Triazine-based porous organic polymer as pipette tip solid-phase extraction adsorbent coupled with HPLC for the determination of sulfonamide residues in food samples

Yuhan Ning,Yixing Ye,Wanliang Liao,Yang Xu,Weiping Wang,Ai-jun Wang

In this work, triazine-based porous organic polymer (TAPT-BPDA) synthesized by simple solvothermal method with stable chemical properties was employed as pipette tip solid-phase extraction (PT-SPE) adsorbent for the extraction of sulfonamides for the first time. Under the optimal conditions, good linearities (1–300 μg L −1 , R 2  ≥ 0.9987) and low limits of detection (0.10–0.28 μg L −1 ) were obtained. The recoveries of sulfonamides in meat, egg and milk samples were in the range of 76.1–114.0 %. The prepared TAPT-BPDA showed good reusability with the recoveries of sulfonamides remained above 80.0 % after eight recycles. The adsorption mechanism between SAs and adsorbent might be the combined effects of electrostatic interactions, hydrogen bonding and π-π interactions. The results demonstrated potential applications of a TAPT-BPDA-based PT-SPE-HPLC method for the analysis of trace sulfonamide residues in food samples.

Preparation of a Novel Resin Based Covalent Framework Material and Its Application in the Determination of Phenolic Endocrine Disruptors in Beverages by SPE-HPLC

Yunjie Ma,Yang Ruan,Xin Gao,Hang Cui,Wei Zhang,Shaoyan Wang

A new type of economical covalent organic framework material(COF), namely resin based covalent organic framework material, was prepared by combining resin and covalent organic framework material by hydrothermal synthesis, which was based on the preparation of traditional COF material(TpBD COF). The properties of the material and covalent organic framework material were compared in the way of characterization, and the possible reaction mechanism was analyzed. The solid phase extraction separation (SPE) ability of this material for four kinds of phenolic endocrine disrupting compounds (bisphenol F, bisphenol A, octylphenol and nonylphenol) in beverage samples was investigated. The results showed that the prepared COF materials had abundant internal channels, ordered structure, large specific surface area (TpBD COF: 814.6 m2/g and resin based COF: 623.9 m2/g) and good thermal stability (pyrolysis temperature was 443 °C and 437 °C, respectively). Solid phase extraction experiments demonstrated that the two COF materials as adsorbent of solid phase extraction column had ideal adsorption separation effect and good anti-interference ability, and had strong anti-interference ability. The SPE effect was superior to the traditional solid phase extraction column. The precision RSD of this method was less than 3%. This SPE method had high recovery and could be reused (carbonated beverage: 98.18–102.18% and beverage: 98.52–101.79%), In addition, the recovery of the material did not change significantly in the 50 cycles of solid phase extraction, indicating that the material had good stability and could be reused, which could meet the requirements for the detection and analysis of trace pollutants in environmental samples. The resin based COF material prepared in this study could reduce the cost of monomer uses and provide a possibility for its industrial production. At the same time, as an efficient SPE adsorbent, it also provided a new research scheme for the enrichment of trace phenolic endocrine disruptors in beverage samples. Keywords:resin based covalent organic framework materials;phenolic endocrine disruptors;solid phase extraction;trace analysis

Magnetic carbon nanotubes-molecularly imprinted polymer coupled with HPLC for selective enrichment and determination of ferulic acid in traditional Chinese medicine and biological samples

Lulu Fu,Qiu Chen,Jintao Chen,Liqin Ren,Lan Tang,Weiguang Shan

A molecularly imprinted polymer (MIP) with magnetic carbon nanotubes (MCNTs) as carrier was synthesized and used for the enrichment and determination of ferulic acid (FA) by high-performance liquid chromatography (HPLC). The morphology and structure of the FA magnetic carbon nanotubes-molecularly imprinted polymers (MCNTs@FA-MIPs) were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy, and X-ray diffraction. The results demonstrated that the prepared MCNTs@FA-MIPs had excellent magnetic properties and uniform appearance. The adsorption properties of the novel MIP were studied by kinetic, and isothermal adsorption experiments. The results showed that the MCNTs@FA-MIPs exhibited relatively good uptake kinetics (equilibrium time, 2 h), high adsorption capacity (50 mg⋅g −1 ), fast separation, and good selectivity for the template molecule with a separation factor α of 1.73. The MCNTs@FA-MIPs combined with HPLC were successfully applied to the separation, enrichment, and determination of FA in a Ligusticum chuanxiong extract and in plasma of rats which had been administered with Taitai beauty essence. The recoveries for FA were 95.53–100.03 % with relative standard deviations (RSDs) less than 5.5%. The results confirmed that the proposed MCNTs@FA-MIPs offered efficient extraction of FA from traditional Chinese medicinal preparations and blood samples and with high specificity.

A sensitive HPLC-FLD method for the quantitative determination of 3-chloro-1,2-propanediol by pre-column fluorescence derivatization with 9-(2-Hydroxypropyl)adenine

Yan Du,Jinhua Hu,Zhixiong Hu,Weinong Zhang,Yutang Qi,Yanpeng Zhang,Xiaomin Li,Yi Liu

In this paper, a sensitive method for the determination of 3-chloro-1,2-propanediol (3-MCPD) by high performance liquid chromatography-fluorescence detection (HPLC-FLD) was established, in which 9-(2-Hydroxypropyl) adenine (HP-Ade) was selected as the fluorescent derivatization reagent because of its good water solubility and hydrophobicity. The principle of the method lies in the origination of chloroacetaldehyde with the periodate oxidation of 3-MCPD and its' fluorescence derivatization by HP-Ade to produce a strong fluorescent substance, name as 1-N6-vinylidene-9-(2-hydroxypropyl)adenine (ε-HP-Ade), used for quantitative determination of 3-MCPD by HPLC-FLD. Some mobile phase conditions and incubation parameters related with the method features were studied and optimized. Under the optimal conditions, the method shows high sensitivity, good linearity (r2 = 0.999) and good repeatability (RSD% between 1.04% and 2.55%), the detection limit (LOD) and quantification limit (LOQ) were 0.02 ng/mL and 0.06 ng/mL respectively. It has been successfully applied for the determination of various edible vegetable oil samples, the chromatogram shows symmetrical peak shape, perfect separation and very “clean” profiles. In comparison with gas chromatography-mass spectrum (GC-MS), the pretreatment process of the proposed method was totally operated in aqueous phase, the instrument requirements and cost are relatively lower, thus it will provide another choice for the routine analysis of 3-MCPD.

Application of core–satellite polydopamine-coated Fe3O4 nanoparticles–hollow porous molecularly imprinted polymer combined with HPLC-MS/MS for the quantification of macrolide antibiotics

Wenjia Fan,Dan Yang,Nan Ding,Puyu Chen,Lei Wang,Guanwei Tao,Feng Zheng,Shunli Ji

Core–satellite-structured magnetic nanosorbents (MNs) used for the selective extraction of macrolide antibiotics (MACs) were prepared in this study. The MNs (core–satellite polydopamine-coated Fe3O4 nanoparticles–hollow porous molecularly imprinted polymer) consisted of polydopamine-coated Fe3O4 nanoparticles (Fe3O4@PDA) “core” linked to numerous hollow porous molecularly imprinted polymer (HPMIP) “satellites” with bridging amine functional groups. It is worth mentioning that HPMIPs act as “anchors” for selectively capturing target molecules. Polymers were characterized using TEM, SEM, FT-IR, VSM, and TGA and applied as magnetic dispersive solid-phase extraction (MDSPE) sorbents for the enrichment of trace MACs from a complex food matrix prior to quantification by HPLC-MS/MS. Nanocomposites revealed outstanding magnetic properties (36.1 emu g−1), a high adsorption capacity (103.6 μmol g−1), selectivity (IF = 3.2), and fast kinetic binding (20 min) for MACs. The multiple advantages of the novel core–satellite-structured magnetic molecularly imprinted nanosorbents were confirmed, which makes us believe that the preparation method of the core–satellite MNs can be applied to other fields involving molecular imprinting technology.

Polyamine-Modified Magnetic Graphene Oxide Nanocomposites and HPLC-MS/MS Allow the Determination of Two Indolic Derivatives in Strong-Aroma Types of Base Baijiu

Wei Dong,Ruonan Guo,Wenxuan Lin,Miao Liu,Caihong Shen,Xiaotao Sun,Baoguo Sun,Bo Deng

Simultaneous detection of 3-indoleacetic acid (IAA) and 3-indolepropionic acid (IPA) in strong-aroma types of base Baijiu (base SAB) is crucial for elucidating the metabolic pathway of 3-methylindole during the base SAB brewing. Herein, a novel magnetic poly(allylamine)-modified graphene oxide (GO@PAA@Fe3O4) was synthesized as extraction sorbent, followed by high performance liquid chromatography mass spectrometry HPLC-MS/MS. As a surface modifier of GO, the introduction of PAA and Fe3O4 provided more adsorption sites for IAA and IPA, mainly through the generation of H-bonding sites. Moreover, modified by an activation step, the capacity of the activated GO@PAA@Fe3O4 for the adsorption of IAA and IPA was 2.1–3.4 times higher than that of unactivated material. The adsorptions of IAA and IPA on GO@PAA@Fe3O4 were fitted to the pseudo-second-order kinetic model and Langmuir isotherm, respectively. Under the optimal conditions, IAA and IPA were determined in 16-base SAB for the first time, and their concentrations ranged from 0.6 to 11.3 and 0.7 to 18.7 μg/L, respectively.

Monodispersed mesoporous SiO2@metal-organic framework (MSN@MIL-101(Fe)) composites as sorbent for extraction and preconcentration of phytohormones prior to HPLC-DAD analysis

Qin Peige,Zhu Wenli,Han Lizhen,Zhang Xiaowan,Zhao Bing,Zhang Xuebin,Lu Minghua

The monodispersed mesoporous SiO 2 @metal-organic framework (MSN@MIL-101(Fe)) composites were prepared by grafting MSN-NH 2 onto MIL-101(Fe) particles with a solvothermal method. The adsorption ability of the composites was greatly improved compared to that of pristine MSNs or MIL-101(Fe) for phytohormones (Phys). The MSN@MIL-101(Fe) composites were characterized by scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, X-ray diffraction, energy dispersive spectrometer, and mapping analysis. Using MSN@MIL-101(Fe) composites as sorbent, a dispersive solid-phase extraction procedure (dSPE) was developed to extract three endogenous Phys (abscisic acid (ABA), indole-3-aceticacid (IAA), and indole-3-butyric acid (IBA)) and two exogenous Phys (1-naphthylacetic acid (1-NAA) and 2-naphthylacetic acid (2-NAA)) prior to HPLC-DAD analysis. The experimental parameters including sample volume, sorbent amount, adsorption time, adsorption pH, desorption time, and desorption solvent on extraction efficiency were optimized and evaluated. Under optimized conditions, the working range of 0.08 to 0.45 ng mL −1 with enrichment factors from 144 to 207 were achieved. The linear range is 0.75–200 ng mL −1 for IAA, 0.20–200 ng mL −1 for ABA, and 1.0–200 ng mL −1 for IBA, 1-NAA, and 2-NAA. With MSN@MIL-101(Fe) as sorbent for extraction of Phys and determination by HPLC-DAD, two endogenous Phys (IAA and ABA) were detected from mung bean sprouts which were made in a laboratory, and the results were further confirmed by high-resolution mass spectrometry. The composites can be applied to extract other small molecules, which have similar chemical structures with Phys in biological, environmental, and food samples. Schematic presentation of a dispersive solid-phase extraction using monodispersed mesoporous SiO 2 @metal-organic framework composites (MSNs@MIL-101(Fe)) as the sorbent for extraction, clean-up, and preconcentration of phytohormones in mung bean sprouts prior to HPLC-DAD analysis.

Effects of temperature and heating time on the formation of aldehydes during the frying process of clam assessed by an HPLC-MS/MS method

Zhong-Yuan Liu,Da-Yong Zhou,Ao Li,Man-Tong Zhao,Yuan-Yuan Hu,De-Yang Li,Hong-Kai Xie,Qi Zhao,Xiao-Pei Hu,Jiang-Hua Zhang,Fereidoon Shahidi

This study aimed at investigating the formation and accumulation of 16 reactive aldehydes in clam ( Ruditapes philippinarum ) during oil frying in both the tissue and the oil using an HPLC-ESI-MS/MS methodology. After processing, the accumulation of acrolein, crotonaldehyde, pentanal, trans -2-hexenal, hexanal, trans , trans -2,4-heptadienal, heptanal, nonanal, trans , trans -2,4-decadienal and 4-hydroxy-2-nonenal was most noticeable in both fried clam and frying oil. Most of the aldehyde species showed a time- and temperature-dependent manner of formation and accumulation during frying due to continuous oxidative degradation under conditions employed. However, several species of aldehyde such as acrolein and trans -2-pentenal slightly decreased at higher temperatures and/or longer frying times, which may be due to the imbalance toward disappearance of aldehydes resulting from their evaporation under the extreme conditions. Presence of natural polyphenols in bamboo leaves significantly prevented the formation of aldehydes in both fried clam and frying oil due to their antioxidant activity ( P  < 0.05).

CD147-SNAP-tag-CMC/HPLC-MS two-dimensional coupling system: a universal platform for screening active components targeting CD147 for anti-liver cancer applications

Wenhao Chen,Haoyun Bai,Quan Zhang,Jingjing Yang,Shengyu Zhou,Du Feng,Shengli Han

Established a universal screening platform for CD147-targeted therapeutics. • Integrated the CMC-HPLC system with mass spectrometry (MS) for the identification of unknown compounds. • Two bioactive constituents from Magnolia officinalis, directly interact with CD147. Hepatocellular carcinoma (HCC) is one of the most prevalent digestive malignancies, with CD147 emerging as a promising therapeutic target for HCC. To date, no natural bioactive components targeting CD147 for anti-HCC therapy have been explicitly reported. Given the low toxicity and favorable efficacy of natural small-molecule drugs, identifying and investigating CD147-targeted natural agents is imperative. In this study, based on cell membrane chromatography (CMC) and SNAP-tag protein labeling technology, we constructed a CD147-SNAP-tag-CMC/HPLC-MS two-dimensional coupled system to screen CD147-binding components from Magnolia officinalis , followed by ligand-receptor affinity verification, in vitro pharmacological evaluation, and downstream molecular mechanism validation. Two bioactive compounds, honokiol and magnolol, were successfully identified: both exhibit strong CD147 binding affinity, effectively suppress HCC cell migration, and exert their functions by regulating the CD147/integrin-FAK-paxillin pathway as confirmed by Western blotting. Collectively, the established two-dimensional system serves as a universal platform for CD147-targeted component screening. This study is the first to reveal the strong CD147 affinity of honokiol and magnolol and their anti-migratory effect on HCC cells via modulating the aforementioned pathway.

A novel sample pretreatment of nanofiber-packed solid-phase extraction of aflatoxin B1, B2, G1 and G2 in foods and simultaneous determination with HPLC

Yunzheng Wang,Lanling Chu,Jiansong Qu,Bo Ding,Xuejun Kang

A novel analytical method based on nanofiber-packed solid-phase extraction (PFSPE) combined with HPLC-FLD has been successfully developed and applied to determine aflatoxin B1, B2, G1, G2 in foods. In order to effectively adsorb analytes from complex food matrices, four types of nanofibers based on skeleton of polystyrene–polyvinyl pyrrolidone were prepared by electrospinning and applied as adsorbent to home-made solid-phase extraction device. The effects of type of nanofibers, pH of sample, type and concentration of salt ion, type of activation solvent, type and volume of eluent, amount of nanofibers, and extraction time on the adsorption/desorption were investigated and optimized. Under optimal conditions, the method showed satisfactory linear relationship, with limits of detection (LODs) of 0.07–0.17 ng g −1 , intra-day and inter-day RSDs for spiked samples of 1.3–8.0 % and 1.9–5.8 %, and absolute recoveries in the range of 60.1–98.4 %. The results presented the great potential to be utilized to determine AFs in foodstuffs.

A novel analytical method based on HPLC-PDA coupled post-column derivatization to evaluate the ability to inhibit tyrosine nitration in lotus leaf extracts

Yin Qian,Xi Chen,Jin Qi,Xuming Liu

Protein tyrosine nitration plays a key role in many inflammatory and cardiovascular diseases and diabetes. Many natural products are used to treat these diseases through their ability to potentially interfere this reaction. Here, we describe a novel method to provide active fingerprinting of inhibition of tyrosine nitration by natural products based on post-column tyrosine nitration reaction analysis using high-performance liquid chromatography coupled to a photometric diode array. Results indicated that lotus leaf extracts exhibited obvious inhibitory activity against tyrosine nitration by peroxynitrite, and that chemical and active fingerprints were simultaneously established, with the active fingerprints indicating the active compounds of the lotus leaves. Additionally, flavonoids were screened as the principal active compounds involved in inhibiting tyrosine nitration in the lotus leaf extracts, with quercetin-3-O-glucuronide and quercetin-3-O-glucoside exhibiting the greatest contributions. Moreover, our results suggested that lotus leaves from three regions (Nanjing, Suzhou, and Hangzhou) exhibited the best inhibitory activity. These findings indicated the usefulness of this method for screening active compounds involved in inhibiting protein tyrosine nitration, and that similar strategies can likely be applied to evaluate the inhibitory activity against tyrosine nitration of other natural products.

Enrichment and sensitive determination of phthalate esters in environmental water samples: A novel approach of MSPE-HPLC based on PAMAM dendrimers-functionalized magnetic-nanoparticles

Yalin Wu,Qingxiang Zhou,Yongyong Yuan,Hongyuan Wang,Yayan Tong,Yali Zhan,Xueying Sheng,Yi Sun,Xianqi Zhou

Phthalate esters (PAEs) are an important kind of environmental endocrine disrupting chemicals , and have attracted great attention in environmental field. Present study described a new method for rapid and sensitive determination of PAEs including dibenzyl phthalate (DPhP), dibutyl phthalate (DnPP), and dicyclohexyl phthalate (DCHP) from aqueous matrices based on magnetic solid-phase extraction. Polyamidoamine (PAMAM) dendrimers-grafted magnetic-nanoparticles were synthesized and characterized, and the expected integration of more multifunctional sites of PAMAM dendrimers and rapid separation property was utilized for method development. To achieve the best extraction efficiency, several important parameters were optimized including the dosage of the adsorbent, sample pH, kind and volume of eluent , extraction time, desorption time, ionic strength . Under the optimal conditions, three phthalate esters were well enriched and simultaneously determined by high performance liquid chromatography with variable wavelength detector (VWD). Excellent linearities were observed in the range of 0.1–600 μg L −1 for DPhP and DnPP and 0.5–600 μg L −1 for DCHP, and all correlation coefficients (R 2 ) were larger than 0.997. The limits of detection (LODs, S/N = 3) were ranged from 0.025 to 0.16 μg L −1 . The spiked recoveries of PAEs in real water samples were in the range of 93.5–101.8% with satisfied relative standard deviations (RSDs) ranging from 0.9 to 4.1%. The prepared magnetic materials have shown good adsorption capability for PAEs and the developed method earned merits such as high sensitivity, simplicity, rapidness and environmental friendliness , which can be used as a robust alternative tool for monitoring PAEs in water samples.
