Encapsulation of selenium-containing peptides in xanthan gum-lysozyme nanoparticles as a powerful gastrointestinal delivery system

Yiqing Zhu,Xinyang Sun,Xieqi Luo,Jian Ding,Fengjiao Fan,Peng Li,Xinchun Shen,Yong Fang

In this study, nanoparticles (NPs) prepared with xanthan gum and lysozyme were established as a powerful delivery system for two Se-containing peptides: TSeMMM (STP) and SeMDPGQQ (SHP). NPs-STP and NPs-SHP had relatively small particle sizes (145 nm and 148 nm) and negative zeta potentials (−47 mV and −49 mV). The encapsulation efficiency of NPs-STP and NPs-SHP was determined to be 34.35% and 41.35%, respectively. The stability and antioxidant activity of Se-containing peptides were greatly enhanced due to encapsulation. NPs-STP and NPs-SHP exhibited controlled release of Se-containing peptides under in vitro gastrointestinal conditions. NPs-STP and NPs-SHP showed low toxicity and entered Caco-2 cells through clathrin-mediated endocytosis, contributing to a significant increase in the apparent permeability coefficient of STP (2.19 × 10 −6 cm/s) and SHP (2.21 × 10 −6 cm/s). Thus, NPs-STP and NPs-SHP are considered promising delivery systems for Se-containing peptides and have good potential applications in the food and pharmaceutical industries.

Effect of Natural Antioxidants from Marigolds (Tagetes erecta L.) on the Oxidative Stability of Soybean Oil

Xiuqiong Huang,Wei Gao,Xuan Yun,Zhixing Qing,Jianguo Zeng

In recent years, synthetic antioxidants that are widely used in foods have been shown to cause detrimental health effects, and there has been growing interest in antioxidants realised from natural plant extracts. In this study, we investigate the potential effects of natural antioxidant components extracted from the forage plant marigold on the oxidative stability of soybean oil. First, HPLC-Q-TOF-MS/MS was used with 1,1-diphenyl-2-picrylhydrazyl (DPPH) to screen and identify potential antioxidant components in marigold. Four main antioxidant components were identified, including quercetagetin-7-O-glucoside (1), quercetagetin (2), quercetin (3) and patuletin (4). Among them, quercetagetin (QG) exhibited the highest content and the strongest DPPH radical scavenging activity and effectively inhibited the production of oxidation products in soybean oil during accelerated oxidation, as indicated by reductions in the peroxide value (PV) and acid value (AV). Then, the fatty acids and volatile compounds of soybean oil were determined with gas chromatography–mass spectrometry (GC-MS) and headspace solid-phase microextraction–gas chromatography–mass spectrometry (HS-SPME-GC-MS). A total of 108 volatile components, including 16 alcohols, 23 aldehydes, 25 ketones, 4 acids, 15 esters, 18 hydrocarbons, and 7 other compounds, were identified. QG significantly reduced the content and number of aldehydes and ketones, whereas the formation of acids and hydrocarbons was completely prevented. In addition, the fatty acid analysis demonstrated that QG significantly inhibited oxidation of unsaturated fatty acids. Consequently, QG was identified as a potential, new natural antioxidant that is believed to be safe, effective and economical, and it may have potential for use in plant extracts feed additives. Keywords:Tagetes erectaL.;quercetagetin;DPPH-HPLC-Q-TOF/MS;HS-SPME-GC-MS;oxidative stability

Preparation, characterization, and biological activity of Cinnamomum cassia essential oil nano-emulsion

Dongyi Liang,Baijian Feng,Na Li,Linhan Su,Zhong Wang,Fansheng Kong,Yongguang Bi

To solve the problems of low bioavailability and unstable properties of Cinnamomum cassia Essential oil (CCEO), encapsulation technology was introduced as an effective means to improve its shortcomings. In this study, Cinnamomum cassia Essential oil nano-emulsion (CCEO-NE) was successfully synthesized by the oil-in-water method and characterized by standard analytical methods, including dynamic light scattering (DLS), Scanning electron microscopy (SEM), and Transmission electron microscopy (TEM). The results show that the synthesized CCEO is spherical, smooth in surface, and uniform in shape, with an average particle size of 221.8 ± 1.95 nm, which is amorphous. In this experiment, by simulating the digestion of CCEO-NE in the gastrointestinal tract, it was found that CCEO-NE was undigested in the oral cavity, mainly in the stomach, followed by the small intestine. By understanding the digestion of CCEO-NE, we can improve the potential of CCEO bioavailability in food and drug applications. In addition, through the study of ABTS and DPPH free radicals by CCEO and CCEO-NE, it was found that the antioxidant activity of CCEO-NE was more potent than that of CCEO. When the concentration of CCEO-NE and CCEO is 400 μg/mL, the DPPH free radical scavenging rate is 92.03 ± 0.548% and 80.46 ± 5.811%, respectively. In comparison, ABTS free radical scavenging rate is 90.35 ± 0.480% and 98.44 ± 0.170% when the concentration of CCEO- NE, and CCEO is 75 μg/mL, respectively. The antibacterial test shows that CCEO-NE can inhibit both Gram-positive and Gram-negative bacteria. Among them, CCEO-NE has a stronger antibacterial ability than CCEO, and the maximum inhibition zone diameter of CCEO can reach 15 mm, while that of CCEO-NE can reach 18 mm. Meanwhile, SEM and TEM showed that CCEO-NE treatment destroyed the ultrastructure of bacteria. Generally speaking, we know the situation of CCEO in the gastrointestinal tract. CCEO-NE has more potent antioxidant and antibacterial ability than CCEO. Our research results show that whey protein is an effective packaging strategy that can improve the effectiveness, stability, and even bioavailability of CCEO in various applications, including food and health care industries.

Ultrasound-assisted preparation of ‘Ready-to-use’ extracts from Radix Paeoniae Rubra with natural deep eutectic solvents and neuroprotectivity evaluation of the extracts against cerebral ischemic/ reperfusion injury

Yu Zhao,Haofang Wan,Jiehong Yang,Yan Huang,Yu He,Haitong Wan,Chang Li

Natural deep eutectic solvent (NaDES) is widely applied in the extraction of nutrients from natural resources as a greener alternative for fossil solvent. In the present work, 27 different NaDESs were screened for the extraction of paeoniflorin (PF) and galloyl paeoniflorin (GPF) from Radix Paeoniae Rubra (RPR). After screening and extraction parameter optimization, the extraction yields of PF and GPF reached up to 182.8 mg/g and 77.4 mg/g with the selected NaDES, ChCl-Sor. Furthermore, the antioxidant activity in vitro and neuroprotectivity in vivo of the ‘ready-to-use’ extracts were evaluated comprehensively. Especially in vivo , the cerebral ischemic/ reperfusion injury model was established in rats and the protective effects of the RPR extracts were determined. The results not only proved that NaDES is a valuable green extraction media, but also indicated the safety and potential pharmaceutical application of NaDES based ‘ready-to-use’ extracts from medical plants.

Dexamethasone-loaded ROS-responsive poly(thioketal) nanoparticles suppress inflammation and oxidative stress of acute lung injury

Zihe Zhai,Wei Ouyang,Yuejun Yao,Yuqi Zhang,Haolan Zhang,Feng Xu,Changyou Gao

Acute lung injury (ALI) is associated with excessive inflammatory response, leading to acute respiratory distress syndrome (ARDS) without timely treatment. A fewer effective drugs are available currently to treat the ALI/ARDS. Herein, a therapeutic nanoplatform with reactive oxygen species (ROS)-responsiveness was developed for the regulation of inflammation. Dexamethasone acetate (Dex) was encapsulated into poly(thioketal) polymers to form polymeric nanoparticles (NPs) ( [email protected] ). The NPs were composed of poly(1,4-phenyleneacetonedimethylene thioketal) (PPADT) and polythioketal urethane (PTKU), in which the thioketal bonds could be cleaved by the high level of ROS at the ALI site. The [email protected] could accumulate in the pulmonary inflammatory sites and release the encapsulated payloads rapidly, leading to the decreased ROS level, less generation of pro-inflammatory cytokines, and reduced lung injury and mortality of mice. RNA sequencing (RNA-seq) analysis showed that the therapeutic efficacy of the NPs was associated with the modulation of many immune and inflammation-linked pathways. These findings provide a newly developed nanoplatform for the efficient treatment of ALI/ARDS.

Novel multifunctional dual-dynamic-bonds crosslinked hydrogels for multi-strategy therapy of MRSA-infected wounds

Pengpeng Deng,Xiao Liang,Feixiang Chen,Yun Chen,Jinping Zhou

Wound healing is a complex dynamic process involving hemostasis, inflammation, proliferation and remodeling stages. It is urgent need to develop multifunctional hydrogels to treat multiple links of wound healing. In this work, we designed novel multifunctional dual-dynamic-bonds crosslinked hydrogels through the electrostatic interaction between phenylboric acid and the amino group of chitosan, and the dynamic borate ester bond between phenylboric acid and the catechol structure of PDA-modified CNTs. This dual dynamic cross-linking mechanism endowed hydrogels with very fast self-healing properties, which could quickly form a whole dressing after being injected into the wound site. The phenylboric acid and catechol structures endowed hydrogels with excellent adhesion properties, the adhesion strength to pig skin was up to 27.6 kPa, could be used for hemostasis in vivo . Moreover, hydrogels displayed the advantages of injectable, rapid shape adaptation, antioxidant, biocompatibility, blood compatibility, electrical conductivity and photothermal antibacterial properties. In the MRSA-infected wound model, hydrogels could effectively reduce the expression level of wound inflammatory factors, accelerate collagen deposition, epithelial tissue and vascular regeneration, and thus promote wound healing. This work provides a new idea for the design of multi-strategy therapy for wound healing and has broad clinical application prospects.

Green Synthesis of Robust Selenium Nanoparticles via Polysaccharide–Polyphenol Interaction: Design Principles and Structure–Bioactivity Relationship

Jiaojiao Zhou,Die Zhang,Xuqin Lv,Xiaoqing Liu,Wei Xu,Lei Chen,Jie Cai,Zia-ud Din,Shuiyuan Cheng

Selenium is an essential micronutrient and is well known for its role in regulating metabolism. However, it is also highly toxic in some of its forms and above certain concentration levels. Amorphous selenium nanoparticles (SeNPs) are extremely unstable and convert into non-bioactive or gray elemental selenium with crystal-type structures due to aggregation, which limits its antioxidant and anti-tumor activities. To overcome these limitations, monodisperse SeNPs were obtained by stabilizing SeNPs with glucan and rosmarinic acid (RA) via Se–O bonds and polysaccharide–polyphenol interactions. The prepared glucan-RA-SeNPs were found much more stable than both SeNPs and glucan-stabilized SeNPs. Meanwhile, glucan-RA-SeNPs displayed enhanced bioactivities including an improved free radical scavenging ability and an increased cytotoxicity toward cancer cells. The underlying mechanism might be related to caspase activation by glucan-RA-SeNPs, thus overproducing intracellular reactive oxygen species and eventually resulting in cancer cell apoptosis. Our work proposes a feasible strategy for improving the structure–bioactivity of SeNPs through polysaccharide–polyphenol interaction.

Investigation of the mechanism of phytate-modified biochar-catalyzed persulfate degradation of Ponceau 2R

Hua Li,Cheng Taozhi,Liang Zhiyong,Wei Ting

The mechanism of oxidation and degradation effect of phytate-modified biochar catalyzed persulfate on Ponceau 2R was investigated. Chemical-structural properties of phytate-modified biochar, such as surface morphology and surface oxygen-containing functional groups were characterized. The results suggest that modified biochar has better oxidation performance than unmodified biochar, and the modified biochar generated at 500 ℃ pyrolysis temperature can catalyze peroxymonosulfate (PMS) system with high efficiency, in large pH and temperature scope. And the degradation mechanism of Ponceau 2R by biochar-catalyzed PMS generation (BC-PMS) system was researched. It revealed that PBC300 (phytate-modified biochar pyrolyzed at 300 °C), PBC500 (phytate-modified biochar pyrolyzed at 500 °C), and PBC700 (phytate-modified biochar pyrolyzed at 700 °C) may have metaphosphoric acid linked to oxygen atoms and metaphosphoric acid linked in a bridging manner on the surface of biochar, catalyzing the production of hydroxyl radicals by PMS. PBC700 catalyzes the production of singlet oxygen by PMS through its structural defects, and singlet oxygen is the main catalytic product of PBC700.

Surface Coating of Pulmonary siRNA Delivery Vectors Enabling Mucus Penetration, Cell Targeting, and Intracellular Radical Scavenging for Enhanced Acute Lung Injury Therapy

Jiaojiao Zhu,Man Guo,Yanhui Cui,Yingcai Meng,Jinsong Ding,Wenbin Zeng,Wenhu Zhou

Pulmonary delivery of anti-inflammatory siRNA presents a promising approach for localized therapy of acute lung injury (ALI), while polycationic vectors can be easily trapped by the negatively charged airway mucin glycoproteins and arbitrarily internalized by epithelial cells with nontargetability for immunological clearance. Herein, we report a material, the dopamine (DA)-grafted hyaluronic acid (HA–DA), coating on an anti-TNF-α vector to address these limitations. HA–DA was simply synthesized and facilely coated on poly(β-amino ester) (BP)-based siRNA vectors via electrostatic attraction. The resulting HA–DA/BP/siRNA displayed significantly enhanced mucus penetration, attributable to the charge screen effect of HA–DA and the bioadhesive nature of the grafting DA. After transmucosal delivery, the nanosystem could target diseased macrophages via CD44-mediated internalization and rapidly escape from endo/lysosomes through the proton sponge effect, resulting in effective TNF-α regulation. Meanwhile, DA modification endowed the coating material with robust antioxidative capability to scavenge a broad spectrum of reactive oxygen/nitrogen species (RONS), which protected the lung tissue from oxidative damage and synergized with anti-TNF-α to inhibit a cytokine storm. As a result, a remarkable amelioration of ALI was achieved in a lipopolysaccharide (LPS)-stimulated mice model. This study provides a multifunctional coating material to facilitate pulmonary drug delivery for the treatment of lung diseases.

A preliminary study on polysaccharide extraction, purification, and antioxidant properties of sugar-rich filamentous microalgae Tribonema minus

Huo Shuhao,Wang Huiying,Chen Jing,Hu Xinjuan,Zan Xinyi,Zhang Cunsheng,Qian Jingya,Zhu Feifei,Ma Haile,Elshobary Mostafa

The total polysaccharide content of the sugar-rich filamentous microalga  Tribonema minus (Xanthophyceae, Tribonematales) under 2% CO 2 was up to 50%. The optimal conditions for the extraction of polysaccharide by hot water extraction-ethanol deposition were as follows: A maximum polysaccharide yield of 27.25% was obtained under the optimized conditions of extraction temperature 80°C, extraction time of 2 h, pH 9, and liquid to raw material 0.5:1 mL mg −1 . The crude polysaccharides were mainly acidic polysaccharides containing sulfate groups and uronic acid, and the contents of which were 14.57% and 42.21 mg g −1 , respectively. The crude polysaccharide was mainly composed of rhamnose, arabinose, xylose, mannose, glucose, and galactose with a molar ratio of 0.31:0.26:0.40:0.42:1.00:0.47. In addition, the polysaccharide had a potential antioxidant activity in vitro. The scavenging effect and reducing power of polysaccharides increased with the increase of polysaccharide concentration. The scavenging effect was 56.11%, 75.6%, and 61.89% for 1,1-diphenyl-2-picrylhydrazyl free radical (DPPH), superoxide, and hydroxyl radicals at a concentration of 2 mg L −1 , respectively. Hence, T. minus polysaccharides could be a source of natural antioxidants that might be used in foods or pharmaceuticals.

Antioxidation of a proteoglycan from Ganoderma lucidum protects pancreatic β-cells against oxidative stress-induced apoptosis in vitro and in vivo

Yanna Pan,Shilin Yuan,Yilong Teng,Zeng Zhang,Yanming He,Ying Zhang,Haohui Liang,Xiao Wu,Jiaqi Li,Hongjie Yang,Ping Zhou

Oxidative stress is one of the major factors in induction of pancreatic β-cell apoptosis and diabetes. Here, we investigated systematically the roles of a proteoglycan (namely, FYGL ) from Ganoderma lucidum in protection and repair of pancreatic β-cells against oxidative stress-induced injury and apoptosis on molecular, cellular and animal basis. FYGL in vitro had antioxidant activity in terms of scavenging of free radicals and reduction power. FYGL improved cells viability, insulin secretion, redox indicator expressions, and mitochondrial membrane potential in H 2 O 2 -induced INS-1 cell via regulating the activations of apoptosis-related mitogen-activated protein kinases (MAPK) and nuclear factor kappa B (NF-κB) pathways as well as the insulin secretion-related pathway. Thrillingly in vivo , FYGL repaired the injured pancreas, reduced the pancreatic β-cells apoptosis, and improved insulin secretion because of regulating the balance of oxidation-reduction, therefore well managed blood glucose in db/db diabetic mice. These results demonstrated that FYGL is promising to be used as a novel natural remedy for protection of pancreatic β-cells against oxidative stress in diabetes treatment.

Potential of phenolic compounds in Ligustrum robustum (Rxob.) Blume as antioxidant and lipase inhibitors: Multi-spectroscopic methods and molecular docking

Hao-Xiang Gao,Heng-Yu Liang,Nan Chen,Bi Shi,Wei-Cai Zeng

Ligustrum robustum (Rxob.) Blume is traditionally served as a functional tea in China. In this work, the antioxidant activities of L. robustum (Rxob.) Blume extract (LRE) were evaluated and its inhibitory effect and mechanism on pancreatic lipase were further investigated. With the high contents of phenols (139.70 ± 1.41 mg gallic acid equivalent/g extract) and flavonoids (326.46 ± 7.36 mg rutin equivalent/g extract), LRE showed significant antioxidant activities ( p < 0.05) for scavenging free radicals and hydrogen peroxide, inhibiting lipid peroxidation and providing strong reducing power. Meanwhile, LRE displayed remarkable inhibitory activity on pancreatic lipase with a low half-effective inhibitory concentration (IC 50 ) of 2.469 ± 0.005 mg/ml which was further determined as non-competitive inhibition. The spectroscopic results showed that LRE inhibited the activity of pancreatic lipase by modifying the tertiary and secondary structures of lipase. Moreover, four phenolic compounds (acteoside, lipedoside A, oleuropein and ligurobustoside C) were identified from LRE by the high performance liquid chromatography-quadrupole- time of flight-mass spectrometry. In addition, according to molecular docking analysis, the four phenols could interact with pancreatic lipase by hydrogen bonds, so as to change the spatial structure of pancreatic lipase and inhibit its catalytic activity. The present results suggest that LRE not only exhibits strong antioxidant capacity but possesses effectively inhibitory activity on pancreatic lipase, which might have the potential to be developed as functional food and nutraceuticals for the prevention of metabolic diseases. Practical Application Ligustrum robustum (Rxob.) Blume extract has been confirmed to possess antioxidant activity and lipase inhibitory activity, which indicates that the L. robustum extract has the potential to prevent oxidative stress and regulate fat metabolism. This work suggests that L. robustum extract can be served as a novel resource to prepare nutraceuticals and functional food in food industries.
