Methods of large-scale controllable production of uniform monodispersed spherical nanoparticles have been one of the research directions of scientists in recent years. In this paper, we report an azeotropic distillation-induced evaporation self-assembly method as a universal method, and monodispersed hydrophobic ordered mesoporous silica nanospheres (MHSs) were successfully synthesized by this method, using triethoxymethylsilane (MTES) as the silica precursor and hexadecyl trimethyl ammonium bromide (CTAB) as the template. SEM and TEM images showed good monodispersity, sphericity, and uniform diameter. Meanwhile, SAXS and N2adsorption–desorption measurements demonstrated a highly ordered lamellar mesostructure with a large pore volume. The model drug, curcumin was successfully encapsulated in MHSs for drug delivery testing, and their adsorption capacity was 3.45 mg g−1, which greatly improved the stability of curcumin. The release time when net release rate of curcumin reached 50% was extended to 6 days. Keywords:drug delivery;curcumin;azeotropic distillation;self-assembly;hydrophobic;mesoporous silica nanospheres

In this work, the multilayer film with polycaprolactone as the outer layers and the curcumin-loaded gelatin film as the inner layer were fabricated by sequential solution blow spinning (SBS). The hydrophobic outer layers improved the water contact resistance and protected the inner layer from being dissolved by water. The interactions of hydrogen bonds between the outer and inner layers enhanced the thermal stability. The hydrophobicity and mechanical properties of the gelatin film were also improved by the sandwich structure. The multilayer film exhibited good slow-release ability, releasing curcumin continuously for 312 h. Meanwhile, the multilayer film was as effective as the single-layer gelatin loaded with curcumin against Escherichia coli and Staphylococcus aureus . The results of this study indicate that multilayer nanofilm fabricated by SBS have promising applications for the encapsulation and controlled release of antibacterial substances.

Practical application of versatile nanomaterials to food engineering is desirable. In this study, highly encapsulated curcumin-loaded shellac (Cur@Lac) nanoparticles are efficiently prepared via a rotating packed bed (RPB). In this modified co-precipitation process, the morphology of Cur@Lac nanoparticles is systematically investigated under experimental parameters involving the high-gravity level, the volume of antisolvent and the concentration of shellac. By virtue of the highly structured polymeric ester matrices of shellac and adequate supersaturation brought from RPB on a microscale, the obtained Cur@Lac nanoparticles with uniform size exhibit high encapsulation efficiency of over 96%. After 2 months of storage at room temperature, the aqueous Cur@Lac nanodispersions exhibit long-term stability with no agglomeration or sedimentation, and the final retention rate of curcumin was over 85%. Colored drinks of fruit syrup (pH = 4.0), fruit soda (pH = 6.0), and soda water (pH = 8.3) are prepared by using the Cur@Lac as the colorant, which possessed orange-yellow color. The nanocomposites of alkali-sensitive indicators based on cellulose paper and gelatin film demonstrated in the work show promising application value for sensing of alkaline solution and gas of perishable food.

Curcumin (CUR) is an extract of Curcuma longa Linn., which has various pharmacological activities. The instability, low water solubility and bioavailability of CUR greatly limit its clinical application. This work prepared Pluronic F127-liposome-encapsulated curcumin (CUR-LIP-F127) and explored its functional role in wound healing. Liposome-encapsulated curcumin (CUR-LIP) and CUR-LIP-F127 were prepared. Human keratinocyte cell line (HaCaT) was treated with CUR, Pluronic F127-liposome (LIP-F127) and CUR-LIP-F127, or combined with ML385 (Nrf2 inhibitor). The expression of mRNAs and proteins was detected by quantitative real-time PCR and western blotting. MTT and wound healing assays were performed to detect cell viability and migration. CUR, LIP-F127 and CUR-LIP-F127 all had no influence on cell viability of HaCaT cells. CUR-LIP-F127 treatment significantly accelerated cell migration and enhanced the expression of nuclear factor erythroid-related factor 2 (Nrf2) and kelch-like erythroid cell-derived protein 1 (Keap1) in HaCaT cells with respect to CUR or LIP-F127 treatment. ML385 treatment impaired CUR-LIP-F127-mediated promotion of migration and up-regulation of Nrf2 and Keap1 in HaCaT cells. This work demonstrated that CUR-LIP-F127 activated Nrf2/Keap1 signaling pathway to promote migration of HaCaT cells, suggesting that CUR-LIP-F127 may contribute to wound healing.

This study aimed to design a Pickering emulsion (PE) stabilized by whey protein isolate nanofibers (WPINs) prepared with subcritical water (SW) to encapsulate and prevent curcumin (Cur) degradation. Cur-loaded WPINs–SW stabilized PE (WPINs–SW–PE) and hydrothermally prepared WPINs stabilized PE (WPINs–H–PE) were characterized using the particle size, zeta potential, Congo Red, CD, and TEM. The results indicated that WPINs–SW–PE and WPINs–H–PE showed regular spherical shapes with average lengths of 26.88 ± 1.11 μm and 175.99 ± 2.31 μm, and zeta potential values were −38.00 ± 1.00 mV and −34.60 ± 2.03 mV, respectively. The encapsulation efficiencies of WPINs–SW–PE and WPINs–H–PE for Cur were 96.72 ± 1.05% and 94.07 ± 2.35%. The bio-accessibility of Cur of WPINs–SW–PE and WPINs–H–PE were 57.52 ± 1.24% and 21.94 ± 2.09%. In addition, WPINs–SW–PE had a better loading effect and antioxidant activities compared with WPINs–H–PE. SW could be a potential processing method to prepare a PE, laying the foundation for the subsequent production of functional foods. Keywords:subcritical water;whey protein isolate nanofibers;high internal phase Pickering emulsion;curcumin deliver system;bioavailability

A co-delivery system of SN38 (7-ethyl-10-hydroxyl camptothecin) prodrug and CUR (curcumin) was designed for the treatment of lung cancer by pulmonary delivery. SN38 was linked to cell-penetrating peptide (CPP) TAT via a polyethylene glycol (PEG) linker to form the SN38 prodrug (TAT-PEG-SN38). Liposomes co-loaded with amphiphilic TAT-PEG-SN38 and curcumin (Lip-TAT-PEG-SN38/CUR) were successfully prepared by a microfluidic method for the treatment of lung cancer via pulmonary delivery. Lip-TAT-PEG-SN38/CUR showed nanometer-sized sphericity and a particle size of 171.21 nm. Besides, Lip-TAT-PEG-SN38/CUR exhibited enhanced antiproliferative effect, increased cell apoptosis induction and improved cell cycle arrest compared to the single agents in vitro. The combination induced significant tumor inhibition in a BALB/c mouse lung cancer model. These results indicated that our SN38 prodrug and curcumin co-delivery system was a promising candidate for lung cancer treatment.

Photodynamic therapy is a promising alternative to antibiotics in the treatment of bacterial infection, which has the advantages of less side effects, no drug resistance and accurate treatment. However, most photosensitizers (PSs) have some disadvantages, such as poor water solubility, serious self-quenching, poor stability and so on. Therefore, solving the limitations of traditional photosensitizers has become a hot issue in photodynamic therapy. On this basis, the lysozyme (Lys)-gold nanocluster (AuNCs)-curcumin (Cur) conjugate was used as a new type of water-soluble photosensitizer and combined with the bacterial surface through electrostatic interaction to form a bacterial surface enriched in PSs. At the same time, real-time bacterial imaging can be achieved by using the fluorescence properties of Lys-AuNCs-Cur. Under 405 nm light and dark conditions, the inactivation effect of Lys-AuNCs-Cur on Staphylococcus aureus and Escherichia coli was higher than that of free Cur. Moreover, Lys-AuNCs-Cur can also produce enough ROS to kill methicillin-resistant Staphylococcus aureus . The excellent antibacterial properties of nanoparticles may be attributed to the synergism of antibacterial protein lysozyme and AuNCs-Cur nanocomplexes and the increase of 1 O 2 production. The results of scanning electron microscope and fluorescence microscopic imaging showed that the cell membrane of E. coli and S. aureus was seriously damaged, and facilitated penetration of Lys-AuNCs-Cur nanocomplexes into the bacteria through the damaged cell membrane, and improve the photoinactivation efficiency of bacteria. More importantly, nanoparticles exhibited almost negligible dark cytotoxicity. The coupling of Cur and gold nanoclusters can significantly improve the stability of free curcumin molecules and the ability to generate ROS. Therefore, this work is of great significance for the development of new antimicrobial agents against bacterial infection.

Introducing natural products to obtain ultraviolet (UV) shielding materials for inhibiting the photo-oxidation of oily foods is environmentally friendly food technology. Instead of traditional physical mixing, curcumin was introduced into castor oil-based waterborne polyurethane as a chain extender and compounded with gelatin for bioactive composite films. With the increase of curcumin-based waterborne polyurethane (CWP), the film with 10 wt% CWP achieved nearly complete ultraviolet shielding. The swelling index and water solubility were reduced by 800% and 12.93%, respectively. The tensile strength and elongation at break increased by 183% and 114%, respectively. In addition, CWP enhanced the antioxidant activity of curcumin in water system by 52.10% and reduced the oxidation degree of soybean oil by 62.03–87.84%. This work provides a new idea for compounding natural active small molecules with polymers.

In order to improve the water solubility and stability of curcumin (CUR) and achieve its sustained release, the ZEIN– N -(2-hydroxy) propyl-3-trimethylammonium chitosan chloride (HTCC) complexes were designed to encapsulate CUR using an antisolvent precipitation (ASP) method. The encapsulation efficiency ( EE ) and loading capacity ( LC ) of curcumin in ZEIN-HTCC complexes were 92.2% and 0.6%, respectively. After encapsulation, the resistance of curcumin to thermal, natural light and UV irradiation, as well as the antioxidative activity were improved significantly ( P < 0.05). ZEIN-HTCC-CUR complexes exhibited good redispersibility and enhanced the water solubility of curcumin. In vitro simulated digestion experiments clarified that the ZEIN-HTCC complexes can control the sustained release behavior of curcumin, and the release kinetics can be fitted by the Higuchi model. Results taken from fluorescence spectra and Fourier transform infrared spectroscopy (FTIR) indicated that hydrophobic interaction, hydrogen bonding and electrostatic interactions were the main driving forces to form the ZEIN-HTCC-CUR complexes. The X-ray diffraction (XRD) results revealed that the encapsulated curcumin was in an amorphous state. The information provided by this study should be of interest to food industry to fabricate robust nanoscale delivery systems with ZEIN-HTCC complexes for CUR and other hydrophobic bioactive compounds.

Different from previously reported co-amorphous systems, a co-amorphous curcumin-magnolol (CUR-MAG CM) system, as compared with its crystalline counterparts, exhibited decreased dissolution due to its aggregation during dissolution. The main purpose of the present study is to deaggregate CUR-MAG CM to optimize drug dissolution and explore the deaggregation mechanism involved. Herein, a small amount of polymer (HPMC, HPC, and PVP K30) was co-formulated at 5% (w/w) with CUR-MAG CM as ternary co-amorphous systems. The polymer addition changed the surface properties of CUR-MAG CM including improved water wettability enhanced surface free energy, and hence exerted a deaggregating effect. As a result, the ternary co-amorphous systems showed faster and higher dissolution as compared with crystalline CUR/MAG and CUR-MAG CM. In addition, the nucleation and crystal growth of dissolved CUR and MAG molecules were significantly inhibited by the added polymer, maintaining a supersaturated concentration for a long time. Furthermore, polymer addition increased the Tgof CUR-MAG CM, potentially involving molecular interactions and inhibiting molecular mobility, resulting in enhanced physical stability under 25 °C/60% RH and 40 °C/75% RH conditions. Therefore, this study provides a promising strategy to optimize the dissolution and physical stability of co-amorphous systems by deaggregation and crystallization inhibition via adding small amounts of polymers.Keywords:curcumin;magnolol;co-amorphous;deaggregation;dissolution;physical stability

Acetylated acid-ethanol hydrolyzed (AHS) and acetylated debranched starch (ADS) were investigated as prospective nanocarriers . Both acid-ethanol hydrolysis and debranching decreased the molecular weight and viscosity of starch. Acid-ethanol hydrolyzed starch remained the original microstructure, which was confirmed by results of scanning electron microscopy. New absorption peaks in FTIR spectra of starch confirmed the occurrence of acetylation . The substitution degree (DS) of ADS could reach up to 1.18, while that of AHS could be improved by increasing the ethanol concentration. The developed nanoparticles showed uniform spherical structure and the size of that approximated 180–260 nm. The critical micelle concentration was 0.049 mg/mL, and the shift in fluorescence spectra confirmed the interaction between starch and curcumin . These results indicate show that high DS of AHS and ADS could be used as a potential carrier for curcumin delivery.

Present study prepared curcumin-loaded nanoliposomes using bovine milk, krill phospholipids and cholesterol; and investigated the effects of cholesterol on membrane characteristics, storage stability and antibacterial properties of the curcumin nanoliposomes. Bovine milk phospholipids which have higher saturation than krill phospholipids resulted in formation of curcumin-loaded nanoliposomes with higher encapsulation efficiency (84.78%), larger absolute value of zeta potential and vesicle size (size: 159.15 ± 5.27 nm, zeta potential: −28.3 ± 0.62 mV). Cholesterol helps to formation of a more hydrophobic, compact and tighter bilayer membrane structure which improved the storage stability of nanoliposomes under alkaline (66.25 ± 0.46%), heat (43.25 ± 0.69%) and sunlight (49.44 ± 1.78%) conditions. In addition, curcumin-loaded nanoliposomes can effectively target infectious bacteria which secrete pore-forming toxins such as Staphylococcus aureus by causing the bacterial cell wall to lysis. Findings from present work can guide future development of novel antibacterial agents for use in food preservation.
