Fabrication and Evaluation of Alginate/Bacterial Cellulose Nanocrystals–Chitosan–Gelatin Composite Scaffolds

Zhengyue Li,Xiuqiong Chen,Chaoling Bao,Chang Liu,Chunyang Liu,Dongze Li,Huiqiong Yan,Qiang Lin

It is common knowledge that pure alginate hydrogel is more likely to have weak mechanical strength, a lack of cell recognition sites, extensive swelling and uncontrolled degradation, and thus be unable to satisfy the demands of the ideal scaffold. To address these problems, we attempted to fabricate alginate/bacterial cellulose nanocrystals-chitosan-gelatin (Alg/BCNs-CS-GT) composite scaffolds using the combined method involving the incorporation of BCNs in the alginate matrix, internal gelation through the hydroxyapatite-d-glucono-δ-lactone (HAP-GDL) complex, and layer-by-layer (LBL) electrostatic assembly of polyelectrolytes. Meanwhile, the effect of various contents of BCNs on the scaffold morphology, porosity, mechanical properties, and swelling and degradation behavior was investigated. The experimental results showed that the fabricated Alg/BCNs-CS-GT composite scaffolds exhibited regular 3D morphologies and well-developed pore structures. With the increase in BCNs content, the pore size of Alg/BCNs-CS-GT composite scaffolds was gradually reduced from 200 μm to 70 μm. Furthermore, BCNs were fully embedded in the alginate matrix through the intermolecular hydrogen bond with alginate. Moreover, the addition of BCNs could effectively control the swelling and biodegradation of the Alg/BCNs-CS-GT composite scaffolds. Furthermore, the in vitro cytotoxicity studies indicated that the porous fiber network of BCNs could fully mimic the extracellular matrix structure, which promoted the adhesion and spreading of MG63 cells and MC3T3-E1 cells on the Alg/BCNs-CS-GT composite scaffolds. In addition, these cells could grow in the 3D-porous structure of composite scaffolds, which exhibited good proliferative viability. Based on the effect of BCNs on the cytocompatibility of composite scaffolds, the optimum BCNs content for the Alg/BCNs-CS-GT composite scaffolds was 0.2% (w/v). On the basis of good merits, such as regular 3D morphology, well-developed pore structure, controlled swelling and biodegradation behavior, and good cytocompatibility, the Alg/BCNs-CS-GT composite scaffolds may exhibit great potential as the ideal scaffold in the bone tissue engineering field. Keywords:alginate;bacterial cellulose nanocrystals;internal gelation;layer-by-layer electrostatic assembly;composite scaffolds;bone tissue engineering

Porous PLGA-PEG nerve conduit decorated with oriented electrospun chitosan-RGD nanofibre

Yuzhe Chen,Wenxuan Zhao,Honglian Dai

Chitosan (CS) is a type of natural polysaccharide, widely used as a biomaterial on account of its great biocompatibility and the multitudes of functional groups on the long chitosan chain, making it easy for property augmentations. Nevertheless, the lack of workability and mechanical capacity limits the applications. Herein, electrospinning was chosen to fabricate oriented chitosan/polyethylene oxide (PEO) nanofibres decorated by integrin-targeting polypeptides (RGD, Arg-Gly-Asp), which is essential for cell adhesion. At the same time, porous PLGA-PEG membranes were taken as a shell of the composite film, which can provide mechanical support for the scaffold. Adequate topological anisotropic information from oriented nanofibres helps the scaffold simulate the natural axon structure, which can induce accelerated regeneration of neurotmesis. Moreover, β-Tricalcium Phosphate (β-TCP) nanoparticles were brought into nanofibers to neutralize the acidic environment produced by the degeneration of PLGA-PEG. Cell viability tests at the in vitro level and dorsal root ganglion (DRG, 1-day sucking SD rat) co-culture detection proved that the scaffold possesses excellent biocompatibility while guiding oriented regeneration of damaged nerves. In this study, a CS-RGD/PEO/β-TCP/PLGA-PEG porous composite scaffold with low cytotoxicity was fabricated. The scaffold can not only lead to the oriented growth of nerves but also has selectivity, which can help nutrient exchange while inhibiting the invasion of fibrous tissue.

Chitosan-Based Glycolipid Conjugated siRNA Delivery System for Improving Radiosensitivity of Laryngocarcinoma

Jing Miao,Liwen Zhang,Peng Gao,Huawei Zhao,Xianji Xie,Junyan Wang

Glucose Transporter-1 (GLUT-1) is considered to be a possible intrinsic marker of hypoxia in malignant tumors, which is an important factor in radioresistance of laryngocarcinoma. We speculated that the inhibition of GLUT-1 expression might improve the radiosensitivity of laryngocarcinoma. GLUT-1 siRNA was designed to inhibit the GLUT-1 expression, but the high molecular weight and difficult drug delivery limited the application. Herein, we constructed a glycolipid polymer chitosan oligosaccharide grafted stearic acid (CSSA) to conjugate siRNA via electrostatic interaction. The characteristics of CSSA and CSSA/siRNA were studied, as well as the radiosensitization effect of siRNA on human laryngocarcinoma epithelial (Hep-2) cells. Compared with the traditional commercial vector LipofectamineTM2000 (Lipo), CSSA exhibited lower cytotoxicity, more efficiently cellular uptake. Incubating with CSSA/siRNA, the survival rates of Hep-2 cells were significantly decreased comparing with either the group before transfection or Lipo/siRNA. CSSA is a promising carrier for efficient siRNA delivery and radiosensitization of laryngocarcinoma.Keywords:GLUT-1;siRNA;chitosan oligosaccharide grafted stearic acid;laryngocarcinoma;radiosensitization

Laccase mediated phenol/chitosan treatment to improve the hydrophobicity of Kraft pulp

Ni Shuzhen,Liu Na,Fu Yingjuan,Gao Hailong,Qin Menghua

Abstract Herein, a great potential approach initiated by laccase is applied to graft the biopolymers (chitosan and phenols) onto the unbleached Kraft pulp fibers to enhance the hydrophobicity and strength. The results showed that the chitosan and laccase/phenols exhibited a synergistic action on the significant improvement of the hydrophobicity of the KP pulp fibers. Compared with the base paper, a high sizing degree value (227.08 s) was achieved when the fibers were treated by the laccase/4-hexyloxyphenol/chitosan (Lac/HP/CTS) system. The water retention value of the corresponding KP fibers reduced from 176.8 to 171.3%, and the Kappa number of treated fibers increased from 26.8 to 29.3, confirming the successful graft of HP onto the fibers. The effect of the reaction parameters, including the temperature, pH, HP and chitosan dosage, reaction time and pulp suspension consistency on the strength and hydrophobicity was investigated. pH value was found as the most remarkable factor influencing the sizing degree and gave a highest sizing value (506.2 s) at pH 5.5. The strength performance presented an opposite trend against the hydrophobicity, since the number of formed hydrogen bonds is the least when the hydrophobicity is the best. This work is extremely significant for providing valuable reference on improving the hydrophobicity and strength performance of papers through a green and biological way. Graphic abstract

A stable biosensor for organophosphorus pesticide detection based on chitosan modified graphene

Jianhua Zhang,Huaying Hu,Pengchang Wang,Chi Zhang,Majiaqi Wu,Lianqiao Yang

An acetylcholinesterase (AChE) biosensor was successfully fabricated with a stable structure and high detection accuracy. Graphene (Gra) nanofragments modified with chitosan (CS) and AChE were successively drip coated on the surface of a glassy carbon electrode via a layer-by-layer assembly method. The concentration range of the sensor to detect dichlorvos was 0.1–100,000 nM, and the limit of detection was 54 pM. CS was used to modify Gra for the first time, which enhanced the mechanical flexibility of these Gra nanostructures, significantly improving the stability and detection accuracy of this sensor.

Lactobionic acid-modified thymine-chitosan nanoparticles as potential carriers for methotrexate delivery

Jun Wang,Zongyong Zhang,Yilong Ai,Fang Liu,Min-Min Chen,Dahai Liu

In order to achieve efficient delivery of methotrexate (MTX), thymine-chitosan nanoparticles (Thy-Cs NPs) were prepared, and further decorated with lactobionic acid (LA) to obtain tumor-targeting nanoparticles (LA-Thy-Cs NPs). These nanoparticles possessed a regular spherical structure with the average size about 190–250 nm and narrow size distribution, which were kinetically stable in the physiological environment. Due to electrostatic interactions and multiple hydrogen-bonding interactions between MTX and carriers, MTX was loaded into Thy-Cs NPs with high drug loading content (~20%). MTX release from Thy-Cs NPs was significantly accelerated in the mildly acidic environment due to the destruction of two types of non-covalent interactions. In vitro cell experiments demonstrated that LA-Thy-Cs NPs could be efficiently internalized into hepatoma carcinoma cells, leading to higher cytotoxicity. Moreover, MTX-loaded LA-Thy-Cs NPs performed an enhanced growth inhibition in three-dimensional multicellular tumor spheroids . Thus, the LA decorated thymine-chitosan nanocarriers can be a promising candidate for efficient delivery of MTX.

Preparation of low molecular chitosan by microwave-induced plasma desorption/ionization technology

Yuting Li,Yuhan Yang,Zhijun Huang,Zewei Luo,Cheng Qian,Yinjun Li,Yixiang Duan

Compared with high molecular weight chitosan (HMWC), low molecular weight chitosan (LMWC) has better solubility and biological activity. However, there is no quick and environmentally friendly to prepare low molecular chitosan. In this study, microwave induced plasma desorption/ionization (MIPDI) was used for the first time to prepare LMWC through the degradation processes of HMWC. The results showed that MIPDI has the most abundant ∙OH content at the gas-liquid interface, and the active particles represented by ∙OH can degrade chitosan with a molecular weight of 540 KDa into soluble chitosan (≤ 10 KDa), and the yield of soluble chitosan can reach 61% in 60 min. Moreover, a series of characterization results showed that the chain structure and crystal structure gradually degraded as the treatment time increased while the chemical structure of chitosan did not change significantly. Antibacterial experiments also indicated that the antimicrobial property of LMWC obtained by MIPDI degradation was improved. In short, this method has proven to be a new, fast and green processing method for the preparation of low molecular chitosan.

Durable antimicrobial cotton fabric fabricated by carboxymethyl chitosan and quaternary ammonium salts

Wang Lujie,Wen Xiaodong,Zhang Xujun,Yuan Shuntao,Xu Qingbo,Fu Feiya,Diao Hongyan,Liu Xiangdong

In this article, carboxymethyl chitosan (CMC) and (3-carboxypropyl) trimethylammonium chloride (CPTC) were used as raw materials to prepare an antibacterial cotton fabric with excellent laundering durability. CMC was first anchored to the surface of cotton fiber via esterification between the carboxyl groups of CMC and the hydroxyl groups of the cellulose molecules on the cotton fiber surface, and then the CPTC was linked to the CMC chains via amidation to the amino groups of the grafted CMC chains. The antibacterial tests showed that the bacteriostatic reduction rate (BR) of the finished cotton fabric against S. aureus and E. coli was above 99.9%, and the BR value of modified cotton fabric remained above 99.9% even after 120 laundering cycles. The results of cytotoxicity, vapor transmissibility, and tensile strength of the fabric samples, as well as the evaluation of water absorption and flexibility show that the modified cotton fabrics are safe and comfortable.

Hypoxia-sensitive micelles based on amphiphilic chitosan derivatives for drug-controlled release

Zhiming Chen,Weiqiu Wen,Jianwei Guo

Natural chitosan (CS) is composed of a larger hydrophilic skeleton with great biocompatibility. A hypoxia sensitive amphiphilic conjugate (CS-NBCF) was synthesized by conjugating CS with nitrobenzyl chloroformate (NBCF). Subsequently, nano micelles with high stability (CMC = 0.0030 mg/ml) were formed by self-assembly of the polymers in deionized water. The hypoxia-responsive function was demonstrated by examining the size variations of polymeric micelles under the hypoxia conditions. Doxorubicin (DOX) was encapsulated into the micellar core, the drug loading and encapsulation efficiency of DOX were 15.0% and 52.6%, respectively. The in vitro release experiments showed that DOX could be released rapidly in hypoxic environment, and the release amount was up to 68% in 24 h. It was found from the cytotoxicity experiment that that the toxicity of the blank micelles was low, while DOX-loaded micelles exhibited a good anti-cancer effect under hypoxic conditions, and the cell inhibition rate of MCF-7 was high up to 63%. As a result, the demonstrated properties endowed as-developed polymeric micelles with promising application as drug delivery system in tumor treatment.

Effect of Curcumin Addition on the Properties of Biodegradable Pectin/Chitosan Films

Qingtong Xie,Xudong Zheng,Liuting Li,Liqun Ma,Qihui Zhao,Shiyuan Chang,Lijun You

A pectin/chitosan matrix-loaded curcumin film (PCCF) with a deep eutectic solvent (DES) as the solvent and plasticizer was prepared in this study. Different quantities of curcumin (identified as PCCF-0, PCCF-1, PCCF-2. PCCF-3) were loaded on the pectin/chitosan film in order to evaluate their effects on the film properties. Results showed that curcumin could interact with the pectin/chitosan matrix and form a complex three-dimensional network structure. PCCF could promote the thickness, tensile strength, thermal properties, antioxidant and antiseptic capacities, but deteriorate the light transmission and elongation at the same time. The addition of curcumin would change the color of the film, without significantly affecting the moisture content. The tensile strength of PCCF-3 reached the maximum value of 3.75 MPa, while the elongation decreased to 10%. Meanwhile, the water-resistance properties of PCCF-3 were significantly promoted by 8.6% compared with that of PCCF-0. Furthermore, PCCF showed remarkable sustained antioxidant activities in a dose-dependent manner. PCCF-3 could inhibit DPPH and ABTS free radicals by 58.66% and 29.07%, respectively. It also showed antiseptic capacity on fresh pork during storage. Therefore, curcumin addition could improve the barrier, mechanical, antioxidant and antiseptic properties of the polysaccharide-based film and PCCF has the potential to be used as a new kind of food packaging material in the food industry. Keywords:chitosan;pectin;curcumin;film;DES

Characterization, antioxidant activity, and biocompatibility of selenium nanoparticle-loaded thermosensitive chitosan hydrogels

Lingying Wu,Yuling Wu,Xun Che,Dandan Luo,Jiaju Lu,Ruibo Zhao,Muhammed Zubair Iqbal,Quan Zhang,Xiumei Wang,Xiangdong Kong

In this study, we recruited chitosan (CS) both for selenium nanoparticles (SeNPs) synthesis and for the development of a thermoresponsive nanocomposite hydrogel with the addition of glycerol phosphate (GP). Considering that SeNPs are toxic at high concentrations, five different ingredients of the nanocomposite hydrogel system with low concentrations of SeNPs (1.25–20 μg/mL) were prepared. The gelation conditions, structural characteristics, and mechanical properties of SeNPs-loaded thermosensitive CS/GP hydrogels were investigated. We also evaluated their antioxidizing activities and biocompatibility of the CS/GP/SeNPs hydrogels. Our study demonstrated that the incorporation of SeNPs in the hydrogel improved its mechanical properties, antioxidant activity, and degree of swelling. According to the properties of SeNPs and CS/GP thermosensitive hydrogels, the combination of these two technologies in an appropriate manner would be a promising antioxidant system for drug delivery and tissue engineering.

Effects of citronellol grafted chitosan oligosaccharide derivatives on regulating anti-inflammatory activity

Shuifang Mao,Bin Wang,Lin Yue,Wenshui Xia

In order to improve the anti-inflammatory activity of chitosan oligosaccharide (COS), chitosan oligosaccharide graft citronellol derivatives (COS-g-Cit1-3) were successfully synthesized via grafting citronellol (Cit) onto COS backbone. The degrees of substitution (DS) of COS-g-Cit1-3 were 0.165, 0.199 and 0.182, respectively. The structure of COS-g-Cit1-3 was confirmed by UV–vis, FT-IR, 1 H NMR and elemental analysis. The in vivo anti-inflammatory activity evaluation results displayed that COS-g-Cit1-3 drastically reduced the paw swelling, and the oedema inhibitions were 22.58 %, 29.03 % and 25.81 %, respectively. The results indicated that the anti-inflammatory effects of COS-g-Cit1-3 were significantly higher than COS and COS-g-Cit2 exhibited the highest anti-inflammatory ability. The results also presented that COS-g-Cit1-3 reduced the expression levels of TNF-α by promoting the secretion of IL-4 and IL-10. Moreover, western blot analysis data proved that COS-g-Cit1-3 inactivated the NF-κB signaling pathway via inhibiting the phosphorylation of p65, IKBα and IKKβ.
