Therapeutic effects of EGF-modified curcumin/chitosan nano-spray on wound healing

Li Yue,Leng QingQing,Pang XianLun,Shi Huan,Liu YanLin,Xiao SuSu,Zhao Ling,Zhou Ping,Fu ShaoZhi

Dermal injury, including trauma, surgical incisions, and burns, remain the most prevalent socio-economical health care issue in the clinic. Nanomedicine represents a reliable administration strategy that can promote the healing of skin lesions, but the lack of effective drug delivery methods can limit its effectiveness. In this study, we developed a novel nano-drug delivery system to treat skin defects through spraying. We prepared curcumin-loaded chitosan nanoparticles modified with epidermal growth factor (EGF) to develop an aqueous EGF-modified spray (EGF@CCN) for the treatment of dermal wounds. In vitro assays showed that the EGF@CCN displayed low cytotoxicity, and that curcumin was continuously and slowly released from the EGF@CCN. In vivo efficacy on wound healing was then evaluated using full-thickness dermal defect models in Wistar rats, showing that the EGF@CCN had significant advantages in promoting wound healing. On day 12 post-operation, skin defects in the rats of the EGF@CCN group were almost completely restored. These effects were related to the activity of curcumin and EGF on skin healing, and the high compatibility of the nano formulation. We therefore conclude that the prepared nano-scaled EGF@CCN spray represents a promising strategy for the treatment of dermal wounds.

Elastic, super-hydrophobic and biodegradable chitosan sponges fabricated for oil/water separation

Fengyan Liang,Tingting Hou,Sidong Li,Lusheng Liao,Puwang Li,Chengpeng Li

Due to easy availability and biodegradability , chitosan (CS)-based porous adsorbents have attracted significant interest in oil-spill and organic solvent removal. Herein, a multi-network CS-based sponge was fabricated via ion and hydrogen bond cross-linking using lemon, followed by covalent cross-linking via heptanal, in-situ loading of Fe 3 O 4 -polydopamine (PDA) particles and polylactic acid (PLA) coating. The as-fabricated sponge was systematically characterized by Fourier transform-infrared spectrometry (FT-IR), X-ray diffraction (XRD), vibrating sample magnetometer (VSM), X-ray photoelectron spectroscopy (XPS), scanning electron microscope (SEM), transmission electron microscope (TEM), static water contact angles (WCA) and so on. The as-fabricated CS-based sponge exhibits excellent elasticity, which could be compressed to large strains (80%) at a relatively high stress (0.685 MPa). The WCA of the targeted sponge can reach to 152.2 ± 1.2º after multiple surface functionalization. In addition, the degradation rate of the as-fabricated sponge can still reach 45.01 ± 1.11% within 15 days. Particularly, the sponge could selectively adsorb oil/organic solvent with up to 50 times of its own weight, and effectively separate the surfactant-stabilized oil-in-water emulsion with a flux of 8753 ± 423.56 L m −2 h −1 . Last but not least, the sponge can be driven and recycled magnetically. In conclusion, this promising multifunctional sponge can be applied as a new absorbent for oil-water separation.

Ru Nanoparticles Immobilized on Chitosan as Effective Catalysts for Boosting NH3BH3 Hydrolysis

Jiaxin Liu,Peiyun Li,Renfeng Jiang,Xiucheng Zheng,Pu Liu

Graphical Catalytic Hydrolysis : Ruthenium nanoparticles with different dosages are immobilized on a naturally cationic polysaccharide polymer (chitosan, CS) via an adsorption-in situ reduction method for boosting AB hydrolysis. Catalyzed by the optimal Ru/CS catalysts, the turnover frequency is 338.1 min −1 in alkaline solution at 303 K and the apparent activation energy is 41.3 kJ mol −1 , which are superior to many results reported previously. Developing an advanced heterogeneous catalyst is essential for realizing high-performance ammonia borane (AB) hydrolysis, which is a safe and efficient way to generate hydrogen. In this work, a series of ruthenium (Ru) nanoparticles (NPs) immobilized on chitosan (CS) natural polymers are synthesized via an adsorption-in situ reduction method. Various analytical techniques, including XRD, XPS, FT-IR, SEM and TEM, are adopted to explore the structure-catalytic performance relationships of the obtained catalysts for hydrolysis of AB. Benefitting from the possible interaction between Ru NPs and the amino and hydroxyl groups in CS, the as-prepared catalysts exhibit excellent catalytic behavior for hydrogen generation from AB hydrolysis. Catalyzed by the optimal Ru/CS, the turnover frequency reaches up to 331.8 min −1 in NaOH solution at 303 K, and the corresponding apparent activation energy is 41.3 kJ mol −1 , which are superior to many results previously reported. This work demonstrates that the obtained Ru/CS is a promising and efficient catalyst for the hydrolysis of solid hydrogen storage materials.

Anisotropic Structure and Properties of Chitin and Chitosan Nanofibril-Supported Starch Foams

Jiaqi Shan,Dagang Liu,Fan Su,Minyu Li,Huafeng Tian,Mengna Guo,Wenhao Qiao,Jie He,Qin Li,Jun Qian

Highly surface-charged rigid bionanofibrils are difficult to be processed into integrated foams or aerogels owing to lack of good interfibrillar adhesion. An alternative solution is to involve flexible hydrophilic polymers for gluing fibril bundles, allowing a perfect alignment under ice induction in the lyophilized foams. In this work, we incorporated the defibrillated chitin/chitosan nanofibrils (CTF/CSF) into gelatinized starch foams and investigated the effects of surface charge, solid content, nanofibril loading, and directional orientation on the dimensional stability, morphology, crystallinity, compressive stress, and flame-retardant properties of lyophilized composite foams. Results show that CTF with low positive surface charges presents stable electroattractions to negatively charged starch and also exhibits sustainable reinforcement effects on the composite foam. What is more, compact, small porous, and lamellar CTF-supported foams exhibit relatively high resistance to flammable combustion. In the case of high loading of polycationic CSF, directional freeze-drying successfully induced the channel orientation along the vertical, horizontal, spiral, or bi/triphase junctional direction in the anisotropic foams, which revealed variable compression properties according to the fibril alignment.

Chitosan oligosaccharide-g-linalool polymer as inhibitor of hyaluronidase and collagenase activity

Shuifang Mao,Xiaoli Liu,Wenshui Xia

In this study, chitosan oligosaccharide (COS) was modified by grafting Linalool (Lin) on its backbone to improve its anti-inflammatory activity. By changing the molar ratios of COS to Lin, three different degrees of substitution COS-g-Lin1-3 were prepared. The degrees of substitution of derivatives were 0.65, 0.80 and 1.14 respectively. The structure of COS-g-Lin1-3 were characterized by UV–vis, FT-IR, 1 H NMR and elemental analysis in order to show the COS-g-Lin1-3 successfully synthesized. Besides, the thermal stability , solubility, pH stability as well as crystallinity were also investigated. The results revealed that the derivatives exhibited higher thermal stability and more remarkable anti-inflammatory property against hyaluronidase and collagenase than that of COS. The good biocompatibility made this novel material a promising and effective compound for anti-inflammatory applications.

Extraction combined oxidation desulfurization of dibenzothiophene using polyoxometalate-supported magnetic chitosan microspheres

Yefeng Liu,Peng Zuo,Fei Wang,Ying Lv,Ruixin Wang,Weizhou Jiao

In this study, Keggin- and Dawson-type polyoxometalates (POMs) were immobilized on chitosan (CS)-modified Fe 3 O 4 @CS by electrostatic interaction for the preparation of heterogeneous catalysts Fe 3 O 4 @CS@POM (POM=PMo 12 , PW 12 , SiW 12 , P 2 W 17 , or P 2 W 18 ). The oxidative desulfurization (ODS) of dibenzothiophene (DBT) by Fe 3 O 4 @CS@POMs was evaluated. It is found that Fe 3 O 4 @CS@POM shows the good ODS performance due to the good dispersion of POM on Fe 3 O 4 surface and the good accessibility of substrate to it leaded by the full swelling of the cross-linked CS in acetonitrile . The catalytic activity follows the order of Fe 3 O 4 @CS@PMo 12 > Fe 3 O 4 @CS@PW 12 > Fe 3 O 4 @CS@P 2 W 17 > Fe 3 O 4 @CS@P 2 W 18 > Fe 3 O 4 @CS@SiW 12 . At 333 K, the desulfurization rate of DBT (500 ppm) reaches 99.7% at 60 min and 100% at 90 min in the presence of 0.1 g of Fe 3 O 4 @CS@PMo 12 and 100 µL of H 2 O 2 . The ODS process can be well described by the pseudo-first-order kinetic model (R 2  > 0.99) with a high kinetic constant (0.0761 min −1 ) and a low apparent activation energy (60.57 kJ / mol). In addition, the as-prepared catalyst can be easily separated and recycled in the external magnetic field, and it still has the good reusability for several times, stemming from the stable encapsulation of POM clusters by cross-linked chitosan on Fe 3 O 4 surface.

Low-temperature 3D printing of collagen and chitosan composite for tissue engineering

Hairui Suo,Jiaying Zhang,Mingen Xu,Ling Wang

Three-dimensional (3D) printing is a promising method to prepare scaffolds for tissue regeneration. Collagen and chitosan composites are superior materials for tissue engineering scaffold but rarely printed due to their poor printability. Here, we prepared a series of tunable hybrid collagen/chitosan bioinks with significantly improved printability through hydrogen bond interaction and printed them into scaffolds by carefully controlling the temperature. Rheological tests proved the printable bioinks had sound shear thinning behavior, dramatical viscosity variation with temperature, and the gelation temperature from 7 to 10 °C. Chitosan could decrease the swelling ratio of the printed scaffolds, while their degradation rate increased with collagen proportion and the values of Young's modulus and tensile strength increased with chitosan proportion. Moreover, the scaffolds containing 2% (m/v) collagen and 2% (m/v) chitosan had a homogeneous and compact honeycomb-like structure, demonstrating the strengthening effect of chitosan. Cell viability assay presented vigorous cell growth on the surface of scaffolds, meanwhile, live cells were also found inside and at the bottom of the scaffolds, indicating the migration of cells. Therefore, chitosan can improve the printability of collagen and the hybrid collagen/chitosan bioinks can be printed into scaffolds with regulated properties, thus can fit different applications in tissue engineering.

Engineering a novel antibacterial agent with multifunction: Protocatechuic acid-grafted-quaternized chitosan

Chen Zhou,Hai-Yong Ao,Xiao Han,Wen-Wen Jiang,Zhi-Fei Yang,Le Ma,Xiao-Yan Deng,Yi-Zao Wan

As a mild cationic antibacterial agent, hydroxypropyltrimethyl ammonium chloride chitosan (HACC) could kill gram-positive bacteria and gram-positive drug-resistant bacteria without cytotoxicity. Nevertheless, it was not effective against gram-negative bacteria. Herein, protocatechuic acid (PA) with broad-spectrum antibacterial properties and pharmacological activities was grafted on HACC. PA-g-HACC showed favourable antioxidant capacity and anti-inflammatory properties. Most importantly, the results of antibacterial assay indicated that the antibacterial rates of all PA-g-HACC groups against Staphylococcus aureus ( S. aureus ) and methicillin-resistant Staphylococcus aureus ( MRSA ) were above 92 %, and the antibacterial rate of PA-g-HACC against E. coli was increased with the amount of grafted PA. Furthermore, the cytocompatibility of PA-g-HACC was improved by appropriate grafting ratio of PA, while excessive grafted PA can lead to toxicity. We believe that PA-g-HACC in optimum grafting ratio of PA with favorable antibacterial properties, pharmacological activities and cytocompatibility will be potential antibacterial agent for treating infections.

Functionalized chitosan as a novel support for stabilizing palladium in Suzuki reactions

Yahao Dong,Jiajun Bi,Shujun Ming,Shoute Zhang,Dajian Zhu,Di Meng,Tao Li

Chitosan is a versatile polysaccharide in different domains due to facile modification and good biodegradability. In this paper, taking advantage of such functional properties, we have developed a stabilizer agent [OCMCS-SB] produced from chitosan, and palladium was successfully immobilized on this designed stabilizer [OCMCS-SB-Pd(II)]. The obtained complex was illuminated by 13 C CP-MAS NMR, FT-IR, TGA, XRD, XPS, SEM, TEM and ICP-OES analyses. Due to the interactions of primary hydroxyl groups on chitosan, Schiff base and carboxy groups, the Pd complex showed excellent reactivity (up to 99 %) and stability towards Suzuki reactions in eco-friendly medium. Subsequently, the reusability experiments for OCMCS-SB-Pd(II) formed from chitosan were examined in five consecutive cycles, which showed no appreciable decrease in activity. Furthermore, a reasonably trifunctional complex structure was proposed. The present bio-based system offers a promising approach in utilizing such biopolymers in organic transformations.

Chitosan-modified halloysite nanotubes as a controlled-release nanocarrier for nitrogen delivery

Chao Wang,Zuyu He,Yunhao Liu,Chuang Zhou,Jing Jiao,Puwang Li,Dequan Sun,Liyun Lin,Ziming Yang

Halloysite nanotubes (Hal) are naturally occurring tubular clay nanomaterials that have a unique tubular structure and surface modifiability. In order to increase the stability and to achieve controlled release of fertilizers, chitosan (CS) was assembled on the surface of Hal as a valve for preparing Hal@CS and to control the mesoporous surface and lumen ports. Transmission electron microscopy (TEM), X-ray diffraction, Fourier transform infrared, thermogravimetric analysis and nitrogen adsorption isotherm analysis were conducted to confirm the successful grafting of CS onto the surface of the Hal. The cumulative release ratio of urea from the Hal@CS in vitro after 96 h was 34.37% without glutathione (GSH). After introducing GSH the rate of release of urea from the Hal@CS increased. The uptake, translocation, and distribution of the Hal@CS in the epidermal layer and endodermis of root tissue were detected by fluorescence labeling and TEM. We conclude that Hal@CS are a potential controlled-release carrier and delivery system for agricultural fertilizers.

Biomemristic Behavior for Water-Soluble Chitosan Blended with Graphene Quantum Dot Nanocomposite

Lei Li

Bionanocomposite has promising biomemristic behaviors for data storage inspired by a natural biomaterial matrix. Carboxylated chitosan (CCS), a water-soluble derivative of chitosan avoiding the acidic salt removal, has better biodegradability and bioactivity, and is able to absorb graphene quantum dots (GQDs) employed as charge-trapping centers. In this investigation, biomemristic devices based on water-soluble CCS:GQDs nanocomposites were successfully achieved with the aid of the spin-casting method. The promotion of binary biomemristic behaviors for Ni/CCS:GQDs/indium-tin-oxide (ITO) was evaluated for distinct weight ratios of the chemical components. Fourier transform infrared spectroscopy, Raman spectroscopy (temperature dependence), thermogravimetric analyses and scanning electron microscopy were performed to assess the nature of the CCS:GQDs nanocomposites. The fitting curves on the experimental data further confirmed that the conduction mechanism might be attributed to charge trapping–detrapping in the CCS:GQDs nanocomposite film. Advances in water-soluble CCS-based electronic devices would open new avenues in the biocompatibility and integration of high-performance biointegrated electronics. Keywords:biomemristic behavior;water-soluble CCS:GQDs nanocomposites;CCS-based binary biomemory

Chitosan-based melatonin bilayer coating for maintaining quality of fresh-cut products

Hangyue Zhao,Lei Wang,Tarun Belwal,Yunhong Jiang,Dong Li,Yanqun Xu,Zisheng Luo,Li Li

This work was designed to develop the chitosan-based melatonin layer-by-layer assembly (CMLLA) via the inclusion method. The structural characterizations and interaction present in CMLLA were investigated by the scanning electron microscope (SEM), X-ray diffraction (XRD) and Fourier Transform-Infrared spectroscopy (FTIR). The ratio of chitosan (CH) to carboxymethylcellulose (CMC) greatly influenced the mechanical properties, including the tensile strength, moisture content and color performance. Results showed that both antioxidant and antimicrobial properties of CMLLA were enhanced with the addition of melatonin (MLT). Furthermore, it was demonstrated that the CMLLA with 1.2 % (w/v) CH, 0.8 % (w/v) CMC and 50 mg/L MLT better contributed to the delay of chlorophyll degradation and the maintenance of shelf-life quality. Results from this study might open up new insights into the approaches of quality improvement of postharvest fresh products by incorporating the natural antioxidant compounds into natural polymers.
