Multifunctional pH-Responsive Gemini Surfactant

Bin Hu,Jiani Liu,Chengwen Deng,Youmei Xing,Min Gong,Zhen Wu,Guojie Wang

Stimuli-responsive surfactants have garnered significant attention as promising candidates for diverse applications in efficient oil recovery, programmable all-liquid devices, and microreactors owing to their dynamic interfacial self-assembly behavior under specific triggers. However, contemporary applications of stimuli-responsive surfactants remain constrained within monofunctional paradigms, significantly impeding their integration into advanced multifunctional manufacturing. Here, an interesting pH-responsive gemini surfactant is reported that can dynamically switch between gemini (GTL, pH ≥ 8.0) and single-chain (TL + , pH ≤ 6.0) conformation, facilitating adaptive functionalities spanning reversible oil/water separation, structured liquid, 3D printing, and thermal insulative aerogels. As triggered by CO 2 /Ar, GTL acts as a smart emulsifier, achieving efficient oil/water separation over 10-cycle reusability. The electrostatic co-assembly of TL + and sulfated cellulose nanocrystals (CNC-OSO 3 − ) at the oil-water interface drives a jammed film, yielding a reconfigurable all-liquid structure. Furthermore, TL + /CNC-OSO 3 − -stabilized emulsions exhibit self-supporting properties, enabling dual functionality as 3D-printable inks for complex patterning in air/oil baths, and as advanced precursors for thermal insulative aerogels. Density functional theory calculations reveal the pH-dependent adsorption of GTL at the oil-water interface and the co-assembly of TL + /CNC-OSO 3 − driven by abundant non-covalent interactions. This pH-responsive surfactant addresses the critical demand for multifunctional manufacturing, establishing an adaptive paradigm for next-generation smart materials.

Micro Electrochemical pH Sensor Applicable for Real-Time Ratiometric Monitoring of pH Values in Rat Brains

Jie Zhou,Limin Zhang,Yang Tian

To develop in vivo monitoring meter for pH measurements is still the bottleneck for understanding the role of pH plays in the brain diseases. In this work, a selective and sensitive electrochemical pH meter was developed for real-time ratiometric monitoring of pH in different regions of rat brains upon ischemia. First, 1,2-naphthoquinone (1,2-NQ) was employed and optimized as a selective pH recognition element to establish a 2H+/2e– approach over a wide range of pH from 5.8 to 8.0. The pH meter demonstrated remarkable selectivity toward pH detection against metal ions, amino acids, reactive oxygen species, and other biological species in the brain. Meanwhile, an inner reference, 6-(ferrocenyl)hexanethiol (FcHT), was selected as a built-in correction to avoid the environmental effect through coimmobilization with 1,2-NQ. In addition, three-dimensional gold nanoleaves were electrodeposited onto the electrode surface to amplify the signal by ∼4.0-fold and the measurement was achieved down to 0.07 pH. Finally, combined with the microelectrode technique, the microelectrochemical pH meter was directly implanted into brain regions including the striatum, hippocampus, and cortex and successfully applied in real-time monitoring of pH values in these regions of brain followed by global cerebral ischemia. The results demonstrated that pH values were estimated to 7.21 ± 0.05, 7.13 ± 0.09, and 7.27 ± 0.06 in the striatum, hippocampus, and cortex in the rat brains, respectively, in normal conditions. However, pH decreased to 6.75 ± 0.07 and 6.52 ± 0.03 in the striatum and hippocampus, upon global cerebral ischemia, while a negligible pH change was obtained in the cortex.

Light/pH dual controlled drug release

Liu Shihe,Zhang Xin,Bai Zhimin,Yang Yibo,Zhang Jia,Li Kun,Liu Zhiwei,Shi Ming,Dong Lixin,Wang Jidong,Li Jian

Photodynamic therapy (PDT) has significant advantages in treating primary tumors. However, the hypoxic tumor microenvironment hinders the generation of sufficient reactive oxygen species during PDT to effectively kill tumor cells, further greatly limiting the applications of PDT in cancer treatment. Herein, we reported a temperature/pH dual controlled drug delivery system LPC@PCN@PDA/Fe 3+ -AS1411 based on a porous coordination network (PCN (Mn)) coated with polydopamine (PDA) and modified with an aptamer AS1411. β-lapachone (LPC) was loaded inside the PCN (Mn) framework, and Fe 3+ was attached to the surface of the PDA coating. These nanoparticles (NPs) exhibited excellent multimodal cancer therapeutic effects and tumor targeting ability with their photo- and chemodynamic properties. The therapeutic effect can be enhanced by the production of sufficient oxygen by the internal hydrogen peroxide, which improves the photodynamic effect of the photosensitizer PCN (Mn) and the chemotherapy effect of β-lapachone. Notably, the conversion of Fe 2+ to Fe 3+ in the tumor cells exerts the Fenton effect, which generates hydroxyl radicals that cause lipid peroxidation in tumor cells and induce apoptosis, thus enhancing the chemodynamic therapeutic effect. In vitro and in vivo experiments revealed that the NPs demonstrated specific tumor targeting, excellent inhibition effect on tumor growth, and biocompatibility. Together, our findings can help develop an intelligent multifunctional therapeutic nanoplatform for cancer therapy. Graphic abstract

pH-Insensitive Hydrogel Fiber Random Lasers

Guosheng Zhang,Zhiyu Wang,Jinhui Shi,Sheng Zhou,Benli Yu

Overcoming the impact of pH on the performance of random lasers (RLs) has emerged as a significant challenge in its application for biomedical therapy. To tackle this issue, this paper proposes a pH-insensitive hydrogel fiber RLs. The RLs consists of Rhodamine B dye-doped p(acrylamide-co-acrylic acid, AM-co-AA) hydrogel as the fiber core and calcium alginate (Ca-alginate) hydrogel as the cladding, termed as p(AM-co-AA)/Ca-alginate hydrogel fiber RLs. The prepared RLs not only operate reliably at different pH levels but also allow for tuning of its emission wavelength by adjusting the temperature, ensuring the reliability and safety of photothermal therapy. Experimental results demonstrate that the threshold of p(AM-co-AA)/Ca-alginate hydrogel fiber RLs is only 0.23 mJ/cm². Moreover, within the range of 20∼80°C, the random laser emission wavelength can be tuned by controlling the temperature, with a maximum tunable range of 5.6 nm and good reversibility. Most importantly, the random laser intensity and wavelength of the p(AM-co-AA)/Ca-alginate hydrogel fiber RLs exhibit excellent stability when the pH value changes within the range of 3.5 to 9.5, indicating its exceptional ability to resist pH interference. We believe that the hydrogel fiber RLs proposed in this paper, leveraging the porous structure of hydrogel to reduce the laser threshold and employing the “core-cladding” structure to enhance stability, holds broad potential applications in the biomedical field.

Ultrafast spectroscopic studies of the pH responsive 9-acridinecarboxylic acid as a ratiometric and fluorescence lifetime pH indicator

Huangmei Zhou,Yu Zhao,Xihang Chen,Sanjun Zhang

pH is a fundamental physiological parameter related to myriad applications, therefore great efforts are continuously made to develop high performance pH indicators. Here, we exhaustively investigated the pH response of an acridine analog, 9-acridinecarboxylic acid with steady state absorption and fluorescence spectroscopies, picosecond resolved fluorescence spectroscopy, and femtosecond resolved transient absorption spectroscopy. The fluorescence intensity ratio (R 450/525 ) of 9-acridinecarboxylic acid showed a 7-fold change in the physiological pH range (pH ∼ 5–8), and it can be served as an excellent ratiometric and lifetime pH indicator. Analysis of the picosecond and femtosecond-resolved spectra combined with theoretical calculation revealed that 9-acridinecarboxylic acid existed in zwitterionic form at acidic condition with emission centered at 480 nm and lifetime of about 2.1 ns and anionic form at alkaline condition with emission centered at 430 nm and lifetime of about 6.6 ns. As pH decreased, the protonation of heterocyclic N atom resulted in the redshift of fluorescence peak and shortening of the fluorescence lifetime. Moreover, 9-acridinecarboxylic acid exhibited good selectivity, photostability and excellent biocompatibility and was successfully applied for intracellular pH imaging by ratiometric fluorescence.

pH-induced aggregation of hydrophilic carbon dots for fluorescence detection of acidic amino acid and intracellular pH imaging

Yu Zhu Yang,Na Xiao,Shi Gang Liu,Lei Han,Nian Bing Li,Hong Qun Luo

Intracellular pH level plays an important role in physiological and pathological processes. The development of nanoprobes for detecting in vivo pH levels is especially important for early diagnosis of disease. Therefore, we develop a hydrophilic carbon points (CDs) using quercetin and ethylenediamine as precursors to monitor intracellular pH. Under optimized conditions, the prepared CDs not only have uniform particle size and morphology, but also possess strong green fluorescence, photostability, and photoreversibility in water medium. The CDs exhibit pH-sensitive fluorescence effect under acidic and alkaline conditions, which is used to achieve “off-on-off” detection pH (from 3.5 to 13.5). Meanwhile, the pH-dependent mechanism is further investigated and explained, which is the fluorescence quenching caused by the pH-induced aggregation. Based on the pH-sensitive characteristics of CDs, it has been applied to the detection of aspartic acid and glutamic acid. More importantly, when applied to live cells, the pH-probe exhibits low cytotoxicity and high sensitivity, and is successfully used in intracellular pH fluorescence imaging. Consequently, this nanoprobe is expected to be used for real-time monitoring of intracellular pH level.

A reversible near-infrared pH probes for optical measurements of pH in complete water system and living cells

Huan Yu,Mingtai Sun,Kui Zhang,Houjuan Zhu,Zhengjie Liu,Yajun Zhang,Jun Zhao,Lijun Wu,Zhongping Zhang,Suhua Wang

The intracellular pH value controls many aspects of cell metabolism and is considered as an integral part of many metabolic reactions by maintaining the structure and function of nearly all proteins. Detection of pH fluctuation covering the whole intracellular pH window including both neutral and acidic is essential but challenging and still needs more sensitive pH probes. We have synthesized a new near-infrared fluorescent and colorimetric probe based on cyanine dye, which can be used for monitoring the pH fluctuation. The probe has a good water solubility, excellent photostability, reversibility, low cytotoxicity and exhibits a good performance to detect pH fluctuation in both solution and live cells. Upon incubation with live cells, it responds sensitively to low pH value in cell medium by exhibiting bright red fluorescence. On the contrary, the fluorescence intensity of probe seems to be very weak in neutral cellular environment. The satisfactory results show that the probe was a highly sensitive pH probe in living cells and further demonstrate its promising biological applications.

Tailor-made magnetic nanocarriers with pH-induced charge reversal and pH-responsiveness to guide subcellular release of doxorubicin

Zhang Han,Xue Yanan,Huang Juan,Xia Xiaoyang,Song Meifang,Wen Kaikai,Zhang Xiaoxiao,Luo Xiaogang,Cai Ning,Long Sihui,Yu Faquan

A charge reversible, magnetic-targeted, and pH-responsive nano therapy system assembled from citraconic-modified dextran (Dex–COOH), glycine-stabilized superparamagnetic iron oxide nanoparticles (SPION–NH 2 ), and doxorubicin (DOX) through electrostatic interaction was designed for delivering therapeutics to tumor cells. Dex–COOH was first synthesized to build the pH-sensitive citraconic amide bond and further render charge reversal upon the cleavage of the linkage. The SPION–NH 2 anchored in the nanocarrier endows the carrier with magnetic migration. The vibrating sample magnetometer analysis exhibits the superparamagnetic behavior, and the magnetic saturation value is estimated to be 39.6 emu (g Fe) −1 . The change of size and the reversal of zeta potential from negative to positive in acidic environment confirmed these expected responsibilities. The DOX-loaded nanocarriers contain DOX up to 13.0 wt%. An in vitro release profile demonstrated an efficient DOX release of 80 % at pH 5.0 while of 27 % at pH 6.8 and 13 % at pH 7.4, suggesting a pH-induced release mechanism. Microscopic images of Prussian blue staining, quantification of cellular iron and protein concentration displayed apparent iron uptake by HeLa cells. Confocal laser scanning microscopy observations revealed that the nanocarriers could efficiently deliver and release DOX into the nuclei of HeLa cells. MTT assays testified that DOX-loaded nanocarrier exhibited high anti-tumor activity with IC 50 of 0.7 μg mL −1 , while plain nanocarriers were practically non-toxic. This drug delivery system has shown the ability to improve the chemotherapeutic efficacy and to reduce the side effects, indicating great potential for delivery of drugs to the targeted sites in patients.

Enabling Rapid pH Equilibration Through Acoustic Microstreaming for Efficient pH Regulation in Microliter-Scale Samples with Screen-Printed Electrodes

Ziyi Xiao,Ruyi Deng,Xin Liu,Jiahuan Zheng,Kaisong Yuan

Real-time pH measurement and regulation of microliter-scale liquid samples are critical for microfluidic biochemical applications. However, traditional magnetic stirring for macroscopic systems is not applicable under such microscale conditions. This work develops an ultrasound-assisted screen-printed electrode (SPE) pH sensor integrated with a 3D-printed ultrasonic reflection cell and piezoelectric excitation unit. Ultrasound-induced acoustic streaming accelerates mass transfer inside microliter-scale samples. Upon acid addition, the ultrasonic-assisted system reaches potentiometric equilibrium far more rapidly than the static non-ultrasonic control. Optimized parameters are 30 s ultrasonic duration and 5 Vpp input amplitude. The sensor shows linearity from pH 3.04 to 4.15 with R2= 0.9919. Phenolphthalein visualization directly confirms ultrasound-enhanced molecular diffusion. Its mass transfer efficiency matches traditional magnetic stirring, and stable fast equilibrium is realized in complex cell culture medium. This acoustofluidic micromixing strategy offers a simple integrated route for pH monitoring and the regulation of micro-volume biological samples.

Breaking the pH limitation of peroxidase-like CoFe2O4 nanozyme via vitriolization for one-step glucose detection at physiological pH

Xiulian Yin,Peng Liu,Xuechao Xu,Jianming Pan,Xin Li,Xiangheng Niu

In recent years, a growing number of nanosized materials that can mimic the catalytic characteristics of some natural enzymes have experienced a period of rapid development. Among these nanomaterials defined as ‘nanozymes’, peroxidase-mimicking materials draw intensive interest because of their potential applications in various fields, including biosensing, medicine, and environmental engineering. Nonetheless, most of currently explored peroxidase nanozymes exhibit favorable activity only in acidic environments, which greatly restricts their use especially in biochemical analysis. Developing peroxidase mimics with good catalytic performance in a broad pH range is highly desired and still challenging. Herein, we report a vitriolization strategy to broaden the catalytic pH scope of peroxidase-like CoFe 2 O 4 . The vitriolization process endows the nanomaterial with sustainable acidity on its surface. As a result, the obtained peroxidase nanozyme (SO 4 2− /CoFe 2 O 4 ) exhibits good catalytic activity in neutral or even weak alkaline solutions. This provides us a one-step assay for colorimetric glucose detection in one pot at physiological pH via combining the proposed peroxidase mimic with glucose oxidase, which shows advantages of easier operation and faster detection while offering comparable analytical performance.

Enhanced pH adaptability in vanadium solvent extraction: Synergistic coordination of octanohydroxamate and D2EHPA for efficient extraction at low pH

Yiping Wang,Tao Liu,Yimin Zhang,Nannan Xue,Pengcheng Hu

Di-(2-ethylhexyl) phosphoric acid (D2EHPA) is the predominant extractant for vanadium recovery from acidic leachates. However, its narrow pH extraction window necessitates pH adjustment prior to extraction, generating significant neutralization residues and causing vanadium loss. Therefore, enhancing the pH adaptability of D2EHPA to enable direct vanadium extraction at low pH is of significant importance. This study proposes an octanohydroxamate (OHA)-D2EHPA extraction system, which broadens the extraction pH range through intermolecular cooperative coordination, providing an innovative solution for the efficient separation of vanadium from impurity ions under low-pH conditions. Under optimized conditions (OHA:V molar ratio of 2:1, pH of 0.6, D2EHPA concentration of 20 vol%, organic-to-aqueous volumetric phase ratio(O/A) (phase ratio(O/A)) of 1:2, 20 min extraction at room temperature), a three-stage countercurrent extraction process achieved a vanadium extraction efficiency of 99.56 %. Using a mixed solution of sulfuric acid and oxalic acid as the stripping agent under optimal conditions (0.4 mol/L H 2 C 2 O 4 +2 mol/L H 2 SO 4 , phase ratio(O/A) of 5:1, 20 min stripping at 25°C), three-stage stripping reached a vanadium stripping efficiency of 99.21 %, enabling highly efficient separation of vanadium from impurities. Studies confirm that the extraction process follows a H⁺-desorbing cation-exchange mechanism, where synergistic dehydrogenation of N-OH groups in OHA and P-OH groups in D2EHPA facilitates vanadium coordination. Vanadium complexes in the organic phase exist as [C 8 H 16 NO 2 ]VO, [C 8 H 16 NO 2 ]VOA, and [C 8 H 16 NO 2 ]VOA(HA). Among these, [C 8 H 16 NO 2 ]VOA(HA) exhibits the lowest binding energy for vanadium and is preferentially formed during extraction. Consequently, the OHA-D2EHPA system achieves highly efficient vanadium extraction at low pH.
