A D-shaped fiber SPR sensor with a composite nanostructure of MoS2-graphene for glucose detection

Haixia Yu,Yang Chong,Penghao Zhang,Jiaming Ma,Dachao Li

Fiber-based techniques make it possible to implant a miniaturized and flexible surface plasmon resonance (SPR) sensor into the human body for glucose detection. However, the miniaturization of fiber SPR sensors results in low sensitivity compared with traditional prism-type SPR sensors due to limited sensing area. In this paper, we proposed a D-shaped fiber SPR sensor with a composite nanostructure of molybdenum disulfide (MoS 2 )-graphene to improve the sensor sensitivity. Compared with the traditional cylindrical fiber, the planar sensing area on the side-polished fiber makes it easier to modify two-dimensional materials. Chemical vapor deposition (CVD) graphene and CVD MoS 2 were modified on the sensor surface to obtain the MoS 2 -graphene composite nanostructure. π-π stacking interactions were used to modify pyrene-1-boronic acid (PBA) on the graphene. The excellent photoelectric properties of the MoS 2 -graphene composite nanostructure and the ability of PBA to specifically bind glucose molecules improved the glucose detection performance of the SPR sensor. The results show that specific detection of glucose was realized and that the highest sensitivity was achieved with three-layer MoS 2 and monolayer graphene.

Metal-to-Ligand Charge Transfer Chirality Sensing of d-Glucose Assisted with GOX-Based Enzymatic Reaction

Junjie Hao,Yiwen Li,Xiaoqian Xu,Fenghuan Zhao,Ruikun Pan,Junzi Li,Haochen Liu,Kai Wang,Jiagen Li,Xi Zhu,Marie-Hélène Delville,Ming Zhang,Tingchao He,Jiaji Cheng

Chiral transition metal oxides nanoparticles (NPs) with tunable optical properties are widely accepted as promising toolbox for chiral recognition, stereoselective synthesis, and chiroptical devices. Herein, chirality-based strategy is presented for discrimination of d -glucose from its enantiomer through a cooperative synergy between chiral cysteine capped MoO 2 NPs and the glucose oxidase nanosystem. The valence-state-dependent chirality induced by metal-ligand charge transfer effect is found to be ultrasensitive to its redox environment such as the presence of hydrogen peroxide, which is a key indicator of the stereoselective enzymatic reaction between glucose oxidase and d -glucose. With this know-how, glucose enantiomers can be precisely quantified with a limit of detection of 0.446 µ m . Such a chiral bio-nanosystem would be an ideal platform for a rational design of new types of biosensors, photocatalysts, and chirality-based nanodevices.

Tunable d-spacing of dry reduced graphene oxide nanosheets for enhancing re-dispersibility in organic solvents

He Chen,Chunhong Qi,Luyan Shen,Qiang Fu,Zhiqiang Wang,Zhiyuan Xiong,Youyi Sun,Yaqing Liu

The reduced graphene oxide (rGO) nanosheets were modified by using polymeric nanospheres to adjust the d-spacings between dry rGO nanosheets. The experimental and computational studies show that the incorporation of polymeric nanospheres can effectively increase the d-spacing between dry rGO nanosheets, and meanwhile provide good compatibility of rGO with organic solvents. Specifically, when the d-spacing between nanosheets is greater than 14.0 nm, the dried rGO nanosheets can be well redispersed and stabilized in various organic solvents. This study thus provides a new technology that can produce dry rGO nanosheets with good re-dispersibility and stability in various organic solvents on a large-scale in a more environmental friendly manner.

Fluorescent aptasensor based on D-AMA/F-CSC for the sensitive and specific recognition of myoglobin

Dongwei Miao,Dongkui Liu,Yanbo Zeng,Guobao Zhou,Wei Xie,Yiwen Yang,Hailong Wang,Jian Zhang,Yunyun Zhai,Zulei Zhang,Lei Li

A novel fluorescent biosensor based on dabcyl [(E)-4-((4-(dimethylamino) phenyl) diazenyl)benzoic acid] -modified anti-Mb aptamer (D-AMA) and 6-FAM(6-carboxyfluorescein) -modified complementary short chain (F-CSC)for the specific and sensitive detection of Mb levels is presented in this study. In PBS buffer solution, D-AMA bound to F-CSC, and then dabcyl quenched the fluorescence of 6-FAM. After adding Mb into the system, D-AMA bound to Mb and separated from F-CSC. The fluorescence of 6-FAM was restored after it separated from dabcyl. The assay exhibited high specificity and sensitivity toward Mb, with a low limit of detection of 0.07 ng/mL (S/N = 3) and linear relationships of 0.1–5 ng/mL. It was further applied to detect Mb levels in spiked human blood sera samples.

A graphene quantum dots-Pb2+ based fluorescent switch for selective and sensitive determination of D-penicillamine

Qi Wang,Lingfang Li,Tingxuan Wu,Xiangpeng Kong,Qingguo Ma,Chunlei Ma

Taking consideration of metal-induced fluorescence quenching and excellent coordination effect of D-penicillamine (D-PA), a graphene quantum dots (GQDs)-based fluorescent switch for D-PA detection was designed and established firstly with the help of lead ions . GQDs obtained from citric acids made them rich in carboxyl and hydroxyl groups , giving GQDs the ability to combine with lead ions. As anticipated, the fluorescence intensity was quenched by Pb 2+ through electron transfer process. Further, the addition of D-PA effectively recovered the fluorescence due to the departure of Pb 2+ from GQDs aroused by the strong coordination between D-PA and Pb 2+ . Thus, a fluorescent switch was activated for D-PA detection. The fluorescence recovery efficiencies were found to be proportional to the concentration of D-PA in the range of 0.6–50 μmol L − 1 and the detection limit was 0.47 μmol L − 1 . The real sample detection was performed in human urea sample and satisfactory recoveries of 96.84%–102.13% were obtained. The GQDs-Pb 2+ based fluorescent switch sensing method was firstly established with low detection limit and wide linear range, making it a supplement and improvement for D-PA detection.

Colorimetric Recognition of 3,4-Dihydroxy-D,L-phenylalanine with Tetrapeptide-modified Copper Nanoparticles as Chiral Nanozymes

Tian Lin,Cheng Cheng,Zhao Zhenwen,Liu Wei,Qi Li

T he construct of artificial nanocatalyts by simulating natural enzymes and thereby bringing new properties for practical applications is still a challenging task to date. In this study, chiral tetrapeptide ( L -phenylalanine- L -phenylalanine- L -cysteine- L -histidine)-engineered copper nanoparticles (FFCH@CuNPs) were fabricated as an artificial peroxidase (POD). More interestingly, the nano-catalysts exhibited chiral identification function. In comparison with other nanocatalysts like L -cysteine-, L -histidine-, chiral dipeptide ( L -cysteine- L -histidine)-, or chiral tripeptide phenylalanine- L -cysteine- L -histidine)-modified CuNPs, FFCH@CuNPs demonstrated a higher POD-mimetic catalytic activity in the 3,3′,5,5′-tetramethylbenzidine (TMB)-H 2 O 2 system and stronger enantioselectivity in the recognition of 3,4-dihydroxy- D , L -phenylalanine ( D , L -DOPA) enantiomers. Considering the strength difference between the intermolecular hydrogen bonding and the π - π interactions, the principle behind the chiral discrimination of D , L -DOPA was explored. Furthermore, contents levels of surface Cu 2+ ions and hydroxyl radicals were found in the FFCH@CuNPs-D-DOPA-TMB-H 2 O 2 system than in the FFCH@CuNPs-L-DOPA-TMB-H 2 O 2 system. Based on these results, a protocol for distinguishing between D , L -DOPA enantiomers through colorimetric recognition was established. This study provides a new insight into design and fabrication of oligopeptides@CuNPs-based chiral nanozymes with improved catalytic performance and features additional to those of natural enzymes.

Cannabidiol-Decorated Berberine-Loaded Microemulsions Improve IBS-D Therapy Through Ketogenic Diet-Induced Cannabidiol Receptors Overexpression

Xinyu Fan,Jiachen Shi,Ye Liu,Mengqiu Zhang,Min Lu,Ding Qu

Background Berberine (BR) shows promise as a candidate for treating irritable bowel syndrome with diarrhea (IBS-D). However, the undesired physicochemical properties and poor oral absorption limit its clinical translation. A ketogenic diet (KD) can induce intestinal overexpression of cannabidiol (CB) receptors, which may offer a potential target for IBS-D-specific delivery of BR.Methods The microemulsions loaded with BR and decorated with cannabidiol (CBD/BR-MEs) were developed through a one-step emulsion method. The pharmaceutical behaviors of the CBD/BR-MEs were measured using dynamic light scattering and high-performance liquid chromatography. The efficacy of the anti-IBS-D therapy was evaluated by assessing fecal water content, Bristol score, and AWR score. The intestinal permeability were assessed through immunofluorescent staining of CB1 and ZO-1, respectively. The signaling of CREB/BDNF/c-Fos was also studied along with immunofluorescent and immunohistochemical examination of brain sections. Results The CBD/BR-MEs, which had a particle size of approximately 30 nm and a surface density of 2% (wt%) CBD, achieved greater than 80% (wt%) encapsulation efficiency of BR. The pharmacokinetics performance of CBD/BR-MEs was significantly improved in the KD-fed IBS-D rats than the standard diet-fed ones, which is highly related to intestinal expression of CB1 receptors. The treatment with CBD/BR-MEs and KD exhibited evident comprehensive advantages over the other groups in terms of anti-IBS-D efficacy. CBD/BR-MEs and KD synergistically decreased intestinal permeability. Moreover, the treatment with CBD/BR-MEs and KD not only blocked the CREB/BDNF/c-Fos signaling in the brain but also decreased the levels of neurotrophic factors, neurotransmitters, and inflammatory cytokines in the serum of IBS-D model rats. Conclusion Such a design represents the first attempt at IBS-D-targeted drug delivery for improved oral absorption and efficacy through KD-induced target exposure, which holds promising potential for the treatment of IBS-D.

D (+)-Glucosamine (DGA) based polyesteramide TFC NF membrane for the pretreatment of reverse osmosis seawater desalination

Ruijun Zhang,Wenqian Zhu,Jiayu Tian,Shanshan Gao,Bart Van der Bruggen

Reverse osmosis is an effective seawater desalination way extending the available sources of fresh water, but challenged by the gypsum scaling. Herein, D (+)-glucosamine (DGA) was rationally selected and utilized to prepare a novel polyesteramide (PEA) NF membrane that can reject SO 4 2− but release Ca 2+ . Then it was employed in the pretreatment of seawater reverse osmosis to spatially isolate SO 4 2− and Ca 2+ , thus removing the “foundation” for forming gypsum. Compared to the referenced NF270, the optimized sample denoted as DGA-PEA-NF has a looser structure with stronger negative charge, as well as a higher resistance to both organic fouling and chlorine attack. During the pretreatment of seawater, both the water permeability and SO4 2− /Ca 2+ selectivity of the DGA-PEA-NF membrane were much higher than those of NF270; the seawater permeance and SO 4 2− /Ca 2+ selectivity of DGA-PEA-NF were 10.25 L m −2 ·h −1 ·bar −1 and 7.62, respectively, while those of NF270 were only 5.11 L m −2 ·h −1 ·bar −1 and 2.08. Consequently, the DGA-PEA-NF membrane effectively reduced the gypsum scaling both in NF and RO stages rather than just transferring the fouling problem forwards to NF stage. The implementation of this research is expected to provide guidance for a targeted fabrication of NF membranes, as well as support for the sustainable development of seawater desalination.

Immobilization of Bio-imprinted Phospholipase D and Its Catalytic Behavior for Transphosphatidylation in the Biphasic System

Zhao Xia,Guo Meijing,Li Xian,Liu Bo,Li Binglin,Wang Jiao

Phospholipase D (PLD) with the higher transphosphatidylation activity was screened from Streptomyces sp. LD0501 basing on the protoplast mutagenesis technology. Then, it was successfully bio-imprinted to form a hyperactivated structure and rigidified by the intramolecular cross-linking, which was immobilized on the nonporous nanoscale silica. Characterization techniques were employed to investigate the structure and physicochemical properties of the catalysts, including Fourier transform infrared (FTIR) spectra and scanning electron microscopy (SEM) analysis. Transphosphatidylation activity and selectivity were improved significantly when immobilized PLD was used. The maximum yield for the production of phosphatidylserine (PS) reached 97% and the side reaction, the hydrolysis, was minimized. These results were further confirmed by the nuclear magnetic resonance (NMR) and mass spectrometry (MS) analysis. The imprint-induced characteristics of PLD was successfully “remembered” even in the present of much water. In addition, this immobilized hyperactivated PLD showed the excellent operational stabilities and environmental tolerances. Graphical abstract

Vitamin D improves hepatic steatosis in NAFLD via regulation of fatty acid uptake and β-oxidation

Tingwan Du,Lian Xiang,Jingjing Zhang,Chunmei Yang,Wenxin Zhao,Jialu Li,Yong Zhou,Ling Ma

Introduction The study aimed to explore the association of serum 25(OH)D 3 and hepatic steatosis in non-alcoholic fatty liver disease (NAFLD) patients and to determine whether the effect of vitamin D (VD) is mediated by activation of the peroxisome proliferator-activated receptor α (PPARα) pathway. Methods The study contained a case-control study, in vivo and in vitro experiments. A case-control study was conducted to compare serum parameters between NAFLD patients and controls and to evaluate the association of 25(OH)D 3 and NAFLD. In vivo study, male Wistar rats were randomly divided into control and model groups, fed a standard chow diet and a high-fat diet (HFD), respectively, for 7 weeks to generate an NAFLD model. Then, the rats were treated with VD and a PPARα antagonist (MK886) for 7 weeks. Tissue and serum were collected and assessed by biochemical assays, morphological analysis, histological analysis, and western blot analysis. In vitro , HepG2 cells were incubated with oleic acid (OA) to induce steatosis, which was evaluated by staining. HepG2 cells were pretreated with MK886 followed by calcitriol treatment, and differences in lipid metabolism-related proteins were detected by western blot. Results NAFLD patients were characterized by impaired liver function, dyslipidemia, and insulin resistance. Serum 25(OH)D 3 was negatively associated with alanine aminotransferase (ALT) in NAFLD. VD deficiency was a risk factor for patients with no advanced fibrosis. Adequate VD status (25(OH)D 3 >20 ng/mL) had a protective effect in patients after adjustment for confounding variables. NAFLD rats showed hyperlipidemia with severe hepatic steatosis, systematic inflammation, and lower serum 25(OH)D 3 . VD treatment ameliorated hepatic steatosis both in NAFLD rats and OA-induced HepG2 cells. Further, MK886 inhibited the anti-steatosis effect of VD. Conclusion The study revealed that an adequate VD level may act as a protective factor in NAFLD and that VD may alleviate hepatic steatosis via the PPARα signaling pathway.

Synergistic inhibition of benzotriazole and sodium D-gluconate on steel corrosion in simulated concrete pore solution

Tengfei Xiang,Yanli Zhang,Linjing Cui,Jiaqi Wang,Depeng Chen,Shunli Zheng,Yujie Qiang

Herein, the synergistic inhibition of two typical corrosion inhibitors benzotriazole (BTA) and sodium D -gluconate (SD) on the corrosion of Q235 steel in simulated concrete pore solution (SCP) were systematically studied. The results demonstrated that the mixed BTA/SD showed an excellent anti-corrosion performance and the inhibition efficiency of (BTA/SD, 1ː1) was as large as 98.33 % which is higher than that of BTA (88.65 %) and SD (92.16 %). Furthermore, the synergistic inhibition effect of BTA/SD was also researched by surface analysis technologies, the X-ray photoelectron spectroscopy (XPS) result verified that the BTA/SD adsorbed on steel, while both scanning electron microscopy (SEM) and atomic force microscopy (AFM) images proved that the mixed BTA/SD can effectively protect the steel from corrosion in SCP solution which containing corrosive ions. In addition, the anti-corrosion mechanism of the mixed BTA and SD was revealed by adsorption model and molecular dynamics (MD) simulation, the mixed BTA/SD corrosion inhibitors showed the largest K ads , the lowest ∆ G ads and the largest binding energy values , demonstrating the strong adsorption on steel and the best anti-corrosion performance. The results of this work can provide experimental and theoretical references for the corrosion protection of reinforced concrete structures while using synergistic inhibitors.

A novel membrane biofouling mitigation strategy of D-amino acid supported by polydopamine and halloysite nanotube

Xiaoyan Guo,Shougang Fan,Yandi Hu,Xiaolin Fu,Huaiqi Shao,Qixing Zhou

D-amino acid (DAA) an environmentally friendly biofilm inhibitor, has low efficiency for membrane biofouling control due to its instability. To address this challenge, a novel nanocomposite was prepared with DAA adhering to polydopamine (PDA)-coated halloysite nanotube (HNTs) through the interactions of H-bonding and π-π stacking between PDA and DAA. Membrane modified with such nanocomposite was fabricated via blending phase inversion, and the nanocomposite was uniformly distributed in the modified membrane matrix. In comparison with the pristine membrane, the addition of nanocomposites resulted in robust mechanical property for modified membrane with the ultimate stress and strain increased by 23.97% and 35.62%, respectively. Moreover, an excellent tradeoff between water flux (2.5 folds of pristine membrane) and selectivity was achieved, probably due to the improved membrane hydrophilicity. Meanwhile, bovine serum protein (BSA) static adsorption as well as dynamic filtration experiments exhibited excellent antifouling ability of the modified membrane. Most importantly, a superior anti-biofouling stability over a 10-day period was obtained for the membrane modified with nanocomposite, indicating that the activity of DAA to mitigate biofouling was effectively maintained. This study developed a novel and promising strategy for membrane biofouling mitigation.
