Effective synthesis of theaflavin-3,3′-digallate with epigallocatechin-3-O-gallate and epicatechin gallate as substrates by using immobilized pear polyphenol oxidase

Shicheng Lei,Minhao Xie,Bing Hu,Li Zhou,Yi Sun,Muhammad Saeeduddin,Hongcheng Zhang,Xiaoxiong Zeng

In the present study, pear polyphenol oxidase (PPO) was purified, immobilized and applied for the synthesis of theaflavin-3,3′-digallate (TF3). Firstly, PPO of pear ( Pyrus bretschneideri Rehd cv. Huangguan) was purified 24.36-fold in specific activity with a recovery of 6.77% by sequential use of precipitation with 70% saturated ammonium sulfate and chromatography of DEAE-Sepharose Fast Flow column. The purified pear PPO, found to be a dimer of identical subunits of molecular mass 35   kDa, was then successfully immobilized onto Fe 3 O 4 /chitosan nanoparticles via glutaraldehyde coupling reaction. Finally, TF3 was effectively synthesized by applying the immobilized PPO as biocatalyst with epicatechin gallate (ECG) and epigallocatechin-3- O -gallate (EGCG) as substrates, and the maximum yield of TF3 was 42.23% based on the amount of ECG added. In addition, the immobilized enzyme still remained 85% of its initial ability after successive usage of 8 cycles and could be stored for 30   days with less loss of activity. All the results suggest that the developed procedure is quite useful and has great potential for synthesis of TF3 and other theaflavins.

Ultrarobust alginate-based temperature sensing fiber across subzero to above zero self-healing performance enabled by polyphenol-nanosphere engineered multiple dynamic bonds

Xiaoqian Li,Xingyu He,Mi Zhou,Zhonghe Yu,Qiaohong Zheng,Huishan Gan,Yudong Wang,Qing Jiang,Hualing He,Zhicai Yu

Polysaccharide-based self-healing sensing fibers offer a promising green and durable pathway for wearable electronics, yet their effective healing ability in subzero environments still faces significant challenges. Herein, we present a subzero self-healing temperature-sensing (SST) fiber by incorporating tannic acid-Fe 3+ self-assembled nanospheres (TAN) into a sodium alginate/oxidized sodium alginate/carboxymethyl chitosan matrix. The synergy of multiple dynamic bonds and a rapid local molecular rearrangement mechanism with low activation energy endows SST sensing fiber with exceptional self-healing capabilities at subzero temperatures, achieving a strength retention of 65.7% at −10 °C after being cut. Specifically, the addition of TAN significantly enhances the stretchability (approximately 65%) of SST sensing fibers and exhibits remarkable water-responsive self-healing efficiency (97.64% at 25 °C). In addition, incorporating Ti 3 C 2 T x MXene nanosheets endows the SST fiber with broad temperature-sensing capability from 50 to 500 °C, attributed to the linear correlation between output voltage intensity and temperature ( U max  =  0.0115T  −  0.1582 , R 2  =  0.99 ). Furthermore, the SST fiber integrated with a wireless system demonstrates rapid response times (1.7 s) and reliable cyclic stability in temperature detection. This work provides a viable approach for designing bio-based temperature-sensing fibers that possess wide-temperature-adaptive self-healing ability for enhanced reliability and durability in practical wearable applications.

Bio-based polyphenol tannic acid as universal linker between metal oxide nanoparticles and thermoplastic polyurethane to enhance flame retardancy and mechanical properties

Xuan Wang,Wei Cai,Daolin Ye,Yulu Zhu,Mulan Cui,Jianchao Xi,Jiajia Liu,Weiyi Xing

It has been well confirmed that metal oxide nanoparticles can simultaneously enhance the flame retardancy and mechanical performance of polymer composites , owning to its catalysis effect and robust characteristic. However, most metal oxide nanoparticles tend to aggregate in polymers due to their high specific surface energy, thus deteriorating their enhancement effect. One simple and effective as well as environmentally friendly functionalization approach is desiderated for the practical application of metal oxide nanoparticles . Tannic acid is a naturally derived polyphenol compound which spontaneously combines with metal ions by coordinate bond. Due to size effect , there are lots of defect situ in the surface of metal oxide nanoparticles which can interact with tannic acid. Therefore, tannic acid was used to modify three kind of representative metal oxide nanoparticles (ZnO, Fe 2 O 3 and Co 3 O 4 ). The SEM photographs of fracture surface show that the modified metal oxide nanoparticles have a favorite dispersion state in thermoplastic polyurethane (TPU) matrix. As a result, the flame retardancy and mechanical properties of TPU composites have been significantly improved. Specifically, with 4% addition of tannic acid modified metal oxides (MO-TA), the peak HRR value showed a remarkable decrease of 22.7%, 14.1% and 19.3%, compared to pure TPU. Meanwhile, the tensile stress of MO-TA@TPU increased to 39.2, 34.0 and 39.8 MPa from 22.1 MPa of pure TPU, suggesting an effective mechanical performance improvement. The universality of polyphenol compounds to enhance the interfacial interaction between metal oxide nanoparticles and polymer resin opens a novel pathway to prepare high performance polymer composite materials.

Plant polyphenol-involved coordination assembly-derived Mo3Co3C/Mo2C/Co@NC with phase regulation and interface engineering for efficient hydrogen evolution reaction electrocatalysis

Yu-Hang Zhao,Tao Zhang,Xiao-Feng Wang,Shao-Jie Li,Yu Pan,Yihan Wang,Xue-Zhi Song,Zhenquan Tan

The rational exploration of highly active and low-cost materials for electrocatalytic hydrogen evolution is extremely imperative, but remains challenging as well. Transition metal carbides (TMCs) as highlighted electrocatalysts usually exhibit unsatisfactory activity and stability. In this work, we demonstrate a two-step strategy for electrocatalyst synthesis involving biomass-derived tannic acid (TA) and coordination assembly preparation followed by subsequent high-temperature carbonization. Interestingly, by simply controlling the feed amount of TA, heterostructured nitrogen-doped graphene-coated Mo3Co3C-based tri- and bi-component electrocatalysts (Mo3Co3C/Mo2C/Co@NC and Mo3Co3C/Co@NC) can be obtained with phase regulation and interface engineering. It is observed that the heterostructured materials possess more active sites than that of the TMC-excluding sample (Co@NC). Moreover, the tri-component Mo3Co3C/Mo2C/Co@NC may enhance the intrinsic activity at each active site due to the strong interaction at the Mo3Co3C/Mo2C interface. These advantages allow the Mo3Co3C/Mo2C/Co@NC catalyst to deliver superior electrocatalytic activity and favorable kinetics towards the HER, with a low overpotential of 211 mV and a Tafel slope of 96.0 mV dec−1. Furthermore, it has good long-term stability. This work provides new inspiration for heterostructured TMC-based electrocatalysts with high performance towards promising catalytic applications.

One-pot green preparation of deep-ultraviolet and dual-emission carbon nanodots for dual-channel ratiometric determination of polyphenol in tea sample

Wang Zhong-Xia,Hu Lei,Wang Wen-Juan,Kong Fen-Ying,Wei Mei-Jie,Fang Hai-Lin,Li Qi-Le,Wang Wei

A novel deep-ultraviolet and dual-emission carbon nanodots (DUCDs)-based dual-channel ratiometric probe was prepared by a one-pot environmental-friendly hydrothermal process using guanidine as the only starting material for sensing polyphenol in tea sample (TPPs). Under the exposure to TPPs, the DUCDs not only provided a characteristic colorimetric response to TPPs, but also displayed TPPs-sensitive ratiometric fluorescence quenching. The detection mechanism was proved to be that enrichment-specific hydroxyl sites (e.g., -NH 2 and -COOH) of DUCDs can specifically react with phenolic hydroxyl groups of TPPs to generate dynamic amide and carboxylate bonds by dehydration and/or condensation reaction. As a result, a new carbon nanomaterial with decrement of surface passivation groups, inherent light-absorbing, and invalid fluorescence emission was generated. The ratio (FL 297nm /FL 395nm ) of fluorescence intensity at 297 nm and 395 nm of DUCDs excited at 275 nm decreased with increasing TPPs concentration. The linearity range was 5.0 ng/mL to 100 µg/mL with a detection limit (DL) of 3.5 ± 0.04 ng/mL for TPPs ( n  = 3, 3σ/k). Colorimetry of DUCDs, best measured as absorbance at 320 nm, was increased linearly in the TPP concentration range 200 ng/mL–200 µg/mL with a DL of 94.7 ± 0.04 ng/mL ( n  = 3, 3σ/k). The probe was successfully applied to the determination of TPPs in real tea samples, showing potential application prospects in food analysis. Graphical abstract

Xiaomin Wang,Xue Huang,Fengjuan Zhang,Fanyun Hou,Fangxuan Yi,Xia Sun,Qingqing Yang,Xiangbo Han,Zhanli Liu

In this study, chitosan/zein composite film combined with tea polyphenol was fabricated, and the effect of tea polyphenol on the structure and physical properties of chitosan/zein film was investigated. Low-field nuclear magnetic resonance spectroscopy indicated that the distribution and content of water molecules in chitosan/zein film were changed with the addition of tea polyphenol. The results of fluorescence measurement and secondary structure analysis proved the enhanced physical and structural properties of tea polyphenol composite film. And the film showed favorable stretching resistance and flexibility. Furthermore, the effect of composite film on quality and physiological indexes of mushroom ( Lyophyllum decastes Sing.) stored at 4 °C for 15 d were systematically evaluated. The results indicated that the tea polyphenol composite film showed the effectiveness of maintaining color, texture as well as integrity of cell membrane evidenced by relatively lower membrane permeability and malondialdehyde content of mushrooms. Besides, weight loss and respiration rate were efficiently retarded. In comparison with control and chitosan/zein film, the tea polyphenol composite film inhibited the activity of polyphenol oxidase and peroxidase, while enhanced the activity of antioxidant enzymes (superoxide dismutase and catalase). In a word, the chitosan/zein composite film incorporated with tea polyphenol showed the potential as an effective technology for maintaining the postharvest quality of mushroom ( Lyophyllum decastes Sing.).

Sodium alginate-based nanocomposite films with strong antioxidant and antibacterial properties enhanced by polyphenol-rich kiwi peel extracts bio-reduced silver nanoparticles

Xinyu Sun,Hui Zhang,Jiao Wang,Mengna Dong,Pei Jia,Tong Bu,Qinzhi Wang,Li Wang

Sodium alginate (SA)-based films with excellent antioxidant and antibacterial activities were prepared, which was modified by polyphenol-rich kiwi peel extracts (PE) bio-reduced silver nanoparticles. In detail, PE was employed as reducing and capping agents to self-assemble silver@PE nanoparticles (Ag@PE NPs), which was further used to modify SA film in its functionality. In addition, the effects of the Ag@PE NPs amount on the physicochemical and biological performances were investigated. Results showed that SA-based films had strong tensile strength, water vapor resistance and good UV barrier properties. The incorporation of Ag@PE NPs drove a color change of SA-based films to redness and yellowness, while still showing good transparencies. Besides, Ag@PE NPs modified SA-based films showed a significant enhancement of antioxidant property, as well as an obvious improvement of antibacterial activity against S taphylococcus aureus and Escherichia coli . Therefore, this study provided a new insight for the reutilization of kiwi peels and a green, cheap and easy approach to prepare multifunctional SA-based food packaging films.

Polyphenol and Cu2+ surface-modified chitin sponge synergizes with antibacterial, antioxidant and pro-vascularization activities for effective scarless regeneration of burned skin

Zhentao Li,Xiuhong Huang,Licheng Lin,Yanpeng Jiao,Changren Zhou,Zonghua Liu

Bacterial overgrowth and high oxidative stress often lead to burn wound infection, hyper-inflammation, and slowed wound healing, eventually causing scar formation. Wound dressings synergized with antibacterial, antioxidant, and pro-vascularization activities can effectively promote wound healing and inhibit scars formation. Herein, chitin sponge consisted of chitin fibers was modified by the coating of proanthocyanidins (PAs) – Cu 2+ for effective scarless skin regeneration in a burn injury. The physicochemical characteristics indicated that PAs and Cu 2+ successfully modified chitin sponges by the coordination between PAs and Cu 2+ . For one thing, PAs improved the antioxidant performance of sponges, and for another, with the help of PAs, Cu 2+ effectively catalyzed S-nitrosothiols (RSNOs) to continuously generate nitric oxide (NO) in vitro/vivo for improving the inhibition of microorganisms and pro-vascularization activities. Also, Ch/PAs-Cu promoted proliferation of human umbilical vein endothelial cells (HUVECs) and rat fibroblasts (NIH-3T3), and the expression of angiogenic-related cytokines VEGF, eNOS, FGF, and MMP9. In animal experiments, Ch/PAs-Cu inhibited the accumulation of macrophages and neutrophils to regulate wound inflammation while up-regulating the expression of CD31 and α-SMA cytokines to promote angiogenesis. Ultimately, it promoted wound healing and inhibited scar formation. Therefore, Ch/PAs-Cu may be a promising dressing for the scarless repair of burned skin.

Polyphenol-Assisted Chemical Crosslinking: A New Strategy to Achieve Highly Crosslinked, Antioxidative, and Antibacterial Ultrahigh-Molecular-Weight Polyethylene for Total Joint Replacement

Yue Ren,Fei-Yu Wang,Ri-Tong Lan,Wan-Qun Fu,Zi-Jian Chen,Hao Lin,Shishu Huang,Rizwan M. Gul,Jing Wang,Jia-Zhuang Xu,Zhong-Ming Li

Highly crosslinked ultrahigh-molecular-weight polyethylene (UHMWPE) bearings are wear-resistant to reduce aseptic loosening but are susceptible to oxidize in vivo/in vitro, as reported in clinical studies. Despite widespread acceptance of antioxidants in preventing oxidation, the crosslinking efficiency of UHMWPE is severely impacted by antioxidants, the use of which was trapped in a trace amount. Herein, we proposed a new strategy of polyphenol-assisted chemical crosslinking to facilitate the formation of a crosslinking network in high-loaded tea polyphenol/UHMWPE blends. Epigallocatechin gallate (EGCG), a representative of tea polyphenol, was mixed with UHMWPE and peroxide. Multiple reactive phenolic hydroxyl groups of tea polyphenol coupled with the nearby free radicals to form extra crosslinking sites. The crosslinking efficiency was remarkably enhanced with increasing tea polyphenol content, even at a concentration of 8 wt %. Given by the hydrogen donation principle, the high-loaded tea polyphenol also enhanced the oxidation stability of the crosslinked UHMWPE. The antioxidative performance was preserved even after tea polyphenol elution. Moreover, superior antibacterial performance was achieved by the in situ tea polyphenol release from the interconnected pathways in the present design. The strategy of polyphenol-assisted chemical crosslinking is applicable for producing highly crosslinked, antioxidative, and antibacterial UHMWPE, which has promising prospects in clinical applications.

Silanization of a Metal–Polyphenol Coating onto Diverse Substrates as a Strategy for Controllable Wettability with Enhanced Performance to Resist Acid Corrosion

Xinxiu Cheng,Ruofei Lu,Xiaoqiang Zhang,Yaxin Zhu,Shaoyin Wei,Yagang Zhang,Xingjie Zan,Wujun Geng,Letao Zhang

Wettability is a crucial characteristic of materials that plays a vital role in surface engineering. Surface modification is the key to changing the wettability of materials, and a simple and universal modification approach is being extensively pursued by researchers. Recently, metal–phenolic networks (MPNs) have been widely studied because they impart versatility and functionality in surface modification. However, an MPN is not stable for long periods, especially under acidic conditions, and is susceptible to pollution by invasive species. Spurred by the versatility of MPNs and various functionalities achieved by silanization, we introduce a general strategy to fabricate functionally stable coatings with controllable surface wettability by combining the two methods. The formation process of MPN and silane–MPN coatings was characterized by spectroscopic ellipsometry (SE), UV–visible–near-infrared (UV–vis–NIR) spectroscopy, X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), atomic force microscopy (AFM), water contact angle (WCA), etc. We found that the stability of the MPN was greatly enhanced after silanization, which is attributed to the cross-linking effect that occurs between silane and the MPN, namely, the cross-linking protection produced in this case. Additionally, the wettability of an MPN can be easily changed through our strategy. We trust that our strategy can further extend the applications of MPNs and points toward potential prospects in surface modification.
