Umami peptides can help reduce the salt content in foods while still maintaining a savory taste. Few studies have reported the bioactivity of umami peptides after consumption. We studied the bioactivities of 12 umami peptides after gastrointestinal digestion. Three umami peptides exhibited angiotensin I-converting enzyme (ACE) inhibitory and antioxidant activity after digestion. Six novel peptides were identified from digestion solutions of the peptides by HPLC-MS/MS. Among them, CC, CCNK, and HCHT had both ACE inhibitory activity (IC50 values were 9.81, 9.00, and 114.99 μM, respectively) and antioxidant activity (strong 1,1-Diphenyl-2-pycryl-hydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) free radical scavenging activities). AHSVRF had strong ACE inhibitory activity. These peptides increased the nitric oxide concentration and decreased the content of endothelin-1 in a medium of human umbilical vein endothelial cells in a dose-dependent manner. Experiments with damaged HepG2 cells showed that peptides CC, CCNK, and HCHT had antioxidant activity through their cytoprotective effects and by reducing the reactive oxygen species content. The results indicated that umami peptides may provide many health benefits after consumption.

Bioactive synthetic scaffolds with 3D porous structure are the most attractive materials among various guided bone regeneration membranes. However, its osteogenic potential is still insufficient. This study was aimed to develop a Calcium surface-anchored Collagen I-PLGA/PCL scaffolds (PP/COL I-pDA-Ca) with enhanced osteogenicity. PP/COL I-pDA-Ca was electrospun from a collagen I-blended PLGA/PCL matrix and then modified by the chelation of Ca-ions via mussel-inspired polydopamine coating. PLGA/PCL, PLGA/PCL-polydopamine, PLGA/PCL-pDA chelated by Ca-ions were used as controls. Osteogenic effects of the scaffolds were examined using MC3T3-E1 cell culture. PP/COL I-pDA-Ca maintained 3D porous architecture with interconnected pores formed by randomly-oriented filamentous fibers and MC3T3-E1 cells cultured on it for 12 h or 24 h were more stretched and spread than those on the controls. PP/COL I-pDA-Ca significantly upregulated α10, α11 and β1 integrin expression after 48 h culture. ALP activity, OCN, OSX, BMP2 and RUNX2 expression of MC3T3-E1 cells cultured on PP/COL I-pDA-Ca scaffolds for 7 and 14 days were substantially enhanced when compared to controls ( p < 0.05). The pDA-based Ca chelation, COL I incorporation and 3D bionic structure promoted cell adhesion and osteogenic differentiation of MC3T3-E1 cells. This novel PP/COL I-pDA-Ca scaffolds is an attractive alternative for guided bone regeneration.

Brucellosis, acting as a typical chronic zoonotic disease, is caused by the invasion ofBrucellainto the human body. Outer membrane protein 25 (Omp25), specifically localized on theBrucellamembrane, is the main virulence factor ofBrucellaand participates in multiple links of the damage process. Omp25c, a porin protein ofBrucella, is a paralog of Omp25 with high sequence identity. NADH dehydrogenase [ubiquinone] complex I assembly factor 2 (Ndufaf2) has a key function in cell energy metabolism, particularly in the formation and activity of the mitochondrial respiratory chain. Loss of Ndufaf2 results in oxidative stress and mitochondrial DNA (mtDNA) deletion. However, the functional relationship between Omp25c and Ndufaf2, the underlying mechanism of the proteins, remains unclear. In this work, we purified the Omp25c and Ndufaf2proteins. Our data revealed that Omp25c directly interacts with Ndufaf2, as determined using Biacore analysis. In addition, assays revealed that Ompa2c reshapes the host cell’s redox environment by decreasing the oxidized nicotinamide adenine dinucleotide/reduced nicotinamide adenine dinucleotide (NAD+/NADH) ratioand adenosine triphosphate (ATP) production, whereas Ndufaf2 exerts an opposing regulatory effect; Co-expression results further revealed an antagonistic relationship between the two during metabolic processes. These findings provide a new perspective for elucidating the mechanisms of mitochondrial functional regulation inBrucella–host interactions and lay the theoretical and experimental foundation for drug development targeting metabolic interventions to eliminate intracellular pathogens.

The degradation of complex plant polysaccharides, particularly Rhamnogalacturonan I (RG-I) pectin, is essential for unlocking the value of biomass. This study focuses on the enzymatic toolbox of Penicillium oxalicum for efficient RG-I breakdown. Here, we recombinantly expressed 15 novel pectinolytic enzymes and characterized their biochemical functions. The enzyme system consists of backbone-cleaving rhamnogalacturonan hydrolases and lyases, along with side-chain–degrading enzymes. Specific endo-β-galactanases (GH30, GH43, GH53) and an exo-β-galactosidase (GH35) collaboratively hydrolyze galactan side chains. Meanwhile, α-arabinanases (GH43) and exo-α-arabinofuranosidases (GH54, GH62) work synergistically to degrade arabinan side chains. Through coordinated action, these enzymes achieve complete degradation of complex RG-I structures. Our findings provide a comprehensive enzyme set for targeted polysaccharide deconstruction, offering valuable resources for biomass conversion and pectin valorization in industrial applications.

Zearalenone (ZEN) is a common mycotoxin widely present in food and agricultural products, posing a serious threat to human health. This work developed a fluorescent/smartphone-assisted colorimetric dual-mode aptasensor for detecting ZEN. Platinum nanowires-supported MXene (PtNWs/MXene) composites exhibit peroxidase-like (POD) activity, which can be inhibited by the adsorbed ZEN-specific DNA aptamers. In the presence of ZEN, the specific binding between ZEN and the aptamer triggers desorption of the aptamer from the PtNWs/MXene surface, thus restoring the POD activity of composites. With hydrogen peroxide introduced, PtNWs/MXene catalyzes the oxidation of colorless o-phenylenediamine into colored 2,3-diaminophenazines, simultaneously generating a fluorescent signal. Exonuclease I mediates target cycling to achieve signal amplification, further boosting the detection sensitivity. The smartphone-based colorimetric mode shows a linear detection range of 10 ~ 900 ng/mL with a limit of detection (LOD) of 6.57 ng/mL, while the fluorescence mode presents a linear range of 1 ~ 1000 ng/mL and a LOD as low as 0.74 ng/mL. This sensing platform can accurately distinguish ZEN-positive and ZEN-negative samples, and the detection results are highly consistent with those of ELISA. It provides an efficient and reliable new solution for highly sensitive on-site ZEN detection, demonstrating significant application potential in food safety monitoring. Gastric cancer (GC) is a common malignant tumour of the digestive tract with a high mortality rate worldwide. However, many patients delay treatment due to the avoidance of the costly and painful procedure of gastroscopy. Therefore, an early convenient screening method is essential to improve the survival rate of GC patients. To address this issue, we constructed an electrochemical immunosensor supported by rhombohedral Cu3Pt and MoS2 nanoflowers (MoS2 NFs) for rapid, painless and quantitative detection of the GC biomarker in vitro. Here, pepsinogen I was employed as a model protein biomarker to analyse the performance of the immunosensor. The rhombohedral dodecahedral Cu3Pt nanoparticles decorated with MoS2-NFs were further functionalized; this allowed the constructed sensor to possess more nano- or micro-structures, thereby improving the detection sensitivity. In specific applications, the corresponding bioactive molecules can be flexibly captured. Under optimal conditions, the immunoassay showed a wide linear range from 500 pg mL−1 to 400 ng mL−1 and a low detection limit of 167 pg mL−1 (S/N = 3). This covers the critical value of 70 ng mL−1, and the results obtained from the analysis of human serum samples were on par with those from the enzyme immunoassay, suggesting significant potential for this new method in daily diagnosis.

Our study aimed to investigate the effect of electroacupuncture pretreatment on the inflammatory response and expression levels of LC3-II/I and Beclin 1 using a model of lipopolysaccharide (LPS)-induced acute lung injury (ALI). Eighteen male Sprague-Dawley (SD) rats were randomly divided into three groups: normal control group (NC, n = 6), LSP modeling group (LM, n = 6), and electroacupuncture group (EA, n = 6). Rats in the EA group received electroacupuncture pretreatment at bilateral Zusanli (ST36) and Chize (LU5) points for five days (30 min each time daily, frequency; 3 Hz/15 Hz, intensity; 1 mA). Rats in the EA and LM groups were then injected with 5 mg/kg LPS (Beijing, Solarbio Company, concentration; 5 mg/mL) through the tail vein, while those in the NC group were injected with 5 mg/kg saline. The animals were sacrificed six hours after LPS or saline injection through cervical vertebrae by dislocation under deep anesthesia. Orbital blood was collected for the analysis of serum inflammatory factors including interleukin-1β (IL-1β) and transforming growth factor-β (TGF-β). The lower left lung was excised, stained with hematoxylin-eosin (HE), and subjected to histopathological analysis. The mRNA and protein expression of Beclin 1 and LC3 II/I in the lower right lung tissues were detected via RT-qPCR and Western blot analyses, respectively. The results showed that lung injury score was significantly higher in the LM group than that of the NC group () and EA group (). The IL-1β contents were significantly decreased in the EA group () than in the LM group. In contrast, the GF-β contents were increased in the EA group significantly when compared with the LM group (). RT-qPCR and Western blot detection showed that the relative gene expression of LC3‐II/I and Beclin 1 was significantly lower in the EA group than in the LM group (). However, the relative protein expression level of LC3‐II/I and Beclin 1 was slightly lower in the EA group than the in LM group (). These results show that electroacupuncture pretreatment reduces the inflammatory response in ALI and can protect lung tissue by inhibiting the gene and protein expression levels of LC3‐II/I and Beclin 1.

The nonradiative carrier recombination at the perovskite/carrier selective layer (CSL) interface was accounted for the inferior power conversion efficiency (PCE) of perovskite solar cells (PSCs), especially rigid all-inorganic perovskite (CsPbI3 and CsPbBr3). In this study, targeting the poor interface, we introduce SbX3 (X = Cl, Br, I) surface passivation at the CsPbBr3/carbon interface. Smoothed compressive strain, reduced defect density, and enhanced energy-level alignment were achieved simultaneously, facilitating carrier extraction at the selective interface. With the simple aqueous solution-based two-step process, the PCE of our SbI3 passivated carbon-based CsPbBr3 PSCs has increased from 7.81% (without passivation) to 9.69%, a ∼25% enhancement. Specifically, Voc (1.657 V) of the SbI3-passivated cells was much higher than that of the control ones (1.488 V), confirming the ameliorated interface. Finally, our unencapsulated SbI3 passivated devices maintain 90% of their initial PCEs while left in the air for 30 days with a relative humidity of 60%. To conclude, we present an interfacial carrier extraction-enhanced strategy for preparing high-performance and stable CsPbBr3-based PSCs.

Quantum dots functionalization has been proven to be a simple modification strategy for improving the electroanalytical performance of two-dimensional electrode materials by increasing the specific surface area and active reaction sites. Herein, a new electrochemical sensing platform was fabricated by SnO2 quantum dot-functionalized Ti3C2 MXene (Ti3C2-SnO2QDs) for the highly sensitive detection of Sudan I in food. Ti3C2-SnO2QDs were prepared via in situ synthesis, which can control the nucleation and growth of SnO2QDs, resulting in the well-dispersed SnO2QDs with 2–3 nm size on the intersheet surface of MXene. Moreover, the formation of Ti3C2-SnO2QDs can effectively restrict the aggregation of Ti3C2 and improve the stability of SnO2QDs in aquatic environment. The prepared nanocomposite can be used as an improved modified material to further increase the electrocatalytic performance and electrochemical signal of Sudan I on the surface of a glassy carbon electrode. Under optimized conditions, the proposed analytical method displayed a linear dependence for Sudan I concentration ranging from 0.008 to 10 μM with a detection limit of 0.27 nM (S/N = 3) by electrochemical cyclic voltammetry. This sensor with excellent selectivity, reproducibility and accuracy was quantitatively validated in commercial ketchup and chili powder. This Ti3C2-SnO2QDs-based Sudan I sensor is expected to expand the application of MXene nanocomposites in electrochemical analysis and is envisioned as a promising candidate for monitoring illegal food additives in real food samples.

Carbon dots (CDs) have emerged as promising nanomaterials for bioimaging-guided photodynamic therapy (PDT). However, designing red-emissive CDs (RCDs) with tunable type I and type II reactive oxygen species (ROS) generation to simultaneously meet PDT applications in aerobic and hypoxic scenarios still remain major challenges. Herein, three types of RCDs with maximum emission at approximately 680 nm are successfully prepared. It is noteworthy that they exhibit the adjustable ROS production with equal superoxide anion (via type I PDT) and incremental singlet oxygen (via type II PDT). Detailed structural and optical characterizations along with theoretical calculation reveal that the unique type I/II ROS formation mainly depends on the core sizes and surface states of RCDs, which determine their identical redox potentials and tapering energy gaps between singlet- and triplet states, respectively. Additionally, due to the inherent mitochondria targeting capability, RCDs enable themselves to induce cell programmed death via activating mitochondrion-mediated apoptotic pathways. This work exploits the unprecedented RCDs with tunable type I and type II ROS generation that could ensure highly efficient tumor eradication both in vitro and in vivo, even under the harsh tumor microenvironment , providing a new prospect for CDs as nanophotosensitizers to conquer the limitations of single type PDT.

Herein, we develop a novel strategy for preparing all-inorganic cesium lead halide (CsPbX3, X = Cl, Br, I) perovskite nanocrystals (NCs)@Zn-based metal-organic framework (MOF) composites through interfacial synthesis. The successful embedding of fluorescent perovskite NCs in Zn-MOFs is due to the in situ confined growth, which is attributed to the re-nucleation of water-triggered phase transformation from Cs4PbBr6 to CsPbBr3. The controllable synthesis of mixed-halide based composites with various emission wavelength can be achieved by adding the desired amount of halide (Cl or I) salts in the re-nucleation process. More importantly, the anion exchange reaction is inhibited among various composites with different halogen atoms by being trapped in MOFs. Besides, a white light-emitting diode (WLED) is produced using a blue LED chip with the green-emitting and red-emitting composites, which has a color coordinate of (0.3291, 0.3272) and a wide color gamut. This work provides a novel route to achieving perovskite NCs growth in MOFs, which also can be extended to the other NCs embedded in frames as well.

Pursuing a selective and stable optical sensor for the reliable detection of perfluorooctane sulfonate (PFOS) is critical to controlling and reducing the risk of exposure to human. Here, we elaborately prepared an atomic copper(I)-carbon nitride (Cu-CN) peroxidase-mimic catalyst and therefore developed a reliable and accessible PFOS detection method. Well-distributed Cu(I) atoms are exclusively anchored within CN nanosized matrix as identified Cu-CN single-site active centers. Benefiting from the high peroxidase-mimic catalytic capability and colloidal stability, Cu-CN exhibits outstanding detection of PFOS selectively in the range of 0.07 to 3 μM coupled with the detection limit of 14.9 nM. The occupation of atomic Cu(I) specific catalytic sites caused by strong PFOS coordination and the fluorophilic interaction between PFOS should be responsible for the damping of TMB oxidation. Moreover, this detection platform was also used successfully to detect PFOS in three real water samples with RSD ≤ 8.37%. This well-established colorimetric sensing system based on peroxidase-mimic capability of single-atom catalyst is promising for the detection of other hazards in aquatic environment.
