Structure-based engineering of O-succinyl-L-homoserine mercaptotransferase with improved substrate tolerance

Xiao-Ling Tang,Kang Zhao,Teng-Kun Huang,Bai-Hua He,Xu-Gang Zhang,Zhi-Qiang Liu,Yu-Guo Zheng

O -Succinyl- L -homoserine mercaptotransferase (MetZ) is a key enzyme for the synthesis of L -methionine. However, the limited tolerance to high concentrations of substrate methyl mercaptan sodium restricts its industrial applications, while the “trade-off” between activity and stability has always been a challenge for protein engineering. To address this problem, a strategy of dynamic network rigidification-local flexibility regulation was applied in this study to engineer the enzyme from Chromobacterium violaceum ( Cv MetZ). Through consensus sequence analysis and structure-guided design, the hotspot amino acid residues were anchored for mutagenesis. The best mutant T59I/T119Y/A241T was obtained with synergistic enhancement of activity and stability. The half-life ( t 1/2 ) of whole cells harboring the mutant was increased from 3.3 and 1.3 h to 20.6 and 7.2 h, in the presence of 5% and 10% methyl mercaptan sodium, respectively. The engineered enzyme was successfully immobilized with ideal usability, which laid the foundation for efficient and scale-up bioproduction of L -methionine.

Neurotoxic impacts of neonicotinoid pesticide residues in Chinese honey on Apis mellifera L.

Haoming Guo,Nan Xiao,Yabo Liang,Xueke Liu,Peng Wang,Donghui Liu

BACKGROUND The threat to bees of neonicotinoid pesticides (NEOs) has raised concern. However, data on the disruption of the bee nervous system following long-term low-dose exposure are lacking. In this study, six NEOs were quantified in 31 honey samples from China, and their behavioral effects on bees were evaluated. RESULTS Neonicotinoid pesticide detection rates ranged from 35.48% to 83.87%, with mean residue concentrations of 2.39–6.73 μg kg⁻¹. Clothianidin showed the highest detection frequency (83.87%), with an average residue of 5.54 μg kg −1 . To assess neurotoxic effects at actual residue levels, bees were fed NEO-contaminated syrup for 21 days. This exposure significantly downregulated eight neurotransmitters, particularly 5-hydroxytryptamine (5-HT) and tryptophan (Trp), impairing normal physiological function. Behavioral assays showed reductions of 31.71% to 68.53% in displacement distance, 27.13% to 40.06% in movement rate, and 64.58% to 77.14% in inter-individual contact time. CONCLUSION This study confirmed that the NEO residues in honey could hinder bees' ability to communicate and forage, with potential adverse effects on the development of bee populations. It demonstrated that bees exposed to real-world residue levels of NEOs exhibit neurological impairments. © 2025 Society of Chemical Industry.

Silk Fibroin Material with Enhanced Performances Using Poly(D, L-Lactide-Co-Glycolide)

Jingwen Lan,Rukuan Liu,Sisi Liu,Airong Xu

Eco-friendly silk fibroin/poly(D,L-lactide-co-glycolide) (SF/PLGA) materials were successfully fabricated using a facile strategy. The materials were characterized by scanning electron micrograph (SEM), X-ray diffraction (XRD) and infrared spectra (IR). Systematic investigations were completed to examine the degradation rates in natural soil, cytocompatibility and thermostability of the materials. It is interesting to find that after SF was hybridized by PLGA, the thermostability and degradation rate increased. Meanwhile, the materials show good cytocompatibility. SEM, IR and XRD results reveal that there is hardly any interaction between SF and PLGA in the SF/PLGA material, and SF is physically mixed with PLGA. This study opens up a new horizon in the design and preparation of SF-based materials for promising applications in medical and biodegradable material fields.

Trace sorbitol-modified nano-silica: Towards nano-nucleation for poly(L-lactic acid)

Biao Yang,Xinyu Wan

Nucleating agents, especially those with small particle sizes, are preferred to boost the nucleation density and crystallinity of poly(lactic acid) (PLA) due to its weak crystallization capability. Organophilicly modified nanofillers hardly alter the nucleation and crystallinity of non-isothermally crystallized PLA. Herein, nano-silica adsorbed trace D -sorbitol (m-SiO 2 ) as a heterogeneous nucleating agent was melt-mixed with poly( L -lactic acid) (PLLA), and the isothermal and non-isothermal crystallization behavior, as well as crystallization kinetics, were investigated. Transmission electron microscopy (TEM) revealed that m-SiO 2 was uniformly dispersed in the PLA matrix as 100–300 nm clusters. Differential scanning calorimetry (DSC) and polarized optical microscopy (POM) showed that the nucleation rate and density of the non-isothermally crystallized PLLA/m-SiO 2 composites were significantly improved. Despite the fact that m-SiO 2 does not raise the overall non-isothermal crystallization rate, the crystallization temperature and crystallinity of the PLLA/3%m-SiO 2 composite increased from 97.2 °C and 6.8 % for neat PLLA to 108.2 °C and 48.6 % (10 °C/min cooling rate), respectively. The Avrami exponent n of isothermal crystallization remains unchanged, while the crystallization rate increases dramatically. Both isothermal and non-isothermal crystallization have increased activation energies. The heat deflection temperature increased from 59 °C of neat PLLA to 152 °C with a 50 % increase in impact strength.

Microneedle-Integrated FePc–MOF–MXene Nanozyme Patch for In Vivo L-Cysteine Monitoring

Ghazala Ashraf,Haonan Wang,Khalil Ahmed,Huiwen Xiong,Jilie Kong,Xueen Fang

Advancing clinical diagnostics requires platforms that combine catalytic efficiency, biocompatibility, and real-time, in vivo accessibility. Herein, this study reports a structurally integrated FePc–ZIF-8–MX nanozyme that combines the redox activity of FePc, the porous confinement of ZIF-8, and the electrical conductivity of MX. Synthesized via a low-energy, ambient-condition process, this hybrid enables efficient electron transfer, enhanced analyte enrichment, and sustained catalytic activity in physiological environments. To translate this functionality into a wearable diagnostic format, the hybrid is seamlessly incorporated into a microneedle array, offering minimally invasive access to interstitial fluid for continuous L-cysteine (L-Cys) monitoring. The resulting platform exhibits high selectivity and sensitivity across complex biological matrices, including serum, urine, cultured cells, and a murine model of myocardial infarction. This study presents a multifunctional electrochemical platform that enables on-body metabolite monitoring through a microneedle-integrated nanozyme interface. To the best of our knowledge, it constitutes the first realization of real-time, in vivo L-Cys sensing in this format, setting a new benchmark for precision biosensing in translational healthcare.

Ultrasonic extraction, Structural Characterization, and Immunoactivity of Polysaccharides From Syzygium jambos (L.) Alston

Shao-Qian Wang,Hui Yuan,Hao-Qiang Qin,Ming-Yue Peng,Ming-Xiao Cui,Ke-Hai Liu

This research emphasized the extraction and separation of polysaccharides derived from Syzygium jambos (L.) Alston (PSJAP-5), as well as analyses of their structural characteristics and immunomodulatory activities. This study initially employed response surface methodology to determine the extraction conditions of polysaccharides. The average molecular weight of PSJAP-5 was 58 102 Da, and its monosaccharide composition was galactose, arabinose, galacturonic acid, glucuronic acid, and rhamnose (39.84:29.13:21.97:6.02:4.17). In vitro studies showed that PSJAP-5 promoted the proliferation and phagocytosis of macrophages. Meanwhile, PSJAP-5 (25–200 µg/mL) promoted the release of NO and ROS, and increased the levels of IL-6, TNF-α, and IL-1β. After pretreating macrophages with TLR4, NF-κB, and MAPK inhibitors, the immunoenhancing effects of PSJAP-5 on macrophages treated with the inhibitors were decreased. Similarly, PSJAP-5 promoted the phosphorylation of p38 in the MAPK signaling pathway and reduced the phosphorylation of IκBα in the NF-κB signaling pathway in RAW264.7 cells. PSJAP-5 might serve as a potential immunomodulator for the treatment of diseases characterized by impaired immune function. Graphical This study focused on extracting and purifying a polysaccharide (PSJAP-5) from Syzygium jambos (L.) Alston, analyzing its structural properties, and evaluating its immunomodulatory effects.

Alectinib causes sinus bradycardia by suppressing L-type calcium current in sinus node

Guo-Xuan Liu,Fan Diao,Guang Lu,Qiang Zheng,Bin Fu,Hua-Chen Jiao,Ke-Zhou Wang,Dong-Hai Liu

Alectinib is the first-line therapy for anaplastic lymphoma kinase rearranged non-small cell lung cancer globally. Sinus bradycardia, as the major adverse cardiac events of alectinib, still widely impact patient’s quality of life. However, its underlying mechanism remains elusive. The aim of this study was to reveal the pathogenesis of the alectinib induced sinus bradycardia (AISB) in a rat model, including the electrophysiology alterations and the molecular mechanism. SD rats were administered alectinib (10 mg / kg / day) by gavage for 7 to 10 days to mimic the clinical AISB. 3-days alectinib treatment did not change heart rate and sinus node recovery time (SNRT) as assessed through in vivo electrophysiology study. Also, alectinib didn’t influence automaticity in isolated heart or single sinus node cardiomyocytes, indicating alectinib cannot decrease sinus node function rapidly. The decreased heart rate and prolonged SNRT was found after 7-days alectinib treatment. The inducibility of atrial fibrillation was not affected under the same condition. The RNA-seq assay revealed the transcriptomic alterations in sinus node of alectinib treated rats, and the dysregulation of genes in cardiac function were observed. The decreased expression of L-type calcium channel Cacna1d was confirmed among the channel candidates identified from RNA-seq assay. Subsequently, the patch-clamp test revealed the reduction in the corresponding L-type calcium current density in 7-days alectinib treated rats. These findings revealed that the AISB was caused by the reduction in Cacna1d expression which resulted in the electro-dysfunction mediated by the suppressed I CaL .

CpGSTd3 is a lambda-Cyhalothrin Metabolizing Glutathione S-Transferase from Cydia pomonella (L.)

Wei Wang,Chao Hu,Xin-Ru Li,Xiao-Qi Wang,Xue-Qing Yang

Little is known about the role of specific delta GST genes in the detoxification of lambda-cyhalothrin in the global quarantine fruit pest codling moth, Cydia pomonella (L.). Real-time quantitative PCR shows that CpGSTd3 was ubiquitously expressed at all developmental stages and is most abundant in the larval stage and lowest in the egg stage; the mRNA level of CpGSTd3 is higher in the midgut and Malpighian tubules of fourth-instar larvae and abdomens of adults than in other tissues. Exposure of fourth-instar larvae to an LD10 dosage of lambda-cyhalothrin significantly induced the transcript of CpGSTd3 at 3 h, but the mRNA level was down-regulated after 12 h of treatment. Recombinant CpGSTd3 expressed in Escherichia coli was able to catalyze the conjugation of 1-chloro-2,4-dinitrobenzene (CDNB) and with an IC50 value of 0.65 mM for lambda-cyhalothrin. Metabolism assays indicate that recombinant CpGSTd3 could metabolize lambda-cyhalothrin. These results suggest that CpGSTd3 is probably a lambda-cyhalothrin metabolizing GST in C. pomonella.

Efficient Synthesis of l-Asparagine by an Immobilized Three-Enzyme Cascade Reaction System

Ran Wang,Wei Song,Hangyuan Xu,Jian Wen,Wanqing Wei,Zhaoyang Chen,Guipeng Hu,Cong Gao,Xiaomin Li,Jia Liu,Jing Wu

l-Asparagine (l-Asn) is an important amino acid with broad applications in food, medicine, fine chemicals, and environmental protection. However, its industrial production is limited by the high cost of raw materials and the low catalytic efficiency of enzymes. In this study, a three-enzyme cascade pathway (FDN) was designed to produce l-Asn, using fumaric acid as the raw material. Within this pathway, EcAsnA was identified as the rate-limiting enzyme and was subsequently engineered using a product rescue strategy to reduce product inhibition by opening the closed gate. The optimal mutant, L109 K/K58R, exhibited a 6.61-fold reduction in product inhibition and a 4.24-fold improvement in catalytic efficiency. This mutant was then integrated into Escherichia coli along with the other two enzymes to construct the optimal recombinant strain E. coli 17. Using diatomite-glutaraldehyde cross-linking immobilized E. coli 17 as a biocatalyst, 267.74 g of l-Asn (with 5.35 g·L–1·h–1 STY, > 99% ee) was produced across 50 batch feedings in 1 L reaction volume. The purity of l-Asn exceeded 99% after isolation and purification. This study demonstrates the effective integration of cascade reaction design, enzyme engineering, and cell immobilization, providing a promising approach for synthesizing high-value products from cost-effective substrates.

Redox-responsive nanomedicine of doxorubicin-conjugated poly-L-glutathione oxidized for cancer therapy

Xiao Duan,Qiang Wang,Wenfeng Che,Ting Li,Ke Zhang,Lingna Han,Lihua Song,Wei Guo

Background Doxorubicin (Dox) is a potent anticancer drug with severe side effects to normal tissues. To fabricate a nanomedicine with Dox is a better method to address the side effects of Dox in normal tissues. However, nanomedicines with unapproved materials could not be easily applied in clinic, hence, a nanomedicine fabricated by endogenous materials is a potential method to increase the possibility of clinical application. Methods In this study, a redox-responsive nanomedicine with clinical-applied anticancer of Dox and endogenous molecule of l -glutathione oxidized (GSSG) was prepared for cancer therapy. : The carboxyl and amino groups in l -glutathione oxidized can be reacted itself to form the poly- l -glutathione oxidized peptide-like polymer. Meanwhile, the Dox were conjugated in poly- l -glutathione oxidized polymer by amnio group in Dox and the structure of Dox-conjugated poly- l -glutathione oxidized polymer (Dox-Poly GSSG) was confirmed by FT-IR, NMR and MALDIT-TOF. Afterward, the drug loading of Dox (22 %) in poly-GSSG was measured by UV–vis and the accumulative Dox release rate of Dox-Poly GSSG is 46 % in 5 mg/mL reduced glutathione (GSH) environment at 72 h The size stability of Dox-Poly GSSG in PBS and PBS with GSH was evaluated by dynamic laser scatter (DLS) and transmit emission microscopy (TEM). Furthermore, cell viabilities of Dox-Poly GSSG were similar with free Dox evaluated by CCK-8 assay and flow cytometry experiment, respectively. The distribution of Dox-Poly GSSG in nuclei and tumor tissues was evaluated by Confocal laser scanning microscope and in vivo imaging system, respectively. Significant Findings The drug release results showed that the Dox could be controllable released in high concentration of GSH in tumor tissues. The tumor inhibition, H&E and TUNEL experiments showed that the Dox-Poly GSSG group can significantly inhibit the tumor growth compared to saline group and avoid major side effects to normal tissues compared to free Dox. This redox-responsive nanomedicine with clinical-applied anticancer of Dox and endogenous molecule of GSSG is a potential candidate for clinical application.

L-Phenylalanine Monomer Coacervation Leads to Well-Controlled Nanocrystal Topochemical Photo-RAFT Polymerization

Yuting Li,Xiyu Wang,Ying Cao,Wenjing Niu,Qing Zheng,Xinhua Lu,Yuanli Cai

We describe self-coacervation and nanocrystal topochemical photo-RAFT polymerization of ionic phenylalanine acrylamide. The charged monomer molecules undergo self-coacervation through nanoclustering, liquid-liquid phase separation, and crystallization in crowded droplets in water at pH 7.0 at 25 °C, leading to ultrathin lamellar nanocrystal-containing droplets capable of well-controlled topochemical photo-RAFT polymerization. Reaction induces pathway-dependent self-assembly involving one-dimensional (non)covalent polymerization of monomer nanoclusters to fibril bundles. Furthermore, monomer molecules within crystal lattice undergo one-dimensional rearrangements guided by growing polymer segments, leading to the nanocrystal transition to perforated lamellar hollow sieves accompanied by decreased crystallinity, and the subsequent interfacial topochemical polymerization of monomer nanoclusters site-specifically along newly activated sites of sieve edges, leading to sieve-centred parallel-growing fibrils. Further reaction leads to densely-charged ultrathin fibril lamellae physically crosslinked by fibril network knots. Consequently, the well-controlled topochemical photo-RAFT polymerization up to >98% conversion was achieved shortly in 1 h, under ecofriendly ambient aqueous conditions. This work provides a robust platform of topochemical photo-RAFT polymerization that proceeds with unprecedentedly high molecular weight controllability.

Periplaneta americana (L.) Extract PAS840 Promotes Ischemic Stroke Recovery by Inhibiting Inflammasome Activation

Xin Yang,Canhui Hong,Tangfei Guan,Chenggui Zhang,Yongshou Yang,Peiyun Xiao,Huai Xiao,Zhengchun He

Simple SummaryIschemic stroke (IS) is a high-mortality cardiovascular disease. This study analyzedPeriplaneta americana(L.) extract (PAS840) components using LC-MS/MS and peptidomics, and predicted its IS-related therapeutic targets by network pharmacology. In BV-2 cells after OGD/R injury, multiple techniques explored PAS840’s effects on cell functions and pathways. In a tMCAO rat model, various methods evaluated its impact on the blood–brain barrier, inflammation, and neural function. Results show PAS840 can act on multiple IS-related pathways. It inhibits the NLRP3 inflammasome pathway in BV-2 cells, reduces oxidative stress, inflammation, and pyroptosis, and improves cell viability. In tMCAO rats, it can reduce the NLRP3 inflammasome pathway, activate growth factor pathways, and protect the blood–brain barrier. These findings suggest that PAS840 is a promising drug worthy of development for the treatment of IS.Ischemic stroke (IS) is a high-mortality, multi-complication cardiovascular disease. Reducing brain injury and promoting neuronal repair after IS onset remain important challenges for current treatments. Our team previously found that PAS840, an extract fromPeriplaneta americana(L.), protects nerve function; this study further uses LC-MS/MS and peptidomics to analyze PAS840’s components and network pharmacology to predict its ischemic stroke (IS) therapeutic targets. We then employed Transwell, a biochemical kit, real-time quantitative polymerase chain reaction (RT-qPCR), and transcriptomics to investigate PAS840’s effects on migration ability, oxidative stress levels, and cellular pathways in mouse microglial cells (BV-2) following oxygen–glucose deprivation/reoxygenation (OGD/R) injury. Finally, using Evans blue staining, immunohistochemical analysis, and RT-qPCR, we investigated PAS840’s effects on the blood–brain barrier, inflammation pathways, and neural function in a transient middle cerebral artery occlusion (tMCAO) rat model. PAS840 components target multiple IS pathways, effectively inhibit NF-κB/NLRP3/Caspase-1/IL-1β inflammasome pathway activation in BV-2 cells following OGD/R, reduce cellular oxidative stress, inflammation, and pyroptosis, and improve cell viability and migration ability. PAS840 decreases NF-κB/NLRP3/Caspase-1/IL-1β inflammasome pathway expression in tMCAO rat brains, reduces inflammation, activates BDNF/VGF/NGR1/Erbb4 neurotrophic factor and vascular endothelial growth factor pathways, enhances neuronal cell viability, and effectively protects and repairs the blood–brain barrier.
