Injectable hyperbranched PEG crosslinked hyaluronan hydrogel microparticles containing mir-99a-3p modified subcutaneous ADSCs-derived exosomes was beneficial for long-term treatment of osteoarthritis

Zhaowei Yin,Chaoren Qin,Shaowei Pan,Chen Shi,Guanfu Wu,Yan Feng,Jing Zhang,Ziyi Yu,Bin Liang,Jianchao Gui

Exosomes (Exos) secreted by adipose-derived stem cells (ADSCs) have shown potential in alleviating osteoarthritis (OA). Previous studies indicated that infrapatellar fat pad (IPFP) derived stem cells (IPFSCs) may be more suitable for the treatment of OA than subcutaneous adipose tissue (ScAT) derived stem cells (ScASCs). However, it remains unclear which type of Exos offers superior therapeutic benefit for OA. This study first compared the differences between Exos derived from IPFP stem cells (Exos IPFP ) and ScAT stem cells (Exos ScAT ) in OA treatment. Results suggested that Exos IPFP significantly inhibit the degradation of cartilage extracellular matrix (ECM) than Exos ScAT , following this, the differences in microRNA (miRNA) expression between the two types of Exos using small RNA sequencing were performed. Subsequently, miR-99 b-3p was chosen and over-expressed in Exos ScAT (Exos ScAT−99b−3p ), both in vivo and in vitro experiments demonstrated its efficacy in inhibiting the expression of ADAMTS4, promoting the repair of the ECM in OA. Finally, microfluidic technology was performed to fabricate a hyaluronan-based hydrogel microparticles (HMPs) for encapsulating Exos (HMPs@exos), the injectability, sustained release of Exos and long-term therapeutic effect on OA were validated. In summary, these results suggest miR-99 b-3p regulates the degradation of cartilage ECM by targeting ADAMTS4, the upregulation of miR-99 b-3p in Exos ScAT would enable them to exhibit comparable or even superior effectiveness to Exos IPFP for OA treatment, making it a promising approach for OA treatment. Considering the abundant resources of ScAT and the limited availability of IPFP, ScAT harvested through liposuction could be genetically engineered to yield Exos for OA treatment. Furthermore, the encapsulation of Exos in HMPs provides an injectable sustained local drug release system, which could potentially enhance the efficacy of Exos and hold potential as future therapeutic strategies.

Rapid on-demand in-situ gelling and dissociation of PEG bottlebrush hydrogel via light-mediated grafting-through polymerization for full-thickness skin wound repair

Chong Chen,Siwei Bi,Changyuan He,Ruiqi Liu,Xueshan Zhao,Jialin Liu,Sheng Chen,Jun Gu,Bin Yan

Developing light-triggered in situ forming dynamical hydrogels with on-demand dissolvable and antioxidation capacities to promote wound healing is highly desired yet challenging. Herein, we prepared an in situ forming bottlebrush hydrogel with on-demand dissolution capacity from two thioctic acid (TA)-functionalized poly(ethylene glycol)s (PEGs) telechelic (TA-PEGs) prepolymers via a light-mediated grafting-through polymerization strategy. In response to light irradiation , TA-PEGs could undergo rapid ring-opening polymerization to generate a highly branched bottlebrush network with dynamic disulfide bonds and simultaneously load water-soluble curcumin complexes with excellent antioxidant activity , enabling fast in situ gelation in 3 min to achieve perfect coverage of wound sites with different shapes. Furthermore, the resulting bottlebrush hydrogel dressing can achieve facile and painless removal of the dressing via rapid on-demand dissolution of the hydrogel during dressing changes. Meanwhile, thanks to its dynamical feature of the disulfide-based network, the hydrogel possesses excellent self-healing ability and shape adaptability that can maintain the interiority of the dressing during application and thus prolong its service life. In vitro evaluation demonstrates that this PEG bottlebrush hydrogel possesses favorable hemocompatibility, cytocompatibility, and antioxidant capacity. More importantly, in vivo experiments confirmed that the hydrogel promotes angiogenesis, dermal collagen synthesis, and alleviates the excessive ROS damage, thus providing more beneficial conditions to accelerate full-thickness skin wound healing.

Controllable synthesis of up-conversion nanoparticles UCNPs@MIL-PEG for pH-responsive drug delivery and potential up-conversion luminescence/magnetic resonance dual-mode imaging

Yana Liu,Cheng Zhang,Hui Liu,Yuebin Li,Zushun Xu,Ling Li,Andrew Whittaker

Rare-earth doped up-conversion nanoparticles (UCNPs) with high uniformity and dispersibility were synthesized by a facile solvothermal method. The conditions of synthesis for NaYF 4 :Yb/Tm were explored, and the NaYF 4 :Yb/Tm particles with the optimal size (about 300 nm) were obtained. Blue emission was observed with the excitation 980 nm near infrared (NIR) laser, indicating that the UCNPs can be potentially used for up-conversion luminescence (UCL) imaging. On basis of the optimal UCNPs, further coated by poly (ethylene glycol) (PEG) functionalized metal-organic frameworks (MOFs), a multifunctional platform UCNPs@MIL-PEG for cancer diagnosis and treatment was established. The core-shell structure was confirmed by TEM images. Doxorubicin (DOX) was selected as drug model and the drug loading of UCNPs@MIL-PEG was found to be 60%. Cytotoxicity indicated that UCNPs@MIL-PEG were highly biocompatible. The DOX release in different pH value revealed an excellent pH-triggered drug release. In addition, the UCNPs@MIL-PEG nanoparticles can be tracked by magnetic resonance imaging (MRI). A clear dose-dependent contrast enhancement in T 2 -weighted MR images indicated the potential act as T 2 MRI contrast agents. The UCNPs@MIL-PEG nanoparticles are expected to be simultaneously used for UCL/MR dual-mode imaging and pH-responsive drug release.

Sustained release of PTX-incorporated nanoparticles synergized by burst release of DOX⋅HCl from thermosensitive modified PEG/PCL hydrogel to improve anti-tumor efficiency

Shuxin Xu,Weiwei Wang,Xijing Li,Jianping Liu,Anjie Dong,Liandong Deng

As drug therapies become increasingly sophisticated, the synergistic benefits of two or more drugs are often required. In this study, we aimed at improving anti-tumor efficiency of paclitaxel (PTX)-incorporated thermo-sensitive injectable hydrogel by the synergy of burst release of doxorubicin hydrochloride (DOX⋅HCl). Thermosensitive injectable hydrogel composed of nanoparticles assembled from amphiphilic copolymer poly(ε-caprolactone-co-1,4,8-trioxa[4.6]spiro-9-undecanone)-poly(ethylene glycol)-poly(ε-caprolaone-co-1,4,8-trioxa[4.6]spiro-9-undecanone) (PECT) was fabricated. Hydrophobic PTX and hydrophilic DOX⋅HCl were loaded simultaneously in the thermo-sensitive injectable hydrogel by a two-stage entrapment. Thermosensitive gelling behaviors of drug-loading PECT nanoparticle aqueous dispersions were studied. In vitro release profiles of PTX and DOX⋅HCl and in vivo anti-tumor effect by dual drugs from PECT hydrogel were investigated. The results showed that hydrophilic and hydrophobic drugs could be successfully entrapped in PECT hydrogel simultaneously without affecting its thermo-sensitive behavior. In vitro release profiles demonstrated the burst release of DOX⋅HCl and the sustained release of PTX. Anti-tumor effect was improved by a fast and tense attack caused by the burst release of hydrophilic DOX⋅HCl from hydrogel, which was continued by the sequent sustained release of PTX-incorporated nanoparticles and remnant DOX⋅HCl. Unintentionally, entrapped in PECT hydrogel, hydrophilic DOX⋅HCl was observed to have a sustained releasing pattern in vitro and in vivo .

Integrated Solar-thermal Conversion, Heat Collection and Antibacterial Properties of Composite Fabrics: A Case Study of PEG@SiO2 Microcapsules Intercalated within MXene Interlayer Coatings

Meng Tao,Wei Guo,Jixiang Zhang,Cui Liu,Nian Li,Min Xi,Shudong Zhang,Zhenyang Wang

Composite fabrics with integrated photothermal conversion, heat collection, and antibacterial properties are urgently needed to enhance personal thermal management in northern cold regions. Herein, a designed sandwich-like structure, in which PEG@SiO2 microcapsules were intercalated into the MXene interlayers (PEG@SiO2/MXene), was proposed to achieve the fast solar-thermal conversion and the surplus solar-generated heat storage and release and even further corresponding fabrics with antibacterial properties for the personal thermal management. Remarkably, the prepared PEG@SiO2/MXene (PSM) exhibited fast solar-to-heat conversion, followed-on heat storage ability, with an average temperature rising rate of 8.1 °C/min under one-sunlight irradiation and a phase change enthalpy of 155.7 J/g. Furthermore, under one-sunlight irradiation, the designed antibacterial composite fabrics with PSM coatings could greatly increase the local working temperature from 1 °C to 27.3 °C compared with the reference traditional cotton fabrics (from only 1 °C to 6.5 °C). This work provides a potential multifunctional composite material/fabric for the development of personal thermal management in outdoor sunlight applications.

Comprehensive pharmacokinetic and tissue distribution study of α,ω-dipropionic acid polyethylene glycol (PA-PEG₁₂-PA) in rats using a validated UPLC-MS/MS method

Meichen Liu,Yalin Yu,Yifei Jing,Yingxia Guo,Chuya Wang,Hongyu Xue,Hong Liu,Lei Yin,Meiyun Shi

While polyethylene glycol (PEG) is an extensively utilized pharmaceutical polymer with established biocompatibility and regulatory acceptance, the in vivo pharmacokinetics of its bifunctional derivatives, such as α,ω-dipropionic acid polyethylene glycol (PA-PEG-PA), remain largely unexplored. This study presents the development and validation of a highly sensitive and selective UPLC-MS/MS method for the accurate quantification of PA-PEG 12 -PA in various biological matrices, utilizing a straightforward protein precipitation protocol. After intravenous administration in rats (10 mg/kg), PA-PEG 12 -PA demonstrated rapid clearance (t 1/2 = 3.99 ± 1.06 h). Tissue distribution analysis revealed a pronounced affinity for renal accumulation, with kidney concentrations (16473 ± 881 ng/g at 2 h) substantially surpassing those in the liver and lungs, followed by rapid depletion within 24 h. Excretion studies indicated renal clearance as the dominant pathway, with 55.93 % of the administered dose recovered unchanged in urine over 72 h, while fecal excretion was minimal (1.76 %). This work provides the first comprehensive pharmacokinetic and biodistribution profile of PA-PEG 12 -PA, underscoring its renal-driven clearance and tissue disposition. The findings offer crucial insights for the rational design of PEGylated drug delivery systems, and the robust UPLC-MS/MS assay established herein serves as a valuable tool for characterizing polymeric excipients in biological environments.

Novel phase-change PDA-Ni@GNS/CNF-C/SA/PEG composites with ultrahigh shape stability and latent heat as thermal management material for microelectronic devices

Xiaohong Wang,Bin Yang,Yue Zhang,Yuqing Yang,Huijie Lu,Hao Wang,Ziyi Wang,Jiasheng Qian,Ru Xia,Ning Jia,Yuchao Ke

Phase-change materials (PCMs) have become a hot spot in the development of modern thermal management materials because of their absorbing and releasing heat while keeping the temperature constant during phase change process. However, their relatively low thermal conductivity (TC) considerably restricts their further application. In this work, graphene nanosheets (GNS) were initially nickel-plated to increase the contact area between adjacent GNS (in the form of Ni@GNS) by constructing a “point-surface” structure. Then, the prepared Ni@GNS was coated with polydopamine (PDA) to prepare PDA-Ni@GNS, which helped to effectively reduce the interface thermal resistance (ITR) between the filler and the matrix, and also made the prepared filler more easily dispersed in the polymer matrix. By constructing well-defined three-dimensional (3D) thermally conductive pathways in the matrix, the PCMs exhibited excellent TC and effectively prevented the leakage of polyethylene glycol (PEG) even at low filler loading. The results of thermogravimetry (TG), differential scanning calorimeter (DSC), and cyclic DSC tests jointly showed that the PDA-Ni@GNS/CNF-C/SA/PEG PCMs displayed excellent thermal storage properties, high latent heat as well as good cyclic thermal stability. The present study provides a facile way of preparing PEG-based PCMs, which are applicable in thermal management area.

The effect of carbonization/silver plating double modification on the thermal properties of PEG/wood-based composite phase change materials: experiments and molecular dynamics simulations

Zhiwen Yin,Chaohua Zhang

To overcome the bottleneck of single modification methods for composite phase change materials struggling to balance high thermal conductivity and high phase change enthalpy, this study employed poplar wood as the raw material to fabricate a wood-based porous carrier through a dual-modulation strategy involving carbonization and silver-plating. This modified carrier was then impregnated with polyethylene glycol phase change material to synthesize a shape-stable composite phase change material with integrated structure and function. A combined experimental and modeling approach was used to investigate the thermal properties and underlying mechanisms of the composite. Experimental results demonstrate that the silver-enhanced carbonized wood/polyethylene glycol composite phase change material achieved 93.2 % photothermal conversion efficiency and its thermal conductivity reached 0.632 W·m −1 ·K −1 , representing a 2.64-fold enhancement compared to pure polyethylene glycol. The material exhibited effective temperature control capability in electronic device thermal management scenarios and maintained stable phase change behavior over 100 heating–cooling cycles. Molecular dynamics simulations elucidated the synergistic mechanism between the silver nanoparticle-modified three-dimensional porous network and polyethylene glycol. Vibrational density of states spectra confirmed that enhanced interfacial phonon coupling reduced interfacial thermal resistance to 3.459 × 10 −9 m 2 ·K·W −1 , corresponding to a 56.23 % reduction relative to non-modified wood/polyethylene glycol composites. This work innovatively combines wood cell wall engineering with interfacial phonon engineering, revealing atomic-scale heat transfer mechanisms at the microscopic level. The integrated experimental and simulation approach provides valuable insights for developing wood-based multifunctional composite phase change materials.

Zi-Wen Zhou,Chun-Xiu Liu,Chun-Xian Cai,Na Wang,Xiao-Qi Yu

In this work, PEG modified photocatalytic covalent organic framework (PEG@CTF-OMe) was co-immobilized with laccase into Cu doping calcium alginate gel beads (Ca/Cu-PEG@CTF-OMe-lac). PEG@CTF-OMe improved the light stability of laccase under blue light irradiation , and external introduction of copper into gel beads stabilized the conformation and improved pH adaptability of laccase. With the help of 2,2-binamine-di-3-ethylbenzothiazolin-6-sulfonic acid (ABTs), the immobilized laccase (Ca/Cu-PEG@CTF-OMe-lac) presented outstanding synergistic removal of pollutants by photocatalysis and enzymatic catalysis, as well as ABTs radicals generation promoting by copper ions in the laccase-mediator system. The removal recyclability performance assay found that the Ca/Cu-PEG@CTF-OMe-lac gel beads had more than 80 % removal efficiency for phenol , 4-chlorophenol, 2,4-dichlorophenol, nonyl phenol, bisphenol A and indole in water at 25 °C after 5 cycles, when the concentration reached as high as 70 mg/L, 60 mg/L, 120 mg/L, 100 mg/L, 100 mg/L and 100 mg/L, respectively. The high synergistic removal performance obtained by the gel beads co-immobilizing laccase and photocatalytic CTFs showed greater practical application value.

In Situ Growth of Zeolitic Imidazolate Framework-L in Macroporous PVA/CMC/PEG Composite Hydrogels with Synergistic Antibacterial and Rapid Hemostatic Functions for Wound Dressing

Hang Yang,Xianyu Lan,Yuzhu Xiong

Although many advances have been made in medicine, traumatic bleeding and wound infection are two of the most serious threats to human health. To achieve rapid hemostasis and prevent infection by pathogenic microbes, the development of new hemostatic and antibacterial materials has recently gained significant attention. In this paper, safe, non-toxic, and biocompatible polyvinyl alcohol (PVA); carboxymethyl cellulose (CMC), which contains several carboxyl and hydroxyl groups; and polyethylene glycol (PEG), which functions as a pore-forming agent, were used to prepare a novel PVA/CMC/PEG-based composite hydrogel with a macroporous structure by the freeze-thaw method and the phase separation technique. In addition, a PVA/CMC/PEG@ZIF-L composite hydrogel was prepared by the in situ growth of zeolitic imidazolate framework-L (ZIF-L). ZIF-L grown in situ on hydrogels released Zn2+and imidazolyl groups. They elicited a synergistic antibacterial effect in hemostasis with PVA and CMC, rendering the PVA/CMC/PEG@ZIF-L hydrogel with a good antibacterial effect againstStaphylococcus aureus. At the same time, the macroporous structure enabled the rapid release of Zn2+and imidazolyl groups in ZIF-L and promoted cell proliferation at an early stage, enhancing the coagulation efficiency. A rat liver injury model was used to confirm its rapid hemostasis capacity. Keywords:in situ growth;macroporous structure;synergistic antibacterial;rapid hemostatic;carboxymethyl cellulose;wound dressing;hydrogel;ZIF-L;polyvinyl alcohol;phase separation technique
