Physalis alkekengi L. is one of the most popular berries in the world, and its calyx is rich in nutrients. Still, the study of soluble dietary fiber (SDF) from Physalis alkekengi L. calyx (PC) has not been reported. This study aimed to extract Physalis alkekengi L. calyx soluble dietary fiber (PCSDF), and the optimal extraction process was investigated using response surface methodology. After ultrasonic pretreatment, the yield of PCSDF reached 8.74 ± 0.17% under the conditions of liquid-solid ratio of 30 mL/g, thermostable α-amylase concentration of 2000 U/mL, dispase concentration of 20,000 U/mL, amyloglucosidase concentration of 40,000 U/mL. For the first time, PCSDF was characterized by UV spectroscopy, infrared spectroscopy, scanning electron microscopy and thermogravimetric analysis, and the composition of the monosaccharides, namely mannose, glucuronic acid, rhamnose, galacturonic acid, glucose, galactose, xylose, arabinose and fucose, was identified by HPLC. For antioxidant activity, the half inhibitory concentration (IC 50 ) against DPPH, ABTS + , ·OH, and ·O 2 − radicals were 3.67, 2.96, 5.45, and 2.90 mg/mL, respectively, and there was a specific Fe 3+ reducing capacity. In terms of hypoglycemic feature, the IC 50 values for the inhibitory ability of α-glucosidase and α-amylase were 1.20 and 9.21 mg/mL, respectively. For lipid-lowering potential, the IC 50 value for pancreatic lipase inhibitory ability for lipid-lowering was 16.95 mg/mL. This paper fills the research gap of PCSDF and provides a basis for further evaluation of PCSDF using in vivo studies. Graphical Abstract

Hydrogenations achieved on supported metal catalysts are normally structure sensitive, and comprehensive understandings on such sensitivity are pivotal for gaining insights into the active sites as well as the design of catalysts. Herein, a serials of differently sized Ru nanoparticles supported on carbon nanotube (CNT) were prepared and employed as catalysts for L-alanine hydrogenation to examine the structure sensitivity of amino acid hydrogenation. The reaction rates for L-alanine conversion and the formation of alaninol are demonstrated to be strongly dependent on the sizes of Ru nanoparticles, highlighting the structure sensitivity of the L-alanine hydrogenation. The activation energies extracted from kinetics studies are insensitive to the sizes of Ru nanoparticles on the Ru catalysts sized ≥1.3 nm with similar electronic properties, pointing to a predominant type of active site for L-alanine hydrogenation. By further combining model calculations with the shape of Ru nanoparticles determined by transmission electron microscopy, the Ru(101) sites are identified as the dominant active sites for L-alanine conversion and alaninol formation, which is further rationalized by density functional theory calculations. The kinetics insights into such structure sensitivity are believed to be important for the design and optimization of catalysts for the reaction.

This paper investigated the static in vitro degradation behavior of poly(L-lactic acid) (PLLA)-oriented microcellular materials. The study compared the influence of different molecular chains and internal morphological structures on water penetration into the material. Also, the relationship between thse self-accelerated degradation caused by ester bond breakage and reduced material molecular weight and mechanical properties was explored. Furthermore, the mechanism behind the static in vitro degradation of oriented microcellular PLLA materials in simulated human body fluid was explored, laying the foundation for regulating the mechanical performance decline of oriented microcellular biomimetic bone repair materials to match specific bone healing periods.

L-threo-methylsulonylphenylserine (L-threo-MSPS) is a crucial intermediate in florphenicol and thiamphenicol synthesis. L-threo-MSPS can be synthesized from L-threonine and p-methylsulfonylbenzaldehyde (p-MSBA) chiral selectively under the catalyse of L-threonine transaldolase . However, the byproduct acetaldehyde formed during the reaction process severely hinders the biosynthesis of high-quality L-threo-MSPS. The accumulated acetaldehyde reacts with L-thero-MSPS and L-threonine to form N-derivatives, and the activity of L-threonine transaldolase decreases to 14.10% even when the concentration of acetaldehyde is as low as 0.5 g/L. The acetaldehyde could be removed by aeration, and the removal efficiency is closely related to the aeration conditions, which was described by the volumetric oxygen transfer coefficient k L a in this paper. Regular aeration through pipes cannot provide enough aeration efficiency, and filters or sintered rods were used to increase the k L a value during the reaction. The residual concentration of acetaldehyde depended on the balance between the formation and removal speed of acetaldehyde. When the optimal enzyme concentration was applied and the k L a value of 5.5 min -1 , the L-threo-MSPS concentration reached 293.28 mM in a 1-L reaction, and the highest conversion reached 90.03%. The need of coenzyme NAD + was cut off in this method, and only one enzyme was applied. Consequently, the reaction was simpler and easier to control.

Alpha-ketoglutaric acid (α-KG), as an intermediate product of the tricarboxylic acid cycle, plays a crucial role in peptide and amino acid synthesis. In order to reduce costs and improve efficiency in the oxidative production of α-ketoglutaric acid, this study successfully synthesized and expressed L-glutamate oxidase (LGOXStr) fromStreptomyces viridosporusR111 and catalase (KatGEsc) fromEscherichia coliH736. Two immobilization methods and the conditions for one-step whole-cell catalysis of α-ketoglutaric acid were investigated. α-Ketoglutaric acid has broad applications in the pharmaceutical, food, and chemical industries. The specific research results are as follows: (1) By fusing the sfGFP tag, L-glutamate oxidase (LGOXStrr) and catalase (KatGEsc) were successfully anchored to the outer membrane ofEscherichia colicells, achieving one-step whole-cell catalysis of α-ketoglutaric acid with a conversion efficiency of up to 75%. (2) Through the co-immobilization of LGOXStrand KatGEsc, optimization of the preparation parameters of immobilized cells, and exploration of the immobilization method usingE.coli@ZIF-8, immobilized cells with conversion rates of over 60% were obtained even after 10 cycles of reuse. Under the optimal conditions, the production rate of α-ketoglutaric acid reached 96.7% in a 12 h reaction, which is 1.1 times that ofE. coli@SA and 1.29 times that of free cells.

A novel polymeric nanosensor (named PEINAC) based on polyethyleneimine (PEI), was designed for the selective fluorescence detection of l -lysine (L-Lys) in aqueous solutions. The sensor was synthesized through a one-step, three-component reaction involving orthophthalaldehyde (OPA), PEI, and acetylcysteine. This reaction simultaneously facilitated the creation of an isoindole fluorophore, which was chemically attached to the PEI backbone. The structural properties, size, and morphology of PEINAC were thoroughly analyzed using various characterization techniques. When introduced into a buffered solution at pH 7.0, PEINAC demonstrated high specificity for L-Lys, inducing a marked fluorescence enhancement at 450 nm upon excitation at 367 nm. The fluorescence intensity exhibited a linear relationship with L-Lys concentration, ranging from 1 μM to 1000 μM, with a detection limit of 0.13 μM. Notably, the sensor exhibited excellent selectivity, showing no significant interference from other biomolecules and common transition metal ions and anions. This sensor was successfully applied for L-Lys quantification in blood and urine samples and for cellular L-Lys imaging, demonstrating its potential in various analytical and biomedical applications.

Smilax chinaL. (Chinaroot) is a natural herb that has multiple uses, such as being used to make tea and food. Both its roots and leaves have different uses due to their unique components. In this study, we analyzed the extract ofS. china.roots using LC-HRMS and evaluated the neuroprotective effects and metabolic regulation ofS. chinaonCaenorhabditis elegans.Chinaroot extract prolonged the life span of healthy nematodes, delayed the paralysis time of transgenic CL4176, and reduced the level of β-amyloid deposition in transgenic CL2006. The comprehensive analysis of metabolomics and qRT-PCR revealed that Chinaroot extract exerted neuroprotective effects through the valine, leucine and isoleucine degradation and fatty acid degradation pathways. Moreover, we first discovered that the expressions ofT09B4.8,ech-7, andagxt-1were linked to the neuroprotective effects of Chinaroot. The material exerted neuroprotective effects by modulating metabolic abnormalities in AD modelC. elegans. Glutamic acid (Glu) plays an important role in controlling and regulating the central nervous system and serves as a biomarker for liver diseases. The present detection methods need sophisticated instruments, professional personnel, or are confined for practical applications due to the issues of chiral recognition and the susceptible sensing signal. Here, an L -Glu imprinted two-dimensional photonic hydrogel (Glu-MIPH) was constructed to achieve highly selective, accurate and facile detection of L -Glu by combining molecular imprinting and two-dimensional photonic crystal (2DPC) hydrogel sensing technology. Upon exposure to L -Glu solution, the hydrogel shrinks because L -Glu enters the nanocavities that are like L -Glu in shape, size and binding sites, then forms hydrogen bonds with the amide and carboxyl groups in the hydrogel and increases the crosslinking density. The contracted hydrogel decreases the particle spacing of the 2DPC embedded in the Glu-MIPH, displaying an increase in the Debye diffraction ring diameter. The spacing decrement is linear when L -Glu changes in 1–10 μM and 20–100 μM. The limit of detection is found to be 0.96 μM. The Glu-MIPH demonstrates excellent selectivity, anti-interference, reproducibility for L -Glu, and has been successfully used to determine L -Glu in soy sauce, providing an alternate strategy for chiral recognition of amino acids.

Background Viral pneumonia is an infection of the lungs caused by numerous different viruses, which can lead to severe respiratory distress and even life-threatening conditions. In the absence of specific treatments for viral pneumonia, natural traditional medicines offer an alternative in terms of innovative drug therapies. Morus alba L. (common name mulberry leaf) is a Chinese medicine that has been used clinically as an antiviral. Purpose The therapeutic effect of M. alba on viral pneumonia was investigated along with its mechanism of action. Methods Network pharmacology and molecular docking were used to analyze the mechanism of action of M. alba in the treatment of viral pneumonia. Histology, immunofluorescence, Western blotting, qPCR, and flow cytometry were used to evaluate the protective effect of MLE (the ethanol extract of Morus alba L. ) on PR8 (A/Puerto Rico/8/1934 H1N1, a murine lung-adapted influenza A virus strain)-induced viral pneumonia. SiRNA was used to validate the relationship between the therapeutic effects of MLE on viral pneumonia and the target Syk (a crucial non-receptor tyrosine kinase). Results MLE alleviated PR8-induced viral pneumonia by reducing inflammatory factor expression in the lungs, decreasing NF-κB pathway activation, slowing oxidative damage in the lungs, and inhibiting lung tissue cell apoptosis. Meanwhile, MLE for viral pneumonia was significantly associated with Syk targets. Notably, knockdown of the Syk gene not only reduced the therapeutic effect of MLE, but also suppressed PR8-induced viral pneumonia. Conclusion MLE can alleviate PR8-induced viral pneumonia through inhibiting the Dectin-1/Syk pathway.

Eco-friendly silk fibroin/poly(D,L-lactide-co-glycolide) (SF/PLGA) materials were successfully fabricated using a facile strategy. The materials were characterized by scanning electron micrograph (SEM), X-ray diffraction (XRD) and infrared spectra (IR). Systematic investigations were completed to examine the degradation rates in natural soil, cytocompatibility and thermostability of the materials. It is interesting to find that after SF was hybridized by PLGA, the thermostability and degradation rate increased. Meanwhile, the materials show good cytocompatibility. SEM, IR and XRD results reveal that there is hardly any interaction between SF and PLGA in the SF/PLGA material, and SF is physically mixed with PLGA. This study opens up a new horizon in the design and preparation of SF-based materials for promising applications in medical and biodegradable material fields.

Plant protoplasts are very useful in plant biotechnology, molecular biology, and cell biology. However, an efficient method for protoplast production remains a challenge for many economically important dicotyledonous vegetables, including lettuce (Lactuca sativa L.). Herein, a protocol was optimized for efficient protoplast production from various tissues (leaf and shoot apex) of different lettuce subtypes (romaine, loose-leaf, and semiheading lettuces) by optimizing the major factors affecting protoplast yielding. The optimized protocol yields protoplasts up to 2.0 × 107/g (fresh weight, FW) with viability more than 85%, which is 3–10 times higher than those previously reported. This optimized protocol was also found to be applicable to other dicotyledonous plants (bok choy (Brassica parachinensis), celery cabbage (Brassica pekinensis), and Arabidopsis thaliana) for efficient protoplast production from leaves and shoot apexes. Moreover, an optimized poly(ethylene glycol)-mediated transient expression system (TES), using lettuce shoot apex protoplasts generated via the aforementioned protocol, exhibited high transfection efficiency exceeding 80%. This was further evidenced by the elevated expression levels and subcellular localization of four representative plasma membrane transporters: AAP2, ABCG22, ALMT10, and OATP. In conclusion, the optimized protoplast production protocol along with TES developed in this study will be useful tools for the functional analyses of genes in lettuce and other important dicotyledonous vegetables.

Nucleating agents, especially those with small particle sizes, are preferred to boost the nucleation density and crystallinity of poly(lactic acid) (PLA) due to its weak crystallization capability. Organophilicly modified nanofillers hardly alter the nucleation and crystallinity of non-isothermally crystallized PLA. Herein, nano-silica adsorbed trace D -sorbitol (m-SiO 2 ) as a heterogeneous nucleating agent was melt-mixed with poly( L -lactic acid) (PLLA), and the isothermal and non-isothermal crystallization behavior, as well as crystallization kinetics, were investigated. Transmission electron microscopy (TEM) revealed that m-SiO 2 was uniformly dispersed in the PLA matrix as 100–300 nm clusters. Differential scanning calorimetry (DSC) and polarized optical microscopy (POM) showed that the nucleation rate and density of the non-isothermally crystallized PLLA/m-SiO 2 composites were significantly improved. Despite the fact that m-SiO 2 does not raise the overall non-isothermal crystallization rate, the crystallization temperature and crystallinity of the PLLA/3%m-SiO 2 composite increased from 97.2 °C and 6.8 % for neat PLLA to 108.2 °C and 48.6 % (10 °C/min cooling rate), respectively. The Avrami exponent n of isothermal crystallization remains unchanged, while the crystallization rate increases dramatically. Both isothermal and non-isothermal crystallization have increased activation energies. The heat deflection temperature increased from 59 °C of neat PLLA to 152 °C with a 50 % increase in impact strength.
