Preparation of pectin-tannic acid coated core-shell nanoparticle for enhanced bioavailability and antihyperlipidemic activity of curcumin

Qihong Zhang,Hui Wang,Zongmiao Feng,Zhaohui Lu,Chen Su,Yunchun Zhao,Jingbo Yu,Alexandr V. Dushkin,Weike Su

As a functional polyphenolic compound, curcumin (CUR) has limited application due to its low solubility and bioavailability. In this work, curcumin loaded micelles self-assembled from disodium glycyrrhizin (Na 2 GA) was coated with pectin and tannic acid to construct a core-shell solid dispersion (SD-CUR). The assembly mechanism of the core-shell nanoparticle was clarified by molecular dynamics simulation. Improved solubility and in vitro release characteristics of SD-CUR in simulated gastric and intestinal juice were achieved. The parallel artificial membrane permeability (PAMPA) test confirmed that the permeability of curcumin in SD-CUR was significantly improved compared with the pure curcumin. Pharmacokinetic studies showed that the bioavailability was increased by 10 times. Intestinal in situ absorption study proved SD-CUR could enhance drug to transport across intestinal epithelial cells. Further hypolipidemic study demonstrated that the SD-CUR medication group could considerably reduce the levels of total cholesterol (TC), triglyceride (TG), and low density lipoprotein (LDL) in hyperlipidemia rats. These findings confirmed that SD-CUR had broad application prospects in the prevention and treatment of hyperlipidemia.

Stability and bioaccessibility of curcumin emulsions stabilized by casein hydrolysates after maleic anhydride acylation and pullulan glycation

Yi Pan,Xiao-Min Li,Ran Meng,Bao Zhang

ABSTRACT The effects of maleic anhydride (MA) acylation and pullulan glycation on casein hydrolysates (CH) and the physicochemical stability of modified or unmodified CH-stabilized emulsions were explored. Compared with casein, the solubility of CH was improved, and CH 1 (hydrolysis degree 4%) exhibited the optimal emulsifying properties. After the acylation of MA, degrees of acylation (DA) increased with increasing addition of MA. Fourier-transform infrared spectroscopy revealed that a covalent bond was formed between MA and CH 1 . The results of pullulan glycation indicated that the degree of glycation decreased with increasing DA. Acylation combined with glycation effectively reduced the surface hydrophobicity of CH. Results of analysis of physicochemical stability and gastrointestinal fate of curcumin in emulsions revealed that CH modified by MA acylation and pullulan glycation played a positive role in enhancing the stability and bioaccessibility of curcumin loaded in emulsions.

Fabrication and characterization of dextran/zein hybrid electrospun fibers with tailored properties for controlled release of curcumin

Shiyuan Luo,Abdullah Saadi,Kai Fu,Maierhaba Taxipalati,Lingli Deng

BACKGROUND In recent years, there has been considerable interest in the use of biopolymer electrospun nanofibers for various food applications due to the biocompatibility, biodegradability, and high loading capacity. Herein, we fabricated and characterized novel hybrid electrospun fibers from dextran (50%, w/v ) and zein (0–30%, w/v ) solutions, and the effects of various zein concentrations on the properties of the hybrid electrospun fibers were investigated. RESULTS When zein was added at low concentrations (5% and 10%), dextran and zein showed poor miscibility, as reflected by significantly decreased viscosity of the solutions, and the poor mechanical properties of the derived fiber membranes. When zein was added at medium concentrations (15–25%), hydrogen bonds were formed between dextran and zein molecules, as indicated by the red shift of Fourier-transform infrared bands and β-sheet to α-helix structural transformations. The fiber membranes electrospun from a solution with 25% zein showed the most hydrophobic surface, with a water contact angle of 116.9°. The homogenous dispersion of dextran and zein resulted in improved mechanical properties for fibers electrospun from a solution with 30% zein. Curcumin encapsulating dextran/zein electrospun fibers exhibited effective radical scavenging activity and ferric reducing power, along with the desired controlled release behavior for curcumin delivery. CONCLUSION Food grade dextran/zein hybrid electrospun fibers demonstrated tunable properties, and appear to be promising as delivery systems for bioactive and edible antimicrobial food packaging. © 2021 Society of Chemical Industry.

Excogitation and Assessment of Curcumin-Vitamin E Self-assembly PEGylated Nanoparticles by the Route of Oral Administration

Zipeng Huang,Xinru Chen,Ying Li,Junqiu Zhai,Yan Ma

Curcumin (CUR) has attracted wide research interests due to its abundant bioactivities and potential advantages in cancer treatment. But the poor water solubility, instability, and quick metabolization and elimination after oral administration severely restrict the efficacy and further clinical application of CUR. Derivation is an approach often used to improve the druggability of active ingredients, so the study aim to prepare a CUR derivate with better stability, satisfactory pharmacokinetics , and inherent self-assembled ability in contrast with CUR. The derivate was designed and evaluated in vitro and in vivo . Vitamin E (VE) was used to perform the esterification reaction with CUR, and the cytotoxicity of derivative CUR-VE ester on MCF-7 tumor cells was similar to CUR. Besides the better stability in simulated gastric and intestinal fluid, plasma and liver homogenate, the self-assembly CUR-VE nanoparticles were fabricated by feasible and controllable nanoprecipitation method. The Transmission Electron Microscope (TEM) showed CUR-VE NPs were spherical with an average particle size of 172.9  nm , and drug loading was up to 93%. CUR-VE NPs exhibited a sustained-release behavior and fitted to Fick's diffusion mechanism. Differential Scanning Calorimeter (DSC), X-ray Powder Diffractometer (XRPD) and Fourier Transform Infrared Spectrometer (FTIR) declared no crystalline substances were formed, and the self-assembly process of CUR-VE relied on driving forces including van der Waals forces , hydrogen bonding forces, intermolecular forces . In pharmacokinetics, C max and AUC 0-∞ of CUR-VE NPs by the route of oral administration were (104.69 ± 40.72) ng/mL and (3496.92 ± 1088.93) ng/mL∗h, which were about three times and 18 times more compared with CUR. The eliminated half-time of CUR-VE extended to 28 h ascribed to the outstanding stability and surface PEGylation of NPs. It prompted that appropriately PEGylated NPs via oral administration was beneficial to prolong systemic circulation similar to intravenous PEGylated NPs. In summary, the study provides a convenient way to fabricate the self-assembled CUR-VE NPs qualified with high drug loading, satisfactory stability and desired pharmacokinetics. CUR-VE has the potent advantage to be an ideal substitute for CUR in the future of healthcare and clinical application.

Green and facile synthesis of polyethyleneimine-protected fluorescent silver nanoclusters for the highly specific biosensing of curcumin

Zhifeng Cai,Liangliang Wu,Jierui Xi,Erxiao Hao,Kaifei Qi

In the present work, silver nanoclusters (Ag NCs) were established as a nanosensor for fluorescence determination of curcumin (Cur) for the first time. The silver nanoclusters were prepared by using polyethyleneimine as the stabilizer and ascorbic acid as the reducing agent. It displayed green emission around 510 nm. The fluorescence quenching mechanism between Ag NCs and Cur was studied and might be attributed to the static quenching and inner filter effect (IFE). Furthermore, it was found that this fluorescence sensor possessed an excellent linear relationship with the Cur concentration in the ranges of 0.1−100 μM. This fluorescence detection method have been successfully used for the detection of Cur with satisfactory results in bovine serum samples. This novel platform was expected to expand the possibility of a new fluorescence detection method.

Cyclodextrin-based metal-organic frameworks for pulmonary delivery of curcumin with improved solubility and fine aerodynamic performance

Yixian Zhou,Yiting Zhao,Boyi Niu,Qiaorong Luo,Yue Zhang,Guilan Quan,Xin Pan,Chuanbin Wu

Pulmonary drug delivery has attracted considerable attention in recent years. However, it is still a major challenge to deliver poorly water-soluble drugs to lungs with good solubility and fine aerodynamic performance. In this study, curcumin was loaded into cyclodextrin-based metal-organic frameworks (CD-MOFs) for pulmonary delivery. Compared with micronized curcumin prepared by jet milling, curcumin-loaded CD-MOFs (Cur-CD-MOFs) exhibited excellent aerodynamic performance, which was attributed to the unique porous structure and lower density of CD-MOFs. The dissolution test showed that the drug release rate of Cur-CD-MOFs was much faster than that of micronized curcumin. The all-atom molecular dynamic simulation showed that curcumin molecules were loaded into the hydrophobic cavities of CD-MOFs or entered into the large hydrophilic cavities to form nanoclusters. The elevated wettability of Cur-CD-MOFs and the unique spatial distribution feature of curcumin in porous interior of CD-MOFs might be favorable for the improved dissolution rate. The DPPH radical scavenging test showed that Cur-CD-MOFs had prominent antioxidant activities. Therefore, CD-MOFs were expected to be promising carriers for pulmonary delivery of poorly water-soluble drugs.

Curcumin-Adapted Nano-Composites within Dissolvable Microneedles for Precision-Tuned Piezo-Sono-Thermal Antibiotic-Free Acne Therapy

Yan Li,Caiqin Zhang,Cuihong Chen,Yuan Li,Wanyu Jin,Xiangmei Liu,Chaofeng Wang,Yufeng Zheng,Talante Juma,Yongping Cao,Shuilin Wu,Congyang Mao

The rational design of bioactive molecules to precisely engineer the energy-conversion interfaces represents an unexplored frontier in advanced therapeutics. Herein, we report a curcumin (Cur)-adapted nano-composites (BiO 2-X /Cur), integrated into a dissolvable microneedles (MNs) patch, for synergistic piezo-sono-thermal therapy against drug-resistant skin infections. The natural compound Cur is engineered not merely as an anti-inflammatory agent but as a critical molecular adapter that precisely tunes the interfacial charge dynamics and energy landscape at the composites interface, which extends carrier lifetime to 0.63 ns and significantly enhances oxygen adsorption capacity ( E ads = -1.821 eV). Under ultrasound irradiation, the patch demonstrates superior piezo-sonocatalytic activity (≈0.6 V piezoelectric output and d 33 value of −2.9 × 10 −12 C/N) to release reactive oxygen species and simultaneously delivers a substantial sono-thermal effect (ΔT > 20 °C). The dissolvable MNs platform ensures precise and painless intradermal delivery of BiO 2-X /Cur, enabling in vitro and in vivo near-complete eradication (>99.97%) of Propionibacterium acnes and methicillin-resistant Staphylococcus aureus , downregulating pro-inflammatory cytokines (IL-6, TNF-α, HIF-1α) while upregulating reparative markers (IL-10, Nrf2). This work establishes an emerging paradigm of using natural molecules as precision adapters for engineering high-performance composites, offering an intelligent and versatile platform for treating complex infectious diseases.

Discovery and evaluation of piperid-4-one-containing mono-carbonyl analogs of curcumin as anti-inflammatory agents

Jianzhang Wu,Yali Zhang,Yuepiao Cai,Jian Wang,Bixia Weng,Qinqin Tang,Xiangjian Chen,Zheer Pan,Guang Liang,Shulin Yang

We previously reported the design and discovery of three series of 5-carbon linker-containing mono-carbonyl analogs of curcumin (MCACs) as excellent anti-inflammatory agents. In continuation of our ongoing research, we designed and synthesized the fourth series of MCACs, whose central linker is a piperid-4-one. Their inhibitory effects against IL-6 production were evaluated in lipopolysaccharide (LPS)-stimulated macrophages. Among them, compounds F8 , F29 , F33 , F35 , and F36 exhibited the IC 50 values under 5   μM. The structure–activity relationship was discussed. Mechanistically, F35 and F36 dose-dependently prevented LPS-induced NF-κB and ERK activation. Finally, pretreatment with F35 and F36 significantly protected the C57B/L6 mice from LPS-induced septic death. Together, these data present a series of new analogs of curcumin as promising anti-inflammatory agents.

Preparation and in vitro evaluation of biological agents based on Zinc-laponite- curcumin incorporated in alginate hydrogel

Karimi Hajishoreh Negar,Dadashpour Mehdi,Akbarzadeh Abolfazl

Due to their outstanding structures and properties, three-dimensional (3D) hydrogels and nanoparticles have been widely studied and indicated a very high potential for medical, therapeutic, and diagnostic applications. However, hydrogels and nanoparticles systems have particular drawbacks that limit their widespread applications. In recent years, the incorporation of nanostructured systems into hydrogel has been developed as a novel way for the formation of new biomaterials with various functions to solve biomedical challenges. In this study, alginate-loaded Zinc- laponite—curcumin (Zn/La/Cur) nanocomposites were fabricated via ionic cross-linking. The prepared nanocomposite hydrogels were characterized via FTIR and FE-SEM. Moreover, energy dispersive x-ray spectroscopy (EDX) was used to study the elements of the Zn/La/Cur nanocomposite. The NIH3T3 fibroblast cell line was utilized for the MTT assay to determine the cell viability of the fabricated alginate-loaded Zn/La/Cur nanocomposites. MTT results demonstrated that there was no evidence of toxicity in the samples. These outcomes suggest that applying Al/Zn/La/Cur nanocomposite as a biological agent could be a novel tissue engineering strategy for treating soft tissue disorders.Graphical Abstract

Curcumin-stabilized silver nanoparticles encapsulated in biocompatible electrospun nanofibrous scaffold for sustained eradication of drug-resistant bacteria

Ling Cai,Xinyi Zhu,Hongjie Ruan,Jing Yang,Wei Wei,Yuan Wu,Liuzhu Zhou,Huijun Jiang,Minghui Ji,Jin Chen

Due to the misuse of antibiotics, the emerging drug-resistance of pathogenic microbes has aroused considerable concerns for the public health, which demands the continuous search for safe and efficient antimicrobial treatment. In this study, curcumin reduced and stabilized silver nanoparticles (C-Ag NPs) were successfully encapsulated into electrospun nanofiber membranes consisted of polyvinyl alcohol (PVA) cross-linked by citric acids (CA), which exhibited desirable biocompatibility and broad-spectrum antimicrobial activities. The homogeneously distributed and sustained release of C-Ag NPs in the constructed nanofibrous scaffolds yield prominent killing effect against Escherichia coli , Staphylococcus aureus and Methicillin-resistant Staphylococcus aureus ( MRSA ), which involved the reactive oxygen species (ROS) generation. Outstanding elimination of bacterial biofilms and excellent antifungal activity against Candida albicans was also identified after treated with PVA/CA/C-Ag. Transcriptomic analysis on MRSA treated by PVA/CA/C-Ag revealed the antibacterial process is related to disrupting carbohydrate and energy metabolism, as well as destroying bacterial membranes. Significant down-regulation of the expression of multidrug-resistant efflux pump gene sdrM was observed pointing to the role of PVA/CA/C-Ag to overcome the bacterial resistance. Therefore, the constructed ecofriendly and biocompatible nanofibrous scaffolds provide a robust and versatile nanoplatform of reversal potential to eradicate drug-resistant pathogenic microbe in environmental as well as healthcare applications.

Ceria nanozyme coordination with curcumin for treatment of sepsis-induced cardiac injury by inhibiting ferroptosis and inflammation

Chenxiao Jiang,Qianzhi Shi,Jing Yang,Hao Ren,Lu Zhang,Shan Chen,Jiayi Si,Yihai Liu,Dujuan Sha,Biao Xu,Jie Ni

Introduction Sepsis-induced cardiac injury is the leading cause of death in patients. Recent studies have reported that reactive oxygen species (ROS)-mediated ferroptosis and macrophage-induced inflammation are the two main key roles in the process of cardiac injury. The combination of ferroptosis and inflammation inhibition is a feasible strategy in the treatment of sepsis-induced cardiac injury. Objectives In the present study, ceria nanozyme coordination with curcumin (CeCH) was designed by a self-assembled method with human serum albumin (HSA) to inhibit ferroptosis and inflammation of sepsis-induced cardiac injury. Methods and results The formed CeCH obtained the superoxide dismutase (SOD)-like and catalase (CAT)-like activities from ceria nanozyme to scavenge ROS, which showed a protective effect on cardiomyocytes in vitro . Furthermore, it also showed ferroptosis inhibition to reverse cell death from RSL3-induced cardiomyocytes, denoted from curcumin. Due to the combination therapy of ceria nanozyme and curcumin, the formed CeCH NPs could also promote M2 macrophage polarization to reduce inflammation in vitro . In the lipopolysaccharide (LPS)-induced sepsis model, the CeCH NPs could effectively inhibit ferroptosis, reverse inflammation, and reduce the release of pro-inflammatory factors, which markedly alleviated the myocardial injury and recover the cardiac function. Conclusion Overall, the simple self-assembled strategy with ceria nanozyme and curcumin showed a promising clinical application for sepsis-induced cardiac injury by inhibiting ferroptosis and inflammation.

Curcumin-loaded polycaprolactone/carboxymethyl chitosan nanofibers with photodynamic inactivation of Staphylococcus aureus developed by solution blow spinning

Chaoyi Shen,Jiahao Zhong,Fengjun Guo,Changfeng Zhang,Changqing Zhu,Di Wu,Kunsong Chen

The purpose of the present study was to utilize solution blow spinning to fabricate curcumin (CUR)-loaded polycaprolactone (PCL)/carboxymethyl chitosan (CMCH) nanofibers with photodynamic inactivation activity for inactivation of S. aureus . The results of ATR-FTIR and XRD spectra demonstrated the successful encapsulation of CUR without disrupting the structure of the nanofibers and increasing the crystallinity of the nanofibers. After the encapsulation of CUR, the thermal stability, tensile strength, and hydrophobicity of the PCL/CMCH nanofibers were improved. Besides, the CUR-loaded PCL/CMCH nanofibers possessed good biocompatibility and low toxicity, and could release CUR sustainably over 120 h. Moreover, the obtained nanofibers showed better inhibition of S. aureus under blue light irradiation at a wavelength of 450 nm compared to the no-light condition. The above results suggested that the developed nanofibers with photodynamic inactivation can be applied as food packaging materials to reduce the impact of S. aureus on human wellness.
