Asymmetric mesoporous silica nanoparticles with anisotropic geometry and dual-compartments are highly desired for loading and release of dual-drugs in separated storage spaces. In this study, an asymmetric lollipop-like mesoporous silica nanoparticle Fe 3 O 4 @SiO 2 &EPMO (EPMO = ethane bridged periodic mesoporous organosilica) was successfully developed via an anisotropic epitaxial growth strategy. The asymmetric nanoparticles show a uniform lollipop shape with a head of spherical Fe 3 O 4 @SiO 2 core-shell that is 200 nm in diameter and a tail of EPMO nanorods with a length of ∼90 nm, and a specific surface area of ∼650.3 m 2 g −1 . Most importantly, the asymmetric nanoparticles possess the unique dual independent (hydrophilic/hydrophobic) spaces with good loading capacities and are significantly more efficient for cancer cell killing than pure drug based on in vitro studies. Additionally, the dual-drug-loaded nanoparticles exhibited excellent antibacterial activity.

Inflammatory bowel disease (IBD) is a continuously increasing chronic inflammation, and it has been a worldwide health threat. Nevertheless, long-term treatments for IBD remain limited in clinical application. Curcumin is an ideal anti-oxidation and anti-inflammatory drug candidate for chronic inflammation and yet is still pharmacologically safe. However, its clinical application is limited by low solubility and poor bioavailability. To this end, we developed a colon-targeted curcumin carrier using β-glucan, sodium alginate (Alg) and chitosan (CS) named bionic yeast (BY) to improve the anti-inflammatory activity in IBD therapy. Saccharomyces cerevisiae -derived β-glucan greatly enhanced the bio-accessibility and colon-targeting release ability of curcumin. Moreover, the cell viability of bionic yeast demonstrated almost no toxicity on Caco-2 and RAW 264.7cells. The NO, iNOS, and proinflammatory factor including TNF-α, IL-1β, IL-6 were measured in LPS-induced RAW 264.7 cell line. These results indicated that bionic yeast containing β-glucan has a more pronounced anti-inflammatory effect than curcumin does. Therapeutic efficacy of bionic yeast was evaluated in DSS-induced IBD mouse model. It maintained the body weight, liver and spleen index triggered by DSS. The colon proinflammatory factor and inflammatory cell infiltration were significantly alleviated after receiving bionic yeast treatment. Furthermore, the bioavailability of bionic yeast was determined by pharmacokinetics. Compared with free curcumin, the concentration of bionic yeast increased with 11.8-fold in bloodstream and 14.1-fold in colon, demonstrating that bionic yeast is readily absorbed after oral administration. This bionic yeast delivery system combines the immune-enhancing properties of β-glucan with the colon-targeting capabilities of Alg and CS, bridging the gap in ‘natural carrier synergistic therapy’ and addressing the challenge of long-term management in acute bowel inflammation.

Diabetic wound healing remains a challenge due to high levels of oxidative stress, excessive inflammation, and bacterial infection. Smart dressings loaded with natural active monomers are proving to be effective strategies for enhancing diabetic wound healing. Herein, the bio-composites (PTIGA-Cur and PTIGA-Cur-Ag) with curcumin (Cur) responsive release were reported for promoting angiogenesis and diabetic wound repair, showing excellent anti-inflammatory, antioxidant, and antibacterial properties. The integration of three-dimensional networks and chemical bonds endowed the bio-composites with superior thermodynamic, mechanical, and self-healing properties. Notably, pH-responsive Schiff base as well as ester groups in the matrix enable the Cur to be released in a controlled manner. A biosensor assembled from PTIGA-Cur-Ag demonstrated electronic conductivity based on in-situ synthesis of AgNPs, which enabled sensitive monitoring of blood glucose levels. In addition, the release of AgNPs enhanced the antibacterial and anti-inflammatory properties. Expectedly, the bio-composites exhibited remarkable biocompatibility, effectively promoting the polarization of macrophages to M2 phenotype, and reducing the expression of proinflammatory cytokines. The full-thickness diabetic wound model revealed that PTIGA-Cur and PTIGA-Cur-Ag were able to effectively promote collagen deposition, neovascularization, and granulation tissue regeneration through their anti-inflammatory, antioxidant, and antibacterial properties. This study provides evidence supporting the potential utility of bio-composites with both pro-healing properties and monitoring functions in the management of diabetic wounds.

Curcumin (Cur) is a highly pleiotropic anticancer agent that inhibits cell proliferation and induces apoptosis in cancer cells. A variety of nano-systems constituted by polymer–drug conjugates have been designed to overcome its shortages on water solubility, chemical instability, and poor bioavailability. However, most of them suffer from ineffective release of Cur in cancer cells in vivo . This work developed a novel flexible acid-responsive micelle formulation by covalently conjugating Cur on the hydrophilic terminals of pluronic F68 chains via cis -aconitic anhydride linkers. The synthesized F68-Cis–Cur conjugates can readily precipitate to form homogeneous micelles with average size about 100 nm in aqueous solution. In acid environments, F68-Cis–Cur conjugates would break down and subsequently release Cur rapidly, for the reason of pH-sensitive cleavage of cis -aconitic anhydride linkers. In vitro anticancer activity tests demonstrated that F68-Cis–Cur micelles induced higher cytotoxicity against both A2780 and SMMC 7721 cells than free Cur. It provided a larger decrease of mitochondrion membrane potential and induced cellular apoptosis. F68-Cis–Cur micelles remarkably increased cellular uptake of Cur than free Cur through caveolae-mediated endocytosis in an energy-dependent manner. This study demonstrates F68-Cis–Cur conjugation as a promising tool for improving intracellular drug delivery in cancer therapy.

Background: Alzheimer's disease (AD) is one of the most common causes of dementia, affecting many old people. Objectives: By designing and synthesizing intracerebral imaging probes, we tried to provide a new solution for the early diagnosis of AD. Methods: We designed and
synthesized bis-iodine-labeled curcumin, and verified its performance through in vivo and in vitro experiments. Results: In this study, bis-iodine-labeled curcumin (7, BICUR) was synthesized. In the in vitro mass spectrum binding assay, Kd values of BICUR with Aβ1-40 and
Aβ1-42 aggregates were 46.29 nM and 64.29 nM, respectively. Aβ plaques in AD brain adjacent sections were positively stained by BICUR, which was similar to some other curcumin derivatives. The Log P value of BICUR was 1.45. In the biodistribution experiment, BICUR showed the highest
initial brain uptake (5.87% compared to the blood concentration) two minutes after the tail vein injection and rapid clearance from the mouse brain. In the acute toxicity experiment, BICUR showed low toxicity, and the LD50 was >100 mg/kg. Moreover, BICUR showed a high stability
in vitro (86.68% unchanged BICUR after incubation for 120min in mouse brain homogenate). Besides, BICUR produced an enhanced CT imaging effect that could be sensitively detected in vitro, but it also showed an obvious differentiation from surrounding tissues after intracerebral injection.
Conclusion: All results suggested that BICUR could probably act as a targeted CT imaging agent for Aβ plaques in the brain.

Bone regeneration in compromised environments, such as infected or inflammatory defects, remains a significant clinical challenge. This study addresses the limitations of curcumin (Cur), including poor solubility, low bioavailability, and limited antibacterial activity, by developing a series of curcumin-metal complexes through chelation reactions. These complexes were encapsulated into nanoliposomes and subsequently incorporated into poly(lactic-co-glycolic acid)/polycaprolactone (PLGA/PCL) electrospun scaffolds to construct multifunctional bone regeneration platforms. The fabricated scaffolds exhibited uniform nanofibrous morphology, enhanced mechanical properties, and improved surface hydrophilicity, which collectively promoted cell adhesion and proliferation. Among the complexes, Cur-Cu demonstrated the highest antioxidant activity and superior osteogenic performance, significantly enhancing alkaline phosphatase (ALP) activity and upregulating integrin β1/α1 gene expression in MC3T3-E1 cells. In contrast, Cur-Zn exhibited broad-spectrum antibacterial efficacy against both Escherichia coli and Staphylococcus aureus , while Cur-Ca showed limited functional benefits due to rapid ion release and potential cellular interference. These findings highlight the tailored functionality of Cur-metal complexes. Cur-Cu scaffolds are promising for repairing bone defects under oxidative stress, whereas Cur-Zn scaffolds are effective in preventing implant-associated infections. This work provides a rational strategy for designing microenvironment-responsive biomaterials with integrated antioxidant, osteogenic, and antibacterial properties for advanced bone tissue engineering.

Bacterial biofilms in infected wounds and implant-associated infections are highly tolerant to antibiotics and often coexist with excessive inflammatory reactive oxygen species (ROS), resulting in treatment failure and delayed healing. Here, we report a photothermal-responsive curcumin-loaded gold nanorod-silica hybrid nanocomposite (Cur@T-Au) for antibiotic-free infection control and infectious microenvironment remodeling. Gold nanorods were electrostatically assembled onto hollow mesoporous silica nanoparticles (H Au) and further coated with polyethyleneimine to yield a positively charged carrier (T-Au) with bacterial-adhesive capability. Upon near-infrared irradiation, Cur@T-Au demonstrated rapid photothermal hyperthermia synergized with photothermal-responsive triggered structural degradation for controlled release of curcumin, exhibiting broad-spectrum antibacterial activity and potent antibiofilm efficacy. Meanwhile, Cur@T-Au effectively scavenged free radicals and reduced intracellular ROS, and markedly modulated inflammatory responses. In vivo , Cur@T-Au significantly reduced bacterial burden and accelerated healing in infected wound and implant-related infection models, accompanied by improved tissue regeneration and inflammation resolution. Collectively, Cur@T-Au provides an on-demand photothermal-responsive sequential therapeutic strategy that integrates bactericidal action, ROS mitigation and degradability, offering a promising non-antibiotic platform for treating biofilm-associated infections.

Difenoconazole (DFZ) is a persistent triazole fungicide that leads to aquatic toxicity and ecosystem damage due to its environmental persistence and high toxicity. Curcumin (CUR), a natural polyphenol derived from turmeric, exhibits strong anti-inflammatory, antioxidant, and antitumor properties. This study assesses CUR’s protective role against DFZ-induced kidney injury in carp and investigates its mechanisms. Carp were exposed to DFZ for 30 days, with dietary supplementation of 400 mg/kg CUR. H&E staining revealed that CUR significantly reduced DFZ-induced histopathological damage in the kidneys. Dihydroethidium staining and biochemical assays demonstrated that CUR effectively lowered Reactive Oxygen Species levels and lipid peroxidation. Through biochemical kits, qPCR, and Western blotting, it was found that CUR decreased T-AOC and iron content, while activating the Nrf2/HO-1 pathway, thereby alleviating ferroptosis. CUR also upregulated the expression of xCT and GPX4, increased GSH levels, and further suppressed ferroptosis. In terms of inflammation, CUR improved the response by reducing pro-inflammatory cytokines (iNOS, IL-1β, IL-6, TNF-α) and increasing anti-inflammatory factors (IL-10, TGF-β1). With regard to apoptosis, CUR inhibited pro-apoptotic proteins (Bax, Cleaved-caspase3, Cleaved-caspase9, Cytochrome c) and restored the anti-apoptotic protein Bcl-2. In conclusion, CUR alleviates DFZ-induced kidney injury in carp by inhibiting ferroptosis through the Nrf2/GPX4 axis, while reducing oxidative stress, inflammation, and apoptosis. This study highlights CUR as a promising natural agent for mitigating environmental toxicity caused by triazole fungicides. Graphical

Optimized emulsion gels were stabilized by crosslinker-free thiolated pectin. • Enhanced interfacial adsorption was verified by quartz crystal microbalance. • Curcumin encapsulation was enhanced by thiolated pectin-based emulsion gels. • Colon targeted and sustained release under in vitro simulated digestion was achieved. In this study, thiolated pectin with tunable modification degrees was successfully synthesized. Structural confirmation was achieved and the thiol group content was determined to range from 321.25 to 565.47 μmol/g. Thiolated pectin effectively stabilized gel-like emulsions with storage modulus (981.6–2027.2 Pa) remarkably higher than that of unmodified ones (46.4 Pa). Microscopic analysis revealed a honeycomb-like network that tightened with higher thiolation levels. Quartz crystal microbalance with dissipation demonstrated improved interfacial adsorption and the formation of a cohesive film via disulfide bond. The curcumin encapsulation was significantly improved to 89.87% by thiolated pectin, and curcumin retention was approximately doubled relative to unmodified pectin. Furthermore, the curcumin release was significantly inhibited in the simulated gastric and small intestinal phases, while its cumulative release during the simulated colonic phase reached up to 71.75%. This work proposed an exogenous crosslinker-free strategy for the encapsulation of lipophilic bioactives, which exhibited considerable potential for colon-targeted delivery. Curcumin has attracted more attention because of its inhibition efficacy on protein amyloid fibrillation. However, the inhibition mechanism was still ambiguous and the clinical application of curcumin was greatly limited because of its poor stability at physiological conditions for the presence of β-diketone moiety. In this paper, a new mono-ketone-containing curcumin analogue (MDHC) was designed and synthesized to realize the possible inhibition mechanism and unveil the important role of β-diketone moiety of curcumin in the inhibition process of amyloid fibrillation using hen egg white lysozyme (HEWL) as model protein. Although all experiment results (ThT, CR, ANS and TEM) showed that the inhibitory capacity of curcumin was better than MDHC, MDHC still could show obvious inhibition effect. Molecular docking showed that both curcumin and MDHC could bind with HEWL by hydrogen bond of phenloic hydroxyl and the binding energy of MDHC was higher than that of curcumin. All the findings inferred that β-diketone group was one of great important groups in the inhibition process of HEWL amyloid fibrillation, which provided more room to construct novel inhibition reagents.

Antibacterial upconversion nanoparticles (UCNP)-based photodynamic-chemical combined therapy (UCNP-aPCCT) provides an ideal method to solve the problem of antibiotic-resistant bacteria in deep-tissue infections. Saponin is a kind of natural product exhibiting promising antibacterial activity. Curcumin is also a natural product that could be used as photosensitizer for antibacterial photodynamic therapy (aPDT). Herein, we have designed the dual-layer silica-coated upconversion nanoparticles combined with UCNP-photodynamic therapy (PDT) and chemotherapy using curcumin as a photosensitizer and the Camellia sapogenin derivative (CSD) as an antibacterial agent. It turned out that the prepared UCNP@mSiO2(Cur)@DmSiO2(CSD)@PVP nanoparticles displayed high antibacterial activity against Escherichia coli and Staphylococcus aureus under near-infrared (NIR) irradiation at 808 nm. Its antibacterial activity is stronger than that of the simple sum of saponin and curcumin aPDT, indicating the existence of a synergetic photodynamic-chemical effect. The current nanoparticle drug system may have potential applications for deep-tissue anti-infection treatment.

Curcumin was encapsulated within dairy milk products (skim, low-fat and whole) using a pH-driven method. The curcumin was incorporated into all milk products at a high encapsulation efficiency (>85%) and was shown to be in an amorphous state. Ultracentrifugation analysis showed that the curcumin was mainly associated with the milk fat globules in the whole and low-fat milk but with the casein micelles or whey proteins in the skimmed milk. The chemical stability of curcumin during storage and heat treatment was higher for the whole milk. Encapsulation of curcumin within milk using the pH-driven method led to a higher in vitro bioaccessibility (after simulated gastrointestinal digestion) and in vivo antioxidant activity (after oral administration to rats) than simply mixing powdered curcumin with milk. This study may lead to the production of nutraceutical-enriched dairy products that could be used as functional foods to improve human health and wellbeing.
