Platycodon grandiflorum root fermentation broth reduces inflammation in a mouse IBD model through the AMPK/NF-κB/NLRP3 pathway

Zhe Wang,Chunhai Li,Xi He,Kang Xu,Zhipeng Xue,Ting Wang,Zhenshang Xu,Xinli Liu

The effect of Platycodon grandiflorum (PG) on colitis and its underlying mechanism were rarely studied. In this study, Lactobacillus rhamnosus 217-1 was used to ferment PG roots, and the concentrations of platycodin-D, flavonoids, and polyphenols and the DPPH free radical scavenging rate were significantly increased. Treatment with a PG root fermentation broth (PGRFB) could reduce dextran sulfate sodium (DSS) induced ulcerative colitis (UC) in mice. Meanwhile, the PGRFB significantly reduced the content of inflammatory factors in mouse serum and the expression of inflammatory factor mRNA in the intestinal tract, regulated the polarization of M1/M2 macrophages, and increased the expression of tight junction protein mRNA in intestinal epithelial cells. In summary, it was proved that the PGRFB could inhibit the nuclear factor kappa B (NF-κB) signaling pathway and the expression of Nod-like receptor protein 3 (NLRP3) inflammasomes by activating AMP-activated protein kinase (AMPK) and lowering the release of pro-inflammatory cytokines.

Drug-free contact lens based on quaternized chitosan and tannic acid for bacterial keratitis therapy and corneal repair

Zehui Jiao,Qiuyi Huo,Xuanyu Lin,Xiaoying Chu,Zhennv Deng,Hanwen Guo,Yaou Peng,Si Lu,Xianjing Zhou,Xinping Wang,Bailiang Wang

Bacterial keratitis (BK) and related inflammatory diseases causes irreversible damage to the corneal tissue. In this study, a novel polyacrylamide semi-interpenetrating network hydrogel including quaternized chitosan and tannic acid (PAM-QCS-TA) were used to construct a novel antibacterial and antioxidant contact lens. The obtained hydrogels showed high water content (>85%), swelling resistance, light transmittance (>90%) and adjustable mechanical property. Both quantitative and qualitative antibacterial experiments against Staphylococcus aureus and Escherichia coli ( E. coli ) indicated excellent sterilization function especially against E. coli (almost 100%). Due to the presence of tannin acid, it showed obvious antioxidant properties, which relieved oxidative stress and protect cells from reactive oxygen species-induced cytotoxicity. Animal experiments also indicated the shortened treatment time of BK (only 3 days) as well as the protection of eye tissue structure. Therefore, such drug-free antibacterial and antioxidant contact lens avoiding the development of drug resistance is a potential candidate in ocular infectious and inflammatory diseases treatment.

Effect of sodium alginate/phosphate-stabilized amorphous calcium carbonate nanoparticles on chitosan membranes

Bin Nie,Hong Wang,Yanwei Zhang,Chaohui Rao,Huifang Wang,Xianghua Gao,Wenfeng Li,Baolong Niu

Biodegradable chitosan (CS) films can meet the demand for sustainable development. However, the performance of pure CS membrane still exists a particular gap compared with the traditional film. Inorganic nanomaterials with the controllable release are added to improve its physical and chemical properties. Herein, a series of CS/phosphate-stabilized amorphous calcium carbonate (CS/ACCP) and CS/sodium alginate/ACCP (CS/Alginate/ACCP) composite films were prepared by the flow method. The effects of ACCP and Alginate/ACCP nanoparticles on the physical and chemical properties of the composite membrane were investigated. The results showed that the composite nanoparticles could significantly improve CS film's compactness, hydrophobicity , and mechanical properties and enhance its ultraviolet (UV) blocking ability, water resistance, and water vapor blocking ability. When the number of nanoparticles was 8%, the mechanical properties of the CS composite membrane reached optimum value, and the comprehensive performance was better. In addition, the controlled-release properties of CS composite membranes were also studied, and the antioxidant, antibacterial, biocompatibility, and fresh-keeping effects of the composite membranes were explored. The results indicated that the CS composite membranes had not only excellent bacteriostatic (72%) properties ( Escherichia coli ) but also presented well fruit preservation (15 days) properties (sugar orange). In particular, the controlled release range of CS composite membrane at 12 h was between 30% and 90%, which provided a theoretical basis for its use as an edible membrane. Therefore, based on ACCP and Alginate/ACCP nanoparticles, CS composite membranes with more excellent application value in the biological field were prepared through further optimization design.

Xiaohui Guo,Yuanfang Luo,Lijuan Chen,Bowen Zhang,Yongjun Chen,Demin Jia

To achieve simultaneous environment protecting, high-efficiency rubber antioxidant is still an enormous and long-term challenge in green rubber industry. Hence, a novel biomass antioxidant (denoted as silica-s-TP) was obtained by silica supported with green tea polyphenols exhibited prominent anti-aging functionality. Silica-s-TP hybrid particles were uniformly dispersed in rubber matrix with remarkable interfacial interaction , and the high-efficiency free radical capturing activity was stem from the construction of polyphenol structure on silica surface. Therefore, silica-s-TP brought to simultaneously enhance the performance of anti-aging property of SBR composites and surpass the achievements of the commercial antioxidants. Interesting, the biomass antioxidant silica-s-TP exhibited remarkable free radical-scavenging activities and stability in SBR matrix than the TP directly added in SBR matrix, due to the chemical anchor TP on silica surface. Such clay surface treatment technology with natural antioxidant may also bring to a new path for the preparation of green and eco-friendly functionalization rubber additives.

The differences in metabolites in male and female individuals of dioeciousMorus albaL. (Moraceae) are usually ignored and lack study. In the present study, 58 leaves and 61 stem barks from male and female individuals were analyzed by untargeted metabolomics via headspace solid-phase microextraction gas chromatography-mass spectrometry (HS-SPME-GC-MS) coupled with chemometrics, including principal component analysis (PCA) and orthogonal partial least squares-discriminant analysis (OPLS-DA). A total of 66 and 44 metabolites were identified from leaves and stem barks, respectively. Four and eight differential metabolites among candidate metabolites in leaves and stem barks from male and female individuals were identified. Moreover, females possessed stronger antioxidant activity than males. This is the first report where untargeted metabolomics coupled with chemometrics was used to analyze the different metabolites and to discriminate the gender of leaves and stem barks of dioeciousM. alba. It provided the basis for further study ofM. albaand reference value for researching dioecious plants. Keywords:Morus albaL.;dioecious plant;leaves;stem barks;untargeted metabolomics;chemometrics

Carboxymethyl chitosan incorporated with gliadin/phlorotannin nanoparticles enables the formation of new active packaging films

Jianbo Zhao,Haixin Jiang,Qiuyu Huang,Jingting Xu,Mengxia Duan,Shan Yu,Zijian Zhi,Jie Pang,Chunhua Wu

Advanced carboxymethyl chitosan (CMCS) based functional films were fabricated by involving some amounts of gliadin/phlorotannin nanoparticles (GPNPs) using a solution casting method. GPNPs were synthesized by an antisolvent precipitation approach, and they presented a spherical morphology with a mean diameter of 145.30 ± 2.06 nm. The effect of GPNPs concentration on the structural, physical, antioxidant and antimicrobial properties of CMCS-GPNPs (C-G) functional films was evaluated. It was found that the added GPNPs were homogeneously distributed over the whole CMCS matrix, allowing to reduce the free volume of the nanocomposite matrix and subsequently improve the physical properties of the final film (evidenced by mechanical and water barrier properties). FT-IR spectra indicated the intermolecular interactions, such as hydrogen bonds and electrostatic interaction, within the matrix of the nanocomposite films were increased. Impressively, the anti-ultraviolet properties, antioxidant activity and antimicrobial behaviors of the as-formed C-G functional films were greatly enhanced compared to the pure CMCS film. All these results suggested that our as-prepared C-G nanocomposite films could be a promising food packaging material.

Solvents effect on phenolics, iridoids, antioxidant activity, antibacterial activity, and pancreatic lipase inhibition activity of noni (Morinda citrifolia L.) fruit extract

Ruimin Wang,Lu Wang,Lin Zhang,Sitong Wan,Congfa Li,Sixin Liu

This study focused on the relationship between content levels of phytochemicals and the biological activities of noni ( Morinda Citrifolia L.) fruit extracts (NFEs) prepared with traditional solvents and deep eutectic solvents (DESs). The results indicated the total phenolic content in Bet-Gly (Betaine: Glycerol) extracts (11.89 mg GAE/g DW) and total iridoid content in 70% ethanol extracts (26.38 mg CE/g DW) were the highest. A total of 17 compounds were identified and quantified in NFEs. Traditional solvent extracts, except ethyl acetate, exhibited higher antioxidant activities than DESs. Three DES extracts showed higher activities against pancreatic lipase than traditional solvent extracts. Multivariate analysis revealed that the type of extraction solvent exerts a significant influence on the phytochemical compositions and biological activities of NFEs. This study provided valuable information on the efficient extraction of phytochemicals from noni fruits and DESs are promising green solvent for the extraction of bioactive compounds from noni fruits.

Efficient extraction of flavonoids from Polygonatum sibiricum using a deep eutectic solvent as a green extraction solvent

Hongli Zhang,Feilong Hao,Zhifang Yao,Junling Zhu,Xu Jing,Xiaowen Wang

Deep eutectic solvents (DESs) are becoming eco-friendly alternatives for extracting biologically-active substances from natural products. In this study, a DES is used as an eco-friendly solvent to extract flavonoids from Polygonatum sibiricum with the assistance of ultrasound. The antioxidant activity of the extracts was then investigated and the recyclability and reusability of the DES evaluated. Optimizing the DES extraction process produced the following optimum conditions: a betaine–acetic acid molar ratio of 1:4, water content of 30%, solid–liquid ratio of 1: 100 g mL −1 , extraction temperature of 50 °C, and extraction time of 30 min. The yield of extracted flavonoids reached 25.4 mg g −1 , which is about eight times that achieved using ethanol extraction. Furthermore, the antioxidant ability of the DES extract was more potent than that of the extract obtained using ethanol. Used DES can be efficiently recovered using ultrahigh capacitance porous activated carbon (the recovery yield of DES was around 92% of that of the original DES after ten reuses). In conclusion, DESs can be used to establish green ultrasound-assisted extraction methods for efficient extraction of plant flavonoids.

Stability profiling and degradation products of dihydromyricetin in Dulbecco's modified eagle's medium

H.L. Zhang,M.L. Wang,L.Z. Yi,P. Högger,R. Arroo,V.K. Bajpai,M.A. Prieto,X.J. Chen,J. Simal-Gandara,H. Cao

Dihydromyricetin has shown many bioactivities in cell level. However, dihydromyricetin was found to be highly instable in cell culture medium DMEM. Here, the underlying degradation mechanism was investigated via UPLC-MS/MS analysis. Dihydromyricetin was mainly converted into its dimers and oxidized products. At lower temperature, dihydromyricetin in DMEM showed higher stability. Vitamin C increased the stability of dihydromyricetin in DMEM probably due to its high antioxidant potential.

Functional Properties and Composition of New “Nut” Oil Obtained from Xanthium sibiricum Seeds

Sheng Zhou,Yuxiu Wen,Yiting Duan,Qi Li,Yuan Gao,Xiuzhu Yu

To develop new high-nutrient oil resources, eight Xanthium sibiricum seeds (XSs) are used as raw materials for the preparation of oil. The properties, chemical composition, and free radical scavenging capacity of XS oils (XSOs) are evaluated in this study. The characteristics of XSOs from different regions are also compared and analyzed. Results show that XSOs contains a high content of unsaturated fatty acids (up to 90.90%). XSOs are rich in phytochemicals, such as tocopherols (consisting of α-, β-, and γ-tocopherols) and phytosterols (including β-sitosterol, campesterol, and stigmasterol). The contents of phytochemicals and free radical scavenging capacity in XSOs from Gansu are higher than those in other regions. Furthermore, the free radical scavenging capacities of XSOs are significantly ( p < 0.05) affected by some minor components, especially in α-tocopherol. XSOs generally have high nutritional value, and they could be used as a new alternative to nut oil. Practical applications : The excellent chemical composition and functional properties of XSOs indicate that it could be used as a new edible oil through proper processing. Due to its high proportion of unsaturated fatty acids, XSOs is expected to become a functional oil for preventing cardiovascular and cerebrovascular diseases. XSOs could also be developed as a new low-cost “nut oil” with similar characteristics and higher active substance content.

Structural analysis of light-colored separated lignin (lignocresol) and its antioxidant properties

Qi Liu,Haonan Zhang,Hao Ren,Huamin Zhai

The use of lignin is limited by its heterogeneity and complexity. This study was processed using different methods to obtain spruce milled wood lignin (SMWL), spruce kraft lignin (SKL), and spruce lignocresol (SLC) for comparative analysis of the structure and antioxidant activity . SMWL has a complete softwood lignin side-chains structure and lignin carbohydrate complexes. SKL contains fewer ether bonds, while more conjugate structures and condensed structures contribute to the color. However, the α-position of the lignin side chain eliminates most of the hydroxyl and ether bonds (β-O-4/α-OH, phenylcoumaran, and dibenzodioxocine structure) and effectively grafts p -cresol in the phase separation reaction. It not only inhibits the self-condensation of lignin, but also forms the 1,1-diarylpropane unit while protecting β-O-4 linkages from not breaking. Importantly, SLC has few conjugate structures that result in the lightest color among all lignin isolated. Besides, SLC has a high yield and contains trace carbohydrates, indicating that the phase separation method can achieve great amounts of purity separated lignin. The antioxidant activity of lignin was evaluated, results show that 85% of the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radicals were scavenged at the end of 60 min. Owing to its unique color, structural properties, and continuous antioxidant activity, SLC has the potential to manufacture antioxidant cosmetics.

Probing the interaction of tert-butylhydroquinone and its β-cyclodextrin inclusion complex with bovine serum albumin

Jiali Gu,Yanxuan Ma,Zimu Li,Hongrui Liu,Qionghua Zhang

Tert-butylhydroquinone (TBHQ) is a synthetic antioxidant found widely in foods such as cooking oil, fried baked goods and meat products, however, if consumed in excessive quantities, it may cause nutritional problems and chronic diseases. The binding of TBHQ to transport protein before and after β-cyclodextrin (β-CD) encapsulation was investigated by multiple spectroscopies and molecular docking technology. At 298 K, the binding constant, enthalpy, and entropy of bovine serum albumin (BSA) with TBHQ were (1.18 ± 1.50) × 10 4 M −1 , (−110.11 ± 15.29) kJ mol −1 and (−293.36 ± 50.87) J mol −1 K −1 at 298 K, respectively. By forming a stable complex with BSA through van der Waals forces and hydrogen bonds, TBHQ altered BSA's secondary structure and microenvironmental polarity. However, the binding affinity of TBHQ with BSA decreased after encapsulation by β-CD, but the binding distance increased. Because of the encapsulation of β-CD, the interference of TBHQ with the BSA conformation was also inhibited. Fourier transform infrared spectroscopy, X-ray diffraction, and scanning electron microscope study confirmed the formation of an inclusion complex between CD and TBHQ. According to UV spectroscopy, TBHQ and β-CD have a binding constant of (1.21 ± 0.03) × 10 3 M −1 . The formation of TBHQ–β-CD inclusion complex inhibited the binding of TBHQ and BSA, possibly because the inclusion of β-CD prevented TBHQ from entering the hydrophobic cavity of BSA, which prevented TBHQ from binding to BSA, alleviating the negative effects of TBHQ on BSA conformation. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging results showed that the antioxidant activity of TBHQ was not affected by β-CD encapsulation.
