Brucellosis, acting as a typical chronic zoonotic disease, is caused by the invasion ofBrucellainto the human body. Outer membrane protein 25 (Omp25), specifically localized on theBrucellamembrane, is the main virulence factor ofBrucellaand participates in multiple links of the damage process. Omp25c, a porin protein ofBrucella, is a paralog of Omp25 with high sequence identity. NADH dehydrogenase [ubiquinone] complex I assembly factor 2 (Ndufaf2) has a key function in cell energy metabolism, particularly in the formation and activity of the mitochondrial respiratory chain. Loss of Ndufaf2 results in oxidative stress and mitochondrial DNA (mtDNA) deletion. However, the functional relationship between Omp25c and Ndufaf2, the underlying mechanism of the proteins, remains unclear. In this work, we purified the Omp25c and Ndufaf2proteins. Our data revealed that Omp25c directly interacts with Ndufaf2, as determined using Biacore analysis. In addition, assays revealed that Ompa2c reshapes the host cell’s redox environment by decreasing the oxidized nicotinamide adenine dinucleotide/reduced nicotinamide adenine dinucleotide (NAD+/NADH) ratioand adenosine triphosphate (ATP) production, whereas Ndufaf2 exerts an opposing regulatory effect; Co-expression results further revealed an antagonistic relationship between the two during metabolic processes. These findings provide a new perspective for elucidating the mechanisms of mitochondrial functional regulation inBrucella–host interactions and lay the theoretical and experimental foundation for drug development targeting metabolic interventions to eliminate intracellular pathogens.

The degradation of complex plant polysaccharides, particularly Rhamnogalacturonan I (RG-I) pectin, is essential for unlocking the value of biomass. This study focuses on the enzymatic toolbox of Penicillium oxalicum for efficient RG-I breakdown. Here, we recombinantly expressed 15 novel pectinolytic enzymes and characterized their biochemical functions. The enzyme system consists of backbone-cleaving rhamnogalacturonan hydrolases and lyases, along with side-chain–degrading enzymes. Specific endo-β-galactanases (GH30, GH43, GH53) and an exo-β-galactosidase (GH35) collaboratively hydrolyze galactan side chains. Meanwhile, α-arabinanases (GH43) and exo-α-arabinofuranosidases (GH54, GH62) work synergistically to degrade arabinan side chains. Through coordinated action, these enzymes achieve complete degradation of complex RG-I structures. Our findings provide a comprehensive enzyme set for targeted polysaccharide deconstruction, offering valuable resources for biomass conversion and pectin valorization in industrial applications.

Zearalenone (ZEN) is a common mycotoxin widely present in food and agricultural products, posing a serious threat to human health. This work developed a fluorescent/smartphone-assisted colorimetric dual-mode aptasensor for detecting ZEN. Platinum nanowires-supported MXene (PtNWs/MXene) composites exhibit peroxidase-like (POD) activity, which can be inhibited by the adsorbed ZEN-specific DNA aptamers. In the presence of ZEN, the specific binding between ZEN and the aptamer triggers desorption of the aptamer from the PtNWs/MXene surface, thus restoring the POD activity of composites. With hydrogen peroxide introduced, PtNWs/MXene catalyzes the oxidation of colorless o-phenylenediamine into colored 2,3-diaminophenazines, simultaneously generating a fluorescent signal. Exonuclease I mediates target cycling to achieve signal amplification, further boosting the detection sensitivity. The smartphone-based colorimetric mode shows a linear detection range of 10 ~ 900 ng/mL with a limit of detection (LOD) of 6.57 ng/mL, while the fluorescence mode presents a linear range of 1 ~ 1000 ng/mL and a LOD as low as 0.74 ng/mL. This sensing platform can accurately distinguish ZEN-positive and ZEN-negative samples, and the detection results are highly consistent with those of ELISA. It provides an efficient and reliable new solution for highly sensitive on-site ZEN detection, demonstrating significant application potential in food safety monitoring. The equilibrium mole fraction solubility of tanshinone I in acetone, N,N-dimethylformamide (DMF), methanol, ethanol, water, ethylene glycol (EG), n-propanol, isopropanol, and 1-butanol as well as methanol/ethanol/DMF/acetone + water was determined by the isothermal saturation method at 278.15–323.15 K under 101.2 kPa. The temperature and the composition of solvent mixtures play a decisive role in the solubility of tanshinone I. Then, four monosolvent models (Apelblat, van’t Hoff, λh, and NRTL) and three cosolvency models (Jouyban–Acree (J-A), van’t Hoff–Jouyban–Acree (V-J-A), and modified Apelblat–Jouyban–Acree (A-J-A)) were utilized to correlate the mole fraction of tanshinone I. The maximum deviation (relative average deviation (RAD) and root-mean-square deviation (RMSD)) between the experimental data and the solubility data calculated by the monosolvent models are 6.21% and 3.02 × 10–4, respectively. The maximum of RAD and RMSD for the mixture systems are 1.58% and 1.35 × 10–4, respectively. The Kamlet and Taft linear solvation energy relationship model was used to analyze the solvent effect, and the nonspecific dipolarity/polarizability interactions between the solute and solvent played a major promoting role. All in all, the experimental and computational data are able to provide basic data for the optimization and improvement of the recrystallization process of tanshinone I.

Hepatic stellate cells (HSCs) activation represents a central pathological mechanism in liver fibrosis, with emerging evidence implicating fatty acid metabolic reprogramming as a critical regulator of this process. Our study established the vitamin D receptor (VDR) as a key transcriptional coordinator of fatty acid metabolism during HSC activation. Genetic VDR deletion in mice exacerbated liver fibrosis progression, which was associated with elevated TGF-β1 levels and increased Smad3 phosphorylation. Mechanistically, VDR deficiency disrupted lipid homeostasis through the upregulation of lipogenic enzymes (fatty acid synthase, acetyl-CoA carboxylase 1, ATP citrate lyase) and desaturases (stearoyl-CoA desaturase-1, fatty acid desaturases 1/2) and the suppression of the β-oxidation gatekeeper carnitine palmitoyltransferase 1A (CPT1A). Pathological VDR downregulation was observed in both TGF-β1-activated HSCs and fibrotic liver tissues, suggesting a disease-associated regulatory circuit. Calcitriol-mediated VDR activation reversed TGF-β1-induced Smad3 phosphorylation and normalized metabolic enzyme expression, effectively reducing lipid accumulation and collagen deposition. We further identified senkyunolide I as a novel natural VDR agonist that rebalances fatty acid metabolism by simultaneously downregulating lipogenesis/desaturation machinery and upregulating CPT1A. The complete abolition of anti-fibrotic effects of senkyunolide I following VDR knockdown confirmed its strict receptor dependency. These findings identify VDR as a master regulator of metabolic reprogramming in HSC activation and validate pharmacological VDR activation as a promising therapeutic strategy for liver fibrosis. The dual metabolic regulatory capacity of senkyunolide I through VDR signaling highlights its potential for targeted antifibrotic intervention.

Polydopamine (PDA) possesses potential as a photothermal agent for tumor treatment owing to its photothermal capacity, chemical modifiability, excellent biocompatibility, and selective biodegradability. However, weak near-infrared (NIR) light absorption seriously hinders its actual utilization, especially in NIR-II. This study presents a novel molecularly modified PDA with remarkably increased NIR absorption and remarkably enhanced photothermal conversion capacity in both NIR-I and NIR-II. Dopamine monomers are covalently bound with elaborately selected 2,6-pyridinedicarboxylic acid (DPA) through an amidation reaction and then undergo an oxidative polymerization to form DPA-doped PDA nanoparticles (DPA-PDA NPs). By constructing donor–acceptor pairs and extending the conjugation length of molecular units, the energy band gap of DPA-PDA between the highest occupied molecular orbital and the lowest unoccupied molecular orbital is diminished from 0.77 to 0.45 eV, facilitating the electron low-energy transition. Meanwhile, the content of carbon-centered free radicals is obviously raised via suppressing their dimerization and quenching by the conjugation effect and steric hindrance effect, where the strengthened electron spin and vibration may promote charge and energy transfers, increasing and accelerating the excited electron nonradiative decay. As a result, the DPA-PDA NPs exhibit the enhanced NIR light absorption (196 and 205% over PDA) and much higher photothermal conversion efficiency (1.4 and 2.1 times that of PDA) under 808 and 1064 nm irradiation, respectively, achieving more effective tumor cells inhibition in vitro and tumor ablation in vivo. Interestingly, photothermal therapies with DPA-PDA for tumor-bearing mice emerge with higher tumor elimination (nearly 100%) under 1064 nm irradiation than that under 808 nm irradiation (94.3%), without recurrence within 20 days, which is attributed to both the enhanced photothermal capacity of DPA-PDA and the stronger tissue penetration ability of NIR-II. This design provides a new option to enhance the photothermal capacity of polydopamine as an applicative photothermal agent for tumor therapy.

Incorporating two-dimensional (2D) perovskite phases at the interfaces of three-dimensional (3D) perovskite solar cells improves device performance. These 2D structures form in the bulk and at interfaces of the perovskite film by adding long-chain ammonium salts into the perovskite, but achieving them exclusively at the buried interface remains challenging. Here we sequentially graft thioglycolic acid and oleylamine onto SnO 2 nanoparticles. The strong chemical bond between thioglycolic acid and oleylamine ensures that cation exchange with formamidinium iodide, a perovskite precursor, occurs only during thermal annealing, creating a 2D/3D perovskite structure solely at the bottom interface. This localized 2D layer accelerates 3D phase formation, enhances perovskite crystallization and reduces defect concentration at the interface by over tenfold. The resulting solar cells achieve power conversion efficiencies of 26.19% (certified 26.04%, 0.09 cm 2 ), 23.44% (aperture area of 21.54 cm 2 , certified 22.68%) and 22.22% (aperture area of 64.80 cm 2 ), respectively.

Inorganic metal halide perovskite quantum dots (PQDs) have recently emerged as promising candidates for color conversion materials in Micro-LED displays, due to their outstanding color purity, high absorption coefficient, tunable emission wavelength . However the inferior stability and undesirable photoluminescence quantum yield (PLQY) restrict their particle applications. Herein, CsPbX 3 /PMMA (X = Br, I) composites with improved stability and optical performance were successfully prepared by a general and mild in-situ polymerization strategy. The ester carbonyl (C O) group in MMA formed complexes with Pb 2+ , leading to the in-situ thermal polymerization of polymethyl methacrylate (PMMA) under the action of the initiator. The multifunctional PMMA can efficiently passivate halogen vacancies while acting as a barrier against the degradation caused by heat, water, and oxygen. As a result, the PLQYs and stability of PMMA-coated CsPbX 3 PQDs have been significantly improved. When implementing CsPbX 3 /PMMA/PDMS composites as color conversion layers in Micro-LEDs, they achieve an impressive color gamut of 127.23% with excellent stability. We believe our work will stimulate the exploration of perovskite/polymer composites for industrial Micro-LED applications.

Differences in the prebiotics of pectin cannot be fully explained by the sources and single structure. Microbiota exhibit distinct preferences for different structure of pectin, influencing the microbial composition by lowering pH and producing short-chain fatty acids (SCFAs) in the fermentation. However, the effect on the pectin fine structure on its fermentation properties and microbiota still controversial. Here, three pectin polysaccharides with different degree of esterification (DE) and rhamngalacturonan-I (RGI) ratio while ensuring that either the degree of RGI or the DE remained constant were prepared. By using in vitro fermentation model and microbiomics, the mechanisms by which the pectin fine structure in modulating the composition of gut microbiota and the metabolism of SCFAs were proposed. Pectins enriched with RGI structures promoted the growth of Phascolarctobacterium and Parabacteroides, and increased concentration of the SCFAs (such as acetic acid and propionic acid) associated with fat loss. Bacteria groups such as Tyzzerella , Collinsella , Prevotella-9 , Dialister and Sutterella were sensitive to DE and thereby promote SCFA production (such as hexanoic acid, butyric acid, etc). As pectin promotes the growth of probiotics and an increase in SCFAs, contributing to a decrease in pH, the growth of Lachnospiraceae_UCG-004 , Escherichia-Shigella and Lachnoclostridium is significantly inhibited. We identified thatkey roles of the DE and RGI within pectin in promoting the growth of probiotic microbiota and production of SCFAs. These results provide new a reference for the prediction of the pectin physiological activity and the development of functional pectin polysaccharide prebiotics.

Glutathione (GSH) plays an important role in the biological system and serves many cellular functions. Since all of the biothiols possess similar functional groups, it is still challenging to selectively detect GSH over cysteine (Cys) and homocysteine (Hcy). In this work, a novel and simple colorimetric method for discriminative detection of glutathione (GSH) over Cys and Hcy is developed. The proposed method is based on the fact that Ag [I] ion could oxidize 3,3′,5,5′,-tetramethylbenzidine (TMB) to the oxidized TMB to induce a blue color and an absorption peak centered at 652 nm. However, the introduction of GSH could cause the reduction of oxidized TMB and it could also combine with Ag + , both of which result in a blue color fading and a decrease of the absorbance at 652 nm. Based on this finding, we propose a method to qualitatively and quantitatively detect GSH by naked eyes and UV–vis spectroscopy, respectively. The proposed method shows a low detection limit of 0.1 µM by naked eyes and 0.05 µM with the help of UV–vis spectroscopy. In addition, this method has great potential in discriminatively detecting GSH over other amino acid and biothiols. More importantly, this method is simple and fast without the preparation of nanomaterials and has also been successfully applied to the detection of GSH in biological fluids.

The all-inorganic perovskite CsPbI 3 quantum dots are difficult to achieve pure red stable luminescence due to their poor lattice thermodynamic stability and non-radiative recombination loss caused by surface halide vacancies. Therefore, the pure red perovskites are usually achieved by mixing the composition of halides (Br and I). In this work, CsPb(Br/I) 3 quantum dots (QDs) were synthesized by the hot-injection method under the synergistic effect of homophthalic acid (HA), 2-bromoethanesulphonic acid sodium salt (SBES), and oleylamine (OAm). By adjusting the molar ratio of passivates, the synergistic effect of multiple ligands can not only passivate the unliganded Pb 2+ -related defects on the surface of NCs, but also increase the formation energy of halide vacancies. Meanwhile, due to the bidentate structure of HA, smaller size of perovskites nanocrystals were obtained. Eventually, synergistic boost in device performance is achieved for color-by-blue white light-emitting diodes (WLEDs) with CIE coordinates of (0.3319, 0.3297) and have an external quantum efficiency (EQE) of 10.13%. This strategy paves an efficient way for improving the efficiency and stability of CsPb(Br/I) 3 NCs.

Extraction and utilization of hot carriers are considered viable approaches to surpass the Shockley–Queisser limit of perovskite solar cells (PSCs). Here, we introduce N,N′-bis(4-trifluoromethoxybenzyl) naphthalene-1,4,5,8-tetracarboxylic acid diimide (denoted as NDI-BOCF3) to modify the buried SnO2/perovskite interface, simultaneously accelerating hot-electron extraction and passivating interfacial defects. The NDI-BOCF3-modified SnO2 (denoted as SnO2–NDI-BOCF3) exhibits improved surface uniformity and optimized energy-level alignment and passivates oxygen vacancies (VO), thereby providing ∼22% enhancement in hot-electron extraction. Nonadiabatic molecular dynamics simulations confirmed that NDI-BOCF3 enhances hot-electron extraction via higher-frequency lattice vibrations and more ordered nonadiabatic couplings at the interface. In addition, NDI-BOCF3 passivates multisite defects across various perovskite terminations. As a result, SnO2–NDI-BOCF3 devices achieve a power conversion efficiency (PCE) of 25.06%, compared to 23.96% for a device with pristine SnO2. An increase of PCE from 8.81 to 17.50% for SnO2-free devices is also achieved. Furthermore, due to passivation, the activation barrier for positively charged iodine vacancies (VI+) increases from 0.80 to 0.87 eV, resulting in an enhancement in the thermal and photostability of the perovskite thin films.
