation of tumor cells, but are limited in the application of photo-thermal therapy (PTT) owing to their low molar extinctions. By coupling the photo-thermal effects of CNTs and the intense localized surface plasmon resonance (LSPR) in the near-infrared (NIR) region of Cu2−xSe nanoparticles (NPs), herein we developed a highly efficient PTT nanohybrids agent by in situ growth of Cu2−xSe NPs on the acid-functionalized CNTs at room temperature. The as-prepared Cu2−xSe/CNT nanohybrids have been found to have intense LSPR in the NIR region and good photo-thermal conversion properties. Moreover, possessing good biocompatibility and dispersibility, the nanohybrids could easily be internalized by cancer cells, and show smart photo-thermal performance, properties which were then successfully applied for photo-thermal therapy in vitro, indicating that the Cu2−xSe/CNT nanohybrids could be a potential photo-thermal therapy agent for the ablation of tumor cells.

Hydrophobic-carbon-dot-based dual-emission micelle for ratiometric fluorescence biosensing and imaging of Cu2+ in liver cells

Linlin Lu,Chongchong Feng,Jie Xu,Fengyang Wang,Haijun Yu,Zhiai Xu,Wen Zhang

Copper is closely related to liver damage, therefore, it is essential to develop a simple and sensitive strategy to detect copper ions (Cu 2+ ) in liver cells. A hydrophobic carbon dots (HCDs)-based dual-emission fluorescent probe for Cu 2+ was prepared by encapsulating HCDs in micelles formed by self-assembly of amphiphilic polymer DSPE-PEG and tetrakis (4-carboxyphenyl) porphyrin (TCPP)-modified DSPE-PEG. The obtained probe showed characteristic fluorescence emissions of HCDs and TCPP with large emission shift of 170   nm with single-wavelength excitation. In the presence of Cu 2+ , the fluorescence of TCPP was quenched and that of HCDs remained unchanged, displaying ratiometric fluorescence response to Cu 2+ . The developed probe exhibited high sensitivity (detection limit down to 36   nM) and selectivity to Cu 2+ over other substances, and the probe was used to image the changes of Cu 2+ level in liver cells successfully.

Ratiometry, Wavelength, and Intensity: Triple Signal Readout for Colorimetric Sensing of Mercury Ions by Plasmonic Cu2-xSe Nanoparticles

Hui Zhang,Yunsheng Xia

In this study, we have reported a new colorimetric platform for sensitive and selective sensing of Hg2+ using near-infrared (NIR) plasmonic Cu2-xSe nanoparticles (NPs) as reporters. Because of ultrahigh affinity between Hg2+ and Se2–, the added Hg2+ can react with Cu2-xSe NPs and exchange their Cu+/Cu2+, yielding a HgSe layer around the host NPs. Accordingly, the absorption profiles of the Cu2-xSe NPs are modulated substantially: The absorbance at 400–600 nm is increased, and the NIR localized surface plasmon resonance dramatically decreases with a more than 150 nm bathochromic shift. Thus, the system possesses triple signal responses, namely, ratiometry, wavelength, and intensity, to the analytes simultaneously. Such uniquely multiple signal output not only provides more choices for the quantification, but also enhances the reliability in the analyte detection. By rationally choosing poly(allylamine hydrochloride) as the NP template, Hg2+ ions can be determined as ranging from 0–800 nM. The detection limit is as low as 2.7 nM, which is nearly 4 times lower than the limit value (10 nM) defined by the U.S. Environmental Protection Agency for drinking water. Other heavy/transition metal ions, such as Cu2+, Ag+, Pb2+, Cd2+, Ni2+, Co2+, Mn2+, Zn2+, Cr3+, Fe2+, and FeF63–, do not interfere with the sensing. Especially, Hg2+ contents can be quantified, even if their concentrations are as low 10 nM in tap water and common environmental water samples. Due to favorable analytical performance, the proposed Cu2-xSe NPs based system has potential applications in monitoring trace Hg2+ in various real samples, even in drinking water.

A self-assembled triphenylamine-based fluorescent chemosensor for selective detection of Fe3+ and Cu2+ ions in aqueous solution

Li Wenfeng,Ma Hengchang,Lu con,Ma Yuan,Qi Chunxuan,Zhang Zhonwei,Yang Zengming,Cao Haiying,Lei ziqiang

A novel triphenylamine-based fluorescent sensor tris((4-amino)phenylduryl)amine (m-TAPA) for Fe3+/Cu2+ ion has been developed. m-TAPA shows high selectivity and sensitivity toward Fe3+/Cu2+ over alkali and transition metal ions in aqueous solution. The possible mechanism of fluorescence quenching was that Fe3+/Cu2+ can be captured by the NH2 groups of m-TAPA to form non-fluorescent complexes, resulting in strong quenching. The detection limits of Fe3+ and Cu2+ were calculated to be 230 nM and 620 nM, respectively. Furthermore, fluorescent test strips have been prepared for convenient detection of Fe3+ and Cu2+ ions in environmental water samples, even in drinking water.
