Two-point immobilization of M3 muscarinic receptor: a method for recognizing receptor antagonists in natural products

Huang Xiaomin,Wang Ting,Wang Ludan,Sun Yantao,Zhang Ziru,Zhang Yajun

In the investigation of active ingredients from natural products, current technologies relying on drug–target affinity recognition analysis face significant challenges. This is primarily due to their limited specificity and inability to provide downstream pharmacodynamic information, such as agonistic or antagonistic activity. In this study, a two-point method was developed by immobilizing M3 acetylcholine receptor (M3R) through the combination of the conformation-specific peptide BJ-PRO-13a and the HaloTag trap system. We systematically assessed the specificity of the immobilized M3R using known M3R antagonists (pirenzepine and atropine) and agonists (cevimeline and pilocarpine). By frontal analysis and nonlinear chromatography, the performance of immobilized M3R was evaluated in terms of binding kinetics and thermodynamics of four drugs to the immobilized M3R. Additionally, we successfully identified two M3R antagonists within an extract from Daturae Flos (DF), specifically hyoscyamine and scopolamine. Our findings demonstrate that this immobilization method effectively captures receptor-ligand binding interactions and can discern receptor agonists from antagonists. This innovation enhances the efficiency of receptor chromatography to determine binding-affinity in the development of new drugs, offering promise for the screening and characterization of active compounds, particularly within complex natural products.

Synergistic effect between biochar and nitrate fertilizer facilitated arsenic immobilization in an anaerobic contaminated paddy soil

Wenli Jia,Zhi Yu,Junfeng Chen,Jian Zhang,Jian Zhu,Wentao Yang,Ruidong Yang,Pan Wu,Shengsen Wang

Nitrate nitrogen fertilizer was usually used to mitigate arsenic (As) release and mobilization in the anaerobic contaminated paddy soil. However, the effect of the interplay between nitrate fertilizer and biochar on As availability as well as the involved mechanism were poorly understood. Herein, the effects and mechanisms of biochar, nitrate fertilizer, and biochar-based nitrate fertilizer on the availability of As in the contaminated paddy soil were investigated via a microcosm incubation experiment. Results indicated that the application of biochar-based nitrate fertilizer significantly lessened the available As concentration in the contaminated paddy soil from 3.01 ± 0.03 (control group) to 2.24 ± 0.08 mg kg −1 , which presented an immobilization efficiency of 26.6 % better than those of individual biochar (13.5 %) and nitrate fertilizer (17.6 %), exhibiting a synergistic effect. Moreover, the biochar-based nitrate fertilizer also facilitated the transformation of more toxic arsenite in the contaminated soil to less toxic arsenate. Further, biochar-based nitrate fertilizer increased soil redox potential (Eh), dissolved organic carbon, organic matter, and nitrate yet decreased soil pH and ammonium, which changed the microbial community in the soil, enhancing the relative abundance of Bacillus , Arthrobacter , and Paenibacillus . These functional microorganisms drove the coupled transformation between nitrate denitrification and Fe(II) or As(III) oxidation, favoring As immobilization in the anaerobic paddy soil. Additionally, the co-application of biochar offset the negative effect of single nitrate fertilizer on microbial community diversity. Overall, biochar-based nitrate fertilizer could be a promising candidate for the effective immobilization of As in the anaerobic paddy soil. The current research can provide a valuable reference to the remediation of As-contaminated paddy soil and the production of safe rice.

Efficient Immobilization of CMlrA on Mesoporous Carriers for Synchronized Harmful Microcystis Blooms Suppression and Microcystin Degradation

Yu Wu,Linna Shao,Cai Cheng,Xianyong Kou,Shi Huang,Liexiong Zhang,Jie Yu,Xuehan Xu,Xingyu Xiang,Xiaoliang Guo,Lingling Feng

Harmful Microcystis blooms (HMBs) and their hepatotoxic metabolites microcystins (MCs) pose escalating threats to aquatic biodiversity and public health due to their recalcitrance to conventional remediation approaches. To address this dual challenge, mesoporous silica (MSN) and hollow mesoporous organosilica (HMON) nanostructures were engineered via structure-guided design principles, enabling the development of the crude extract microcystinase A (CMlrA)-tailored immobilized platform (CMlrA@MSN and CMlrA@HMON) for synchronized HMBs suppression and MCs detoxification. Systematic optimization of carrier physicochemical properties (including specific surface area, hydrophilicity, and pore size) significantly enhanced the immobilization efficiency and catalytic durability of CMlrA. Kinetic modeling revealed 201.3% and 44.1% K cat / K m enhancements in MC-LR degradation for CMlrA@MSN and CMlrA@HMON relative to free CMlrA. Immobilized enzyme systems achieved 77.06% and 83.03% growth inhibition rates in Microcystis aeruginosa FACHB-905 after six-day treatment. This biocatalytic immobilization strategy establishes an eco-efficient framework for synergistic HMB suppression and MC degradation, providing the technical groundwork for practical water remediation systems.

Supercritical water gasification of hyperaccumulators: Study on hydrogen production, heavy metal immobilization, and environmental risk assessment

Chengwei Huang,Shanke Liu,Xu Bao,Jialing Xu,Xuanyu Cai,Chao Liang,Lijun Yu

Phytoremediation is an effective technology for the removal of heavy metals (HMs) from contaminated soils. However, improper post-treatment of hyperaccumulators can lead to secondary environmental hazards. This study explores the application of supercritical water gasification (SCWG) for the treatment of Cu-enriched hyperaccumulator Elsholtzia splendens ( ES ), with the dual aims of H 2 production and HM immobilization. SCWG process yielded a maximum H 2 output of 16.68 mol/kg after 20 min, demonstrating its potential for waste valorization. Within the temperature range of 500–650°C, the migration and transformation mechanisms of HMs were investigated through detailed biochar characterization. The results demonstrate that SCWG significantly reduces the leaching rate of Cu from 25.18 % to below 0.08 %, with over 99 % of HMs being immobilized in the resultant biochar. Furthermore, higher temperatures and prolonged residence times were found to promote the complexation of HMs with inorganic components in the biochar, resulting in their transformation into stable F4 fractions. Then, HMs in liquid products and solid residues were evaluated in terms of Nemerow index and potential ecological risk. The results indicated that the pollution risks of liquid products were safe while the original hyperaccumulators showed high risk, underscoring the promise of SCWG as an effective method for the concurrent production of H 2 -rich syngas and the immobilization of HMs.

Bi@PVP/PVA aerogels with 3D porous networks: High-performance trapping and immobilization of iodine vapor

Chunhui Wu,Qingwei Ping,Xueru Sheng,Na Li,Bing Wang,Jian Zhang

During the reprocessing of irradiated nuclear fuel, radioactive iodine vapor—a hazardous substance threatening environmental safety and human health—is released in substantial quantities, creating an urgent need for adsorbents with high-efficiency iodine trapping capabilities. This study synthesizes BiOBr aggregates via hydrothermal method and loads them onto a three-dimensional (3D) porous aerogel, which is composed of crosslinked polyvinylpyrrolidone (PVP) and polyvinyl alcohol (PVA) (denoted as Bi@PVP/PVA). Compared with Bi@PVP and Bi@PVA, Bi@PVP/PVA shows significantly enhanced iodine adsorption performance: Bi@PVP/PVA-3, featuring a well-developed 3D porous network with optimized pore structure, achieves an exceptional iodine trapping capacity of 2482.2 mg/g at 85 °C, outperforming the single-polymer-supported counterparts. Characterizations confirm that crosslinking between PVP and PVA constructs a macroporous framework, which not only provides abundant sites for uniform loading of BiOBr aggregates but also facilitates iodine diffusion. Kinetic model analysis reveals the dual adsorption mechanism: the PVP/PVA porous network enables rapid trapping of iodine vapor, while chemical reactions between BiOBr and iodine ensure its firm immobilization. This synergistic effect of the carrier’s porous structure and the loaded BiOBr endows Bi@PVP/PVA aerogels with superior iodine treatment efficiency, highlighting their practical value in iodine capture during nuclear fuel reprocessing.

Xin Wang,Lihong He,Jianrong Huang,Nanjing Zhong

In this study, SBA-15 was modified by halogen & haloalkanes and later used to immobilize lipases. The hydrolysis activity and the glycerolysis performance of the immobilized lipases was carefully studied. Highest activity of the immobilized Candida antarctica lipase B (CALB), Lipase from Aspergillus oryzae (AOL), Lecitase® Ultra (LU) and lipase from Rhizomucor miehei (RML) was respectively at 5577, 12000, 2822 and 11,577 U/g; in addition, the highest activity was obtained from the lowest or moderate lipase loading, at 25.73, 90.72, 89.52 and 30.56 mg/g respectively. The mechanism of lipase immobilization was studied and it was through interfacial activation. The halogen & haloalkanes modification of SBA-15 afforded considerable glycerolysis activity for diacylglycerols (DAG) preparation. CALB@SBA-15-CH 2 CH 2 (CF 2 ) 5 CF 3 and CALB@SBA-15-CH 2 CH 2 (CF 2 ) 7 CF 3 were suitable for DAG production, they both exhibited good reusability in glycerolysis reaction, with 117.36% and 93.06% of their initial glycerolysis activity retained respectively after ten cycles of reuse. The relationships between temperature with triacylglycerols (TAG) conversion were ln V 0  = 3.13–3.07/T and ln V 0  = 7.90–4.64/T respectively for CALB@SBA-15-CH 2 CH 2 (CF 2 ) 5 CF 3 and CALB@SBA-15-CH 2 CH 2 (CF 2 ) 7 CF 3 ; in addition, their activation energy ( Ea ) was respectively at 25.50 and 38.54 kJ/mol.

Resource recovery from iodine-containing silver-loaded silica gel: recycling of silver and immobilization of iodine

Lei Liao,Shilong Song,Gaoling Liang,Xiaoqiang Wang,Ning Pan,Hao Lei,Yongde Zhang,Hao Zou,Jiajia Cheng,Jie Wen,Weifeng Zhang,Tingting Deng

Silver-loaded silica gel is widely used in spent fuel reprocessing plant due to its high adsorption efficiency of iodine and low influence of humidity and NO x . This study proposed a novel method for the disposal of iodine-loaded silver-silica gel. The elution of iodine using Na 2 S solution, the leaching of silver through HNO 3 solution. Subsequently, Zero-valent silver was synthesized via reduction of Ag + with NaBH 4 to recover silver. Additionally, the eluted iodide ions were transformed into iodosodalite and then combined with borosilicate glass powder for secondary immobilization. The results demonstrated that the elution efficiency of Na 2 S was 99.03%, the leaching efficiency of HNO 3 was 99.20%, and the recovery efficiency of Ag reached 86.93%. The iodine static saturated adsorption capacity of the silver-loaded silica gel prepared using recycled silver was 129.95 ± 3.46   mg/g, higher than original silver-loaded silica gel. In addition, the secondary immobilization body had a dense structure. The iodine leaching rate was 1.2×10 -4   g ⋅ m -2 ⋅ d -1 , which showed a good chemical stability. This work successfully realized the recycling of silver in iodine-containing silver-loaded silica gel and the safe disposal of iodine, which can provide new ideas for the disposal of iodine waste in spent fuel reprocessing plant.

Linker Peptide Engineering Combined With SA-PVA Immobilization in Fed-Batch Biocatalysis for High-Efficiency Carcinine Synthesis

Man Zhao,Mengying Yu,Huiru Yuan,Yiting Shen,Zhiqiang Liu,Yuguo Zheng

Carcinine, a valuable imidazole dipeptide with antioxidant and therapeutic properties, faces biosynthesis challenges due to enzyme aggregation and substrate inhibition. In this study, an integrated strategy combining linker peptide engineering and immobilization was applied to address these challenges and enhance carcinine production. Rational design of linker peptides (D 5 , L 2 , L 3 ) in the sfp-Ebony fusion protein enabled its highest soluble expression in WSL 2 E strain, achieving 93.1% conversion efficiency—3.5-fold higher catalytic efficiency than WSGE strain. Response surface methodology optimized sodium alginate-polyvinyl alcohol (SA-PVA) immobilization parameters (5% PVA, 3% SA, 2.3% CaCl₂), yielding excellent immobilized WSL 2 E@SA-PVA cells with 95.93% activity recovery. Structural characterization by scanning electron microscopy (SEM), Fourier-transform infrared spectroscopy (FT-IR), and X-ray diffraction (XRD) confirmed the formation of a porous SA-PVA matrix that protected cells from harsh conditions. The immobilized biocatalyst exhibited superior operational stability (retaining > 80% activity after 7 cycles) and storage stability (maintaining 44.89% activity after 14 days at 4°C). Fed-batch scale-up (50 mL) achieved a record carcinine titer of 71.13 mM, mitigating the inhibitory effect of high substrate concentrations through phased substrate feeding. This study provides a scalable biocatalytic platform for industrial carcinine production, effectively addressing key bottlenecks in biocatalyst stability and substrate tolerance.

Development of nano-hydroxyapatite@biochar composite from low-cost precursors for enhanced immobilization of lead and cadmium

Ling Zhang,Qiao Li,Yubo Yan,Xinbai Jiang,Jinyou Shen

The composite of nano-hydroxyapatite (nHAP) and biochar offers dual advantages: enhancing biochar’s heavy metal adsorption capacity while improving the dispersion of nHAP, positioning nHAP@biochar as a promising candidate for heavy metal immobilization. In this study, ammonia-soda residue (ASR) leachate and Doudan skin waste (DDS) were explored for the first time as low-cost precursors to synthesize nHAP@biochar for efficient adsorption of lead (Pb(II)) and cadmium (Cd(II)). The optimal mass ratio of nHAP to biochar, 2:1 (HMD2), was determined. The results showed that the uptake of both metal ions by HMD2 was favorable and spontaneous, with Pb(II) exhibiting a faster adsorption rate than Cd(II). Molecular dynamics (MD) simulations were applied to model the adsorption kinetics of metal ions on the pure HAP surface, offering a molecular-level interpretation for the experimental results. Furthermore, HMD2 demonstrated exceptionally high theoretical adsorption capacities of 1250 and 172.4 mg/g for Pb(II) and Cd(II), respectively, outperforming many other similar adsorbents. Mechanistic analysis revealed multiple processes involved in the adsorption of metal ions on HMD2, including pore filling, ion exchange, cation-π interaction, precipitation, complexation, and electrostatic interactions. Density functional theory (DFT) calculations further supported that surface oxygen atoms in HAP serve as the primary active sites for adsorption. This study not only provides new insights into the valorization of ASR and DDS but also lays a theoretical foundation for the application of nHAP@biochar composites in the immobilization of heavy metals in contaminated soils.

Waste Algae-Derived Biochar Composites for Synergistic Soil Cd–As Immobilization: Feasibility, Dose–Response Thresholds, and Mechanism

Xue Zhou,Lianfang Li,Mengqi Kang

The antagonistic geochemical behaviors of cadmium (Cd) and arsenic (As) in co-contaminated soils complicate their simultaneous remediation. This study aimed to develop a synergistic immobilization strategy by convertingSpirulinaresidue into a magnetic biochar-layered double hydroxide composite (FSRBL). The composite was applied to both acidic red and calcareous black soils, and its effects on Cd and As, immobilization efficiency, and ecotoxicity were evaluated. The results showed that FSRBL effectively transforms Cd and As from mobile fractions to stable residual forms. At a 2.5% application rate, FSRBL achieved remarkable immobilization efficiencies of 39.2% for Cd and 57.5% for As, representing effectiveness 3.55 and 5.97 times higher than that of unmodified biochar, respectively. A dose–response relationship between the application amount of FSRBL and the immobilization efficiency of As and Cd was observed and further quantified using a logistic model. The results indicate that while increased FSRBL application enhances immobilization efficiency, the marginal benefit of each additional unit diminishes as the application rate increases, demonstrating a significant diminishing marginal effect. According to the ecotoxicity assessment experiment, the soil leachate from FSRBL-amended soil remarkably decreased the ecological toxicity to rice (Oryza sativaL.). Mechanistic investigations employing SEM/TEM-EDS, XRD, and XPS revealed that the synergistic immobilization could be ascribed to the multi-component cooperation within FSRBL, which resolved the conflicting pH/Eh requirements for the immobilization of Cd and As: (1) the LDH phase efficiently immobilized As oxyanions through anion exchange and isomorphic substitution; (2) the magnetic Fe phase concurrently immobilized Cd2+and As oxyanions via redox transformation and coprecipitation, resulting in the formation of precipitates such as Fe/Ca/Cd–As(V). This work demonstrates a feasible approach to upcycle biomass waste into a value-added material for sustainable remediation of Cd–As co-contaminated soil.

Integration of Medium Engineering, Immobilization, and Rotating Bed Reactor for Intensifying Enzymatic Reduction of Halogenated Aryl Ketones

Shuling Zhang,Jiajing Yan,Quan Yuan,Yanjun Jiang,Lihui Wang,Yunting Liu

Enzymatic reduction of halogenated aryl ketones (HAKs) is an important reaction for the synthesis of pharmaceutical-related enantiopure halohydrins, but its preparative application remains restricted due to low substrate solubility and poor mass transfer in aqueous media. Herein, a multiengineering approach involving medium optimization, enzyme immobilization, and a rotating bed reactor was applied for intensifying the enzymatic reduction of HAKs. A deep eutectic solvent (DES) composed of betaine, glucose, and water (Bet-Glc-H2O) was demonstrated to be effective in improving enzymatic activity and stability, enabling efficient reduction of high-concentration HAKs (1.0 M, 85% conversion). In addition, enzyme immobilization was conducted using DES-modified resin supports, markedly enhancing the operational stability and reusability of the biocatalysts. Finally, a SpinChem rotating bed reactor (RBR) was implemented for further intensifying the enzymatic process, leading to dramatically shorter reaction times and increased overall productivity.
