In this study, an injectable dopamine-modified alginate hydrogel combined with nano-hydroxyapatite or strontium hydroxyapatite was cross-linked via the in situ slow release of divalent ions from the hydrolysis of D-glucono-δ-lactone (GDL). The cross-linking mechanism involved the chelation of an alginate with divalent ions and the hydrogen-bonding interaction between alginate and catechol groups. As the GDL concentration at 0.5% (w/v) regulated the release of the divalent ions, which enhanced the chelation effect, it was investigated, and inorganic nanoparticles were homogeneously distributed into the hydrogel system to improve the mechanical properties and stabilize the hydrogel network. Furthermore, the effects of dopamine on the improvement of the mechanical properties were investigated. Results demonstrated that the compressive strength of this injectable hydrogel is enhanced by 4.6 times compared with that of the hydrogel without the addition of GDL, and the compressive strength is enhanced by 4.0 times compared with that of the unmodified alginate hydrogel at strain values ranging from 10% to 50%. The hydrogels combined with strontium hydroxyapatite exhibited a more compact, suitable pore size, with a three-dimensional network structure, and the particle size was 50–100 μm; this range of values is considered as optimal for the growth of bone tissues. In addition, the gelation time can be adjusted from a few minutes to a few seconds by adjusting experiment variables. Our study provides a potential method for improving the mechanical properties of bone tissue engineering materials. 1HNMR, FTIR, and UV–vis spectroscopy measurements; XRD and SEM; and pH, ion release, and mechanical tests were carried out.

The purpose of this study was to compare the properties of exenatide-loaded poly ( d , l -lactic-co-glycolic acid) microparticles (Ex-PLGA-MPs) prepared by a novel ultra-fine particle processing system (UPPS) and spray drying. UPPS is a proprietary technology developed by our group based on the disk rotation principle. Characteristics of the MPs including morphology, particle size distribution, drug content, encapsulation efficiency and in vitro release were comparatively studied. Cytotoxicity of the MPs was examined on A549 cells and the pharmacodynamics was investigated in vivo in type 2 diabetes Sprague-Dawley (SD) rats. Ex-PLGA-MPs prepared by UPPS showed larger particle size, denser surface, greater encapsulation efficiency, less initial burst release, and stable sustained release for more than one month in vitro as compared with the spray drying MPs. Meanwhile, the UPPS MPs effectively controlled the body growth rate and blood glucose in diabetes rats for at least three weeks after a single injection, while the spray drying MPs showed effective control period of about two weeks. UPPS technology was demonstrated to manufacture Ex-PLGA-MPs as a potential sustained release protein/polypeptide delivery system, which is an alternative method for the most commonly used spray drying. This comparative research provides a new guidance for microparticle preparation technology.

Organic fluorescence dyes with long wavelength emission are attracting people’s interests in bioimaging, cells tracing and medical diagnosis due to their many advantages. This contribution exhibited a novel red fluorescent monomer BDPPA with traditional tetraphenylethylene (TPE) and phenothiazine (PHT) as structural units, which showed aggregation-induced emission enhancement (AEE) effects together with the twisted intramolecular charges transfer (TICT) owing to its D-A-D’ structure. The maximal emission wavelength of BDPPA located at 592 nm with a shoulder peak at 629 nm. Afterwards, the amphiphilic PEG-BD copolymers was acquired by RAFT polymerization of BDPPA and PEGMA. When the feeding ratio of BDPPA increased from 10.0 % to 20.0 %, the ratio of BDPPA in PEG-BD copolymers correspondingly increased from 9.6 % to 20.9 %. The obtained amphiphilic PEG-BD could self-assemble into fluorescence organic nanoparticles (FONs) with about 150 nm diameter in H 2 O solution. Owing to their TICT effect and D-A-D’ structure, both BDPPA and PEG-BD copolymers showed obvious fluorescence in THF solvent, and their intensity firstly decreased and then grew with the increase of water concentration, indicative of the obvious aggregation-induced emission enhancement (AEE). From the calculations of energy quantum, the difference ΔE of BDPPA decreased to 3.09 eV from 3.53 eV of its substrate PHT, which was very favourable for the long wavelength emission and cell imaging. Moreover, due to the long wavelength emission, excellent water solubility and biocompatibility, and AEE effect, the obtained PEG-BD FONs revealed the promising application in biological imaging field.

Background
Vitamin D deficiency is a global public health concern, and women of reproductive age represent a particularly vulnerable population. Accurate measurement of serum 25-hydroxyvitamin D [25(OH)D] is therefore essential for reliable clinical assessment. This study aimed to evaluate the analytical performance of the Innodx vitamin D assay for 25(OH)D measurement and to investigate the prevalence of vitamin D deficiency among women of reproductive age in southeastern China.
Methods
This cross-sectional study enrolled women of reproductive age (20–45 years) who sought medical services at Xiamen Maternal and Child Health Hospital in China between November 2023 and September 2025. Serum 25(OH)D levels were measured to determine vitamin D status using the Innodx vitamin D assay (Xiamen Innodx Biotech Co., Ltd., Xiamen, China). The analytical performance of the assay was validated according to Clinical and Laboratory Standards Institute (CLSI) guidelines, covering precision, accuracy, linearity, and interference. A method comparison was performed against liquid chromatography-tandem mass spectrometry (LC-MS/MS) and the Roche electrochemiluminescence assay. Based on the measured levels, vitamin D status was categorized as deficient (< 20 ng/mL), insufficient (≥20, < 30 ng/mL), or sufficient (≥ 30 ng/mL).
Results
The intra-assay coefficients of variation (CV) were 3.18%, 2.09%, and 1.86% at three concentration levels (5, 25, and 100 ng/mL), while intermediate precision CVs were 4.34%, 3.64%, and 3.17%, respectively. Accuracy testing showed bias of 4.20% and 3.26% at 20 and 50 ng/mL. The assay demonstrated excellent linearity (R 2 = 0.998) across the range of 4.0-150.0 ng/mL. No significant interference was observed from hemoglobin (100 mg/dL), bilirubin (40 mg/dL), triglycerides (400 mg/dL), or rheumatoid factor (600 IU/mL). In method comparison with LC-MS/MS, the Innodx vitamin D assay demonstrated strong concordance with a Pearson correlation coefficient of r = 0.970. When compared with the Roche vitamin D assay, the Innodx showed better correlation with LC-MS/MS. Among 495 women, the mean 25(OH)D concentration was 20.50 ± 7.88 ng/mL, with 88.9% (95% CI: 86.1%, 91.7%) having vitamin D deficiency or insufficiency. Women aged ≤ 25 years showed significantly lower levels (18.38 ± 6.26 ng/mL) compared to older groups (31–35 years: 23.31 ± 8.97 ng/mL; > 35 years: 23.72 ± 8.56 ng/mL) ( P < 0.05).
Conclusion
The Innodx vitamin D chemiluminescence assay demonstrated satisfactory analytical performance for clinical use. Our findings reveal a high prevalence of vitamin D deficiency among reproductive-age women in southeastern China, particularly in younger age groups, highlighting the need for public health interventions.

This work described a sensitive and economical HPLC-UV method with FDAA derivatization to simultaneously detect 36 D/ l -amino acids, which provides higher sensitivity and lower cost than other HPLC-based methods. It was validated for linearity range (8–1000 µmol/L), limits of detection (2.68–62.44 pmol/L), limits of quantification (2.93 to 208.13 pmol/L), intraday precision (0.30 % − 5.31 %), interday precision (1.96 % − 8.04 %) and accuracy (86.53 % − 121.46 %). This method was then applied in the determination of D/L- amino acids abundance in fermented and unfermented food materials and showed the characteristics of each type of foods. The method also demonstrated good performance in another application case for the discrimination of different types of food materials based on D/L- amino acids profile. It emphasizes the ability of the method to study the characteristics, distribution and abundance of d -amino acids in foods and their potential application in food quality control.

Shikonin (SHK) has various biological and pharmacological activities, including anticancer, antibacterial and anti-inflammation activities. However, the protective effects and mechanism of SHK on lipopolysaccharide (LPS) and D-galactosamine (D-GalN) induced acute liver injury remain unclear. In this study, LPS/D-GalN caused acute liver injury by intraperitoneal injection. SHK was administrated for 1 h. Then, LPS/D-GalN was given to C57BL/6 mice for 3 h. Our results showed that SHK treatment distinctly decreased serum TNF-α, IL-1β, IL-6 and IFN-γ inflammatory cytokine production, reduced serum ALT, AST, hepatic MPO and ROS production levels, and tissue histology harmful effects, inhibited JNK1/2, ERK1/2, p38 and NF-κB (p65) phosphorylation, and suppressed IκBα phosphorylation and degradation. Furthermore, our research showed that SHK could dramatically increase SOD and GSH production, as well as reduce ROS production, through up-regulating the protein expression of HO-1, Nqo1, Gclc and Gclm, which was related to the induction of Nrf2 nuclear translocation. These results showed that SHK exerted anti-inflammatory activity, which was associated to the inhibition of inflammatory production via down-regulation of the MAPK and NF-κB signaling pathways, and anti-oxidative effects were connected with GSH and SOD activation through up-regulation of the Nrf2/HO-1 signaling pathways.

Amino acids are commonly used as nutritional fortification substances in functional foods, and their chiral configuration is an important determinant of food function. Rapid chiral screening methods are urgently needed in food analysis but are limited by the long-time chiral separation and matrix interference. In this study, we show a kinetic method coupled to thermal-assisted paper spray ionization mass spectrometry for direct determination of enantiomeric excess (ee) of multiple d/l-amino acids in complex food matrixes without sample pretreatment. 3-(2-Naphthyl)-l-alanine was selected as a new chiral reference ligand for the kinetic method to achieve efficient chiral differentiation (discrimination degree is 8.7 for d/l-phenylalanine and 10.2 for d/l-tyrosine). An additional thermal-auxiliary device was developed for paper spray ionization mass spectrometry to facilitate the enantiomeric purity determination. The developed method allowed a rapid simultaneous enantiomeric purity determination of multiple chiral amino acids (d/l-phenylalanine and d/l-tyrosine) within 30 s. Good linearities were achieved for the quantitation of ee (R2 = 0.9996 for phenylalanine and 0.9995 for tyrosine) with unknown amino acid concentrations ranging from 10 μM to 600 μM. The developed method was successfully applied for the enantiomeric purity determination of multiple chiral amino acids in functional capsules and beverages and showed great potential for efficient enantiomer-related food safety screening and nutrition analysis.

Near-infrared-II (NIR-II) fluorescence imaging-guided photothermal therapy (PTT) represents a cutting-edge approach for precise tumor diagnosis and treatment, providing real-time therapeutic efficacy evaluation. However, a significant challenge lies in the creation of phototheranostic agents that provide robust imaging and efficient photothermal conversion. To address this, a donor- π -acceptor (D- π -A) structure heptamethine cyanine derivative, IR1116, that confers a strong intramolecular charge transfer effect is developed. To enhance its applicability, IR1116 is formulated with DSPE-PEG to create a water-dispersible NIR-II phototheranostic nanoheater, NP IR1116 . This nanoheater benefits from the incorporation of an electron-withdrawing group, leading to reduced photooxidation activity and significantly improved photostability. NP IR1116 exhibits strong NIR-II absorption and fluorescence, peaking at 1004 and 1116 nm, respectively, as well as a photothermal conversion efficiency of 79% under 1064 nm lasers. In vitro and in vivo studies showed the efficacy of NP IR1116 in tumor imaging via NIR-II fluorescence and its ability to effectively induce tumor cell apoptosis under 1064 nm laser irradiation. These findings underscore the potential of NP IR1116 in NIR-II fluorescence imaging-guided PTT for tumor treatment, paving the way for further advancements in NIR-II dye development and bioimaging technologies.

Despite advancements in treatments such as atropine, the pathological changes in high myopia still lead to vision loss, highlighting the need to identify novel therapeutic targets. Clinically, axial elongation are closely related to reduced choroidal blood flow. This study revealed that the pyroptosis-associated protein Gasdermin D (GSDMD) may play a key role in high myopia. Experiments showed that chemical inhibition of GSDMD significantly reduced axial elongation and choroidal blood flow reduction. Furthermore, by screening pharmacologically active traditional Chinese medicine compounds, we identified that Artemisia argyi (Aiye) and its active ingredient apigenin act as effective GSDMD inhibitors, capable of replicating the protective effects observed with chemical inhibition. In all, this study suggests that GSDMD is a potential target mediating axial elongation and choroidal blood flow changes in high myopia, and apigenin is a potential therapeutic strategy for intervening in high myopia.

2,4-Dichlorophenoxyacetic acid (2,4-D) is an extensively applied organic compound, primarily for agricultural weed control and plant growth agents. Although 2,4-D usually exists in the environment in low volumes, the detection of 2,4-D is critical for human health and environmental safety. In this work, a biophotonic biosensor was fabricated by coating the surface of a tapered optical fiber with gold nanoparticles (AuNPs) to excite the localized surface plasmon resonance (LSPR) and functionalizing the fiber with molybdenum disulfide nanosheets (MoS2-NSs)/zinc oxide nanowires (ZnO-NWs) to extend the effective sensing area. Due to the inhibitory effect of 2,4-D on the hydrolytic activity of ALP, the refractive index (RI) around the sensor surface changes, leading to a shift in the LSPR peak wavelength. According to this sensing technique, the sensor can detect concentrations in the range of 1–10 mg/L, with a limit of detection (LOD) of 0.29 mg/L. The stability, repeatability and selectivity tests show that the sensor has good stability and selectivity. In the actual sample detection experiment, the recovery rates of apples and Chinese cabbage were 96.2–100.4% and 83.8–108.8%, respectively, which indicated that the detection method had good accuracy for the detection of target substances in actual samples. Thus, the proposed sensor has an important application in the detection of 2,4-D herbicides.

Coupling glycerol oxidation with hydrogen production offers a promising approach to reducing the energy consumption of hydrogen production and upgrading biomass. However, the catalytic performance is often limited by insufficient interfacial charge transfer and poorly defined active sites. Herein, we report a sulfur-modulated NiFe layered double hydroxide catalyst integrated with a Metal-organic frameworks (MOF)-derived Cu framework on nickel foam, in which well-defined heterogeneous interfaces are deliberately constructed to regulate the local electronic structure and catalytic microenvironment. Sulfur incorporation reconstructs the metal coordination environment and induces electron redistribution. As a result, the catalyst exhibits outstanding glycerol oxidation performance toward formate with high selectivity over a broad potential window, along with remarkable durability at high current densities. In situ Fourier transform infrared (FTIR) spectroscopy identifies the key intermediates involved in glycerol oxidation, while density functional theory (DFT) calculations reveal that sulfur-induced electronic coupling lowers the reaction energy barrier and facilitates charge transfer.

A low-cost Cu-Mo dual-doped carbon nitride nanozyme was synthesized. • It shows excellent peroxidase-like activity via superoxide radical pathway. • A smartphone-based platform enables portable and visual D-PA detection. D-Penicillamine (D-PA), a sulfur-containing amino acid, is used to treat various diseases such as Wilson's disease, fibrotic lung disease, and cystinuria. However, excessive intake of D-PA can cause adverse effects, necessitating the development of accurate quantitative detection methods. This study aimed to establish a visual colorimetric method for D-PA detection using a synthesized Cu-Mo dual-doped carbon nitride nanomaterial ( Cu - Mo - VC - CN ). The synthesis capitalizes on the simplicity and cost-effectiveness of a one-pot thermal polymerization reaction, utilizing only inexpensive and commercially available precursors, namely copper chloride, ammonium molybdate, urea, and vitamin C. The material exhibited exceptional peroxidase-like activity, catalyzing the oxidation of TMB by H 2 O 2 via a superoxide radical pathway. The optimal reaction conditions were: temperature 30 °C, time 30 min, pH 5.5, TMB concentration 0.2 mM, and H₂O₂ concentration 0.2 mM. Steady-state kinetic analysis revealed that the Cu - Mo - VC - CN nanozyme had Michaelis-Menten constants ( K ₘ) of 0.083 mM for TMB and 0.603 mM for H 2 O 2 . Under optimal conditions, the system enabled highly sensitive D-PA quantification with a linear range of 0.5–100 μM and a detection limit of 0.45 μM. The nanozyme was successfully applied to determine D-PA content in pharmaceutical tablets with satisfactory recovery (from 97.73 to 98.27%). Furthermore, a smartphone-based testing platform was established to provide a portable and practical solution for the real-time monitoring of D-PA. This work provides both an efficient strategy for developing multi-metal-doped nanozymes and a reliable colorimetric platform for D-PA monitoring.
