Effects of plastics as hydrogen donor on catalytic pyrolysis of cellulose to produce monocyclic aromatics

Hengshuo Gu,Ting Zhao,Jian Li,Yufei Gu,Hucheng Ge,Yuke Liu,Fuwei Li,Zhixia Li,Hongfei Lin,Jiangfei Cao

Hierarchical ZSM-5 (Z-40) was synthesized via a simple two-step hydrothermal crystallization without using any organic template. Low-density polyethylene (LDPE) as hydrogen donor was added into the catalytic pyrolysis of cellulose to enhance deoxygenation and the formation of BTX (benzene, toluene, xylene). Cellulose and LDPE showed the strongest synergistic effect at a feed ratio of 1, and the obtained experimental BTX yield (22.9 %) was 52.7 % higher than the theoretical yield (15 %). Co-catalytic pyrolysis (CCP) of two polymers followed similar cracking behavior to CCP of their pyrolysis vapor model compounds (furfural and n -octane), mainly including pyrolysis, catalytic cracking of pyrolysis fragments in external surface and hydrocarbons pool reactions of small cracking fragments in the internal micropores of zeolite. Z-40 showed better catalytic performance than the commercial ZSM-5 (higher BTX yield and less coke deposition) due to the appropriate acidic properties and distinctive pore structure. The optimal condition for CCP of cellulose/LDPE was temperature at 550 °C, feed ratio at 1:1 and catalyst/feed ratio at 2:1, and the achieved BTX yield reached 24.5 % with high BTX selectivity of 86.7 %. This work provided a cost-effective method for synthesis of ZSM-5 and production of high value-added BTX via a green non-oil route.

Efficient Etherification of 2,5-Bis(hydroxymethyl)furan to 2,5-Bis(propoxymethyl)furan by an Amorphous Silica-Alumina Catalyst in a Fixed-Bed Reactor

Liangqi Chen,Tingting Xue,Hualei Hu,Chunlin Chen,Dazhi Wang,Weijie Cai,Yong Yang,Jian Zhang

Graphical Efficient etherification of 2,5-bis(hydroxymethyl)furan (BHMF) to 2,5-bis(propoxymethyl)furan (BPMF) was achieved by using low-cost amorphous silica-aluminas (ASA) catalysts in a fixed-bed reactor. Benefiting from the appropriate acidity and better mass transfer, ASA-30 catalyst exhibited superior performance with a maximum BPMF yield of 85.1 %. This work provides a useful reference to develop the efficient ASA catalysts for the large-scale production of BPMF. The efficient etherification of 2,5-bis(hydroxymethyl)furan (BHMF) to 2,5-bis(propoxymethyl)furan (BPMF) was achieved by using low-cost amorphous silica-aluminas (ASA) catalysts in a fixed-bed reactor. A considerable yield of BPMF up to 85.1 % was obtained over ASA-30 catalyst under the reaction conditions of 140 °C, 2.0 MPa of N 2 , and 0.015 h −1 of WHSV. The excellent performance of ASA-30 catalyst could be attributed to the relatively stronger acidity (>375 °C) and larger mesoporous size (6 nm), thereby facilitating the conversion of BHMF to BPMF. In addition, the lower ratio of Brønsted/Lewis acid sites for ASA catalyst was found to efficiently suppress the occurrence of side reactions.

One-Pot Method for the Synthesis of 2,5-Furandicarboxylic Acid from Fructose: In Situ Oxidation of 5-Hydroxymethylfurfural and 5-Acetoxymethylfurfural over Co/Mn/Br Catalysts in Acetic Acid

Heng Ban,Youwei Cheng,Lijun Wang,Xi Li

2,5-Furandicarboxylic acid (FDCA) derived from biomass is a versatile platform chemical for producing various degradable polymers, especially polyethylene 2,5-furandicarboxylate (PEF). To reduce the high cost of feedstock, fructose instead of expensive 5-hydroxymethylfurfural (HMF) was used as the feedstock for producing FDCA in the acetic acid (HOAc) solvent by a one-pot, two-step method. Fructose was first catalytically converted into HMF and 5-acetoxymethylfurfural (AMF) in HOAc over the Amberlyst-15 catalyst. After the separation of only Amberlyst-15, the in situ-formed HMF and AMF were further catalytically oxidized to FDCA by air (oxygen) over homogeneous Co/Mn/Br catalysts following the free-radical chain reaction mechanism. HOAc was not only used as the solvent for the dehydration and following oxidation process but also served as the effective co-catalyst for the dehydration of fructose. The effects of catalyst species, dosage, and reaction temperature on the dehydration process and oxidation temperature on FDCA yield and purity were evaluated systematically. Under the optimized reaction conditions, a total yield of HMF and AMF of 50% and a final FDCA yield of 35% were achieved. Combined with experimental results and theoretical calculation, the reaction mechanism for fructose dehydration side reactions and the HMF and AMF oxidation reactions was proposed correspondingly. The proposed two-step, one-pot method using fructose as the starting material is a promising route for the commercial production of FDCA with the simple operation and viable yield.

Catalytic CO2 Cycloaddition and Methanation by ZSM-5 Synthesized from Coal Gangue

Yuxia Zhu,Yuze Zu,Wei Xu,Yinghui Sun,Ying Li,Jie Bai

Coal gangue, the primary solid waste generated during coal mining, is mainly composed of silicon and aluminum oxide. Achieving its high-value utilization is crucial for resource recycling and sustainable development. In this study, we developed a method to synthesize ZSM-5 zeolite (ZSM-5-CG) using coal gangue as the sole silica–alumina source. The synthesis process, crystal growth mechanism, and catalytic performance of the resulting zeolites were systematically investigated. Compared with ZSM-5 prepared from commercial raw materials (ZSM-5-C), the coal gangue-derived synthesis required a significantly lower amount of template due to the favorable interaction between gangue-derived species and the template. Although crystallization in both systems follows the “solid–liquid phase transformation” mechanism, coal gangue, as a natural mineral source, exhibits significant differences in Si and Al coordination environment compared to commercial inorganic raw materials. These differences alter the dissolution and migration behaviors of Si and Al species in the mother liquor, leading to a synchronous release of these components during crystallization. As a result, nucleation and crystal growth processes happen more slowly, yielding a lower overall growth rate than that observed in systems derived from commercial raw materials. The obtained ZSM-5-CG was further applied in CO2 cycloaddition and methanation reactions, exhibiting catalytic activity comparable to that of ZSM-5-C, owing to its abundance of acid sites and large surface area. A cost analysis revealed that the total production cost of ZSM-5-CG was 29.7% lower than that of ZSM-5-C. This work provides a novel and economically viable route for the high-value utilization of coal gangue and highlights the potential of ZSM-5-CG in CO2 catalytic conversion.

METTL3 antagonizes 5‑FU chemotherapy and confers drug resistance in colorectal carcinoma

Min Li,Mingyue Xia,Ziyu Zhang,Yanyin Tan,Enjie Li,Zhigang Guo,Mingzhi Fang,Yong Zhu,Zhigang Hu

Colorectal cancer (CRC) is one of top five leading causes of cancer‑associated mortalities worldwide. 5‑Fluorouracil (5‑FU) is the first‑line chemotherapeutic drug in the treatment of CRC; however, its antineoplastic efficiency is limited due to acquired drug resistance. The regulatory mechanism underlying 5‑FU chemotherapeutic response and drug resistance in CRC remains largely unknown. The present study identified that silencing of methyltransferase‑like 3 (METTL3) suppressed the proliferation and migration of CRC HCT‑8 cells. Using cell survival assays, flow cytometric and colony formation analyses, it was revealed that inhibition of METTL3 sensitized HCT‑8 cells to 5‑FU by enhancing DNA damage and inducing apoptosis in HCT‑8 cells under 5‑FU treatment. Furthermore, the expression of METTL3 was upregulated in 5‑FU‑resistant CRC cells (HCT‑8R), which contributed to drug resistance through regulation of RAD51 associated Protein 1 (RAD51AP1) expression. Western blotting, immunofluorescence staining and drug sensitivity assays demonstrated that knockdown of METTL3 augmented 5‑FU‑induced DNA damage and overcame 5‑FU‑resistance in HCT‑8R cells, which could be mimicked by inhibition of RAD51AP1. The present study revealed that the METTL3/RAD51AP1 axis plays an important role in the acquisition of 5‑FU resistance in CRC, and targeting METTL3/RAD51AP1 may be a promising adjuvant therapeutic strategy for patients with CRC, particularly for those with 5‑FU‑resistant CRC.

OsNSUN2-Mediated 5-Methylcytosine mRNA Modification Enhances Rice Adaptation to High Temperature

Yongyan Tang,Chun-Chun Gao,Ying Gao,Ying Yang,Boyang Shi,Jia-Li Yu,Cong Lyu,Bao-Fa Sun,Hai-Lin Wang,Yunyuan Xu,Yun-Gui Yang,Kang Chong

Summary Extreme weather events can cause heat stress that decreases crop production. Recent studies have demonstrated that protein degradation and rRNA homeostasis as well as transcription factors are involved in the thermoresponse in plants. However, how RNA modifications contribute to temperature stress response in plant remains largely unknown. Herein, we identified OsNSUN2 as an RNA 5-methylcytosine (m 5 C) methyltransferase in rice. osnsun2 mutant displayed severe temperature- and light-dependent lesion-mimic phenotypes and heat-stress hypersensitivity. Heat stress enhanced the OsNSUN2-dependent m 5 C modification of mRNAs involved in photosynthesis and detoxification systems, such as β-OsLCY , OsHO2 , OsPAL1 , and OsGLYI4 , which increased protein synthesis. Furthermore, the photosystem of osnsun2 mutant was vulnerable to high ambient temperature and failed to undergo repair under tolerable heat stress. Thus, OsNSUN2 mutation reduced photosynthesis efficiency and accumulated excessive reactive oxygen species upon heat treatment. Our findings demonstrate an important mechanism of mRNA m 5 C-dependent heat acclimation in rice.

A multifunctional 5-fluorouracil prodrug: synthesis, biological evaluation, and antitumor mechanism investigation

Zhang Ran,Tang Liqing,He Jianglong,Jiang Guanyu,Su Yingfei,Jiang Xueping,Wu Ping,Zhang Dongyang,Chen Sitong,Li Hao,Gui Zhongzheng

As FDA-approved chemotherapeutic agents, paclitaxel (PTX) and 5-fluorouracil (5-FU) are widely used in the clinic but suffer from dose-limiting toxicities. To overcome these limitations, we designed and synthesized two novel 5-FU prodrugs conjugated with the microtubule inhibitor PTX. The hybrids exhibited potent cytotoxicity against cancer cell lines while demonstrating favorable selectivity over normal human cells, outperforming the individual agents or their combinations. As a representative, FP2 potently induced DNA damage through downregulation of thymidylate synthase (TS), arrested the cell cycle at S-phase, and suppressed metastasis of HCT-116 cells. Moreover, FP2 triggered mitochondrial dysfunction, characterized by loss of mitochondrial membrane potential, increased ROS generation, downregulation of Bcl-2, and upregulation of Bax. FP2 also triggered oxidative stress, elevating MDA and NO levels, depleting GSH and SOD, downregulating Nrf2, and upregulating HO-1. Additionally, FP2 activated the PERK/ATF4/CHOP signaling pathway, leading to endoplasmic reticulum (ER) stress. Furthermore, FP2 induced autophagic cell death, as evidenced by the formation of autophagosomes, an increased LC3 and decreased p62 expression. Collectively, these findings established FP2 as a novel compound with a promising in vitro anticancer profile that promotes HCT-116 cell death through an integrated mechanism involving DNA damage, mitochondrial dysfunction, ER stress, oxidative stress, and autophagy.

Dual-site Engineering of Ga-Modified HZSM-5 towards Efficient Propane Aromatization

Zhaoshuo Ge,Lixia Bao,Yangping Lv,Yiheng Wu,Daxin Shi,Qi Liu,Qin Wu,Kangcheng Chen,Guiyuan Jiang,Hansheng Li,Yaoyuan Zhang

Light alkane aromatization represents a pivotal catalytic process for valorizing abundant light alkanes into high-value aromatics, holding significant industrial promise. Herein, a series of Ga-HZSM-5 catalysts were synthesized by the hydrothermal method. The effects of Ga content and Si/Al ratio on the catalytic performance of propane aromatization over Ga-HZSM-5 were investigated to discern the structure-activity relationship, reaction pathway, and coke formation. The optimized 1%Ga-HZSM-5(50) catalyst exhibited superior performance, with propane conversion and BTX formation showing a volcano-shaped trend with increasing Ga content. Conversely, for a fixed Ga loading, aromatization performance was enhanced with decreasing Si/Al ratio, with 1%Ga-HZSM-5(25) delivering the highest aromatization activity. A positive correlation was identified between catalytic activity and Lewis acid density, underscoring the role of Lewis sites in facilitating propane dehydrogenation and subsequent BTX formation. Furthermore, TG and TPO analyses revealed that the deposited coke in Ga-HZSM-5 was predominantly highly graphitized, which could not be removed by the air-regeneration treatment under reaction temperature.

Preparation and in vitro evaluation of photodynamic-responsive nanoliposome loaded PL-5

Wen Lin,Qiong-zhi Shi,Xiang-ru Liao,Yuan Zeng,Xiang-yang Xie,Gang-jian Ji,Yin-ke Li

Burn wound infections are frequently complicated by biofilm-forming and multidrug-resistant pathogens, particularly methicillin-resistant Staphylococcus aureus (MRSA), posing major therapeutic challenges. Antimicrobial peptides (AMPs) such as PL-5 (peceleganan) exhibit broad-spectrum activity but are limited by instability, poor biofilm penetration, and reduced efficacy in complex wound environments. Here, a red-light-responsive, porphyrin-phospholipid (PoP)-containing cationic liposomal system for PL-5, aiming to enhance its antibacterial and antibiofilm performance was developed. Optimized liposomes achieved high encapsulation efficiency (~73%), uniform nanoscale size (~50 nm), narrow polydispersity, and positive surface charge. They demonstrated good storage stability and controlled peptide release under red-light irradiation (635 nm). In vitro , red-light activation significantly enhanced antimicrobial activity against MRSA and methicillin-susceptible S. aureus (MSSA), reducing minimum inhibitory concentration (MIC) values fourfold and accelerating bactericidal kinetics compared with free PL-5 and non-irradiated liposomes. Additionally, red-light-activated liposomes markedly inhibited biofilm formation. These results indicate that light-responsive liposomal delivery enables spatiotemporally controlled release of PL-5, significantly potentiating its antibacterial and antibiofilm efficacy. This approach offers a promising localized treatment strategy for biofilm-associated burn wound infections and a foundation for future translational studies.

5-HMF inhibits glucocorticoid-induced osteoporosis through the VEGFR2/PI3K/AKT pathway

Liu Siqi,Fang Fei,Jiang Yu

Chronic administration of glucocorticoids are being linked to an increased risk of osteoporosis. 5-Hydroxymethylfurfural (5-HMF) is a natural compound that possesses various biological activities, including osteogenic regulation. In this research, we sought to investigate the potential of 5-HMF to exert protective effects against glucocorticoid-induced osteoporosis. In cellular investigations, we deployed Western Blot (WB), Real-Time Quantitative real-time PCR (qRT-PCR), Alkaline phosphatase (ALP) and Alizarin S-red (ARS) staining to scrutinize the expression of osteogenic differentiation markers within MC3T3-E1 pre-osteoblasts and BMSCs. We used dexamethasone to establish a mouse model for glucocorticoid-Induced osteoporosis and administered 5-HMF to evaluate its influence on bone density and architecture through the use of micro-CT and histological analysis. In addition, we employed network pharmacology to elucidate the potential pathways and targets of 5-HMF. Finally, we explored the effect of AKT knockout on the treatment of 5-HMF. We found that 5-HMF significantly enhanced VEGFR2 phosphorylation, reestablishing angiogenesis and activating the PI3K/AKT pathway in vitro and in vivo. 5-HMF also inhibited Dexamethasone -induced apoptosis by regulating Bax and Bcl-2 expression. MK2206, an AKT inhibitor, abrogated 5-HMF’s protective properties against GCs. These results indicate that 5-HMF counteracts the negative effects of GCs on osteoblasts and has good bone-promoting differentiation effects.

Pd/DUT-5 for mild and selective hydrogenation of nitrile butadiene rubber

Shukang Ti,Dongmei Yue

Pd/DUT-5 is an efficient catalyst for NBR hydrogenation to HNBR. • High hydrogenation activity was achieved under mild conditions. • Metal-support interactions stabilize electron-enriched Pd δ+ species. • Pd δ+ species enhance catalytic activity and chemoselectivity. • DFT reveals low-barrier heterolytic H 2 activation at Pd δ+ –μ 2 O - sites. Hydrogenated nitrile butadiene rubber (HNBR), produced via the selective hydrogenation of nitrile butadiene rubber (NBR), has attracted considerable interest because of its excellent overall performance. However, the development of efficient and selective catalytic systems for NBR hydrogenation under mild conditions remains challenging, particularly when both activity and cost-effectiveness are required. Herein, Pd nanoparticles were immobilized on the Al-based metal-organic framework DUT-5, and the influences of Pd precursor salts and pretreatment conditions on catalytic hydrogenation performance were systematically investigated. The results show that using Pd(OAc) 2 as the precursor in combination with relatively low reduction temperatures promotes the formation and stabilization of electron-enriched Pd δ+ species through metal-support interactions, leading to excellent low-temperature activity and good reusability, with the hydrogenation degree remaining 94.5% after three cycles. Density functional theory calculations using C 2 H 4 as a model substrate further reveal that Pd δ+ sites cooperatively interact with μ 2 O - moieties to facilitate H 2 activation and subsequent hydrogen transfer through a heterolytic cleavage pathway with reduced energy barriers. These results provide a mechanistic basis for the efficient hydrogenation of NBR under mild conditions. Overall, this work demonstrates an effective and economical strategy for HNBR production and offers new insights into the design of highly active and chemoselective catalysts for NBR hydrogenation. The functions of a 5′ tRNA-Ala-derived fragment in gene expression

Li Yuanyuan,Gao Junping,Wang Ying,Cai Jun,Wu Dousheng,Wang Long,Pu Wenxuan,Yu Feng,Zhu Sirui

Transfer RNA (tRNA) can produce smaller RNA fragments called tRNA-derived fragments (tRFs). tRFs play critical roles in multiple cellular programs, although the functional mechanisms of tRFs remain largely unknown in plants. In this study, we examined the phenotype associated with 5′ tRF-Ala (tRF-Ala, produced from tRNA-Ala) overexpression and knockdown lines (tDR-Ala-OE and tDR-Ala-kd, respectively) and the mechanisms by which tRF-Ala affects mRNA levels in Arabidopsis (Arabidopsis thaliana). We investigated the candidate proteins associated with tRF-Ala by quantitative proteomics and confirmed the direct interaction between tRF-Ala and the splicing factor SERINE-ARGININE RICH PROTEIN 34 (SR34). A transcriptome sequencing analysis showed that 318 genes among all the genes (786) with substantial alternative splicing (AS) variance in tDR-Ala-OE lines are targets of SR34. tRF-Ala diminished the binding affinity between SR34 and its targets by direct competition for interaction with SR34. These findings reveal the critical roles of tRF-Ala in regulating mRNA levels and splicing.
