Chronic wound infection often leads to irregular tissue closure and accompanies delayed healing and economy issues. Developing an ideal wound dressing that can control the occurrence of antibacterial infections and biological responses is highly desirable. In this study, a multifunctional hybrid hydrogel (GS@EG–Cu–CA NPs) containing synthesized thiolated gelatin, methacrylated silk fibroin, and (−)-epigallocatechin gallate–copper ionic–carrageenan nanoparticles (EG–Cu–CA NPs) was engineered by a thio–ene click reaction. The metal–polyphenol EG–Cu–CA NPs were encapsulated with kappa-carrageenan to enhance its aqueous-soluble, mechanical, and bioactive properties and endowed the hydrogel dressing with fascinating antibacterial, antioxidation, and nitric oxide (NO) generation by catalyzing. The hybrid hydrogels also illustrated a favorable cytocompatibility. Benefiting from the thio–ene click reaction, the hybrid hydrogels were injected and photocured rapidly in situ to cover an irregular wound. In an SD rat full-thickness skin-wound-infected model, the methicillin-resistant Staphylococcus aureus-infected wound covered with GS@EG–Cu–CA NPs was almost completely healed after 10 days. This study presents a facile design of hydrogel dressing incorporating metal–polyphenol nanoparticles, which demonstrates a promising potential way for dealing with effective wound infection management and other complicated wound healings.

The chemical degradation of proton exchange membranes (PEMs) suffering from radical attack is a crucial issue in the development of proton exchange membrane fuel cells (PEMFCs), while incorporating cerium oxide (CeO 2 ) nanoparticles (NPs) with regenerative redox properties to eliminate radicals could effectively alleviate degradation. However, the low stability and restricted activity of CeO 2 in the PEMFC operating condition and the poor compatibility with PEM due to CeO 2 agglomeration stimulate the urgency for structural modulation of CeO 2 . Achieving a balance of membrane performance and oxidation resistance through a rational surface modification strategy of CeO 2 is important for expanding PEMFC applications. Herein, polyphenols surface functionalized CeO 2 core-shell structure (CeO 2 @TP) were constructed via assembling oxidation-induced coupling tea polyphenols (TP) on CeO 2 NPs. The TP oligomeric shell as a protective layer enhances the stability of CeO 2 , mitigating radical scavenging activity degradation and improving compatibility with PEMs. Physicochemical characterisation shows that the overall performance of the nanocomposite membrane is improved due to the interaction of CeO 2 @TP nanoparticles with the polymer matrix. Gratifyingly, the phenolic hydroxyl-rich reductive TP oligomeric shell accelerates the regeneration of Ce(IV) to Ce(III), increasing the proportion of Ce(III) and oxygen vacancies on the CeO 2 surface, thus boosting antioxidation efficiency. As a result, the CeO 2 @TP-based PEMs exhibited an OCV decay rate of 0.22 mV h −1 , a maximum power density of 1.06 W cm −2 , an H 2 crossover value of 2.18 mA cm −2 , thickness retention (91.1%), and negligible Ce migration after 404 h of accelerated degradation testing. It is proven that the desired consequences of doping antioxidants in PFSA matrix can be intensified by surface modulation of the physicochemical properties of CeO 2 .

Enzyme-assisted aqueous two-phase extraction (EA-ATPE) using ethanol/ammonium sulfate system was investigated for total polyphenol (TP) and lutein from marigold flowers. The important factors were investigated by single factor experiment and response surface methodology combined with Box–Behnken design to optimize the operating parameters of EA-ATPE. The maximum yields of TP and lutein were 83.56 ± 0.69 mg g−1 and 5.59 ± 0.13 mg g−1, respectively. Compared with aqueous two-phase extraction and Soxhlet extraction (SE), the extraction yield of TP by EA-ATPE is 64.91% higher and the extract of EA-ATPE has better antioxidant activity. The pretreatment effect was also researched by scanning electron microscopy. Thus, EA-ATPE is an efficient method for extracting bioactive components from plants.

Pickering emulsions were prepared by using zein/chitosan nanoparticles as stabilizer and then incorporated into chitosan coatings. To improve the stability and performances, tea polyphenol and cinnamaldehyde (CA) were used to modulate the formation and functionalities of Pickering emulsions. The oil phase in Pickering emulsions were set at 5 % and 20 % to alter the hydrophobicity of chitosan coatings. Physical, structural, antioxidant and antibacterial activities of chitosan coatings with Pickering emulsions were characterized. Tea polyphenol significantly enhanced antioxidant capacity of chitosan coatings from 2.09 % to 57.61 % of DPPH value and from 2.63 % to 38.85 % of ABTS value. CA effectively increased the antibacterial activity of chitosan coatings against S. aureus and E. coli . Under 20 % oil content, the inhibition zones on S. aureus and E. coli increased from 3.03 ± 0.23 mm to 18.39 ± 1.22 mm and 7.66 ± 1.61 mm to 15.70 ± 1.75 mm, respectively. The preservative effect of chitosan coatings on fresh pork was further confirmed that the shelf-life of fresh pork could be extended by >4 days. These results suggested a great potential application of Pickering emulsion-incorporated chitosan coatings in the preservation of fresh pork.

The development of nano-encapsulated materials with responsive properties for oral targeted delivery systems is innovative and promising. The aim of this study using corn starch (CS) and β-cyclodextrin (β-CD) to prepare composite nanoparticles (CNPs) that can efficiently encapsulate Tea polyphenol (-)-Epigallocatechin gallate (EGCG) with pH-responsive properties for targeted delivery. The results demonstrated that the CNPs prepared with a 1:1 mass ratio of CS and β-CD exhibited excellent stability to pH, ionic concentration, heating and storage conditions, which was mainly driven by hydrogen bonding and hydrophobic interactions. The characterization results showed that tea polyphenol was effectively loaded into the CNPs and the encapsulation and loading rates were 86.96% and 10.87%, respectively. The CNPs encapsulated tea polyphenol exhibited excellent photothermal stability and maintained significant antioxidant activity after 15 days. Additionally, bio-accessibility experiments revealed that the CNPs can resist low pH environments, can be fully released and reach the target site under alkaline conditions, with potential pH-responsive and intestinal-targeted delivery properties. Overall, the CNPs prepared in this study will support to develop pH-responsive characterization of oral targeted delivery systems for the food industry and biomedicine fields.

Sustainable carboxymethyl cellulose (CMC)-based active composite films were developed through the addition of polyphenol-rich extract from coffee husk (CHE) and carbon dots (CDs) prepared using the biowaste residue of CHE extraction. The influences of various CDs contents on the physicochemical and functional characteristics of composite films have been researched. The 6% (w/w) CHE and 3% (w/w) CDs were uniformly dispersed within the CMC matrix to produce a homogenous film with enhanced mechanical properties. The CMC/CHE/CDs3% film exhibited outstanding UV-light blocking, improved water and gas barriers, potent antioxidant activity with above 95% DPPH and ABTS scavenging rates, and effective antibacterial capabilities against L. monocytogenes and E. coli . The food packaging experiment demonstrated that this active composite film slowed the rotting of fresh-cut apples and extended their shelf-life to 7 days at 4 °C storage. Therefore, the obtained multifunctional film showed promise as an environmentally friendly food packaging material.

Ultra-high molecular weight polyethylene (UHMWPE) is a versatile polymer with various industrial applications, particularly in orthopedic implants and structural materials. However, its susceptibility to oxidation limits its utility in demanding environments. This study investigates the potential of d -sorbitol (DS), a natural polyol, in enhancing the oxidation stability and crosslink density of UHMWPE when stabilized with tea polyphenols. Tea polyphenols, such as epigallocatechin gallate (EGCG), have been previously evaluated as a promising candidate to stabilize UHMWPE against oxidation without affecting its crosslink density. However, the higher content of EGCG has adverse effect on its mechanical properties. It is hypothesized that addition of DS to EGCG can improve the oxidation and crosslink density of UHMWPE blends. For this purpose, EGCG/DS was blended with UHMWPE at low antioxidant concentrations (≤ 1 wt%), and its comprehensive performance was evaluated. Results demonstrate that the incorporation of d -sorbitol effectively retards oxidation-induced degradation in tea polyphenol-stabilized UHMWPE. The presence of d -sorbitol enhances the crosslink density within the polymer matrix, thereby improving its resistance to oxidative stress. Furthermore, the study elucidates the underlying mechanisms through which DS interacts with EGCG and UHMWPE, elucidating the formation of stable complexes that mitigate oxidative damage. The in vitro biocompatibility of the blends was also very promising for use in artificial joints.

Oral ulcers (OU) represent a highly prevalent mucosal disease; however, treatment using existing dressings is often challenging due to insufficient wet adhesion and limited efficacy. In this study, a polyphenol-functionalized silk fibroin (SF) adhesive incorporating a drug delivery platform was developed for the treatment of OU. Silk fibroin (SF) was functionalized with epigallocatechin gallate (EGCG) to form dry granules (termed pwd#ES). Subsequently, drug delivery platforms loaded with dexamethasone (denoted CMC/CS@Ca@Dex) were doped into these granules, resulting in a composite dry granular SF dressing [pwd#ES-(CMC/CS@Ca@Dex)] designed for ulcerated mucosal tissue. The CMC/CS@Ca@Dex platform was prepared by self-assembling chitosan (CS) and carboxymethyl cellulose (CMC) layer-by-layer onto CaCO₃ cores, followed by dexamethasone loading. The resultant composite SF dressing exhibited strong wet tissue adhesion (32.8 kPa), mediated by synergistic interactions within the polyphenol-functionalized SF matrix, alongside excellent reactive oxygen species (ROS) scavenging and bactericidal activity attributed to EGCG. In a mouse model of OU, the pwd#ES-(CMC/CS@Ca@Dex) dressing significantly accelerated healing and promoted faster re-epithelialization. This pwd#ES-(CMC/CS@Ca@Dex) dressing is anticipated to improve the efficiency of OU treatment and inspire the rational design of wet tissue adhesives for the repair of moist tissue defects.

Tea polyphenols have been reported as potential α-amylase inhibitors. However, the quantitative structure–activity relationship (QSAR) between tea polyphenols and human pancreas α-amylase (HPA) is not well understood. Herein, the inhibitory effect of twelve tea polyphenol monomers on HPA was investigated in terms of inhibitory activity, as well as QSAR analysis and interaction mechanism. The results revealed that the HPA inhibitory activity of theaflavins (TFs), especially theaflavin-3′-gallate (TF-3′-G, IC 50 : 0.313 mg/mL), was much stronger than that of catechins (IC 50 : 18.387–458.932 mg/mL). The QSAR analysis demonstrated that the determinant for the inhibitory activity of HPA was not the number of hydroxyl and galloyl groups in tea polyphenol monomers, while the substitution sites of these groups potentially might play a more important role in modulating the inhibitory activity. The inhibition kinetics and molecular docking revealed that TF-3′-G as a mixed-type inhibitor had the lowest inhibition constant and bound to the active sites of HPA with the lowest binding energy (−7.74 kcal/mol). These findings could provide valuable insights into the structures-activity relationships between tea polyphenols and the HPA inhibitors.

Diabetes remains a critical global public health challenge, posing a growing threat to human health and well-being. White tea is a lightly fermented tea and one of the six traditional tea categories in China. Owing to its rich content of bioactive compounds such as catechins and alkaloids, it has demonstrated potential anti-diabetic properties. However, its precise bioactive components, mechanisms of action, and relevant molecular targets require further investigation. In this study, an integrated approach combining polyphenol-targeted metabolomics, in vitro antioxidant assays,α-glucosidase inhibition tests, network pharmacology analysis, GEO database exploration, molecular docking, and molecular dynamics simulations was employed to identify the potential anti-diabetic compounds, targets, and mechanisms of white tea. The findings revealed that white tea is particularly abundant in 10 bioactive compounds, including epigallocatechin gallate, epicatechin gallate, and catechin, all of which exhibit significant anti-diabetic potential. These compounds were found to exert their effects by interacting with core molecular targets, namely cathepsin V (CTSV) and nucleotide-binding oligomerization domain-containing protein 1 (NOD1), and engaging in pathways related to signal transduction, apoptosis, and immune responses. This study establishes a strong theoretical basis for advancing white tea research and underscores new opportunities for applying natural products in diabetes therapy.

Cisplatin, a frontline chemotherapeutic agent against cancer, faces challenges in clinical application due to significant toxicities and suboptimal efficacy. Renal toxicity, a dose-limiting factor of cisplatin, results from multifactorial processes including cisplatin-induced cellular pyroptosis, oxidative damage, and inflammatory responses. Our findings reveal that Tea Polyphenols Nanoparticles (TPNs) derived from Epigallocatechin gallate (EGCG) effectively could address these diverse mechanisms, comprehensively alleviating cisplatin-induced nephrotoxicity. Leveraging TPNs as carriers, chemical conjugation enables the encapsulation of tetravalent cisplatin prodrug, extending its systemic half-life, enhancing tumor tissue accumulation, while simultaneously mitigating renal toxicity. Concurrently, employing a CRISPR/Cas9 kinase library, we identified CSNK2A1 as a target sensitizing tumor cells to cisplatin, enabling specific siRNA sequences to augment cisplatin susceptibility, thereby minimizing the dosage requirement. Benefiting from the versatile carrier properties of TPNs to codeliver cisplatin prodrug and anti-CSNK2A1 siRNA, we developed a codelivery system, Pt-TPNs/siRNA. Pt-TPNs/siRNA not only enhances the anticancer effects but also mitigates cisplatin-induced renal toxicity, achieving efficacy while reducing toxicity. Mechanistic and safety assessments of these nanoparticles were conducted at both cellular and animal levels, opening new avenues for improved clinical utilization of cisplatin.
