The solid-state architecture of iontronics osmotic power source (IOPS) significantly enhances volumetric power density and eliminates risks associated with leakage and drying up.
By making deep use of the ion-electron coupling properties of MXene, the highly integrated electrode design enables the IOPS device to be assembled into a compact osmotic power source using only three building blocks.
IOPS arrays can efficiently power commercial electronic devices and show great potential in portable, responsive power systems.

Metal–organic framework (MOF) membranes offer great potential to address the energy penalties of energy-intensive gas separations 1 , 2 , 3 , 4 , 5 . Deriving convenient scalable protocols is essential for the successful translation and deployment of MOF-based membranes into practical applications 6 , 7 , 8 . Here we report a new membrane architecture, the quasi-pure MOF membrane (>90 vol% MOF), manufacturable at scale using industry-reliable solution-processing techniques and offering separation performance approaching that of associated pure MOF membranes, fully realizing the MOF intrinsic potential. This advance is enabled by a merged-phase approach fusing MOFs and polymers into a single pseudo-continuous phase and proffering MOFs with polymer-like surface properties, allowing rheologically controlled flocculated networks even at extreme solid concentrations. Representative benchmark MOFs (ZIF-67, CALF-20 and CuBDC) were fabricated into quasi-pure membranes and evaluated for essential separations, including propylene/propane, ethylene/ethane, carbon capture and hydrogen purification. Demonstrating industrial relevance, continuous roll-to-roll fabrication of quasi-pure (110)-oriented ZIF-67 membranes was achieved at an industrial plant. For propylene/propane separation, these membranes achieve a propylene permeability of about 160 barrer and mixed-gas selectivity of about 100—sufficient to produce polymer-grade propylene, offering an 80% purification cost reduction compared with distillation based on techno-economic analysis. This study addresses the long-standing gap between performance and scalability of crystalline membranes for demanding molecular separations.

Realizing high-performance perovskite/silicon tandem solar cells requires precise control of wide-bandgap perovskite crystallization. Solvent engineering is the most direct lever for this task; yet, its intricate, multi-variable mechanisms defy intuition-driven design. Herein, we overcome this bottleneck by pioneering a retrieval-augmented large language model to screen > 8000 solvents, identifying γ -valerolactone (GVL) as a non-toxic, high-performance cosolvent. It is found that the GVL strongly coordinates FA + , thus precisely modulating crystallization kinetics, retarding nucleation, and promoting oriented, micrometer-scale grain growth. The resulting films exhibit not only superior crystallinity, reduced non-radiative recombination, but also improved scalability to large area and the tolerance to increased film thickness. Consequently, both the single-junction and tandem devices achieve efficiencies of 23.3% and 32.5%, respectively, along with excellent stability under moisture and illumination. This study establishes the first artificial intelligence (AI)-guided cosolvent strategy for 1-μm-thick perovskite layers in perovskite/silicon tandem architectures, underscoring the transformative role of generative AI in advancing high-performance photovoltaics.

Regional epithelial lineages of the human respiratory system reside within an extracellular matrix (ECM) whose mechanics vary along the airway–alveolar axis, yet how ECM stiffness directs epithelial fates remains unclear. Here, utilizing human pluripotent stem cell-derived lung organoids embedded in stiffness-tunable hydrogels as an in vitro model, we show ECM stiffness governs region-specific epithelial differentiation. Stepwise softening of ECM stiffness yields airway organoids with proximal-to-distal airway epithelial compositions and biomimetic physiological functions. During alveolar differentiation, increased stiffness promotes alveolar type 2 (AT2) and type 1 (AT1) maturation and drives AT2-to-AT1 transition. Furthermore, RNA sequencing reveals ECM stiffness regulates epithelial fates primarily through mechanotransduction pathways. Finally, these organoids reproduce the infection tropisms of SARS-CoV-2 variants. Together, this research elucidates ECM stiffness as a critical determinant of epithelial cell fate specification and region-specific lung organoid generation, which offers a valuable in vitro model for studying region-specific lung development, diseases pathogenesis, and drug screening.

Asthma is a chronic inflammatory respiratory disease worldwide. Its severe phenotypes, particularly the glucocorticoid-resistant ones, pose a major challenge, with neutrophilic inflammation being the key driver. However, no effective treatments are currently available. Herein, we report a macromolecular therapeutic strategy capable of targeting multiple key cells involved in the pathogenesis of neutrophilic asthma. The macromolecular therapy named PMART is constructed by conjugating artesunate and polyethylene glycol to the triazine scaffold. It is highly water-soluble and can self-assemble into nanomicelles, which enables efficient targeting to inflamed lung tissues and localization in inflammatory cells. In a murine model of neutrophilic asthma, PMART has proven to have in vivo efficacy equivalent to or greater than that of two clinically used anti-asthma drugs. Mechanistically, PMART reduces succinate levels by inhibiting branched-chain amino acid transaminase 1 (BCAT1) overexpression, thereby suppressing neutrophilic inflammation via blocking the NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome and the production of pro-inflammatory cytokines. Notably, PMART exhibits a favorable safety profile in mice. Taken together, these data support the potential of PMART as a novel and highly effective therapeutic agent for neutrophilic asthma and other neutrophil-mediated diseases. Graphical

Despite the systemic spread of cancer, the ventricular myocardium is one of the least common sites of metastasis—a phenomenon that remains poorly understood. To examine this, we develop the Multi-organ Invasion Device (MInD), an organ-on-a-chip platform that enables multi-organ culture under flow. Organ compartments are connected in MInD using PermeoTubes—3D-printed porous conduits that support cancer cell intravasation, migration, and extravasation. In dual-organ devices, where highly aggressive breast cancer cells are co-cultured with either hepatic or cardiac tissue, invasion into cardiac tissue is significantly suppressed relative to hepatic co-culture. Importantly, in cardiac–hepatic–cancer tri-culture, the presence of cardiac tissue reduces overall invasion, with cancer cells preferentially migrating toward hepatic compartments. Cytokine profiling and RNA sequencing reveal that cardiac co-culture suppresses cell metastasis and invasion, while inducing immune activation. Overall, this platform presents an approach for uncovering organ-specific drivers of metastasis, accelerating future discovery of metastasis-inhibiting therapies.

Osteoporosis is a major public health concern characterized by reduced bone mass and increased fracture risk, and Cyathulae Radix has been used in China for its prevention, yet its active constituents and mechanisms remain unclear. This study integrated network pharmacology with in vitro experiments to identify key bioactive components and underlying pathways. Using the TCMSP database, we screened the major compounds of Cyathulae Radix and, through intersection with osteoporosis-related targets from OMIM and GeneCards, obtained 29 overlapping genes. Subsequent protein–protein interaction network analysis via STRING revealed core targets including AKT1, TNF, IL-6, and MMP2 , among which quercetin emerged as a principal active component. To validate its effects, we employed H₂O₂ to induce oxidative damage in MC3T3 E1 pre-osteoblasts, and then treated cells with quercetin based on CCK8 and ROS assay results. The CCK8 assay showed that quercetin restored cell viability to near-normal levels. Quercetin treatment significantly improved cell proliferation and restored osteogenic differentiation, as shown by elevated alkaline phosphatase staining intensity, increased mineralized nodules in alizarin red staining, and markedly upregulated mRNA levels of RUNX2, OPN , and BMP-2 . Furthermore, quercetin attenuated oxidative stress by increasing SOD and GSH activities, decreasing MDA production, and activating the NRF2/HO-1/GPX4 antioxidant cascade, as confirmed by western blotting. Collectively, these results demonstrate that quercetin, a key constituent of Cyathulae Radix, exerts anti osteoporotic effects through both promoting osteogenesis and enhancing antioxidant defense, offering a mechanistic rationale for its clinical use against osteoporosis.

A novel biosensing platform is proposed that integrates catalytic hairpin assembly (CHA) and hybridization chain reaction (HCR) cascade isothermal amplification with the CRISPR/Cas12a system, enabling ultrasensitive detection of microRNAs (miRNAs) targets. Within this platform, two modules are integrated: a target recognition and signal amplification module constructed by the cascade of CHA and HCR, and a signal transduction module in which the CRISPR/Cas12a system acts in concert with DNA probes loaded onto gold nanoparticles (AuNPs). This design achieves cascaded amplification from target recognition to signal output, thereby conferring high signal gain. Experimental results demonstrate that the proposed biosensor had high sensitivity toward the target miRNA, with a detection limit as low as 37 fM. Moreover, it shows remarkable single-base discrimination capability, effectively distinguishing sequences with single-nucleotide mismatches. Notably, the sensor maintains stable and reliable performance in complex biological matrices, including serum samples and lysates from various tumor cells. This strategy effectively couples signal amplification with the CRISPR system, achieving both high sensitivity and specificity, making it a useful tool for miRNA detection and early cancer screening. Background Doxorubicin (DOX) is a widely used broad-spectrum chemotherapy drug, but its severe organ toxic and side effects limit its clinical application. Tetrahydropalmatine (THP) protects against DOX-induced renal injury, yet its underlying mechanism remains unclear. This study aimed to verify THP’s protective effect and explore if it acts via the SIRT3-mediated Nrf2/HO-1 signaling pathway. Methods This study adopted an integrated in vivo and in vitro research system to explore the relevant mechanisms. In vivo, a DOX-induced mouse renal injury model was established with THP intervention; renal function was evaluated by detecting serum creatinine (Scr) and blood urea nitrogen (BUN), while renal injury severity was assessed via urine microalbumin (mALB) and urine albumin/creatinine ratio (UACR). Renal histopathological changes including glomerular injury and tubulointerstitial fibrosis were observed, and subcellular structures such as podocyte integrity and mitochondrial morphology were analyzed by transmission electron microscopy (TEM); additionally, the expression of pathway-related proteins was detected. In vitro, a DOX-induced cell injury model was constructed using mouse podocytes (MPC-5), and Western blot and immunofluorescence techniques were employed to determine the expression of key molecules in the Sirt3-Nrf2/HO-1 pathway, combined with in vivo and in vitro experiments to clarify the underlying mechanism. Results THP significantly attenuated DOX-induced renal injury, improved renal function (reduced Scr/BUN), mitigated histopathological/mitochondrial abnormalities, and preserved podocyte integrity. Molecularly, THP upregulates podocyte markers Nephrin/Podocin, enhances antioxidant capacity (increased GSH, SOD, and CAT, decreased MDA), inhibits inflammation (downregulated IL-6, IL-1β, and TNF-α), and suppresses apoptosis (downregulated Bax/caspase-3, upregulated Bcl-2). Notably, both in vivo and in vitro data link THP’s protection to SIRT3-mediated Nrf2/HO-1 pathway activation. Conclusion THP protects against DOX-induced renal injury in mice. Mechanistically, this may involve activating the SIRT3-mediated Nrf2/HO-1 pathway, improving podocyte function, and inhibiting oxidative stress, inflammation, and apoptosis.

Endoplasmic reticulum autophagy (ER-phagy) is a selective autophagy pathway in which receptor proteins target ER membranes and proteins for degradation, yet its role in Alzheimer’s disease (AD) remains unclear. Here, we identify FAM134B/RETREG1 as a specific ER-phagy receptor mediating amyloid precursor protein (APP) degradation. FAM134B directly interacts with ER-localized wild-type and familial mutant APP via their C-terminal domains and recruits LC3 through its LC3-interacting region (LIR) to promote APP delivery to phagophores for lysosomal degradation. In AD, epigenetic silencing at the FAM134B promoter suppresses its transcription by limiting TFEB/TFE3 binding despite their nuclear enrichment. This transcriptional suppression impairs ER-phagy, leading to APP accumulation and exacerbated AD pathology. AAV-mediated hippocampal expression of wild-type, but not LIR-mutant, FAM134B in 5XFAD mice restores ER-phagy, enhances APP clearance, reduces Aβ deposition, preserves synaptic and myelin integrity, and improves cognitive performance. These findings establish FAM134B downregulation as an upstream pathogenic event in AD, suggesting ER-phagy enhancement as a promising strategy to suppress Aβ generation at its source.

Silver is an extensively investigated electrode material for electrochemical CO 2 reduction owing to its high electrical conductivity and structural stability. However, a key limitation of silver catalysts is their weak adsorption of the *CO intermediate. This intrinsic constraint restricts the reaction primarily to two-electron pathways, hindering the formation of multi-electron products. Here we demonstrate that surface molecular modification can address this limitation. By anchoring bromothymol blue molecules onto the silver surface, we engineer a localized hydrogen-bonding network that stabilizes *CO intermediates via O···H–O interactions, prolonging their surface residence time. In situ spectroscopy and theoretical simulations reveal that this local microenvironment thermodynamically stabilizes *CO against desorption while kinetically facilitating its subsequent hydrogenation and C-C coupling. Consequently, the retained *CO undergoes deeper reduction pathways, generating CH 4 , C 2 H 4 , C 2 H 5 OH, and CH 3 COOH. At a current density of 400 mA cm −2 , the Faradaic efficiency for multi-electron products reaches 24.2%. This work shifts the design paradigm from metal-centric electronic tuning to local microenvironment engineering, offering an alternative strategy for enabling multi-electron transfer on non-copper catalysts.

Background Laser-assisted hatching (LAH) has been reported to improve implantation and pregnancy rates, yet its efficacy and safety profile require rigorous evaluation. To assess the impact of LAH on pregnancy-related outcomes and safety metrics within Assisted Reproductive Technologies (ART) procedures, we conducted a systematic review and meta-analysis. Methods Electronic databases were searched from inception to November 30, 2023. Studies, comparing LAH with other hatching methods or no intervention within ART frameworks, were included. The quality of the included studies was rigorously evaluated using the Risk of Bias In Non-randomized Studies-of Interventions (ROBINS-I) tool with a Weighted Cohen’s kappa coefficient (κ). Summative data, subgroup analyses, and evaluation of heterogeneity were analyzed via Stata 15. Results 47 eligible studies were included, covering 24,876 couples. When comparing the effectiveness of LAH to other methods or no intervention, the results revealed enhancements in implantation and clinical pregnancy rates. For implantation rates, the pooled effect size showed an OR of 1.22 (95% CI 1.09–1.36), indicating a modest improvement with LAH. As for clinical pregnancy rates, LAH was associated with an OR of 1.12 (95% CI 1.03–1.22), demonstrating its positive impact. However, term delivery rates showed a modest but statistically detectable improvement, with an OR of 1.07 (95% CI 1.01–1.14), and sensitivity analysis revealed this effect to be study-dependent. The abortion rate remained statistically unchanged with LAH, OR 0.95 (95% CI 0.85–1.06; P = 0.34; I 2 : 0%). Subgroup analyses, particularly focusing on age and embryo status (fresh vs. frozen), elucidated some of the observed heterogeneity, offering a deeper understanding of LAH's varying impacts across different patient demographics and treatment specifics. Conclusion LAH represents a valuable adjunct in ART, particularly beneficial for specific patient groups. It modestly improves implantation and clinical pregnancy rates without compromising safety. However, its effect on term delivery is small, sensitive to individual studies, and uncertain overall. Graphical
