Background. Liangxue Tongyu prescription (LTP) is a commonly used formula for acute intracerebral hemorrhage (AICH) in clinical practice that has significant ameliorative effects on neurological deficits and gastrointestinal dysfunction, yet the mechanism remains elusive. The aim of this study was to investigate the pathway by which LTP alleviates brain damage in AICH rats. Methods. The AICH rat models were established by autologous caudal arterial blood injection. The neurological function scores were evaluated before and after treatment. The water content and the volume of Evans blue staining in the brain were measured to reflect the degree of brain damage. RT-PCR was used to detect the inflammatory factors of the brain. Western blotting was used to detect the expression of the tight junction proteins zonula occludens 1 (ZO-1), occludin (OCLN), and claudin (CLDN) in the brain and colon, followed by mucin 2 (MUC2), secretory immunoglobulin A (SIgA), and G protein-coupled receptor 43 (GPR43) in the colon. Flow cytometry was used to detect the ratios of helper T cells 17 (Th17) and regulatory T cells (Treg) in peripheral blood, and the vagus nerve (VN) discharge signals were collected. Results. LTP reduced the brain damage of the AICH rats. Compared with the model group, LTP significantly improved the permeability of the colonic mucosa, promoted the secretion of MUC2, SigA, and GPR43 in the colon, and regulated the immune balance of peripheral T cells. The AICH rats had significantly faster VN discharge rates and lower amplitudes than normal rats, and these abnormalities were corrected in the LTP and probiotics groups. Conclusion. LTP can effectively reduce the degree of brain damage in AICH rats, and the mechanism may be that it can play a neuroprotective role by regulating the function of the intestinal mucosal barrier.

The slow release of hydrogen sulfide has been shown to be beneficial to plants by protecting them from environmental stressors, increasing germination, and extending the lifetime of harvested fruits. A major challenge in this field is controlling the amount and location of release of hydrogen sulfide so that it is available for use by plants at optimal amounts. This article reports a dual method to release hydrogen sulfide near the roots of plants by controlling its release using the hydrolysis of a dithiophosphate and the degradation of poly(lactic acid) [PLA]. Di(t-butanol)dithiophosphate phenylethylamine (tBDPA) was dissolved in a solution of PLA, and the solvent was allowed to evaporate. The resulting solid was crushed in a blender and separated into microparticles with two different size distributions of 250–500 or 500–2000 μm. The microparticles were characterized by powder X-ray diffraction to measure the presence of microcrystals of tBDPA within PLA, and images obtained using scanning electron microscopy with energy dispersive X-ray analysis confirmed the presence of these crystals. Microparticles of tBDPA loaded within PLA were characterized for their release of phosphorus and hydrogen sulfide, which both showed a burst release within 3 days, followed by a steady release. Radish plants grown with microparticles of PLA loaded with tBDPA had up to a 141% increase in harvest yield compared to plants grown in the presence of free tBDPA not loaded into PLA, PLA microparticles without tBDPA, and control plants grown without PLA or tBDPA. These experiments showed that loading hydrogen sulfide-releasing chemicals into PLA is a promising method to improve the effect of hydrogen sulfide on plants.

The health risks posed by chloroneb (CHL), a representative
fungicide,
in agricultural products and the environment necessitate rapid detection
methods. Here, computational chemistry-aided rational hapten design
yielded a high-performance monoclonal antibody against CHL (IC 50 = 2.63 ng/mL), guided by analysis of its spatial conformation
and electronic properties. A dual-T-line gold nanoparticle-based immunochromatographic
assay (AuNP-ICA) was established, achieving 2.6-fold higher sensitivity
and a 2.9-fold higher upper detection limit compared with traditional
single-T-line formats. The assay enables rapid (<20 min) on-site
detection of CHL in lake water, soil, tea, soybeans, and mangoes,
with limits of detection ranging from 2.51 to 4.54 μg/kg. Recovery
and blind sample analyses demonstrated excellent agreement with instrumental
methods. This work not only provides a practical tool for field detection
of CHL but also establishes a generalizable technological framework
that integrates rational hapten design with a dual-T-line configuration
for developing sensitive immunoassays targeting other small-molecule
contaminants.

Background T-2 toxin, a highly toxic trichothecene mycotoxin contaminating cereal grains, poses serious health threats. However, its specific effects on hepatic drug-metabolizing enzymes (DMEs) remain poorly characterized. This study aimed to investigate the impact of T-2 toxin on key cytochrome P450 (CYP) and carboxylesterase (CES) enzymes in mouse liver. Methods Male C57BL/6 mice were randomly divided into control and T-2 toxin-treated groups, receiving either normal saline or T-2 toxin (0.2 mg/kg) by oral gavage daily for four weeks. Histopathological and ultrastructural alterations were assessed by hematoxylin and eosin staining and transmission electron microscopy. The activity, mRNA, and protein levels of target enzymes in liver tissues or AML12 cells were determined via enzymatic assays, quantitative real-time PCR (qRT-PCR), and western blotting. Transcriptomic profiling (RNA-seq) was performed to identify differentially expressed genes (DEGs) and enriched pathways. Results T-2 toxin exposure induced hepatocellular injury, including focal necrosis and mitochondrial damage. It significantly altered the activity and expression of multiple phase I DMEs, notably CYP1A2, CYP2B10, CYP3A11, and CES isoforms, at both transcriptional and protein levels. Transcriptomic analysis identified 277 DEGs, predominantly enriched in xenobiotic and drug metabolism pathways, including drug metabolism-cytochrome P450 and xenobiotic metabolic process. Conclusion T-2 toxin induces hepatotoxicity and concurrently alters the expression and activity of key hepatic DMEs. These findings provide insight into T-2 toxin-induced disturbances in xenobiotic metabolism and may contribute to future studies on metabolic biomarkers and toxicological mechanisms.

Bone defect repair remains a significant challenge faced in clinical practice. Bone tissue engineering has emerged as an effective approach for bone defect repair due to its ample donor source and remarkable repair efficacy. In this study, we combined Abaloparatide (ABL) with bone marrow-derived mesenchymal stem cells (BMSCs) and utilized methacrylated gelatin (GelMA) as a matrix to successfully develop a bio-scaffold with dual activity (ABL@GelMA/BMSCs). BMSCs regulate the bone microenvironment by secreting cytokines, which promote bone repair through a paracrine mechanism. The addition of ABL not only facilitated the osteogenic differentiation and paracrine function of BMSCs but also significantly enhanced the bone induction capability of the scaffold, as evidenced by the marked upregulation of osteogenic markers (ALP, RUNX2, OCN). Furthermore, this bio-scaffold was able to upregulate the Foxp3 gene, inducing Foxp3+ regulatory T cells, thereby modulating the local immune microenvironment to promote bone formation. After implantation of this bio-scaffold into a critical-sized cranial defect model, histological results demonstrated a higher bone mineral density (BMD), and CT reconstruction indicated good healing of the defect. In summary, this bio-scaffold exhibits a powerful capacity for bone defect repair while improving the bone microenvironment through the synergistic modulation of regulatory T cells, providing a new strategy for bone defect repair.

Highly concentrated boron is widespread in salt lake brine and nuclear wastewater. Treatment of these aqueous solutions involves the solubility of extensive boron-containing minerals. Recently, we concentrated on the temperature-dependent solubility of the H3BO3 + salt + water system. Here, we systematically determined the ternary solubility isotherms in the H3BO3 + RbCl + H2O and H3BO3 + NH4Cl + H2O systems at T = (273.15, 298.15, 323.15, 348.15 and 363.15) K, using an isothermal equilibrium method. The results showed that both ternary systems belonged to a simple eutectic case. The solubility isotherms consisted of the solubility branches of H3BO3(cr) and RbCl(cr) or NH4Cl(cr). Using a revised Pitzer–Simonson–Clegg (rPSC) model implemented in the ISLEC software, these ternary solubility data were represented by fitting the interaction parameters between H3BO3 and RbCl and H3BO3 and NH4Cl. With the help of these models, polythermal phase diagrams of the two ternary systems are predicted.

It has been proven that alcohols can regulate the performance of aviation fuels. In this work, the densities (ρ) and viscosities (η) for the ternary system of isopropylcyclohexane (1) + n-tridecane (2) + n-butanol (3) and the corresponding binary systems have been measured to understand the physical properties of endothermic hydrocarbon fuel with alcohol over the whole composition range at temperatures from 293.15 to 333.15 K under atmospheric pressure (p = 0.1 MPa). The Redlich–Kister equation has been used to fit the excess molar volumes (VmE) and viscosity deviations (Δη) of binary systems, while four semiempirical equations (Singh, Cibulka, Nagata–Tamura, and Redlich–Kister) have been applied to correlate VmE and Δη of the ternary system. For all binary systems, the values of VmE are positive. The values of Δη for binary systems containing n-butanol are all negative. For the ternary system, the values of VmE are all positive and the values of Δη are all negative over the entire concentration range. All results have been interpreted with the perspective of intermolecular interaction and structural effect.

Separating the mixture of the target product n-propyl acetate (NPAC) and the unreacted reactant n-propanol (NPA) is necessary during the production of NPAC. NPA and NPAC cannot be separated by simple distillation because the two could form an azeotrope under atmospheric pressure. Liquid–liquid extraction is an energy-saving and environment-friendly method for separating azeotropic mixtures. Ionic liquids (ILs) are promising alternatives to conventional extractants for separating azeotropes due to the advantages of designability and negligible vapor pressure. In this study, the liquid–liquid equilibrium (LLE) data for the ternary systems of NPAC + NPA + 1-butyl-3-methylimidazolium dihydrogen phosphate ([BMIM][H2PO4]) or 1-butyl-3-methylimidazolium thiocyanate ([BMIM][SCN]) or 1-butyl-3-methylimidazolium tetrafluoroborate ([BMIM][BF4]) were determined at 303.15 K and 101.3 kPa. The accuracy of the experimental LLE data was checked by the Bachman, Hand, and Othmer–Tobias equations. The selectivity and partition coefficient were calculated to evaluate the extraction capability of the selected extractants. The experimental LLE data were correlated with the nonrandom two-liquid model, and the binary interaction parameters were regressed. Finally, the separation mechanism of the three ILs was explored through σ-profile analysis.

In the study, the solubility of l-serine were measured in eight common pure solvents and binary mixed solvents (acetonitrile + water, isopropanol + water, methanol + water, and acetone + water) from 278.15 to 333.15 K under atmospheric pressure by gravimetric method (acetone was measured from 278.15 to 328.15 K). The solubility of l-serine increases with increasing temperature, and the order in eight common pure solvents is water > acetonitrile > methanol > acetone > ethyl acetate > ethanol > n-butanol > isopropanol. In binary mixed solvents, the solubility of l-serine is dependent on temperature and solvent composition. Also, the solubility of l-serine increases with increasing temperature, and it decreases as the concentration of acetonitrile, isopropanol, methanol, or acetone increases at a constant temperature. The experimental data in eight pure solvents were fitted by the modified Apelblat model, Buchowski–Ksiazaczak λh model. The modified Apelblat equation, combined nearly ideal binary solvent/Redlich–Kister (CNIBS/R–K) model, and Jouyban–Acree model were used to correlate the experimental data in four binary solvents. Among the four models, the modified Apelblat model is more suitable with a higher accuracy than the other three models. The experimental data can be used to optimize the industrial manufacturing process of l-serine.

Densities (ρ) and viscosities (η) of a ternary system cyclopropanemethanol (1) + 2,2,4-trimethylpentane (2) + decalin (3) and three corresponding binary systems were measured at temperatures from 293.15 K to 323.15 K and atmospheric pressure. Densities were obtained by using a vibrating-tube densimeter. Viscosities were determined by an automatic microviscometer based on the rolling ball principle. The excess molar volumes (VmE) and viscosity deviations (Δη) of the ternary system were derived from the experimental data and then were fitted to Clibuka, Nagata–Tamura and Redlich–Kister equation, respectively. The binary subsystems were correlated by Redlich–Kister equation. The values of VmE and Δη were used to discuss the nature of mixing behaviours between mixture components.

The enthalpies of dissolution of 2HNIW·HMX co-crystal in N,N -dimethylformamide at different temperatures were measured by a DC08-1 Calvet microcalorimeter under standard atmospheric pressure, and it is indicated that the dissolutions are exothermic process. The empirical formulae for the calculation of the molar enthalpy ( \(\Delta_{\text{diss}} H\) ) of dissolution, relative partial molar enthalpy ( \(\Delta_{\text{diss}} H_{\text{partial}}\) ), relative apparent molar enthalpy ( \(\Delta_{\text{diss}} H_{\text{apparent}}\) ), and enthalpy of dilution ( \(\Delta_{\text{dil}} H_{1,2}\) ) at 298.15 K are obtained. The differential enthalpies ( \(\Delta_{\text{dif}} H\) ) and kinetic equations describing the dissolution process at different temperatures are deduced. Furthermore, the apparent activation energy E = 10.54 ± 0.22 kJ mol -1 and pre-exponential constant A = 0.34 ± 0.03 s −1 of 2HNIW·HMX co-crystal are obtained. The standard molar Gibbs free energy of activation ( \(\Delta G_{ \ne }^{\theta }\) ) at different temperatures is 86.44 ± 0.02 kJ mol −1 (298.15 K), 88.02 ± 0.03 kJ mol −1 (303.15 K), 89.61 ± 0.01 kJ mol −1 (308.15 K), 91.18 ± 0.01 kJ mol −1 (313.15 K), and 92.75 ± 0.02 kJ mol −1 (318.15 K), respectively. The standard molar entropy of activation ( \(\Delta S_{ \ne }^{\theta }\) ) and standard molar enthalpy of activation ( \(\Delta H_{ \ne }^{\theta }\) ) are − 262.55 ± 0.72 J mol −1 K −1 and 7.98 ± 0.22 kJ mol −1 , respectively.

Background Anti-malarial drug artesunate (ART), a semi-synthetic derivative of artemisnin, has immunosuppressive effects on several autoimmune diseases, including Systemic lupus erythematosus (SLE), Rheumatoid arthritis (RA), and Colitis . However, molecular mechanisms of ART, especially on follicular helper T cells (Tfh), central players in SLE pathology, are far from clear. Purpose The object for this work is to investigate the therapeutic effect of ART on lupus-prone MRL/ lpr mice and its regulatory function on Tfh cells . Study design and methods MRL/ lpr mice were used to explore therapeutic effects of ART on lupus-prone MRL/ lpr mice and its regulatory functions on Tfh cells. Then, experiments of renal function were accomplished using the biochemical kits. Effects of ART on histopathology of kidneys, inflammatory factors and autoantibodies were examined using H&E staining, ELISA and real-time PCR. Flow cytometry and western blot analysis were used to examine effects of ART on Tfh differentiation and Jak2–Stat3 signaling pathway . Results Upon oral administration , ART significantly prolonged the survival of MRL/ lpr mice, ameliorated the lupus nephritis symptoms, decreased the levels of anti-dsDNA antibodies deposited in the kidney, and the levels of pathogenic cytokines (IL-6, IFN-γ and IL-21). After ART treatment , T-cell compartment in the spleen of MRL/ lpr mice was restored in terms of reduction in the number of Tfh cells and in the maintenance of the ratio of Tfr to follicular regulatory T cells (Tfh). In addition, ART has significantly inhibited the phosphorylation levels of Jak2 and Stat3 in the MRL/ lpr mice. Conclusion ART showed therapeutic effects on lupus-prone MRL/ lpr mice by inhibiting the differentiation of Tfh cells as well as altering the activation status of Jak2–Stat3 signaling cascade.
